A study was conducted to investigate the effects of wheat straw ammonisation and supplementation with a rumen undegradable protein (UDP) source on nutrient digestion and nitrogen balance by lambs while diets were supplemented with kibbled carob pods as energy source. Ammonisation increased the crude protein content of wheat straw by nearly 100% and decreased the contents of neutral detergent fibre and acid detergent fibre by 7% and 1.7% respectively. Treating the straw with ammonia resulted in significant (P<0.01) increase in nitrogen (N) intake and intakes of organic matter (OM) and dry matter (DM) tended toward significance (P<0.1). The UDP source had no effect (P>0.05) on DM and OM intakes but resulted in an increase (P<0.05) of N intakes. Both, ammonization and UDP supplementation increased (P<0.01) the DM, OM and N digestibility. In conclusion, the results of this study suggest that ammonisation and UDP supplementation is a practical dietary manipulation option to improve the nutritional status of ruminants fed on roughage-based diets.

The galE gene of Streptomyces lividans was used to probe a cosmid library harbouring Brucella melitensis 16M DNA and the nucleotide sequence of a 2.5 kb ClaI fragment which hybridised was determined. An open reading frame encoding a predicted polypeptide with significant homology to UDP-galactose-4-epimerases of Brucella arbortus strain 2308 and other bacterial species was identified. DNA sequences flanking the B. melitensis galE gene shared no identity with other gal genes and, as for B. abortus, were located adjacent to a mazG homologue. A plasmid which encoded the B. melitensis galE open reading frame complemented a galE mutation in Salmonella typhimurium LB5010, as shown by the restoration of smooth lipopolysaccharide (LPS) biosynthesis, sensitivity to phage P22 infection and restoration of UDP-galactose-4-epimerase activity. The galE gene on the B. melitensis 16M chromosome was disrupted by insertional inactivation and these mutants lacked UDP-galactose-4-epimerase activity but no discernible differences in LPS structure between parent and the mutants were observed. One B. melitensis 16M galE mutant, Bm92, was assessed for virulence in CD-1 and BALB/c mice and displayed similar kinetics of invasion and persistence in tissues compared with the parent bacterial strain. Plant traits – the morphological, anatomical, physiological, biochemical and phenological characteristics of plants and their organs – determine how primary producers respond to environmental factors, affect other trophic levels, influence ecosystem processes and services and provide a link from species richness to ecosystem functional diversity. Trait data thus represent the raw material for a wide range of research from evolutionary biology, community and functional ecology to biogeography. Here we present the global database initiative named TRY, which has united a wide range of the plant trait research community worldwide and gained an unprecedented buy-in of trait data: so far 93 trait databases have been contributed. The data repository currently contains almost three million trait entries for 69 000 out of the world's 300 000 plant species, with a focus on 52 groups of traits characterizing the vegetative and regeneration stages of the plant life cycle, including growth, dispersal, establishment and persistence. A first data analysis shows that most plant traits are approximately log-normally distributed, with widely differing ranges of variation across traits. Most trait variation is between species (interspecific), but significant intraspecific variation is also documented, up to 40% of the overall variation. Plant functional types (PFTs), as commonly used in vegetation models, capture a substantial fraction of the observed variation – but for several traits most variation occurs within PFTs, up to 75% of the overall variation. In the context of vegetation models these traits would better be represented by state variables rather than fixed parameter values. The improved availability of plant trait data in the unified global database is expected to support a paradigm shift from species to trait-based ecology, offer new opportunities for synthetic plant trait research and enable a more realistic and empirically grounded representation of terrestrial vegetation in Earth system models.

The self-assembly and bioactivity of the peptide–polymer conjugate DGRFFF–PEG3000 containing the RGD cell adhesion motif has been examined, in aqueous solution. The conjugate is designed to be amphiphilic by incorporation of three hydrophobic phenylalanine residues as well as the RGD unit and a short poly(ethylene glycol) (PEG) chain of molar mass 3000 kg mol-1. Above a critical aggregation concentration, determined by fluorescence measurements, signals of b-sheet structure are revealed by spectroscopic measurements, as well as X-ray diffraction. At high concentration, a self-assembled fibril nanostructure is revealed by electron microscopy. The fibrils are observed despite PEG crystallization which occurs on drying. This suggests that DGRFFF has an aggregation tendency that is sufficiently strong not to be prevented by PEG crystallization. The adhesion, viability and proliferation of human corneal fibroblasts was examined for films of the conjugate on tissue culture plates (TCPs) as well as low attachment plates. On TCP, DGRFFF–PEG3000 films prepared at sufficiently low concentration are viable, and cell proliferation is observed. However, on low attachment surfaces, neither cell adhesion nor proliferation was observed, indicating that the RGD motif was not available to enhance cell adhesion. This was ascribed to the core–shell architecture of the self-assembled fibrils with a peptide core surrounded by a PEG shell which hinders access to the RGD unit.

Bis-triazinylphenanthroline ligands (BTPhens), which contain additional alkyl (n-butyl and sec-butyl) groups attached to the triazine rings, have been synthesized, and the effects of this alkyl substitution on their extraction properties with Ln(III) and An(III) cations in simulated nuclear waste solutions have been studied. The speciation of n-butyl-substituted ligand (C4- BTPhen) with some trivalent lanthanide nitrates was elucidated by 1 H-NMR spectroscopic titrations. These experiments have shown that the dominant species in solution were the 1:2 complexes [Ln(III)(BTPhen)2], even at higher Ln(III) concentrations, and the relative stability of 2:1 to 1:1 BTPhen-Ln(III) complexes varied with different lanthanides. As expected, sec-butylsubstituted ligand (sec-C4 BTPhen) showed higher solubility than C4-BTPhen in certain diluents. A greater separation factor (SFAm/Eu = ca. 210) was observed for sec-C4-BTPhen compared to C4-BTPhen (SFAm/Eu = ca. 125) in 1-octanol at 4 M HNO3 solutions. The greater separation factor may be due to the higher solubility of the 2:1 complex for sec-C4-BTPhen at the interface than the 1:1 complex of C4-BTPhen.

At the beginning of the Medieval Climate Anomaly, in the ninth and tenth century, the medieval eastern Roman empire, more usually known as Byzantium, was recovering from its early medieval crisis and experiencing favourable climatic conditions for the agricultural and demographic growth. Although in the Balkans and Anatolia such favourable climate conditions were prevalent during the eleventh century, parts of the imperial territories were facing significant challenges as a result of external political/military pressure. The apogee of medieval Byzantine socio-economic development, around AD 1150, coincides with a period of adverse climatic conditions for its economy, so it becomes obvious that the winter dryness and high climate variability at this time did not hinder Byzantine society and economy from achieving that level of expansion. Soon after this peak, towards the end of the twelfth century, the populations of the Byzantine world were experiencing unusual climatic conditions with marked dryness and cooler phases. The weakened Byzantine socio-political system must have contributed to the events leading to the fall of Constantinople in AD 1204 and the sack of the city. The final collapse of the Byzantine political control over western Anatolia took place half century later, thus contemporaneous with the strong cooling effect after a tropical volcanic eruption in AD 1257. We suggest that, regardless of a range of other influential factors, climate change was also an important contributing factor to the socio-economic changes that took place in Byzantium during the Medieval Climate Anomaly. Crucially, therefore, while the relatively sophisticated and complex Byzantine society was certainly influenced by climatic conditions, and while it nevertheless displayed a significant degree of resilience, external pressures as well as tensions within the Byzantine society more broadly contributed to an increasing vulnerability in respect of climate impacts. Our interdisciplinary analysis is based on all available sources of information on the climate and society of Byzantium, that is textual (documentary), archaeological, environmental, climate and climate model-based evidence about the nature and extent of climate variability in the eastern Mediterranean. The key challenge was, therefore, to assess the relative influence to be ascribed to climate variability and change on the one hand, and on the other to the anthropogenic factors in the evolution of Byzantine state and society (such as invasions, changes in international or regional market demand and patterns of production and consumption, etc.). Aims Glycosylation with beta-N-acetylglucosamine (O-GlcNAcylation) is one of the most complex post-translational modifications. The cycling of O-GlcNAc is controlled by two enzymes: UDP-NAc transferase (OGT) and O-GlcNAcase (OGA). We recently reported that endothelin-1 (ET-1) augments vascular levels of O-GlcNAcylated proteins. Here we tested the hypothesis that O-GlcNAcylation contributes to the vascular effects of ET-1 via activation of the RhoA/Rho-kinase pathway. Methods and results Incubation of vascular smooth muscle cells (VSMCs) with ET-1 (0.1 mu M) produces a time-dependent increase in O-GlcNAc levels. ET-1-induced O-GlcNAcylation is not observed when VSMCs are previously transfected with OGT siRNA, treated with ST045849 (OGT inhibitor) or atrasentan (ET(A) antagonist). ET-1 as well as PugNAc (OGA inhibitor) augmented contractions to phenylephrine in endothelium-denuded rat aortas, an effect that was abolished by the Rho kinase inhibitor Y-27632. Incubation of VSMCs with ET-1 increased expression of the phosphorylated forms of myosin phosphatase target subunit 1 (MYPT-1), protein kinase C-potentiated protein phosphatase 1 inhibitor protein (protein kinase C-potentiated phosphatase inhibitor-17), and myosin light chain (MLC) and RhoA expression and activity, and this effect was abolished by both OGT siRNA transfection or OGT inhibition and atrasentan. ET-1 also augmented expression of PDZ-Rho GEF (guanine nucleotide exchange factor) and p115-Rho GEF in VSMCs and this was prevented by OGT siRNA, ST045849, and atrasentan. Conclusion We suggest that ET-1 augments O-GlcNAcylation and this modification contributes to increased vascular contractile responses via activation of the RhoA/Rho-kinase pathway.

Optimized experimental conditions for extracting accurate information at subpixel length scales from analyzer-based X-ray imaging were obtained and applied to investigate bone regeneration by means of synthetic beta-TCP grafting materials in a rat calvaria model. The results showed a 30% growth in the particulate size due to bone ongrowth/ingrowth within the critical size defect over a 1-month healing period.

Nicotinic acetylcholine receptors (nAChRs) were studied in detail in the past regarding their interaction with therapeutic and drug addiction related compounds. Using fast kinetic whole-cell recording, we have now studied effects of tacrine, an agent used clinically to treat Alzheimer`s disease, on currents elicited by activation of rat alpha(3)beta(4) nAChR heterologously expressed in KX alpha(3)beta(4)R2 cells. Characterization of receptor activation by nicotine used as agonist revealed a K(d) of 23 +/- 0.2 mu M and 4.3 +/- 1.3 for the channel opening equilibrium constant, Phi(-1). Experiments were performed to investigate whether tacrine is able to activate the alpha(3)beta(4) nAChR. Tacrine did not activate whole-cell currents in KX alpha(3)beta(4)R2 cells but inhibited receptor activity at submicromolar concentration. Dose response curves obtained with increasing agonist or inhibitor concentration revealed competitive inhibition of nAChRs by tacrine, with an apparent inhibition constant, K(I), of 0.8 mu M. The increase of Phi(-1) in the presence of tacrine suggests that the drug stabilizes a nonconducting open channel form of the receptor. Binding studies with TCP and MK-801 ruled out tacrine binding to common allosteric sites of the receptor. Our study suggests a novel mechanism for action of tacrine on nAChRs besides inhibition of acetylcholine esterase.

The main objective of this thesis work is to develop communication link between Runrev Revolution (IDE) and JADE (Multi-Agent System) through Socket programming using TCP/IP layer. These two independent platforms are connected using socket programming technique. Socket programming is considered to be newly emerging technology among these two platforms, the work done in this thesis work is considered to be a prototype.A Graphical simulation model is developed by salixphere (Company in Hedemora) to simulate logistic problems using Runrev Revolution (IDE). The simulation software/program is called “BIOSIM”. The logistic problems are complex, and conventional optimization techniques are unlikely very successful. “BIOSIM” can demonstrate the graphical representation of logistic problems depending upon the problem domains. As this simulation model is developed in revolution programming language (Transcript) which is dynamically typed and English-like language, it is quite slow compared to other high level programming languages. The object of this thesis work is to add intelligent behaviour in graphical objects and develop communication link between Runrev revolution (IDE) and JADE (Multi-Agent System) using TCP/IP layers.The test shows the intelligent behaviour in the graphical objects and successful communication between Runrev Revolution (IDE) and JADE (Multi-Agent System).

A S100B é uma proteína ligante de cálcio, de massa molecular de 21kDa, expressa principalmente por astrócitos. Esta proteína tem sido implicada em atividades funcionais tanto intra quanto extracelulares. Muitos estudos têm sugerido que intracelularmente ela está envolvida na modulação de proteínas do citoesqueleto e na regulação do ciclo celular. A proteína S100B pode ser secretada pelos astrócitos e desenvolver atividades extracelulares, que parecem depender de sua concentração. Em concentração nanomolar ela atua como fator trófico às células neurais, enquanto que em concentrações micromolar ela pode ser neurotóxica. A quantificação da proteína S100B no sangue e líquor se correlaciona com a extensão e intensidade do dano ao sistema nervoso central (SNC) o que permite sua utilização em estudos como marcador bioquímico de dano ou disfunção cerebral. Esta tese está dividida em três partes. A primeira parte propõe a utilização clínica da proteína S100B em patologias com envolvimento do SNC como a síndrome de Down, mielopatia associada ao vírus HTLV-I, lupus eritematoso sistêmico, epilepsia secundária a neurocisticercose e, além disso demonstramos a curva de ontogenia da S100B no sangue. Na segunda parte descrevemos uma atividade de nucleotidases presente em líquor de ratos, e finalmente, na terceira parte abordamos as perspectivas para futuros trabalhos. Os resultados obtidos pelo nosso grupo e por outros grupos internacionais relatam que a proteína S100B é um marcador inespecífico para evidenciar dano ou disfunção em doenças agudas e crônicas com envolvimento do SNC. Apesar de ser um marcador inespecífico, medidas dos níveis da proteína S100B tem grande sensibilidade para detectar uma resposta celular cerebral inespecífica. Além disso, demonstramos que estudos clínicos com esta proteína necessitam controles pareados por idade e sexo. A atividade nucleotidásica descrita no líquor de ratos hidrolisa preferencialmente o GDP e UDP comparado aos outros nucleotídeos. Nas perspectivas, os resultados mostrados são referentes a experimentos preliminares o que torna prematuro qualquer tipo de conclusão.

A área de gerência de rede cresce à medida que redes mais seguras e menos vulneráveis são necessárias, e as aplicações que concorrem pelo seu uso, necessitam de alta disponibilidade e qualidade de serviço. Quando estamos focando a gerência da infra-estrutura física das redes de computadores, por exemplo, a taxa de uso de um segmento de rede, podemos dizer que esse tipo de gerenciamento encontra-se em um patamar bastante sedimentado e testado. Por outro lado, há ainda lacunas para pesquisar na área de gerenciamento de protocolos de alto nível. Entender o comportamento da rede como um todo, conhecer quais hosts mais se comunicam, quais aplicações geram mais tráfego e, baseado nessas estatísticas, gerar uma política para a distribuição de recursos levando em consideração as aplicações críticas é um dever nas redes atuais. O grupo de trabalho IETF RMON padronizou, em 1997, a MIB RMON2. Ela foi criada para permitir a monitoração de protocolos das camadas superiores (rede, transporte e aplicação), a qual é uma boa alternativa para realizar as tarefas de gerenciamento recém mencionadas. Outro problema para os gerentes de rede é a proliferação dos protocolos de alto nível e aplicações corporativas distribuídas. Devido a crescente quantidade de protocolos e aplicações sendo executados sobre as redes de computadores, os softwares de gerenciamento necessitam ser adaptados para serem capazes de gerenciá-los. Isso, atualmente, não é fácil porque é necessário usar linguagens de programação de baixo nível ou atualizar o firmware dos equipamentos de monitoração, como ocorre com os probes RMON2. Considerando este contexto, esse trabalho propõe o desenvolvimento de um agente RMON2 que contemple alguns grupos dessa MIB. O agente baseia-se na monitoração protocolos de alto nível e aplicações que são executados sobre o IP (Internet Protocol) e Ethernet (na camada de enlace). Além da implementação do agente, o trabalho apresenta um estudo de como obter estatísticas do agente RMON2 e usá-las efetivamente para gerenciar protocolos de alto nível e aplicações.

A informática vem adquirindo papéis cada vez mais importantes na vida cotidiana. Um dos papéis mais significativos, hoje, é o suporte a comunicações; atualmente, é muito difícil pensar em comunicações – mesmo interpessoais – sem fazer associação às áreas de informática e redes. Dentre as aplicações que utilizam informática e redes como suporte, a tecnologia de videoconferência tem recebido papel de destaque. Os avanços na tecnologia de redes e conectividade, aliados à padronização e à crescente oferta de produtos de videoconferência, têm aumentado a aplicabilidade e a popularidade destes produtos, sobretudo utilizados sobre arquitetura de redes TCP/IP. Trata-se de uma tecnologia atraente em termos de resultado, por agregar, além do áudio – recurso comum há muito tempo como suporte à comunicação – os recursos de vídeo e aplicações integradas (como quadro-branco compartilhado, Chat, troca de arquivos e outros). Contudo, essas aplicações são bastante exigentes, tanto em termos de banda quanto de qualidade de serviço (QoS) da rede. O primeiro item se justifica pelo volume de dados gerados pelas aplicações de videoconferência; o segundo, pela significativa influência que os problemas de qualidade da infraestrutura de rede (como elevada latência, jitter e descartes) podem exercer sobre tais aplicações. A busca para as soluções destes problemas não é tarefa simples, pois muitas vezes envolve investimentos que desencorajam a adoção da tecnologia de videoconferência – principalmente para uso pessoal ou por empresas pequenas. Este trabalho propõe uma solução aos problemas mencionados, visando proporcionar uma melhor aceitação e maior disseminação da tecnologia de videoconferência, valendo-se de recursos com pouca demanda de investimento. A estratégia abordada é a adaptação de tráfego, com um enfoque diferenciado: o de levar em conta, para cada aplicação, o comportamento que o processo de adaptação apresentasse. A partir dessa orientação, é proposto um modelo de adaptação de tráfego orientado ao perfil da aplicação, voltado ao interesse do usuário, e que disponibilize uma forma ao mesmo tempo simples e eficiente para que o usuário realize a adequação do mecanismo de adaptação do sistema às suas necessidades e expectativas. A partir desta proposta, foi implementado um protótipo de aplicação, com o objetivo de verificar a funcionalidade do modelo em termos práticos. As observações dos resultados dos testes, bem como as conclusões geradas, serviram como validação da proposta.
