Blood constituents labelled with technetium-99m (99mTc) has been used with radiobiocomplexes in several procedures in nuclear medicine. Some natural and sintetic drugs are capable to interfere on the labeling of blood constituents with 99mTc, on the morphology of red blood cells (RBC) and on the biodistribution of radiobiocomplexes. The aim of this study was evaluate the effect of an extract of Passiflora edulis f. flavicarpa on the labeling of blood constituints with 99mTc, on the morphology of RBC and on the biodistribution of the radiopharmaceutical sodium perthecnetate in Wistar rats. On the in vitro studies the Passiflora edulis f. flavicarpa decreased significantly (p<0.05) the %ATI on plasma proteins and on the in vitro morhology of RBC, the passion fruit peel flour altered the shape and the perimeter/área ratio. On the in vivo estudies the extract did not altered the %ATI in blood constituents, and did not altered the shape of RBC. Although, on the biodistribution of the radiobiocomplex sodium perthecnetate (Na99mTcO4) this extract decreased significantly (p<0.05) the uptake in duodenum, spleen, pâncreas and blood, and increased the uptake in stomach. It can be suggested that the effects presented by this extract could be a result of some substances contained in this extract that could alter the binding of 99mTc to plasma proteins, the morphology of RBC and the biodistribution of the radiobiocomplex sodium perthecnetate. Fucan is a term used to denominate a family of sulfated L-fucose-rich polysaccharides. The brown alga Spatoglossum schröederi (Dictyotaceae) has three heterofucans namely fucan A, B and C. The 21 kDa fucan A is composed of a core of β (1-3) glucuronic acid-containing oligosaccharide of 4.5 kDa with branches at C4 of fucose chains α (1-3) linked. The fucose is mostly substituted at C4 with a sulfate group and at C2 with chains of β (1-4) xylose. This fucan has neither anticoagulant (from from 0.1 to 100µg) nor hemorrhagic activities (from 50 to 800 µg/mL). The antithrombotic test in vivo showed the fucan A has no activity in any of the concentrations (from 0.2 to 20µg/g/day) tested 1h after polysaccharide administration. However, when fucan A was injected endovenously 24h before the ligature of the venae cavae, we observed a dose-dependent effect, reaching saturation at around 20g/g of rat weight. In addition, this effect is also time-dependent, reaching saturation around 16h after fucan administration. In addition, regardless of administration pathway, fucan A displayed antithrombotic action. The exception was the oral pathway. Of particular importance was the finding that fucan A stimulates the synthesis of an antithrombotic heparan sulfate from endothelial cells like heparin. The hypothesis has been raised that in vivo antithrombotic activity of fucan A is related to the increased production this heparan. Taken together with the fact that the compound is practically devoid of anticoagulant and hemorrhagic activity suggests that it may be an ideal antithrombotic agent in vivo

The topical corneal application of antimitotic mitomycin-C (MMC) during refractive surgery is still characterized by a lack of standardization and considerable empirism. For this reason the creation of a system capable of reliable drug delivery represents a beneficial innovation for patients submitted to these procedures. Objective: Elaborate a new MMC delivery system during the transoperatory period of photorefractive keratectomy (PRK) followed by patent application. Methods: The project consists of an in vitro experimental study to create an MMC (0.02%) release system. The drug was impregnated in sterile Whatman® 41 paper filter discs with a diameter of 8 mm. After drying, the discs were applied to antibiogram plates seeded with Staphylococcus epidermidis (American Type Culture Collection ATCC 12228), followed by the addition of a drop of sterile water. At the end of 1 minute, the discs were removed and the plates incubated for 48 hours at 35oC. Mean drop volume in the collyrium flasks was measured using analytical balance weighing. The inhibition halo (mm) was correlated with the MMC impregnated into the disc. After completion of the invention design a patent application was lodged at the National Institute of Industrial Property. Results: The correspondence between MMC-produced inhibition halos indicated that a dose of 16μg was ideal for impregnating into the discs. The mean drop volume obtained from the collyrium flasks was 37.7 μL. A minute after the application of one drop of balanced saline solution, the system released an adequate concentration for PRK surgery. Conclusion: A new MMC delivery system was created for transoperatory application in photorefractive keratectomy (PRK). Publication of the patent application (number PI 0704739-8) gives the authors exclusive intellectual property rights. Introduction: The chronic immunosuppression promotes the development of changes in the oral cavity of the kidney transplant recipients (KTR), however with the exception of gingival overgrowth, little is known regarding the prevalence of oral lesions in this population. Objective: To evaluate the prevalence of clinical and microbiological conditions of the oral cavity of the recipients of kidney transplantation and the associated factors. Methods: This was a cross-sectional study which examined 96 clinical KTR and experimental where collected saliva stimulated them to identify Candida sp. Data collection consisted of evaluation stomatologic, socio-demographic, clinical aspects of transplantation, condition of oral hygiene and dental caries, yonder to questions about knowledge of oral alteration after transplantation. Results: Of the total, 66.7% of KTR had some type of oral manifestation. The most common was saburral tongue, followed by gingival overgrowth, with both oral manifestations related to gender and concomitant use of cyclosporine and nifedipine (p <0.05). Tacrolimus showed a protective effect for gingival overgrowth (OR = 0.13). The oral hygiene was associated with saburral tongue(p = 0.03) and severity of gingival overgrowth (p = 0.0001). Oral candidiasis was diagnosed in 17.7% of patients and Candida albicans was isolated most frequently in the saliva of RTR with a colonization of 58.3%. The average DMF-T increased with age. The method of oral hygiene was most used brush and toothpaste to 61.5%. Changes in the oral cavity was seen in 54.2% of KTR, citing as the main growth and gingival ulcers. Instructions for oral hygiene after transplantation were neglected for 61.5% of RTR. Dry mouth and halitosis were reported in 30.2% and 36.5% respectively. Conclusions: More the half of the KTR had at least one injury of the mouth, the immunosuppressive drugs and oral hygiene are associated with these alterations. Prospective cohort studies are needed to elucidate the relationship between oral manifestations and levels of drug and risk of oral manifestations occur over time. The kidney transplant recipients showed to be aware of oral alterations occurred after transplantation and uninformed about the oral hygiene instructions. With regard to hygiene, the incidence of caries was considered high, conditions of risk were identified and improvements in primary attention should be encouraged and reflected in the monitoring of renal transplant

The effects of four light intensities (0; 2.8 +/- 0.9; 5,5 +/- 1,8 e 7,8 +/- 2,5 mu mol s(-1) m(-2), about 136.5 +/- 87.5; 273 +/- 43.8 e 390 +/- 125 lux, respectively) on survival, productivity, weight gain and larval development of Macrobrachium amazonicum were investigated. Four treatments with three replicate tanks were evaluated. Newly hatched larvae were held in black tanks (80.2 +/- 0.6 larvae L(-1)) filled with 50-L-brackish water (salinity of 10), in a recirculating system. Tanks were covered with shadow cloth allowing 35% and 70% light, respectively, to reach light intensities of 2.8 +/- 0.9 and 5.5 +/- 1.8 mu mol s(-1) m(-2) at the water surface. Complete absence of light (0 mu mol s(-1) m(-2)) was obtained covering the tanks with opaque black plastic, and full-light condition used no covering (7.8 +/- 2.5 mu mol s(-1) m(-2)). Observations showed that the survival rate was not affected by light intensity. Productivity and weight gain were higher under 7.8 +/- 2.5 mu mol s(-1) m(-2) light intensity than under 0 and 2.8 +/- 0.9 1 mu mol s(-1) m(-2) intensities (P<0.05). The larval development index was similar among the treatments under the different light intensities. However, from stage VII this index was increased slightly in the treatment under 7.8 +/- 2.5 mu mol s(-1) m(-2) light intensity. In conclusion, light intensity affects larval development of M. amazonicum. Values as high as 7.8 mu mol s(-1) m(-2) (about 390 lux) improve the larval performance by enhancing development, productivity and weight gain compared to lower values.

This study aimed to evaluate the effect of food type on growth performance and water quality of angelfish juvenile. A total of 168 angelfish Pterophyllum scalare var. marble (w(i) = 151.3 +/- 37.9 mg e l(i) = 2.2 +/- 0.07 cm) were distributed in 12 aquaria 14 L (1.0 fish L(-1)). The experiment was conducted in a complete randomized design with three treatments and four replicates. Foods evaluated were: Artemia nauplii, commercial flakes diet and commercial powder diet. Fish weight and length were recorded in the beginning and the end of 60 experimental days. Water temperature, dissolved oxygen, pH and total ammonia were monitored during experiment. Best averages of final weight, final length, weight gain and condition factor were observed on fish fed powder diet. Specific growth rate, weight uniformity and survival were not influenced (p > 0.05) by food type. Just on length uniformity fish fed Artemia showed better averages than fish fed flakes diet and powder diet. Foods evaluated did not influenced (p > 0.05) on water quality parameters. In conclusion, for juvenile angelfish, the food type influences growth performance without affect water quality parameters. Powder diet resulted in better growth performance.

Drogas naturais ou sintéticas podem ser capazes de alterar a sobrevivência de culturas bacterianas, interferir na marcação de estruturas sanguíneas com tecnécio- 99m (99mTc) e alterar a morfologia das hemácias. De acordo com as instruções do fabricante, a formula denominada de Três Bailarinas (TB) é sugerida para ser usada, como bebida, por pessoas que desejam ajustar o peso sem dieta. Os ingredientes dessa fórmula são a Cassia angustifolia e a Malva verticellate. Informações cientificas sobre TB não foram encontradas no indexador PubMed, e esse fato tem estimulado nossas investigações sobre seus efeitos biológicos. O objetivo deste estudo foi avaliar, em diferentes modelos experimentais, o efeito de um extrato aquoso de Três Bailarinas: (i) na sobrevivência de culturas de E. coli AB1157, ii) efeito do SnCl2 em culturas bacterianas, iii) na marcação das hemácias e proteínas plasmáticas e celulares com 99mTc e iv) na morfologia de hemácias de sangue de ratos Wistar. Os resultados encontrados demonstram que o extrato de TB não alterou a sobrevivência de cultura de E. coli AB1157 e aboliu o efeito letal do SnCl2 na sobrevivência dessa cultura bacteriana. Na marcação de estruturas sangüíneas com 99mTc o extrato de TB reduziu a percentagem de atividade (%ATI) referente ao 99mTc no compartimento celular e nas proteínas plasmáticas, mas não alterou a %ATI nas proteínas celulares. O extrato de TB não foi capaz de alterar a morfologia das hemácias. Os modelos experimentais realizados mostram a importância dos mesmos na avaliação de efeitos biológicos de agentes químicos, e contribui para um melhor entendimento das propriedades do extrato de Três Bailarinas. Esse trabalho abrange varias áreas do conhecimento, tais como: radiobiologia, botânica, fitoterapia e hematologia

Este trabalho teve como objetivo avaliar o efeito da levedura desidratada de álcool (Saccharomyces cerevisae) como substituto do suplemento vitamínico, em dietas de tilápia-do-nilo (Oreochromis niloticus). Trezentos alevinos machos, sexualmente revertidos, com peso médio de 1,5 g, foram distribuídos igualmente em 20 aquários de fibra de vidro, equipados com filtro biológico. Cinco dietas isoprotéicas com 32% de proteína bruta na primeira fase (40 dias) e com 28% de proteína bruta na segunda fase (74 dias) foram fornecidas ad libitum, duas vezes ao dia. A dieta-controle não conti-nha levedura, e as demais apresentavam 10% de levedura. O delineamento foi inteiramente casualizado, com cinco tratamentos e quatro repetições. Não houve influência significativa dos tratamentos sobre o ganho de peso e taxa de crescimento específico. Foi observada, nos peixes alimentados com dietas sem suplemento vitamínico, alta mortalidade. Os peixes que receberam levedura apresentaram menor conteúdo corporal de proteína, e maior, de lipídeos. A levedura desidratada de álcool pode ser usada como fonte de vitaminas hidrossolúveis em dietas para alevinos de tilápia-do-nilo.

O objetivo deste estudo foi avaliar o efeito do horário, taxa e freqüência de alimentação no desempenho do matrinxã (Brycon amazonicus) em tanques de cultivo. O trabalho foi realizado no Centro de Aqüicultura da UNESP, Jaboticabal, SP, no período de outubro de 1997 a janeiro de 1998 sendo realizados três ensaios, em tanques de 200m² subdivididos em 4 de 50 m². No ensaio I foram medidos em 3 períodos (manhã-m; meio do dia-md e tarde-t) o consumo de ração, índice de ingestão, tempo de saciação e velocidade de ingestão em peixes com peso médio de 232,13 g, alimentados com ração extrusada (32% de PB). Não foi observada diferença significativa nos parâmetros analisados. No ensaio II, em peixes com peso médio de 233,98 g, foi medido o consumo médio de ração, em intervalos de duas horas, das 07 às 19 horas. O maior consumo ocorreu quando o matrinxã foi alimentado às 17 horas. No ensaio III, durante 57 dias, os peixes foram alimentados uma vez ao dia (m); uma vez ao dia (t); duas vezes ao dia(m/t) e três vezes ao dia (m,md,t). Peixes com peso médio inicial de 322,25 g receberam ração com 32% de PB, na quantidade de 2% do PV. Não foram observadas diferenças significativas no ganho de peso diário (3,17; 2,80; 3,04 e 2,81 g) e na conversão alimentar aparente (2,11; 2,48; 2,16 e 2,31:1). Concluiu-se que a freqüência de alimentação de uma vez ao dia, em qualquer horário, mostrou ser suficiente.

Aim: The aim of this work was to investigate the hypothesis that catechol and 3MC inhibit FADH2-linked basal respiration in mitochondria isolated from rat liver and brain homogenates. Moreover, catechol ability to induce DNA damage in rat brain cells through the comet assay (alkaline single-cell gel electrophoresis assay) was also observed. Methods: Two different catechols were evaluated: pirocatechol (derived from benzene) and 3-methylcatechol (derived from toluene); rat liver and brain homogenates were incubated with 1mM catechol at pH 7.4 for up to 30 minutes. After that, mitochondrial fractions were isolated by differential centrifugation. Basal oxygen uptake was measured using a Clark-type electrode after the addition of 10 mM sodium succinate for a period of 12 minutes. In additional experiments, rat brain cells were treated with 1, 5 and 10mM pirocatechol for up to 20 minutes at 37º C, and submitted to electrophoresis. Results: Catechols (pirocatechol and 3methylcatechol) induced a time-dependent partial inhibition of FADH2-linked basal mitochondrial respiration. Indeed, pirocatechol was able to produce a dosedependent DNA oxidative damage in rat brain cells by 2 and 4 injury levels. These results suggest that reactive oxygen species generated by the oxidation of catechols, induced an impairment on mitochondrial respiration and a DNA damage, which might be related to their citotoxicity. Conclusion: Catechols produced an inhibition of basal respiration associated to FADH2 in isolated liver and brain mitochondria; 3-methylcatechol, at the same concentration, produced similar toxicity in the mitochondrial model. Indeed, pirocatechol induced a DNA damage in rat brain cells, mainly observed in comets formation and consequent DNA degradation

Diabetes mellitus has been associated with bone metabolism alterations, such as osteopenia and osteporosis. So, the search of new anabolic agents promote bone mass gain can be important to prevent osteoporosis. The aim of this study was evaluate zinc anabolic effect over bone in diabetic and post-menopausal osteopenic models. Diabetes was induced by STZ (45mg/Kg of body weight) administration and post-menopausal by bilateral ovariectomy. Adults female Wistar rats (n=65) were divided in 5 groups: control group (n=15), ovariectomized without (n=15) and with zinc supplementation (n=10) groups, diabetic and ovarioctomized without (n=15) and with zinc supplementation (n=10) groups. Studied periods had been untill 90 days. Diabetic condition was confirmed hiperglicemic state and alterations of state with polyuria, polyphagia, polydipsia and glucosuria. Histomorphometric analysis showed that zinc supplementation increased trabecular thickness and reduced trabecular distance significantly in diabetic groups with similar values to those showed in control group. Correlation analysis of histomorphometric parameters with serum glucose concentration showed that more time in hyperglycemia more bone damage, as well as, zinc supplementation contributed to prevent this damage. Elevated serum glucose caused hyperzincuria, phosphaturia and calciuria. Zinc supplementation promoted increased levels of calcium and phosphorous ions in 90th days diabetic group. No alteration was observed by ovariectomy in mineral (Ca, P and Zn) serum and urine concentrations. Total serum Alkaline Phosphatase activity increased in diabetic groups, supplemented or not, compared with control group. However, Tartarate-Resistant Acid Phosphatase, magnesium and serum zinc did not altered in studied groups. Serum albumin was reduced only in diabetic groups. Serum creatinine was unaltered. These results support the hypotesis that zinc can be used to prevent and treat diabetic and post-menopausal osteopenia

O presente trabalho foi realizado com o objetivo de quantificar as interações competitivas e os índices de competitividade entre plantas de tomate industrial (Lycopersicon esculentum cv. Heinz 9553) e maria-pretinha (Solanum americanum). Usou-se como método um experimento substitutivo, com densidade total de 40 plantas m-2 e 11 proporções, além das monoculturas em densidades, que variaram de 20 a 100 plantas m-2, em intervalos de 20 plantas, conduzidos no delineamento de blocos casualizados com três repetições. Os resultados obtidos foram analisados pelo método convencional de análise de experimentos substitutivos e pela produção recíproca total. A maria-pretinha mostrou ser um competidor mais agressivo que o tomate, sendo mais importante a competição interespecífica para a planta cultivada. Para a biomassa seca total, as duas espécies não competiram pelos mesmos fatores de crescimento. Já para a área foliar, as duas espécies se mostraram competidoras pelos mesmos fatores de crescimento, mostrando ser essa a característica mais sensível à interferência.

O objetivo deste trabalho foi avaliar a interferência de densidades de tubérculos de tiririca no crescimento inicial de plantas de algodão, cultivar Delta Opal. Para isso, utilizaram-se caixas de cimento-amianto (0,60 x 0,60 x 0,25 m) com Latossolo Vermelho Escuro, no centro das quais foram semeadas seis sementes de algodão, em linha, espaçadas de 0,10 m; em seguida, plantaram-se tubérculos de tiririca nas densidades de 0, 5, 10, 15, 25, 50, 75, 100, 125, 150, 175 e 200 tubérculos por caixa, que foram nestas distribuídos aleatoriamente. O delineamento experimental foi o de blocos casualizados, em quatro repetições. Aos 50 dias após a semeadura, foram determinados, nas plantas de algodão, a altura e o teor relativo de clorofila total. Ao término do período experimental (65 dias após a emergência do algodão), foram determinados: altura, teor relativo de clorofila total, área foliar e matéria seca do caule e das folhas. Nas plantas de tiririca foi feita a contagem do número final de plantas (partes aéreas) por caixa. A interferência da tiririca no crescimento inicial do algodoeiro iniciou-se a partir de 5 tubérculos/caixa, reduzindo em até 71% as características analisadas na mais alta densidade (200 tubérculos/caixa). A competição intra-específica na tiririca acentuou-se a partir de 75 tubérculos/caixa, quando obteve mais do que 1,86 brotação por tubérculo. As características avaliadas nas plantas de algodão mais sensíveis ao efeito dos tubérculos de tiririca foram: área foliar, massa seca de folhas, caule e altura das plantas, nesta ordem.
