Foram realizados dois experimentos com o objetivo de avaliar a utilização da silagem de grãos úmidos de sorgo (SGUS) de alto ou baixo conteúdo de tanino na alimentação de leitões na fase de creche. No ensaio de digestibilidade, foram utilizados 12 suínos mestiços, machos castrados (peso inicial de 15,9±1,9 kg), alojados em gaiolas para estudos de metabolismo, distribuídos em um delineamento experimental inteiramente casualizado, com quatro repetições, em que a unidade experimental foi constituída por um leitão. O método utilizado foi o de coleta total de fezes e urina. Os valores de MS, proteína, amido, MO e energia digestíveis e os de EM, na matéria natural (68,75% de MS), foram, respectivamente, 43,94; 5,45; 46,67; 63,46%; 2.700 e 2.674 kcal/kg para a SGUS de alto teor de tanino e 40,72; 5,92; 44,08; 63,75%; 2.641 e 2.608 kcal/kg, para SGUS de baixo conteúdo de tanino. O segundo experimento foi conduzido para avaliar o desempenho de leitões e a viabilidade econômica da utilização de rações com diferentes níveis de substituição do milho seco por SGUS de alto ou baixo teor de tanino. Foram utilizados 56 suínos mestiços (14,7±1,9 e 29,2±3,0 kg de pesos inicial e final) distribuídos em um delineamento inteiramente casualizado, com quatro repetições e dois leitões por unidade experimental. Os tratamentos consistiram de uma ração à base de milho e farelo de soja e de outras seis com 33, 66 e 100% de substituição do milho seco por SGUS de alto ou baixo conteúdo de taninos, com base nos valores de energia digestível (ED) desses ingredientes. Não foram observadas diferenças no desempenho dos animais entre os tratamentos, entretanto, houve aumento linear no consumo diário de ração para a SGUS de baixo teor de tanino e redução linear para o custo da ração por quilograma de peso vivo ganho para a SGUS de alto teor de tanino.

Dois experimentos foram realizados para avaliar a digestibilidade aparente da EB, MS e PB em alimentos energéticos e protéicos utilizados para cães adultos. Foram utilizados quatro cães adultos (13,1 ± 2,0 kg), dois machos e duas fêmeas, sem raça definida, na avaliação de cada alimento. Os animais receberam a mesma quantidade de ração por unidade de peso metabólico. No experimento 1, foi determinado o valor nutritivo dos alimentos energéticos e, no experimento 2, os coeficientes de digestibilidade dos alimentos protéicos. Os coeficientes de digestibilidade da EB do milho extrusado (ME), do milho gelatinizado (MG), da gordura de coco e do óleo de soja, em dois níveis de inclusão (OS1 e OS2), e da gordura suína foram, respectivamente, 85,1; 84,4; 92,5; 92,1; 96,2 e 98,6%. Os coeficientes de digestibilidade da MS e PB do ME e MG foram, respectivamente, 84,2 e 65,3 e 84,5 e 65,0%. Na soja integral extrusada e nas farinhas de carne, de carne extrusada, de vísceras, de vísceras extrusada, de peixe extrusada e de pena extrusada, foram obtidos, respectivamente, os seguintes coeficientes de digestibilidade: 80,0; 73,3; 80,7; 87,6; 91,2; 91,1 e 79,8% da EB; 80,0; 68,4; 87,8; 86,7; 88,1; 85,2 e 76,0% da MS; e 83,7; 74,7; 82,3; 88,0; 88,9; 91,9 e 82,3% da PB.

A produção industrial de aves e suínos mostrou enormes avanços nos últimos anos, principalmente devido ao aumento dos conhecimentos na área de genética e de nutrição. Na área de avaliação dos alimentos e das exigências nutricionais de animais monogástricos, as melhoras se dão a passos largos, devido á seriedade com que os profissionais encaram a responsabilidade de fazer pesquisa de qualidade dentro e fora do país. Neste trabalho serão abordadas as metodologias que permitem melhorar a utilização dos alimentos de maneira mais eficiente e econômica. Serão citados alguns cuidados e procedimentos essenciais para executar adequadamente experimentos de desempenho com aves e suínos. Na atualidade, para a realização de experimentos com monogástricos, é necessário: definir claramente os objetivos, utilizar animais com peso inicial uniforme, usar número adequado de repetições e de animais por unidade experimental. Testes de médias devem ser usados para as variáveis qualitativas e quando a variável independente for quantitativa aplicar análise de regressão. O nível de significância utilizado (5, 7, 10%) pode variar conforme a importância econômica da característica estudada. A adequada condução dos ensaios, sejam de crescimento ou de digestibilidade, é fundamental para que as ferramentas apresentadas possam ter efetividade, resultando na melhora da produtividade e na redução dos custos e da excreção de nutrientes.

Objective: This in situ blind crossover study investigated the effect of calcium (Ca) rinse prior to the use fluoride (F) dentifrice on remineralisation of artificially demineralised enamel and on the composition of biofilm. Design: During four phases of 14 days, 10 volunteers wore appliances containing two artificially demineralised bovine enamel blocks. Three times a day, they rinsed with 10 mL, of Ca (150 mM) or placebo rinse (1 min). A slurry (1:3, w/v) of F (1030 ppm) or placebo dentifrice was dripped onto the blocks. During I min, the volunteers brushed their teeth with the respective dentifrice. The appliance was replaced into the mouth and the volunteers rinsed with water. The biofilm formed on the blocks was analysed for F and Ca. Enamel alterations were evaluated by the percentage of surface microhardness change (%SMHC), cross-sectional microhardness (% mineral volume) and alkali-soluble F analysis. Data were analysed by ANOVA (p < 0.05). Results: the use of the Ca pre-rinse before the F dentifrice produced a six- and four-fold increase in biofilm F and Ca concentrations, respectively. For enamel, the remineralisation was significantly improved by the Ca pre-rinse when compared to the other treatments. There was a significantly higher concentration of alkali-soluble F in enamel when the F dentifrice was used, but the Ca pre-rinse did not have any significant additive effect. Conclusions: According to our protocol, the Ca pre-rinse significantly increased biofilm F concentration and, regardless the use of F dentifrice, significantly enhanced the remineralisation of artificially demineralised enamel. The influence of sample preparation strategy of vegetables on the electrothermal behaviour of Se without and with chemical modifiers such as Pd(NO3)(2), Pd(NO3)(2) + Mg(NO3)(2), Pd(NO3)(2) + Cd(NO3)(2), pre-reduced Pd, Mg(NO3)(2), and Ni(NO3)(2) was investigated. Acid digestates and slurries of vegetables (0.1% m/v in 1% m/v HNO3 + 0.005% v/v of Triton X-100) were used to prepare reference solutions or slurries. For 10 mul of each modifier tested, pyrolysis and atomization temperatures were evaluated using pyrolysis and atomization curves, respectively. Best conditions, such as thermal stability, signal profile, repeatability and sensitivity were attained using Pd(NO3)(2) as chemical modifier. The following heating program (temperature, ramp/hold time) of the graphite tube of the Varian SpectrAA-800Z atomic absorption spectrometer was used: dry step (85 degreesC, 5/0 s; 95 degreesC, 40/0 s; 120 degreesC, 10/.5 s); pyrolysis step (1400 degreesC, 10/3s); atomization step (2200 degreesC, 1/2 s); clean step (2600 degreesC, 2/0 s). This pyrolysis temperature is 800 degreesC higher than when measuring without any modifier. For 20 muL sample volume and 10 mug Pd(NO3)(2), analytical curves in the 3.0-30 mug Se 1(-1) range were obtained. The method was applied for Se determination in acid digestates and slurries of 10 vegetable samples and one standard reference material (rice flower) and results were in agreement at 95% confidence level. Recoveries varied from 89 to 95% for spiked samples. The lifetime of the graphite tube was ca. 250 firings and the relative standard deviations (n = 12) for a typical acid digestate and slurry containing 20 mug Se 1(-1) were 3.8% and 8.3%, respectively. The limits of detection were 2.0 mug Se 1(-1) and 0.6 mug Se 1(-1) Se for digestates and slurries, respectively. (C) 2002 Elsevier B.V. Ltd. All rights reserved.

In the present study 21 young swine females, sexually mature, reared and kept under an industrial management system were assessed for cortisol and oestradiol-17 beta serum prolifes during the oestrus cycle. Blood sampling was performed always at the same interval, from 8:00 to 10:00 a.m. Each animal was submitted to 14 venal punctions at days 0, 2, 4, 6, 8, 10, 12, 14, 16, 18, 20, 21, 22 and 23 of the oestrous cycle. The first day of the oestrous phase was assumed to be day 0 and the 23(rd) as the first of the next cycle. Hormone determinations were performed using a solid phase radioimmunoassay (RIA). Regarding to cortisol concentrations, there was a variation from 3.5 to 8.0 mu g/dl and for oestradiol-17 beta the mean values detected varied from 3.5 to 14.9pg/ml.

The use of Saccharomyces cerevisiae as a substrate to selectively retain Sn(II) and Sn(IV) has been investigated. Several factors affecting the retention of the analytes by yeast, such as pH, amount of biomass, temperature and time of contact were evaluated. Based on this study, a method for determination of Sn(II) and Sn(IV) combining inductively coupled plasma optical emission spectrometry (ICP OES) and solid phase extraction using Saccharomyces cerevisiae is proposed. The procedure consists of the selective retention of Sn(IV) by yeast at pH = 2.0 while Sn(II) remains in solution. Determination of tin in the solid phase was easily carried out by submitting a slurry of the yeast (0.5 g/40 mL) directly to ICP OES. The precision of the extraction procedure was characterized by an RSD lower than 4%. The detection limits of tin (3 sigma) in the solid phase and the liquid phase were 1.1 and 0.7 mu g L-1, respectively. The proposed approach was evaluated for determination of Sn(II) and Sn(IV) in spiked river water and real samples of industrial waste water (untreated and treated). For all samples, recoveries of spiked Sn(II) and Sn(IV) were between 85 and 112%.

The use of Saccharomyces cerevisiae as a sorbent material to separate Cd(II) and Cd-metallothionein complex (Cd-MT) has been explored. Solid-liquid phase extractions were carried out in batch mode and the main parameters of the process (pH, temperature, time of incubation, amount of biomass and analyte) were evaluated. Under optimized conditions, the yeast quantitatively retain (94 +/- 5%) the Cd(II) while 97 +/- 2% of the Cd-MT remain in the supernatant. on base of the findings of this study, a simple method is proposed to determine Cd(II) and Cd-MT in cytosols extracted from mouse kidney and crab hepatopancreas. Inductively coupled plasma optical emission spectrometry was used to quantify the analytes in solid and liquid phase. Determination of Cd in the solid phase was carried out by introducing a slurry of the yeast (0.0625 g/10 mL) directly to the inductively coupled plasma optical emission spectrometer. Mixed standards solutions, which also have been submitted to the extraction procedure, were used to quantify the analytes in the samples. Thus, matrix effects due to nebulization of the slurry were overcame. Limits of detection (3 sigma) for Cd(II) and Cd-MT were 1.5 and 1.2 mu g L-1, respectively. Relative standard deviations of signals were 4.2% for measurements in the slurry of solid phase and 2.1% for measurements in the liquid phase. Recoveries of the analytes in cytosol samples were between 76 and 114%. The concentrations of Cd(II) (2.4 +/- 0.5 mu g L-1) and Cd-MT (3.0 +/- 0.5 mu g L-1) found by using the proposed approach were close to those found by tangential-flow ultrafiltration technique (2.6 +/- 0.7 mu g L-1 for Cd(II) and 3.7 +/- 1.7 mu g L-1 for Cd-MT).

The purpose of this study was to describe the anatomy of the lungs of wild boars for comparison with those of domestic swine. It was found that the right lung of the wild boar is divided into four lobes: cranial, median, caudal and accessory, whereas the left lung is divided into two lobes: cranial and caudal. In 93.4% of the cases, right pulmonary artery separates into the ascendant, descendant, median, accessory and caudal branches. In 73.3% of the cases, left pulmonary artery separates most frequently to form three branches to the cranial lobe, whereas the median lobe is generally supplied by only one arterial branch. There is a single pattern of bronchial distribution: in the right lung a tracheal bronchus leads to the cranial lobe, where it separates into the cranial and caudal bronchi and there are also bronchi to the median, caudal and accessory lobes. In the left lung, the large bronchus separates to form two branches, one of which further separates to form two branches to the cranial lobe whereas the other forms a single branch to the caudal lobe.

Starch extraction from roots and tubers uses grating with water and sieves to separate the starch slurry from residual mass. The starch is recovered by decantation or centrifugation. The yam starch extraction is difficult due to high viscosity of the slurry caused by non-starch polysaccharides (NSP). The establishment of an efficient extraction process may turn yam into a competitive raw material. In this paper Dioscorea alata starch extracted by four methods was characterized in order to establish the impact of treatments. When the tubers were digested with an aqueous oxalic acid/ammonium oxalate (OA/AO) 1/1 solution, it was easier to separate the starch slurry from residual mass, because viscosity was reduced. For all the others methods tested, the viscosity remained almost the same. The nitrogen present in yam tubers was removed during the different extractions to a different extent. The largest nitrogen reduction was observed with ONAO followed by the control (water). The spectrum of starch granules sizes obtained also varied according to the treatment. Results proved that NSP carries small starch granules over to the waste water. The smaller starch granules diameter varied from 1.9 mu m (OA/AO extraction) to 13.5 mu m (water and pectinase extractions). The larger diameter varied from 41.0 mu m (NaOH treatment) to 67.7 mu m (ONAO). All starches extracted showed a RVA behavior in agreement with literature for yam starch, but with small differences due to the influence of methods. ONAO extraction showed the best recovery (18 g of starch/100 g tuber yam) and granular variation but it interfered with the rheological behavior of starch.

This work describes an analytical procedure for vanadium determination in human hair slurries by electrothermal AAS using longitudinal heating (LHGA) and transversal heating (THGA) graphite furnace atomizers. The samples were powdered using cryogenic grinding and the hair slurries containing 0.2% (m/v) were prepared in three different media for determination of vanadium: 0.14 mol L-1 HNO3, 0.1% (v/v) Triton X-100 and 0.1% (v/v) water soluble tertiary amines (CFA-C, pH 8). The limits of detection (LOD), limits of quantification (LOQ), and characteristic masses obtained were 0.28, 0.95 mu g L-1 and 35 pg (LHGA) and 0.34, 1.13 mu g L-1 and 78 pg (THGA), respectively. The accuracy of the analytical results obtained by the proposed procedure in both equipments was confirmed by a paired t-test at the 95% confidence level and compared with a conventional procedure based on acid digestion. Conjugated Linoleic Acids (CLAs) comprise a family of positional and geometric isomers of linoleic acid. The main form of CLA, cis-9, trans-11-C18:2 show positive effects in cancer prevention and treatment. The major dietary sources of these fatty acids are derived from ruminant animals, in particular dairy products. In these animals, the endogenous synthesis mainly occurs in mammary gland by the action of enzyme Stearoyl CoA Desaturase (SCD). Different levels of expression and activity of SCD in mammary gland can explain partially the variation of CLA levels in fat milk. Considering a great fat concentration in bubaline milk and the benefit of a high and positive correlation between fat milk and CLA production, this study was carried on with the intention of sequencing and characterizing part of the gene that codifies SCD in buffaloes. Genomic DNA was extracted from blood samples of lactating bubaline which begins to the breed Murrah. After the (acho que nao precisa desse the) extractions, PCR (Polymerase Chain Reaction) reactions were made by using primers Z (sic) (sic) D1 and E1 (sic) (sic) F1. The fragments obtained in PCR were cloned into T vectors and transformed in competent cells DH10B line. After this, three samples of each fragment were sequenced from 5' and 3' extremities using a BigDye kit in an automatic sequencer. Sequences were edited in a consensus of each fragment and were submitted to BLAST-n / NCBI for similarity comparisions among other species. The sequence obtained with Z (sic) (sic) D1 primers shows 938 bp enclosing exons 1 and 2 and intron 1. The primers E1 (sic) (sic) F1 show 70 bp corresponding to exon 3 of bubaline SCD gene. Similarities were obtained between 85% and 97% among bubaline sequences and sequences of SCD gene described in human, mouse, rat, swine, bovine, caprine and ovine species. This study has permitted the identification and partial characterization of SCD codifing region in Bubalus bubalis specie.

Acicular monodispersed Fe1-xREx (RE= Nd, Sm,Eu,Tb;x=0, 0.05, 0.10) metallic nanoparticles (60 +/- 5 nm in length and axial ratio similar to6) obtained by reduction of alumina-coated goethite nanoparticles-containing rare earth (RE) under hydrogen flow are reported. Alumina and maghemite thin layers on particle surface were used to protect the goethite particles against sintering and oxidation, respectively. Al and RE additions were obtained by successive heterocoagulation reactions. Aluminum sulfate (10 at.% based on Fe) was dissolved in water and the pH adjusted to 12.5 with NaOH solution. Goethite particles were suspended in this solution and CO2 gas was blown into the slurry to neutralize it to a pH 8.5 or less. Particles were purified and dehydrated to effect transformation to alumina-coated hematite nanoparticles, which were re-suspended in aqueous solution in which RE sulfate (0-0.15 at.% based on Fe) has been dissolved, and the pH increased by ammonia aqueous solution addition. Resulted alumina-coated RE-doped hematite nanoparticles were reduced to metal at 450 degreesC/12 h under hydrogen flow and passivated with nitrogen-containing ethanol vapor at room temperature. Acicular monodispersed metallic nanoparticle systems were obtained and the presence of Al and RE were confirmed by induced-coupled plasma spectrometry analysis. X-ray diffraction, Mossbauer spectroscopy, and magnetization data are in agreement with the nanosized alpha-Fe core in a bcc structure, having a spinel structure, gammaFe(2)O(3), with thickness similar to1.5 run on particle surface. Main magnetic parameters showed saturation magnetization decreases and significant increasing in the coercive field with the RE composition increases. Magnetic properties of these particles, similar to40% smaller than those commercially available, suggest a decrease in the bit-size for high-density magnetic or magneto-optics recording media application. Statement of problem. Acrylic resin denture teeth soften upon immersion in water, and the heating generated during microwave sterilization may enhance this process.Purpose. Six brands of acrylic resin denture teeth were investigated with respect to the effect of microwave sterilization and water immersion on Vickers hardness (VHN).Material and Methods. The acrylic resin denture teeth (Dentron [D], Vipi Dent Plus [V], Postaris [P], Biolux [B], Trilux [T], and Artiplus [A]) were embedded in heat-polymerized acrylic resin within polyvinylchloride tubes. For each brand, the occlusal surfaces of 32 identical acrylic resin denture posterior teeth were ground flat with 1500-grit silicon carbide paper and polished on a wet polishing wheel with a slurry of tin oxide. Hardness tests were performed after polishing (control group, C) after polishing followed by 2 cycles of microwave sterilization at 650 W for 6 minutes (MwS group), after polishing followed by 90-day immersion in water (90-day Wim group), and after polishing followed by 90-day storage in water and 2 cycles of microwave sterilization (90-day Wim + MwS group). For each specimen, 8 hardness measurements were made and the mean was calculated. Data were analyzed with a 2-way analysis of variance followed by the Bonferroni procedure to determine any significance between pairs of mean values (alpha=.01).Results: Mircrowave sterilization of specimens significantly decreased (P <.001) the hardness of the acrylic resin denture tooth specimens P (17.8 to 16.6 VHN, V (18.3 to 15.8 VHN), T (17.4 to 15.3 VHN), B (16.8 to 15.7 VHN), and A (17.3 to 15.7 VHN). For all acrylic resin denture teeth, no significant differences in hardness were found between the groups Mws, 90-day Wim, and 90-day Wim + MwS, with the exception of the 90-day Wim + MwS tooth A specimens (14.4 VHN), which demonstrated significant lower mean values (P <.001) than the 90-day Wim (15.8 VHN) and MwS (15.7 VHN) specimens.Conclusions. For specimens immersed in water for 90 days, 2 cycles of microwave sterilization had no effect on the hardness of most of the acrylic resin denture teeth.
