The dynamicity and heterogeneity that characterize pervasive environments raise new challenges in the design of mobile middleware. Pervasive environments are characterized by a significant degree of heterogeneity, variability, and dynamicity that conventional middleware solutions are not able to adequately manage. Originally designed for use in a relatively static context, such middleware systems tend to hide low-level details to provide applications with a transparent view on the underlying execution platform. In mobile environments, however, the context is extremely dynamic and cannot be managed by a priori assumptions. Novel middleware should therefore support mobile computing applications in the task of adapting their behavior to frequent changes in the execution context, that is, it should become context-aware. In particular, this thesis has identified the following key requirements for novel context-aware middleware that existing solutions do not fulfil yet. (i) Middleware solutions should support interoperability between possibly unknown entities by providing expressive representation models that allow to describe interacting entities, their operating conditions and the surrounding world, i.e., their context, according to an unambiguous semantics. (ii) Middleware solutions should support distributed applications in the task of reconfiguring and adapting their behavior/results to ongoing context changes. (iii) Context-aware middleware support should be deployed on heterogeneous devices under variable operating conditions, such as different user needs, application requirements, available connectivity and device computational capabilities, as well as changing environmental conditions. Our main claim is that the adoption of semantic metadata to represent context information and context-dependent adaptation strategies allows to build context-aware middleware suitable for all dynamically available portable devices. Semantic metadata provide powerful knowledge representation means to model even complex context information, and allow to perform automated reasoning to infer additional and/or more complex knowledge from available context data. In addition, we suggest that, by adopting proper configuration and deployment strategies, semantic support features can be provided to differentiated users and devices according to their specific needs and current context. This thesis has investigated novel design guidelines and implementation options for semantic-based context-aware middleware solutions targeted to pervasive environments. These guidelines have been applied to different application areas within pervasive computing that would particularly benefit from the exploitation of context. Common to all applications is the key role of context in enabling mobile users to personalize applications based on their needs and current situation. The main contributions of this thesis are (i) the definition of a metadata model to represent and reason about context, (ii) the definition of a model for the design and development of context-aware middleware based on semantic metadata, (iii) the design of three novel middleware architectures and the development of a prototypal implementation for each of these architectures, and (iv) the proposal of a viable approach to portability issues raised by the adoption of semantic support services in pervasive applications.

Immunreaktionen gegen Antigene können eine zelluläre Immunantwort, eingeleitet durch TH1-Zellen induzieren oder aber eine antikörpervermittelte humorale Immunantwort induzieren, die von Zellen des TH2-Typs bestimmt sind. Diese unterschiedlichen Immunreaktionen regulieren sich gegenseitig, indem sie sich wechselseitig unterdrücken. Dadurch stellt sich ein Gleichgewicht zwischen den TH1- und TH2-Immunantworten ein. Die Allergie vom Soforttyp ist TH2-dominiert und wird ausgelöst durch die Produktion spezifischer IgE-Antikörper gegen eigentlich harmlose Antigene. Durch die frühe Ausschüttung des Zytokins IL-4 von T-Zellen differenzieren naive allergenspezifische T-Zellen zu TH2-Zellen aus. Diese sezernieren dann ebenfalls IL-4 und auch IL-13, was die B-Zellen zu einer vermehrten Produktion von IgE-Molekülen stimuliert. Die Untersuchung "enklisis-Modusterminologie und Modusbegriff in der antiken griechischen Grammatik" stellt dar, auf welchen antiken griechischen Termini die über das Lateinische vermittelte Modusterminologie der neuzeitlichen Traditionellen Grammatik („Schulgrammatik“) beruht. Sie ergründet auf der Basis des zugrundeliegenden Modusbegriffs die jeweilige Bezeichnungsmotivation der Termini und versucht aufzuzeigen, inwieweit diese formalen bzw. funktionalen Motive einen aus moderner Sicht problematisch wirkenden Terminus im Kontext der antiken Betrachtung verständlich und angemessen erscheinen lassen. Vor Beginn der eigentlichen Untersuchung werden im einleitenden Teil zunächst grammatisch-sprachwissenschaftliche Grundlagen gelegt. Ausgehend von einer allgemeinen Begriffsbestimmung des Modus wird zunächst sein Standort in der antiken Grammatik skizziert und die zu zugehörige Terminologie aufgeführt. Der zweite Teil der Einleitung hat die terminologisch-methodischen Voraussetzungen zum Gegenstand, auf die sich die Untersuchung der griechischen Modustermini stützt. Darin werden auch zentrale Begriffe wie ‘Terminus’ und ‘Bezeichnungsmotivation’ diskutiert und auf die antiken Verhältnisse angewendet. Der erste Hauptteil behandelt die Geschichte der griechischen Modusterminologie. Ausgehend von den Sprachphilosophen und frühen Grammatikern werden Verwendung und Bedeutung des Terminus enklisis und seiner Alternativen in der antiken grammatischen Literatur untersucht und dem lateinischen Terminus modus gegenübergestellt. Desweiteren werden die Termini für einzelnen Modi ‘Indikativ’, ‘Imperativ’, ‘Optativ’, ‘Konjunktiv’ und ‘Infinitiv’, den die griechischen Grammatiker auch der Kategorie ‘Modus’ zuordnen, untersucht, soweit sie in den antiken Quellen benutzt und diskutiert werden. Das ursprüngliche Motiv eines Terminus wird vor dem Hintergrund seiner Entstehung gedeutet und im Hinblick auf die Entwicklung des Terminus im schulgrammatischen Gebrauch auf seine Universalität hin überprüft. Dabei zeigt sich, daß einige Termini so exakt an die griechischen Verhältnisse angepaßt sind, daß bereits die Übertragung ins Lateinische Verluste mit sich bringt. Im zweiten Hauptteil sind Textpassagen aus der griechischen grammatischen Literatur, die in den vorangehenden Teilen für die terminologische und begriffliche Auswertung immer wieder herangezogen werden, als Testimonien zusammengestellt. Sie sind mit einem textkritischem Apparat sowie einer Kommentierung versehen und – wie alle anderen angeführten Stellen aus lateinischen und griechischen Quellen – ins Deutsche übersetzt.

The quark condensate is a fundamental free parameter of Chiral Perturbation Theory ($chi PT$), since it determines the relative size of the mass and momentum terms in the power expansion. In order to confirm or contradict the assumption of a large quark condensate, on which $chi PT$ is based, experimental tests are needed. In particular, the $S$-wave $pipi$ scattering lengths $a_0^0$ and $a_0^2$ can be predicted precisely within $chi PT$ as a function of this parameter and can be measured very cleanly in the decay $K^{pm} to pi^{+} pi^{-} e^{pm} stackrel{mbox{tiny(---)}}{nu_e}$ ($K_{e4}$). About one third of the data collected in 2003 and 2004 by the NA48/2 experiment were analysed and 342,859 $K_{e4}$ candidates were selected. The background contamination in the sample could be reduced down to 0.3% and it could be estimated directly from the data, by selecting events with the same signature as $K_{e4}$, but requiring for the electron the opposite charge with respect to the kaon, the so-called ``wrong sign'' events. This is a clean background sample, since the kaon decay with $Delta S=-Delta Q$, that would be the only source of signal, can only take place through two weak decays and is therefore strongly suppressed. The Cabibbo-Maksymowicz variables, used to describe the kinematics of the decay, were computed under the assumption of a fixed kaon momentum of 60 GeV/$c$ along the $z$ axis, so that the neutrino momentum could be obtained without ambiguity. The measurement of the form factors and of the $pipi$ scattering length $a_0^0$ was performed in a single step by comparing the five-dimensional distributions of data and MC in the kinematic variables. The MC distributions were corrected in order to properly take into account the trigger and selection efficiencies of the data and the background contamination. The following parameter values were obtained from a binned maximum likelihood fit, where $a_0^2$ was expressed as a function of $a_0^0$ according to the prediction of chiral perturbation theory: f'_s/f_s = 0.133+- 0.013(stat)+- 0.026(syst) f''_s/f_s = -0.041+- 0.013(stat)+- 0.020(syst) f_e/f_s = 0.221+- 0.051(stat)+- 0.105(syst) f'_e/f_s = -0.459+- 0.170(stat)+- 0.316(syst) tilde{f_p}/f_s = -0.112+- 0.013(stat)+- 0.023(syst) g_p/f_s = 0.892+- 0.012(stat)+- 0.025(syst) g'_p/f_s = 0.114+- 0.015(stat)+- 0.022(syst) h_p/f_s = -0.380+- 0.028(stat)+- 0.050(syst) a_0^0 = 0.246+- 0.009(stat)+- 0.012(syst)}+- 0.002(theor), where the statistical uncertainty only includes the effect of the data statistics and the theoretical uncertainty is due to the width of the allowed band for $a_0^2$.
The use of flat panel angioCT (DynaCT) for navigation through a deformed and fractured carotid stent

Navigation through a previously deployed and deformed stent is a difficult interventional task. Inadvertent navigation through the struts of a stent can potentially lead to incomplete secondary stent extension and vessel occlusion. Better visualisation of the pathway through the stent can reduce the risks of the procedural complications and reduce the reluctance of the interventionalist to navigate through a previously deployed stent. We describe a technique of visualisation of the pathway navigated by a guidewire through a previously deployed deformed and fractured carotid stent by the use of DynaCT. Three-dimensional reconstruction of the stent/microwire allows excellent visualisation of the correct pathway of the microwire within the stent.

Objectives The purpose of this study was to assess the impact of renal insufficiency (RI) on the distribution pattern of peripheral arterial disease (PAD). We hypothesised that RI is associated with a distally accentuated involvement of the peripheral arterial tree. Design This is a retrospective analysis. Materials and Methods Analysis was based on a consecutive series of 2709 patients with chronic PAD of atherosclerotic origin undergoing primary endovascular treatment of lower-extremity arteries. Atherosclerotic pattern was grouped into femoropopliteal (n = 2085) and infragenicular (n = 892) disease according to target lesions treated while using iliac disease (n = 1133) as reference. Univariable and multivariable multinomial regression analyses were performed to assess relation with RI. Results are shown as relative risk ratio (RRRs) with 95% confidence intervals (95% CIs). A p < 0.05 was considered statistically significant. RI was defined as glomerular filtration rate (GFR) < 60 ml min−1 1.73 m−2. Results Presence of RI was an independent risk factor for a centrifugal lesion pattern (RRR 1.48, 95% CI: 1.17–1.86, p = 0.001). Moreover, a decrease in GFR by 10 ml min−1 1.73 m−2 was associated with an RRR of 1.08 for below-the-knee arterial disease (95% CI: 1.03–1.13, p = 0.003). Conclusion Presence and severity of RI are independent predictors of a distal obstructive pattern in patients with symptomatic PAD.

Glucocorticoids (GC) have important anti-inflammatory and pro-apoptotic activities. Initially thought to be exclusively produced by the adrenal glands, there is now increasing evidence for extra-adrenal sources of GCs. We have previously shown that the intestinal epithelium produces immunoregulatory GCs and that intestinal steroidogenesis is regulated by the nuclear receptor liver receptor homolog-1 (LRH-1). As LRH-1 has been implicated in the development of colon cancer, we here investigated whether LRH-1 regulates GC synthesis in colorectal tumors and whether tumor-produced GCs suppress T-cell activation. Colorectal cancer cell lines and primary tumors were found to express steroidogenic enzymes and regulatory factors required for the de novo synthesis of cortisol. Both cell lines and primary tumors constitutively produced readily detectable levels of cortisol, as measured by radioimmunoassay, thin-layer chromatography and bioassay. Whereas overexpression of LRH-1 significantly increased the expression of steroidogenic enzymes and the synthesis of cortisol, downregulation or inhibition of LRH-1 effectively suppressed these processes, indicating an important role of LRH-1 in colorectal tumor GC synthesis. An immunoregulatory role of tumor-derived GCs could be further confirmed by demonstrating a suppression of T-cell activation. This study describes for the first time cortisol synthesis in a non-endocrine tumor in humans, and suggests that the synthesis of bioactive GCs in colon cancer cells may account as a novel mechanism of tumor immune escape.

Autophagy (literally self-eating) is a catabolic mechanism involved in the recycling and turnover of cytoplasmic constituents. Although often referred to as type II programmed cell death, autophagy is primarily a survival rather than a cell death mechanism in response to different stress stimuli. Autophagy is a process in which part of the cytoplasm or entire organelles are sequestered into double-membrane vesicles, called autophagosomes, which ultimately fuse with lysosomes to degrade their contents. Studies show that autophagy is associated with a number of pathological conditions, including cancer, infectious diseases, myopathies and neurodegenerative disorders. With respect to cancer, it has been suggested that the early stages of tumourigenesis are associated with downregulation of autophagy-related (ATG) genes. Indeed, several ATG genes display tumour suppressor function, including Beclin1, which is frequently hemizygously deleted in breast cancer cells. Conversely, in advanced stages of tumourigenesis or during anticancer therapy, autophagy may promote survival of tumour cells in adverse environmental conditions. Therefore, a thorough understanding of autophagy in different cancer types and stages is a prerequisite to determine an autophagy-activating or autophagy-inhibiting treatment strategy.

A few supergravity solutions representing configurations of NS5-branes admit exact conformal field theory (CFT) description. Deformations of these solutions should be described by exactly marginal operators of the corresponding theories. We briefly review the essentials of these constructions and present, as a new case, the operators responsible for turning on angular momentum.
