Um equipamento triaxial convencional originalmente utilizado para ensaios em amostras de solos saturados saturada foi modificado para que fosse possível a realização de ensaios triaxiais em solos não saturados. As principais modificações foram a instalação de uma pedra cerâmica de alto valor de entrada de ar (300 kPa) na nova base da câmara triaxial e uma nova linha de aplicação de pressão para aplicação de pressão de ar na amostra. A técnica de translação de eixos é utilizada para evitar cavitação no sistema de aplicação de pressão. O solo estudado nesta pesquisa consiste de um colúvio de arenito localizado nas encostas da Formação Serra Geral, entre os municípios de Timbé do Sul (SC) e São José dos Ausentes (RS). Ensaios triaxiais convencionais foram realizados nas condições drenado e não drenado em amostras indeformadas para tensões de confinamento iniciais variando de 50 até 500 kPa; ensaios triaxiais convencionais foram realizados na condição drenada em amostras remoldadas para as tensões de confinamento iniciais de 50, 100 e 200kPa. Ensaios triaxiais com sucção controlada foram realizados na condição drenado em amostras indeformadas para as tensões normais líquidas de 50, 100, 150 e 200 kPa em níveis de sucção variando de 25 até 150 kPa. A partir dos resultados dos ensaios triaxiais determinou-se os parâmetros de resistência ao cisalhamento: c’ – intercepto coesivo efetivo; f´ - ângulo de atrito interno efetivo; fb – ângulo de atrito interno que quantifica a contribuição da sucção na resistência ao cisalhamento. O comportamento encontrado foi de uma envoltória de resistência ao cisalhamento não saturada bilinear com acréscimo na resistência ao cisalhamento para baixos níveis de sucção até aproximadamente 75 kPa, e após um decréscimo na resistência ao cisalhamento. A tensão normal líquida não influencia no valor de fb e a sucção não provoca alteração no valor de f´. O valor de fb apresenta valores superiores ao valor de f´, quando se utiliza o critério de ruptura clássico de resistência ao cisalhamento. Alguns critérios de determinação dos valores máximos de ruptura foram aplicados para um melhor entendimento do comportamento resistente deste material.

Experiente na missão de fazer da Linguística um objeto de estudo atraente e acessível, José Luiz Fiorin põe à disposição dos interessados – sejam alunos de Letras, estudantes de áreas afins e curiosos em geral – um novo e atualizado manual de introdução aos estudos de linguagem. E, como sempre, com a preocupação em suscitar prazer na imersão nos conceitos fundamentais de uma ciência que se desenvolveu e se ramificou, sobretudo, no século XX. Com Linguística? Que é isso?, lançado no país em maio passado pela editora Contexto, Fiorin – e quatro professores articulistas – recupera questões já abordadas em manuais anteriores, em especial nos dois volumes de Introdução à Linguística1 (organizados por Fiorin, da mesma editora), e propõe uma divisão precisa das principais noções e correntes linguísticas que ganharam luz e forma a partir das escolas histórico-comparativas, na segunda metade do século XIX. Tradição da qual parte para fazer a expansão da Linguística e apresentá-la por intermédio de cinco grandes objetos científicos: a língua, a competência, a variação, a mudança e o uso.

This work, developed within the Master's Degree Program in Public Administration and Government of Eaesp-FGV, consists of an exploratory study on performance measurement in the public sector. It is focused on the identification of complexity elements embedded in the implementation of public policies, to which the goals and targets of a target-oriented system relate, and especially on the implementation of this system as a management tool itself, from the case study of the then ongoing "Programa de Metas da Cidade de São Paulo 2013-2016". All information was obtained from municipal managers, in Government Departments responsible for strategic support activities to the implementation of the system, and sectoral Government Departments whose scope also included crosscutting actions, through interviews conducted from a starting question, and then followed by the free talk of the respondents. Through discourse analysis, an assortment of information was conducted, concerning to: 1) the impacts on management, articulation, and governance of the use of performance measurement and result-oriented management; 2) the flow and publicity of information on performance, produced within the monitoring process of this given target-oriented system, and also concerning the use given to this information; 3) the recording and register of experiences and learning related to the Programa de Metas, and its potential to result as a legacy for the public administrative structure of the City of São Paulo. Assuming that the stories managers tell, in this field of Applied Social Sciences, are as valid as Science or the theoretical constructs on the subject, all information gathered from the point of view of the interviewed public officials was segmented and problematized under the light of the literature on performance measurement. The main objective of this research is to contribute to the creation of parameters for qualitative analysis of the information generated in the implementation of a target-oriented system in Public Management at the local level. It is, from then on, expected that these parameters may help managers and researchers on the monitoring and evaluation of performance of a given government, by favoring the translation of merely arithmetic data on percentage of fulfillment of specific commitments, into eloquent elements more enabled to express the evolution of a mandate. A elevada competição e conturbação do Mundo Actual em relação a tudo (status social, nível intelectual, escolar, profissional, consumismo, fanatismo e corrupção política, desportiva, religiosa,tráfego de drogas, abuso de menores, maus tratos e mais... muito mais...), assume uma proporção tal que muitas vezes nem nos apercebemos da sua gravidade. Esta competição incide essencialmente nos grupos etários mais vulneráveis, particularmente os adolescentes. Vários problemas podem surgir nesta etapa da vida, tais como a marginalidade, a exclusão social, a toxicodepêndencia e outros, para os quais diversos autores consideram a ansiedade, quer como factor desencadeante, quer como consequência. A autora desenvolveu um estudo transversal, de tipo metodológico, correlacional e inferencial que pretende não só avaliar as propriedades psicométricas de uma escala de ansiedade, o Endler Multidimensional Anxiety Scale (EMAS), bem como avaliar o nível de ansiedade duma amostra representativa de jovens (n=2310), de ambos os sexos, com idades compreendidas entre os 12 e os 18 anos, que frequentam escolas básicas e secundárias (rurais, urbanas e suburbanas) da Região Autónoma da Madeira. Por conseguinte, procedeu à tradução, adaptação e validação do EMAS na versão portuguesa, visando o desenvolvimento de um instrumento de medida da ansiedade válido e fidedigno, que permita ser utilizado por profissionais de saúde e docentes por forma a contribuir, não só para a prevenção do fenómeno ansiedade, mas também que proporcione a difusão dos seus dados à comunidade, em especial a científica. Os resultados da avaliação das características psicométricas mostram valores elevados nos diferentes coeficientes e correlações encontrados, quer nos estudos de fidedignidade, quer nos estudos de validade.A versão portuguesa do EMAS é um teste de auto-avaliação promissor (válido e fidedigno) para avaliar a ansiedade em adolescentes, sendo um instrumento que poderá ser utilizado em diferentes contextos (clínicos e pedagógicos) por profissionais que se dediquem ao estudo ou à mensuração daquele conceito.No que concerne ao grupo etário existem diferenças significativas entre todas as sub-escalas da Ansiedade Estado sendo os valores médios de ansiedade mais elevados na etapa da adolescência propriamente dita (grupo etário dos 15 aos 18 anos de idade). Em contrapartida, na Ansiedade Traço os valores mais elevados de ansiedade, constatou-se no sexo masculino e apenas nas situações de avaliação social e de perigo físico e na ansiedade de percepção não se verificaram diferenças significativas. Quanto ao sexo existem diferenças significativas na ansiedade estado global e na sua componente cognitiva, encontrando-se os valores de ansiedade mais elevados no sexo feminino, o que vem confirmar diversas teorias que defendem que este género psicologicamente estrutura-se mais precocemente. Em contrapartida essas diferenças somente se verificaram nas situações de avaliação social e de perigo físico da Ansiedade Traço e a média de ansiedade mais elevada é no sexo masculino, o que vem reforçar o defendido por diversos autores de que os rapazes só mais tardiamente estruturam a sua personalidade . Na ansiedade de percepção, só existem diferenças significativas no item referente ao perigo físico, sendo a média de ansiedade mais elevada nos rapazes, o que revela, o já descrito, que estes adolescentes arriscam mais o perigo do que as raparigas. Mediante a utilização da Análise de Correspondências encontrou-se o paradigma da família “Tipo” Madeirense.

Este trabalho aborda a crescente necessidade de verificação e testes das redes antes da sua implementação no mundo real. Variáveis como o investimento, estudo qualidade-custo, questões de gestão, fiabilidade e tempo de vida útil são parte das principais preocupações associadas a existentes e futuras redes. É sobretudo neste âmbito que este projecto opera, fornecendo uma solução para uma optimização da autoria de cenários de redes visando a sua execução na ferramenta de simulação de redes NS-3. As tecnologias emergentes são os alvos mais comuns de ferramentas de simulação de redes e este facto é determinante ao decidir que simulador de redes usar para simular os seus cenários. Com este trabalho, o principal objectivo é fornecer aos utilizadores uma ferramenta colaborativa que permite descrever os seus próprios cenários de redes, baseando-se numa linguagem XML denominada por NSDL e traduzir esta estrutura XML para um script C++ que possa ser executado num ambiente evolucionário, como é o caso do NS-3. A solução proposta permite a rápida criação de cenários de redes através de estruturas de dados XML para posterior tradução para C++ e então, execução no NS-3. A maior problemática associada a esta solução é a necessidade de actualização constante em estruturas de validação e de tradução de documentos XML, aquando da actualização da ferramenta NS-3, o que tem vindo a acontecer muito frequentemente devido à sua própria evolução.

Las políticas de salud destinadas a las mulheres de la comunidad quilombola de Boa Vista son, de manera general, las mismas políticas destinadas al resto de las mujeres de la región rural del Seridó norterriograndense y también las que se corresponden con regiones marginales del Brasil entero. Aquí, el cuerpo femenino es concebido bajo parámetros universalizantes que lo toman como una entidad homogénea y comparable con otros cuerpos femeninos a partir de su traducción en índices, tasas y estadísticas. En este sentido, decimos que son cuerpos desnudos, cuya intervención no considera los rasgos exteriores, aquellos llamados de culturales, como marcadores de identidad. Por otro lado, la noción de Salud de la Mujer Negra propuesta por recientes políticas de Estado a nivel nacional, se muestra inexistente en la comunidad. El cuerpo que se se exalta hoy a partir de los parámetros de reivindicación étnica es un cuerpo negro, pero también bello, jovem e sobre todo, fuerte; donde la noción de salud no penetra. De esta forma, las dos políticas conciben sujetos sociales diferentes. Sin embargo, existe otro espacio, que es el espacio de las prácticas vernáculas, en el que las mujeres experimentan la articulación entre feminilidad y negritud, pero a partir de otros parámetros local e históricamente delineados. Aquí, tanto las trayectorias de las mujeres como las redes de parentesco y cuidado locales se muestran especialmente significativas, ayudando a comprender las concepciones particulares sobre el cuerpo que imaginan y practican las mujeres de esta comunidad, y revelando la importancia de la maternidad como principio ordenador de identidades sociales. Para eso, hemos realizado un trabajo de observación participante, una serie de 30 entrevista com mujeres de Boa Vista y un estudio de las redes de parentesco organizadas alrededor del término mãe. Con esto, demostramos que existe un espacio cargado de significados sobre el cuerpo femenino y la feminilidad que es construido a partir de una interpretación local de la triple condición de mulher, de mãe y de negra

The metropolitan regions of Northeast Brazil are being gradually included in a scenario of international investments, which are motivated by the restructuring of both touristic and real-estate sectors. The new capital, real-estate developers and space configurations that result from this process indicate the need for the creation and implementation of public tools which should, at least, allow the mitigation of the urban impacts and environmental losses resulting from this situation. The effects on landscape and on the socio-spatial configuration result from the intensification caused by the dynamism of the "real estate-tourism" sector. There is a regional integration as an expression of the urban expansion of the metropolitan area of Natal. This study investigates the uniqueness of the restructuring and territorial integration of coastal areas and the strategies of the circuit of capital accumulation formed by linking the real estate to tourism. It is intended to increase the understanding about the strategies of tourism, real estate and public policy agents involved in this territorial reconfiguration and in the fund-raising needed for the investments, to understand the existing social and environmental effects and their future trends and also to understand the forms of spatial production as results from the practices of approaching the land transformation and the tourism valorization of the landscape, in a synchronous manner, first in the Northeast region and, as a focal study, in the Metropolitan Area of Natal. Likewise, it is intended to apprehend the current processes of metropolization of the eastern coast of Rio Grande do Norte, in addition to indicate its physical-territorial transformation and the types of projects/developments promoted by the market in the recent period. Based upon analysis undertaken for the Metropolitan Region of Natal RN, this piece of work presents some considerations on possible legal instruments that can be adjusted to the municipalities which are experiencing the impact of this peculiar and recent phenomenon in the region, caused by the arrival of the real estate-touristic capital. Chromobacterium violaceum is a free-living bacillus, Gram-negative commonly found in water and sand of tropical and subtropical regions. One of its main characteristic it's the ability to produce the purple pigment named violacein, that shows countless biological activities. In 2003, the genome of this organism was totally sequenced and revealed important informations about the physiology of this bacteria. However, few post-genomics studies had been accomplished. This work evaluated the protein profile of C. violaceum cultivated in LB medium at 28ºC that allowed the identification and characterization of proteins related to a possible secretion system that wasn't identified and characterized yet in C. violaceum, to the quorum sensing system, to regulatory process of transcription and translation, stress adaptation and biotechnological potential. Moreover, the response of the bacteria to UVC radiation was evaluated. The comparison of the protein profile, analyzed through 2-D electrophoresis, of the control group versus the treatment group allowed the identification of 52 proteins that arose after stress induction. The obtained results enable the elaboration of a stress response pathway in C. violaceum generated by the UVC light. This pathway, that seems to be a general stress response, involves the expression of proteins related to cellular division, purine and pirimidine metabolism, heat chock or chaperones, energy supply, regulation of biofilm formation, transport, regulation of lytic cycle of bacteriophages, besides proteins that show undefined function. Despite the response present similarities with the classic SOS response of E. coli, we still cannot assert that C. violaceum shows a SOS-like response, mainly due to the absence of characterization of a LexA-like protein in this organism

The β-proteobacterium Chromobacterium violaceum is a Gram-negative, free-living, saprophytic and opportunistic pathogen that inhabits tropical and subtropical ecosystems among them, in soil and water of the Amazon. It has great biotechnological potential, and because of this potential, its genome was completely sequenced in 2003. Genome analysis showed that this bacterium has several genes with functions related to the ability to survive under different kinds of environmental stresses. In order to understand the physiological response of C. violaceum under oxidative stress, we applied the tool of shotgun proteomics. Thus, colonies of C. violaceum ATCC 12472 were grown in the presence and absence of 8 mM H2O2 for two hours, total proteins were extracted from bacteria, subjected to SDS-PAGE, stained and hydrolysed. The tryptic peptides generated were subjected to a linear-liquid chromatography (LC) followed by mass spectrometer (LTQ-XL-Orbitrap) to obtain quantitative and qualitative data. A shotgun proteomics allows to compare directly in complex samples, differential expression of proteins and found that in C. Violaceum, 131 proteins are expressed exclusively in the control condition, 177 proteins began to be expressed under oxidative stress and 1175 proteins have expression in both conditions. The results showed that, under the condition of oxidative stress, this bacterium changes its metabolism by increasing the expression of proteins capable of combating oxidative stress and decreasing the expression of proteins related processes bacterial growth and catabolism (transcription, translation, carbon metabolism and fatty acids). A tool with of proteomics as an approach of integrative biology provided an overview of the metabolic pathways involved in the response of C. violaceum to oxidative stress, as well as significantly amplified understanding physiological response to environmental stress. Biochemical and "in silico" assays with the hypothetical ORF CV_0868 found that this is part of an operon. Phylogenetic analysis of superoxide dismutase, protein belonging to the operon also showed that the gene is duplicated in genome of C. violaceum and the second copy was acquired through a horizontal transfer event. Possibly, not only the SOD gene but also all genes comprising this operon were obtained in the same manner. It was concluded that C. violaceum has complex, efficient and versatile mechanisms in oxidative stress response

Toxoplasmosis is a zoonosis of worldwide distribution caused by the protozoan Toxoplasma gondii, triggering dangerous complications in immunocompromised patients and pregnant women, as well as having great economic impact for the livestock. So far the control of toxoplasmosis is made primarily by chemotherapy. However, most drugs used routinely have some limitations. In order to control this disease, several research groups, including ours, has been working to develop a medical-veterinary vaccine based on parasite antigens, vectors and protocols of immunization. In this study were implemented and standardized methodologies for amplification and cloning of recombinant immunogens in the system for the development of a prototype vaccine, based on the surface antigens of T. gondii and recombinant adenovirus encoding these antigens. Genes encoding BAG1, GRA2 and SAG1 proteins were amplified. We established a strategy for cloning SAG1, SAG2, SAG3 and TgAMA1- genes in recombinant system. The genes encoding SAG1 and SAG2 were cloned and their sequences showed high similarity with sequences from GenBank. The virtual translation of these proteins showed polymorphisms in the amino acid sequence, which can be correlated with levels of antigenicity. Simultaneously, the adenovirus encoding the SAGs (HAdSAGs) were expanded, purificated and characterizated. Immunization of C57bl/6 mice, using viral supernatant was not enought to elicit immune responses at high levels, being required HAdSAGs titration for future immunizations. Therefore, this study allowed the cloning of the two genes important for the development of a prototype vaccine. This study approaches the topic of humanization in health that involves the set of policies implemented by the Ministry of Health in Brazil. Its aims are directed towards a reflection on the guiding theoretical and organizing axes of the National Humanization Policy (NHP) and their repercussions on municipal health policy of Natal, Brazil; an analysis of the results of the policy at the local level; knowledge of the views and experiences of the humanization agents in the daily work process and identification of the main challenges of the policy. The empirical field of investigation was the Family Health Strategy (FHS) of the city of Natal. The assumption of the study is that the FHS has produced local experiences with potentialities that must not be wasted, in which there are difficulties and discrepancies between the real and proposed model. The contradictions and challenges in the social and political context of Brazil in the early XXI century and their consequences in the field of health reflect anti-utilitarian aspects anchored strongly in the theoretical concepts of Boaventura de Sousa Santos about the sociology of privations and emergencies as well as of the work of translating. The predominantly qualitative approach collects some complementary quantitative data. The study procedures used were the following: bibliographic research; documental research; interviews; and direct observation. Interpretation of the information obtained was based on documental analysis and on the symbolic cartography of the social representations. Cartographic evidence suggests that practices still take place under dehumanizing conditions that compromise the quality of care given. However, there is a movement aimed at changing the work process that has been strengthening the link and widening the measures developed, incorporating new directions in diversity, integrality and solidarity. The map drawn shows a reality manifested by explicit intentions in a political agenda, by concrete solutions marked by an assortment of difficulties and expressed in the words of the agents and by latent clues identified in successful local experiences, posing many challenges for the consolidation of the proposed changes

This thesis is a translation of work of the Brazilian doctor, Pedro da Silva Nava (1903-1984), in particular, his memoirs and chronicles, articulated with the writings of medicine history, aiming to defend that the autobiographical narratives are sources of research capable of promoting discussions on the expansion of the present at the confluence of complex and unequal society in constant changing process as the Brazilian. The theoretical and methodological support circulates around studies, proposals and thesis by Boaventura Santos about empowering past, destabilizing subjectivity, sociology of absences, cosmopolitan reason and translation work. The empirical support drawn from the literature produced by Nava were analyzed with reference this reasoning and studies that have facilitated the flow of translation among others, the studies of Antonio Candido, Arrigucci Jr., Boris Cyrulnik, Beatriz Sarlo, Ecléa Bosi, Ítalo Calvino, José Willington Germano, José Maria Cançado, Lev Vygotsky, Marilena Chauí, Paul Ricöeur and Walter Benjamim, without neglecting what we consider indispensable to scientific research, the production of relevant knowledge and prudent, in view of a decent life. The initial inflections reflect the subject of the Memoirs and its education/training, to then place the Memoir subject in the literary context, scientific, historical and Brazilian poetic (1972-2010), bringing great interpreters and discussing the rationale used by the Narrator that we defend stand closer to the cosmopolitan, showing the formation of narratives whose presence insert itself beforehand to modernist verve, linked to the discursive array against the literature as domination space, disseminated in Brazil in the early twentieth century. So, it articulate with those in which the concerns adjust the construction of the social formation of Brazil as a national heritage through literary narrative that focuses on a historical principle that becomes the past empowering, allowing his rereading, whose converge to memory, the lifestyles, the plurality of language and Brazilian culture, formed by several people, converging into a design not of culture but multiculturalism in Brazil. The memory issue was addressed in the space-time of experiences of being that narrates, shaped by a destabilizing subjectivity that sought to order the testimony of a time, a history and society, retelling them by creative imagination, almost fictional, to make circulate his knowledge about Brazil attached to his medical knowledge, as well as other subjects in his living group and other groups with whom they maintained contact. Thus, he portrayed both tangible and intangible cultural assets of the country as a form of preservation, giving them meanings and sense. It approaches, therefore, from the perspective of sociology of absences, the expansion of the present and by the logic inherent in his narratives of self and Brazil
