Os compostos orgânicos de estanho (OTs), de entre os quais se destaca o tributilestanho (TBT), encontram-se amplamente dispersos no meio aquático devido à sua intensa utilização como agente biocida em tintas antivegetativas. Estudos anteriores sobre a poluição por organoestanhos em Portugal demonstraram que estes compostos se encontram presentes não só na linha de costa mas também em zonas da plataforma, sendo as zonas portuárias (onde se incluem portos comerciais, portos de pesca, marinas e estaleiros navais) os principais focos de poluição. A presente tese tem como objectivo investigar o estado actual da poluição por organoestanhos na costa Portuguesa confirmando se os padrões espaciais acima descritos se mantêm, por meio da quantificação de diversos OTs, nomeadamente, butilestanhos, fenilestanhos e octilestanhos. Assim, os níveis destes compostos foram avaliados em populações de Mytilus galloprovincialis e Nassarius reticulatus ao longo da costa continental Portuguesa, com particular incidência na Ria de Aveiro onde se quantificaram os níveis de OTs em mexilhões, gastrópodes e sedimentos, recolhidos numa malha de amostragem mais densa. A distribuição espacial dos organoestanhos foi determinada utilizando o bivalve M. galloprovincialis como espécie bioindicadora. Os níveis totais de estanho (SnT) foram quantificados nos tecidos do mexilhão e relacionados com os níveis totais de OTs nos mesmos tecidos, incluindo monobutilestanho (MBT), dibutilestanho (DBT), tributilestanho (TBT), difenilestanho (DPhT), trifenilestanho (TPhT), monoctilestanho (MOcT) e dioctilestanho (DOct). A contribuição dos OTs para os valores de estanho total (SnT) foi superior nas estações de amostragem localizadas no interior de portos onde atingiram proporções próximas dos 50%. De entre estes, os butilestanhos (BuTs=MBT+DBT+TBT) contribuíram em média com 98.6% para o valor total de OTs, tendo sido detectados em todas as amostras analisadas. Os valores mais elevados foram registados no interior ou na proximidade de portos, corroborando a ideia anterior de que constituem importantes focos de poluição. A variação das concentrações de TBT no mexilhão situou-se entre os 0,9 e 720 ng Sn.g-1 de peso seco (ps). Estes valores são, em 69% das estações amostradas, superiores ao valor do proposto pela OSPAR (4,9 ng TBT-Sn.g-1 ps) para tecidos de mexilhão o que sugere a forte probabilidade de ocorrência de efeitos adversos sobre os ecossistemas. Os níveis de OTs foram também quantificados em tecidos de N. reticulatus recolhidos ao longo da costa em 2008 Os butilestanhos representaram a maioria dos compostos organoestânicos quantificados e os níveis mais elevados foram novamente detectados no interior ou nas imediações de portos. Os valores de TBT nos tecidos deste gastrópode variaram entre 3,5 e 380 ng Sn.g-1 ps, representando uma percentagem média de 50,4% do total de butilestanhos. Simultaneamente, os níveis de imposex foram também avaliados e relacionados com os valores deste composto nos tecidos. As distribuições espaciais de imposex e de TBT seguiram a mesma tendência, sendo que em todos os locais amostrados foram encontradas fêmeas afectadas. Os valores de VDSI (índice da sequência do vaso deferente) variaram entre 0,2 e 4,4. Em 91% dos locais os valores de VDSI foram superiores a 0,3 (definido pela OSPAR como o valor de VDSI em N. reticulatus acima do qual o objectivo de qualidade ecológica não é atingido), confirmando a suspeição da existência de efeitos adversos nos ecossistemas. Em todos os compartimentos analisados na Ria de Aveiro, os butilestanhos foram os principais contribuintes para estanho orgânico total. A utilização do imposex em N. reticulatus como biomarcador da poluição por TBT permitiu determinar um gradiente decrescente desde o interior da Ria (onde se situa a zona portuária) até zonas costeiras adjacentes. O mesmo gradiente foi observado relativamente às concentrações de TBT em tecidos de mexilhão. Para os sedimentos, as concentrações de TBT são bastante variáveis com valores entre 2,7 e 1780 ng Sn.g-1 ps encontrando-se significativamente correlacionadas com o conteúdo em matéria orgânica da amostra. Em todas as amostras analisadas os níveis de TBT são elevados e superiores ao valor inferior (provisório) de EAC (critério de avaliação ambiental) proposto pela OSPAR (0,004 ng TBT-Sn.g-1 ps). A análise da evolução temporal da poluição por TBT ao longo da costa foi concretizada por meio da comparação entre níveis de organoestanhos em N. reticulatus em amostras de 2008 e 2003 e também através da comparação dos níveis de imposex registados em campanhas realizadas naqueles dois anos. Os resultados obtidos indicam a ocorrência de reduções significativas nas concentrações de TBT, DBT e MBT, assim como uma diminuição significativa nos valores de VDSI entre 2003 e 2008. Os resultados obtidos sugerem que a redução verificada se deve à implementação do Regulamento 782/2003 da Comunidade Europeia, que tem por objectivo a erradicação das descargas e emissões de TBT para o ambiente a partir dos sistemas antivegetativos A diminuição da poluição por TBT ao longo dos últimos anos foi acompanhada por um aumento no número de fêmeas com um vaso deferente, mas sem pénis (imposex do tipo b): 3,5% em 2000, 11% em 2003 e 24% em 2008. Um aumento no número de locais onde se registou o fenómeno também é evidente: dois em 2000, sete em 2003 e treze em 2008. A proporção de fêmeas b no estádio 1 de VDS apresentou igual tendência com aumento de 38% em 2000 para 65% em 2008. O aumento no número de fêmeas com esta via parece estar associado à diminuição da poluição por TBT. Face à esperada diminuição da presença do composto no meio ambiente, devido à sua proibição, o aumento de fêmeas com imposex do tipo b é previsível. A ocorrência de compostos xenoestrogénicos no ambiente aquático foi também estudada e os níveis de estrona (E1), 17α-e 17β-estradiol (E2), 17α- etinilestradiol (EE2), bisfenol-A (BPA) e nonilfenol (NP) foram quantificados em efluentes de estações de tratamento de águas residuais (ETARs) localizadas na região de Aveiro, bem como no efluente final descarregado no Oceano Atlântico, através de um emissário submarino (S. Jacinto), sendo amostras recolhidas na entrada da Ria e ao largo usadas como referência. Os níveis de hormonas esteróides e compostos fenólicos registados nos locais de referência são baixos. De entre as hormonas esteróides os níveis mais elevados foram registados para a estrona, com valores máximos de 85.3 ng.L- 1 . Os níveis mais elevados de compostos fenólicos foram detectados em efluentes industriais (máximos de NP e BPA de 2410 ng.L-1 e 897 ng.L-1 , respectivamente). Os resultados obtidos sugerem que os níveis de compostos xenoestrogénicos em locais de referência são baixos e não parecerem acarretar risco ecológico, no entanto o mesmo não será verdadeiro para as imediações do emissário de S. Jacinto que liberta efluentes com concentrações muito elevadas de E1, NP e BPA. Foram realizadas experiências laboratoriais de forma a elucidar o papel do receptor retinóico X (RXR) no mecanismo de indução de imposex (presentemente o mecanismo que demonstra maior promessa na explicação do desencadear deste fenómeno). Fêmeas de Nucella lapillus e N. reticulatus foram injectadas com TBT em etanol ou com ácido 9-cis- retinóico em FBS (soro fetal bovino) tendo-se procedido à sua observação nos 30 dias subsequentes. Tanto o TBT como o 9CRA induziram o desenvolvimento de imposex em N. lapillus e N. reticulatus. Aumentos significativos nos valores de VDSI e FPL entre o controlo de etanol e o tratamento de TBT e o controlo de FBS e o tratamento de 9CRA foram registados. Os resultados obtidos fornecem novas provas do envolvimento da via de sinalização associada ao RXR no desenvolvimento de imposex em ambas as espécies.

A new electrochemical methodology to study labile trace metal/natural organic matter complexation at low concentration levels in natural waters is presented. This methodology consists of three steps: (i) an estimation of the complex diffusion coefficient (DML), (ii) determination at low pH of the total metal concentration initially present in the sample, (iii) a metal titration at the desired pH. The free and bound metal concentrations are determined for each point of the titration and modeled with the non-ideal competitive adsorption (NICA-Donnan) model in order to obtain the binding parameters. In this methodology, it is recommended to determine the hydrodynamic transport parameter, α, for each set of hydrodynamic conditions used in the voltammetric measurements. The methodology was tested using two fractions of natural organic matter (NOM) isolated from the Loire river, namely the hydrophobic organic matter (HPO) and the transphilic organic matter (TPI), and a well characterized fulvic acid (Laurentian fulvic acid, LFA). The complex diffusion coefficients obtained at pH 5 were 0.4 ± 0.2 for Pb and Cu/HPO, 1.8 ± 0.2 for Pb/TPI and (0.612 ± 0.009) × 10−10 m2 s−1 for Pb/LFA. NICA-Donnan parameters for lead binding were obtained for the HPO and TPI fractions. The new lead/LFA results were successfully predicted using parameters derived in our previous work.

Purpose To analyse pacing strategies displayed by athletes achieving differing levels of performance during an elite level marathon race. Methods Competitors in the 2009 IAAF Women’s Marathon Championship were split into Groups 1, 2, 3, and 4 comprising the first, second, third, and fourth 25% of finishers respectively. Final, intermediate, and personal best (PB) times of finishers were converted to mean speeds, and relative speed (% of PB speed) was calculated for intermediate segments. Results Mean PB speed decreased from Group 1 to 4 and speed maintained in the race was 98.5 + 1.8%, 97.4 + 3.2%, 95.0 + 3.1% and 92.4 + 4.4% of PB speed for Groups 1-4 respectively. Group 1 was fastest in all segments and differences in speed between groups increased throughout the race. Group 1 ran at lower relative speeds than other groups for the first two 5 km segments, but higher relative speeds after 35km. Significant differences (P<0.01) in the percentage of PB speed maintained were observed between Groups 1 and 4, and 2 and 4 in all segments after 20 km, and Groups 3 and 4 from 20-25 km and 30-35 km. Conclusions Group 1 athletes achieved superior finishing times relative to their PB than athletes in other Groups who selected unsustainable initial speeds resulting in subsequent significant losses of speed. It is suggested that psychological factors specific to a major competitive event influenced decision making by athletes and poor decisions resulted in final performances inferior to those expected based on PB times.

We used1H-magnetic resonance spectroscopy to noninvasively determine total creatine (TCr), choline-containing compounds (Cho), and intracellular (IT) and extracellular (between-muscle fibers) triglycerides (ET) in three human skeletal muscles. Subjects' (n = 15 men) TCr concentrations in soleus [Sol; 100.2 ± 8.3 (SE) mmol/kg dry wt] were lower (P < 0.05) than those in gastrocnemius (Gast; 125.3 ± 9.2 mmol/kg dry wt) and tibialis anterior (TA; 123.7 ± 8.8 mmol/kg dry wt). The Cho levels in Sol (35.8 ± 3.6 mmol/kg dry wt) and Gast (28.5 ± 3.5 mmol/kg dry wt) were higher (P < 0.001 andP < 0.01, respectively) compared with TA (13.6 ± 2.4 mmol/kg dry wt). The IT values were found to be 44.8 ± 4.6 and 36.5 ± 4.2 mmol/kg dry wt in Sol and Gast, respectively. The IT values of TA (24.5 ± 4.5 mmol/kg dry wt) were lower than those of Sol (P < 0.01) and Gast (P < 0.05). There were no differences in ET [116.0 ± 11.2 (Sol), 119.1 ± 18.5 (Gast), and 91.4 ± 19.2 mmol/kg dry wt (TA)]. It is proposed that the differences in metabolite levels may be due to the differences in fiber-type composition and deposition of metabolites due to the adaptation of different muscles during locomotion.

Science gateways can provide access to distributed computing resources and applications at very different levels of granularity. Some gateways do not even hide the details of the underlying infrastructure, while on the other end some provide completely customized high-level interfaces to end-users. In this chapter the different granularity levels at which science gateways can be developed with WS-PGRADE/gUSE are analysed. The differences between these various granu-larity levels are also illustrated via the example of a molecular docking gateway and its four different implementations.

Pesticide exposure during brain development could represent an important risk factor for the onset of neurodegenerative diseases. Previous studies investigated the effect of permethrin (PERM) administered at 34 mg/kg, a dose close to the no observable adverse effect level (NOAEL) from post natal day (PND) 6 to PND 21 in rats. Despite the PERM dose did not elicited overt signs of toxicity (i.e. normal body weight gain curve), it was able to induce striatal neurodegeneration (dopamine and Nurr1 reduction, and lipid peroxidation increase). The present study was designed to characterize the cognitive deficits in the current animal model. When during late adulthood PERM treated rats were tested for spatial working memory performances in a T-maze-rewarded alternation task they took longer to choose for the correct arm in comparison to age matched controls. No differences between groups were found in anxiety-like state, locomotor activity, feeding behavior and spatial orientation task. Our findings showing a selective effect of PERM treatment on the T-maze task point to an involvement of frontal cortico-striatal circuitry rather than to a role for the hippocampus. The predominant disturbances concern the dopamine (DA) depletion in the striatum and, the serotonin (5-HT) and noradrenaline (NE) unbalance together with a hypometabolic state in the medial prefrontal cortex area. In the hippocampus, an increase of NE and a decrease of DA were observed in PERM treated rats as compared to controls. The concentration of the most representative marker for pyrethroid exposure (3-phenoxybenzoic acid) measured in the urine of rodents 12 h after the last treatment was 41.50 µ/L and it was completely eliminated after 96 h.

Considering tobacco smoke as one of the most health-relevant indoor sources, the aim of this work was to further understand its negative impacts on human health. The specific objectives of this work were to evaluate the levels of particulate-bound PAHs in smoking and non-smoking homes and to assess the risks associated with inhalation exposure to these compounds. The developed work concerned the application of the toxicity equivalency factors approach (including the estimation of the lifetime lung cancer risks, WHO) and the methodology established by USEPA (considering three different age categories) to 18 PAHs detected in inhalable (PM10) and fine (PM2.5) particles at two homes. The total concentrations of 18 PAHs (ΣPAHs) was 17.1 and 16.6 ng m−3 in PM10 and PM2.5 at smoking home and 7.60 and 7.16 ng m−3 in PM10 and PM2.5 at non-smoking one. Compounds with five and six rings composed the majority of the particulate PAHs content (i.e., 73 and 78 % of ΣPAHs at the smoking and non-smoking home, respectively). Target carcinogenic risks exceeded USEPA health-based guideline at smoking home for 2 different age categories. Estimated values of lifetime lung cancer risks largely exceeded (68–200 times) the health-based guideline levels at both homes thus demonstrating that long-term exposure to PAHs at the respective levels would eventually cause risk of developing cancer. The high determined values of cancer risks in the absence of smoking were probably caused by contribution of PAHs from outdoor sources.

Background: The role of persistent organic pollutants (POPs) with endocrine disrupting activity in the aetiology of obesity and other metabolic dysfunctions has been recently highlighted. Adipose tissue (AT) is a common site of POPs accumulation where they can induce adverse effects on human health. Objectives: To evaluate the presence of POPs in human visceral (vAT) and subcutaneous (scAT) adipose tissue in a sample of Portuguese obese patients that underwent bariatric surgery, and assess their putative association with metabolic disruption preoperatively, as well as with subsequent body mass index (BMI) reduction. Methods: AT samples (n=189) from obese patients (BMI ≥35) were collected and the levels of 13 POPs were determined by gas chromatography with electron-capture detection (GC-ECD). Anthropometric and biochemical data were collected at the time of surgery. BMI variation was evaluated after 12 months and adipocyte size was measured in AT samples. Results: Our data confirm that POPs are pervasive in this obese population (96.3% of detection on both tissues), their abundance increasing with age (RS=0.310, p<0.01) and duration of obesity (RS=0.170, p<0.05). We observed a difference in AT depot POPs storage capability, with higher levels of ΣPOPs in vAT (213.9±204.2 compared to 155.1±147.4 ng/g of fat, p<0.001), extremely relevant when evaluating their metabolic impact. Furthermore, there was a positive correlation between POP levels and the presence of metabolic syndrome components, namely dysglycaemia and hypertension, and more importantly with cardiovascular risk (RS=0.277, p<0.01), with relevance for vAT (RS=0.315, p<0.01). Finally, we observed an interesting relation of higher POP levels with lower weight loss in older patients. Conclusion: Our sample of obese subjects allowed us to highlight the importance of POPs stored in AT on the development of metabolic dysfunction in a context of obesity, shifting the focus to their metabolic effects and not only for their recognition as environmental obesogens.

The aim of this work was to assess ultrafine particles (UFP) number concentrations in different microenvironments of Portuguese preschools and to estimate the respective exposure doses of UFP for 3–5-year-old children (in comparison with adults). UFP were sampled both indoors and outdoors in two urban (US1, US2) and one rural (RS1) preschool located in north of Portugal for 31 days. Total levels of indoor UFP were significantly higher at the urban preschools (mean of 1.82x104 and 1.32x104 particles/cm3 at US1 an US2, respectively) than at the rural one (1.15x104 particles/cm3). Canteens were the indoor microenvironment with the highest UFP (mean of 5.17x104, 3.28x104, and 4.09x104 particles/cm3 at US1, US2, and RS1), whereas the lowest concentrations were observed in classrooms (9.31x103, 11.3x103, and 7.14x103 particles/cm3 at US1, US2, and RS1). Mean indoor/outdoor ratios (I/O) of UFP at three preschools were lower than 1 (0.54–0.93), indicating that outdoor emissions significantly contributed to UFP indoors. Significant correlations were obtained between temperature, wind speed, relative humidity, solar radiation, and ambient UFP number concentrations. The estimated exposure doses were higher in children attending urban preschools; 3–5-year-old children were exposed to 4–6 times higher UFP doses than adults with similar daily schedules.

Gamma radiations measurements were carried out in the vicinity of a coal-fired power plant located in the southwest coastline of Portugal. Two different gamma detectors were used to assess the environmental radiation within a circular area of 20 km centred in the coal plant: a scintillometer (SPP2 NF, Saphymo) and a high purity germanium detector (HPGe, Canberra). Fifty urban and suburban measurements locations were established within the defined area and two measurements campaigns were carried out. The results of the total gamma radiation ranged from 20.83 to 98.33 counts per second (c.p.s.) for both measurement campaigns and outdoor doses rates ranged from 77.65 to 366.51 Gy/h. Natural emitting nuclides from the U-238 and Th-232 decay series were identified as well as the natural emitting nuclide K-40. The radionuclide concentration from the uranium and thorium series determined by gamma spectrometry ranged from 0.93 to 73.68 Bq/kg, while for K-40 the concentration ranged from 84.14 to 904.38 Bq/kg. The obtained results were used primarily to define the variability in measured environmental radiation and to determine the coal plant’s influence in the measured radiation levels. The highest values were measured at two locations near the power plant and at locations between the distance of 6 and 20 km away from the stacks, mainly in the prevailing wind direction. The results showed an increase or at least an influence from the coal-fired plant operations, both qualitatively and quantitatively.

It is well recognized that professional musicians are at risk of hearing damage due to the exposure to high sound pressure levels during music playing. However, it is important to recognize that the musicians’ exposure may start early in the course of their training as students in the classroom and at home. Studies regarding sound exposure of music students and their hearing disorders are scarce and do not take into account important influencing variables. Therefore, this study aimed to describe sound level exposures of music students at different music styles, classes, and according to the instrument played. Further, this investigation attempted to analyze the perceptions of students in relation to exposure to loud music and consequent health risks, as well as to characterize preventive behaviors. The results showed that music students are exposed to high sound levels in the course of their academic activity. This exposure is potentiated by practice outside the school and other external activities. Differences were found between music style, instruments, and classes. Tinnitus, hyperacusis, diplacusis, and sound distortion were reported by the students. However, students were not entirely aware of the health risks related to exposure to high sound pressure levels. These findings reflect the importance of starting intervention in relation to noise risk reduction at an early stage, when musicians are commencing their activity as students.
