There is a worldwide interest in the development of processes for producing colorants from natural sources. Microorganisms provide an alternative source of natural colorants produced by cultivation technology and extracted from the fermented broth. The aim of the present work was to study the recovery of red colorants from the fermented broth of Talaromyces amestolkiae using the technique of colloidal gas aphrons (CGA) comprising surfactant-stabilized microbubbles. Preliminary experiments were performed to evaluate the red colorants’ solubility in different organic solvents, octanol/water partitioning, and their stability in surfactant solutions, namely hexadecyl trimethylammonium bromide (CTAB), sodium dodecyl sulfate (SDS), and polyoxyethylenesorbitan monolaurate (Tween 20), which are cationic, anionic and nonionic surfactants, respectively. The first recovery experiments were carried out using CGA generated by these surfactants at different volumetric ratios (VR, 3–18). Subsequently, two different approaches to generate CGA were investigated at VR values of 6 and 12: the first involved the use of CTAB at pH 6.9–10.0, and the second involved the use of Tween 20 using red colorants partially dissolved in ethanol and Tween 20. The characterization results showed that red colorants have a hydrophilic nature. The highest recoveries were obtained with Tween 20 (78%) and CTAB (70%). These results demonstrated that the recovery of the colorants was driven by both electrostatic and hydrophobic interactions. The VR was found to be an important operating parameter and at VR 12 with CTAB (at pH 9) maximum recovery, partitioning coefficient (K = 5.39) and selectivity in relation to protein and sugar (SP = 3.75 and SS = 7.20 respectively) were achieved. Furthermore, with Tween 20, the separation was driven mainly by hydrophobic interactions. Overall CGA show promise for the recovery of red colorants from a fermented broth. Although better results were obtained with CTAB than with Tween 20 the latter may be more suitable for some application due to its lower toxicity.

The aim of the present study was to analyze the mycobiota, occurrence of mycotoxins (aflatoxins and cyclopiazonic acid), and production of phytoalexin (trans-resveratrol) in two peanut varieties (Runner IAC 886 and Caiapo) during plant growth in the field. Climatic factors (rainfall, relative humidity and temperature) and water activity were also evaluated. The results showed a predominance of Fusarium spp. in kernels and pods, followed by Penicillium spp. and Aspergillus flavus. Aflatoxins were detected in 20% and 10% of samples of the IAC 886 and Caiapo varieties, respectively. Analysis showed that 65% of kernel samples of the IAC 886 variety and 25% of the Caiapo variety were contaminated with cyclopiazonic acid. trans-Resveratrol was detected in 6.7% of kernel samples of the IAC 886 variety and in 20% of the Caiapo variety. However, trans-resveratrol was found in 73.3% of leaf samples in the two varieties studied. (C) 2011 Published by Elsevier Ltd.

The objective of the present study was to evaluate the mycoflora and occurrence of aflatoxins in stored peanut samples (hulls and kernels) from Tupa, State of Sao Paulo, Brazil. The samples were analyzed monthly over a period of one year. The results showed a predominance of Fusarium spp. (67.7% in hulls and 25.8% in kernels) and Aspergillus spp. (10.3% in hulls and 21.8% in kernels), and the presence of five other genera. The growth of Aspergillus flavus was mainly influenced by temperature and relative humidity. Analysis of hulls showed that 6.7% of the samples were contaminated with AFB(1) (mean levels 15-23.9 mu g/kg) and AFB(2) (mean levels = 3.3-5.6 mu g/kg); in kernels, 33.3% of the samples were contaminated with AFB(1) (mean levels 7.0-116 mu g/kg) and 28.3% were contaminated with AFB(2) (mean levels = 3.3-45.5 mu g/kg). Analysis of the toxigenic potential revealed that 93.8% of the A. flavus strains isolated were producers of AFB(1) and AFB(2). mu 2007 Elsevier Ltd. All rights reserved.

Melanin pigments are substances produced by a broad variety of pathogenic microorganisms, including bacteria, fungi, and helminths. Microbes predominantly produce melanin pigment via tyrosinases, laccases, catecholases, and the polyketide synthase pathway. In fungi, melanin is deposited in the cell wall and cytoplasm, and melanin particles (""ghosts"") can be isolated from these fungi that have the same size and shape of the original cells. Melanin has been reported in several human pathogenic dimorphic fungi including Paracoccidioides brasiliensis, Sporothrix schenckii, Histoplasma capsulatum, Blastomyces dermatitidis, and Coccidioides posadasii. Melanization appears to contribute to virulence by reducing the susceptibility of melanized fungi to host defense mechanisms and antifungal drugs.

The ethanol extract from Xyris pteygoblephara aerial parts was evaluated against five microorganism strains, by the microdilution and agar diffusion methods. Extract fractionation led to the isolation of three compounds, whose structures were assigned by spectrometric data (113 and 2D NMR, IR, MS and UV) as (3R,4R)-(-)-6-methoxy-3,4-dihydro-3-n-pentil-4-acethoxy-1H-2-benzopyran-1-one (1), moronic acid and quercetin. The absolute configuration of I was defined by circular dichroism spectroscopy and comparison with data reported for other dihydroisocoumarins. Assay of 1 (100 mu g/disc) by the agar diffusion method against clinical isolates of the dermatophytes Epidermophyton floccosum (inhibition zone, mm +/- s.d.: 4.5 +/- 0.8), Trichophyton mentagrophytes (4.8 +/- 0.4) and Trichophyton rubrum (10.2 +/- 0.8) revealed similar inhibition zones to the positive control amphotericin B (32 mu g/disc; 5.0 +/- 0.2; 5.0 +/- 0.6 and 8.8 +/- 1.2, respectively). The fungi Aspergillus sydowii Ce15, Aspergillus sydowii Ce19, Aspergillus sydowii Gc12, Bionectria sp. Ce5, Penicillium miczynskii Gc5, Penicillium raistrickii Ce16 and Trichoderma sp. Gc1, isolated from marine sponges Geodia corticostylifera and Chelonaplysylla erecta, were evaluated for their ability to grow in the presence of DDD pesticide. Increasing concentrations of DDD pesticide, i.e., 5.0 mg (1.56 x 10(-12) mmol), 10.0 mg (3.12 x 10(-2) mmol) and 15.0 mg (4.68 x 10(-2) mmol) in solid and liquid culture media were tested. The fungi Trichoderma sp. Gc1 and Penicillium miczynskii Gc5 were able to grow in the presence of up to 15.0 mg of DDD, suggesting their potential for biodegradation. A 100% degradation of DDD was attained in liquid culture medium when Trichoderma sp. Gc1 was previously cultivated for 5 days and supplemented with 5.0 mg of DDD in the presence of hydrogen peroxide. However, the quantitative analysis showed that DDD was accumulated on mycelium and biodegradation level reached a maximum value of 58% after 14 days.

The asymmetric reduction of 2-chloro-1-phenylethanone (1) by seven strains of marine fungi was evaluated and afforded (S)-(-)-2-chloro-1-phenylethanol with, in the best case, an enantiomeric excess of 50% and an isolated yield of 60%. The ability of marine fungi to catalyse the reduction was directly dependent on growth in artificial sea water-based medium containing a high concentration of Cl(-) (1.2 M). When fungi were grown in the absence of artificial sea water, no reduction of 1 by whole cells was observed. The biocatalytic reduction of 1 was more efficient at neutral rather than acidic pH values and in the absence of glucose as co-substrate.

This project was performed at Rochester Institute of Technology to get more understanding and knowledge about AM and FM screenings similarities and differences with considerations of the mottle. By designing a test form conformed to the specific measurements and printing it on Heidelberg's Sunday 2000 press, the project group has evaluated the questions that already existed and the ones that occurred during the project. Hence the first press run left some unexpected phenomenon therefore another press run was performed. Measurements were performed and graphs produced in Excel. The project group evaluated the results and from that able to establish facts and draw conclusions. It has been a great experience for the project group and they have learnt a lot.

Um posicionamento de sucesso permite afiliar uma marca especificamente a alguma categoria que os consumidores possam claramente compreender e reconhecer de imediato. Para isso deve possuir imagem positiva, apresentar benefícios e possuir legitimidade. No mercado de bens simbólicos a imagem positiva de uma determinada organização pode ser utilizada para fornecer a lógica que proporciona aos receptores motivos para acreditar que essa marca possui o benefício esperado e portanto, torna-se uma entidade legítima, uma vez que comercializa bens intangíveis. Essas estratégias são acompanhadas de procedimentos retóricos (figuras, gestos, palavras,posturas) que concretizam o alcance dos objetivos à serem alcançados possuem.

Os Fungos Micorrízicos Arbusculares (FMAs) desempenham um papel importante na sustentabilidade dos ecossistemas, devido a importância da simbiose que formam com a maioria das plantas. A associação destes fungos com as raízes possibilita uma melhor nutrição das plantas, além de promover o crescimento e a resistência a fatores causadores de estresse. Desta forma, auxiliam no aumento da produtividade das mesmas, e na redução do uso de insumos químicos, como fertilizantes e agrotóxicos, na agricultura. Em função ao caráter de simbiontes obrigatórios, é de grande interesse o desenvolvimento de processos que permitam o isolamento, caracterização, manutenção de isolados e produção de inóculo desses fungos. Assim, com o objetivo de produzir um método de cultivo axênico, e que também possa auxiliar o estudo destes em áreas como a biologia molecular, testou-se protocolos de micropropagação para a produção in vitro de Glomus etunicatum W. N. Becker & Gerd., utilizando-se como hospedeiro raízes de tomateiro, videira e menta. Além destas culturas, que foram propagadas vegetativamente para a manutenção das culturas estoques e enraizadas in vitro com a aplicação exógena do regulador de crescimento ácido indolbutírico (AIB), trabalhou-se também com a cultura da menta, mas devido a problemas que ocorreram durante a etapa de enraizamento, não foi possível efetivar a associação do inóculo Na inoculação de esporos em raízes da videira cultivadas in vitro, não se obteve êxito. Com o cultivo in vitro de raízes de tomateiro foi possível conduzir os trabalhos até a etapa de colonização do FMA. No enraizamento in vitro das três culturas trabalhadas foi avaliado o número e o comprimento médio das raízes cultivadas nos meios de enraizamento nas doses de 0, 1,0 e 2,0 mg.L-1 de AIB. Para o tomateiro e a menta, o número e o comprimento médio de raízes por explante foram significativamente maiores com a dose de 1,0 mg.L-1 de AIB. Entretanto, para a cultura da videira, embora o uso de AIB, em ambas as doses de 1,0 e 2,0 mg.L-1 induziram um maior número de raízes, o comprimento médio de raízes foi maior quando AIB não foi adicionado no meio de enraizamento.
