Fasciolosis is an important foodborne, zoonotic disease of livestock and humans, with global annual health and economic losses estimated at several billion US$. Fasciola hepatica is the major species in temperate regions, while F. gigantica dominates in the tropics. In the absence of commercially available vaccines to control fasciolosis, increasing reports of resistance to current chemotherapeutic strategies and the spread of fasciolosis into new areas, new functional genomics approaches are being used to identify potential new drug targets and vaccine candidates. The glutathione transferase (GST) superfamily is both a candidate drug and vaccine target. This study reports the identification of a putatively novel Sigma class GST, present in a water-soluble cytosol extract from the tropical liver fluke F. gigantica. The GST was cloned and expressed as an enzymically active recombinant protein. This GST shares a greater identity with the human schistosomiasis GST vaccine currently at Phase II clinical trials than previously discovered F. gigantica GSTs, stimulating interest in its immuno-protective properties. In addition, in silico analysis of the GST superfamily of both F. gigantica and F. hepatica has revealed an additional Mu class GST, Omega class GSTs, and for the first time, a Zeta class member.

A new approach to determine the local boundary of voltage stability region in a cut-set power space (CVSR) is presented. Power flow tracing is first used to determine the generator-load pair most sensitive to each branch in the interface. The generator-load pairs are then used to realize accurate small disturbances by controlling the branch power flow in increasing and decreasing directions to obtain new equilibrium points around the initial equilibrium point. And, continuous power flow is used starting from such new points to get the corresponding critical points around the initial critical point on the CVSR boundary. Then a hyperplane cross the initial critical point can be calculated by solving a set of linear algebraic equations. Finally, the presented method is validated by some systems, including New England 39-bus system, IEEE 118-bus system, and EPRI-1000 bus system. It can be revealed that the method is computationally more efficient and has less approximation error. It provides a useful approach for power system online voltage stability monitoring and assessment. Kumphawapi, which is Thailand’s largest natural freshwater lake, contains a >10,000-year-long climatic and environmental archive. New data sets (stratigraphy, chronology, hydrogen isotopes, plant macrofossil and charcoal records) for two sedimentary sequences are here combined with earlier multi-proxy studies to provide a comprehensive reconstruction of past climatic and environmental changes for Northeast Thailand. Gradually higher moisture availability due to a strengthening of the summer monsoon led to the formation of a large shallow lake in the Kumphawapi basin between >10,700 and c. 7000 cal. BP. The marked increase in moisture availability and lower evaporation between c. 7000 and 6400 cal. BP favoured the growth and expansion of vegetation in and around the shallow lake. The increase in biomass led to gradual overgrowing and infilling, to an apparent lake level lowering and to the development of a wetland. Multiple hiatuses are apparent in all investigated sequences between c. 6500 and 1400 cal. BP and are explained by periodic desiccation events of the wetland and erosion due to the subsequent lake level rise. The rise in lake level, which started c. 2000 cal. BP and reached shallower parts c. 1400 cal. BP, is attributed to an increase in effective moisture availability. The timing of hydroclimatic conditions during the past 2000 years cannot be resolved because of chronological limitations.

Indústrias metalúrgicas de produção de ligas metálicas podem por em risco toda a área envolvente, nomeadamente a qualidade das águas (superficiais e subterrâneas), dos solos e do ar, sempre que não existam infraestruturas e planos adequados de gestão destes resíduos. No caso em estudo, a unidade industrial situa-se na bacia do rio São Francisco, no estado brasileiro de Minas Gerais, e nos mais de quarenta anos de funcionamento tem produzido inúmeros problemas ambientais. Este estudo baseia-se nos primeiros resultados referentes aos solos da envolvente, tendo como objetivos a identificação dos principais contaminantes e a definição da área contaminada, de forma a avaliar a eficácia de projetos futuros de recuperação.

We studied a group of 174 Portuguese children (aged 2 mo-16 y) who mostly traveled to tropical Portuguese-speaking countries and found an attack rate of 21.8% for travelers' diarrhea, much lower than previously described. We also showed that African rate analysis by region may hide significant differences between countries.

A utilização crescente das tecnologias de informação provocou uma revolução do conhecimento. O resultado deste novo paradigma trouxe mudanças significativas no modo de gerir e difundir a informação arquivística. A digitalização de parte ou da totalidade de acervos tem sido uma prática constante nos arquivos, bibliotecas e centros de documentação como forma de permitir ao utilizador o acesso remoto e alargado à informação. Neste sentido, o presente estudo, o qual enquadra-se na Sociedade da Informação, incide sobre preservação a longo prazo de documentos digitalizados e tem como objeto de estudo os representantes digitais inseridos no Arquivo Científico Tropical Digital (ACTD), pertencente ao Instituto de Investigação Científica Tropical (IICT). Propõe-se aqui entender o processo de digitalização dos documentos inseridos no ACTD, analisar se é realizada alguma preservação digital a longo prazo, bem como analisar a perceção dos profissionais da instituição em estudo relativa à esta temática e a partir deste conhecimento fazer recomendações de preservação digital a longo prazo ao Arquivo digital. O método de recolha e análise de dados utilizado diz respeito à análise de documentos institucionais do IICT, das páginas web do IICT e do ACTD, a fim de compreender a sua génese e estrutura e, a análise qualitativa de entrevistas semiestruturadas aplicada a cinco colaboradores do IICT que trabalham direta ou indiretamente com o ACTD. Para ajudar na análise dos dados recolhidos em entrevista construiu-se uma grelha de análise qualitativa por tema e categoria. A partir do tratamento e análise dos dados identificou-se algumas dificuldades que o ACTD apresenta relativamente á preservação digital a longo prazo, e recomendações foram sugeridas. O IICT não possui plano de preservação digital, nem ações de preservação definidas para serem aplicadas aos objetos digitais do ACTD. A perceção dos profissionais entrevistados relativa à temática é superficial, por isso no que diz respeito à preservação dos objetos digitais recomenda-se ao ACTD e aos profissionais que estão a trabalhar diretamente com as coleções documentais digitalizadas a criação de um grupo de trabalho, de um projeto de digitalização formalizado e de um plano de preservação digital, além da utilização de metadados de preservação, assim como a definição de normas e estratégias de preservação com base em políticas nacionais e internacionais.

OBJECTIVE: Skeletal Muscle Biopsy is a minor surgical procedure for the diagnosis of different neuromuscular pathological conditions and has recently gained popularity also in the research field of age-related muscular modifications and sarcopenia. Few studies focused on the application of mini-invasive muscular biopsy in both normal and pathological conditions. The aim of our study was to describe a mini invasive ultrasound-guided skeletal muscular biopsy technique in complete spinal cord injured (SCI) patients and healthy controls with a tri-axial end-cut needle. PATIENTS AND METHODS: Skeletal muscle biopsies were collected from 6 chronic SCI patients and 3 healthy controls vastus lateralis muscle with a tri-axial end cut needle (Biopince© - Angiotech). Muscle samples were stained for ATPase to determine fibers composition, moreover, gene expression of cyclooxygenase-1 (COX-1) and prostaglandin E2 receptor has been analyzed by Real Time RT-PCR. RESULTS: All the procedures were perfomed easily without failures and complications. Control tissue was macroscopically thicker than SCI one. Control specimen displayed an equal distribution of type I and type II fibers, while SCI sample displayed a prevalence of type II fibers SCI specimen displayed a significant reduction in COX-1 gene expression. This mini-invasive approach was easy, accurate and with low complication rate in performing skeletal muscle biopsy in both SCI patients and controls. CONCLUSIONS: This technique could be useful in conditions in which the overall quantity of specimen required is small like for molecular biology analysis. For histological diagnostic purposes and/or conditions in which the original tissue is already pathologically modified, this technique should be integrated with more invasive techniques.
