O objetivo deste trabalho foi avaliar as características florais e carpométricas, a viabilidade dos grãos de pólen e a capacidade germinativa de sementes de cultivares de amoreira‑preta. Foram avaliadas as cultivares Brazos, Caingangue, Cherokee, Choctaw, Comanche, Guarani, BRS Tupy e Ébano. Utilizou-se um delineamento experimental inteiramente casualizado com as oito cultivares e dez repetições. Foram feitas avaliações quanto a: número de estames, carpelos, pétalas e sépalas; composição do meio de cultura; número de grãos de pólen por antera e por flor; taxa de germinação de grãos de pólen; percentagem de frutificação; massa de matéria fresca, dimensões e número de sementes dos frutos; massa e dimensões dos drupetes; e percentagem de emergência de plântulas. O meio de cultura estabelecido para as cultivares de amoreira‑preta foi acrescido de 90 g L‑1 de sacarose e 400 mg L‑1 de ácido bórico, com pH aferido para 6,5 e meio solidificado com 8 g L‑1 de ágar. 'Caingangue', 'Ébano' e 'BRS Tupy' apresentaram baixa germinação de grãos de pólen. 'Ébano' apresentou frutos arredondados e com poucos drupetes. 'Choctaw' e 'Comanche' apresentaram baixa percentagem de emergência de plântulas. 'Brazos' destacou-se na maioria das características avaliadas e é considerada como boa progenitora para programas de melhoramento genético.

The objective of this work was to isolate strains of lactic acid bacteria with probiotic potential from the digestive tract of marine shrimp (Litopenaeus vannamei), and to carry out in vitro selection based on multiple characters. The ideotype (ideal proposed strain) was defined by the highest averages for the traits maximum growth velocity, final count of viable cells, and inhibition halo against nine freshwater and marine pathogens, and by the lowest averages for the traits duplication time and resistance of strains to NaCl (1.5 and 3%), pH (6, 8, and 9), and biliary salts (5%). Mahalanobis distance (D²) was estimated among the evaluated strains, and the best ones were those with the shortest distances to the ideotype. Ten bacterial strains were isolated and biochemically identified as Lactobacillus plantarum (3), L. brevis (3), Weissella confusa (2), Lactococcus lactis (1), and L. delbrueckii (1). Lactobacillus plantarum strains showed a wide spectrum of action and the largest inhibition halos against pathogens, both Gram-positive and negative, high growth rate, and tolerance to all evaluated parameters. In relation to ideotype, L. plantarum showed the lowest Mahalanobis (D²) distance, followed by the strains of W. confusa, L. brevis, L. lactis, and L. delbrueckii. Among the analyzed bacterial strains, those of Lactobacillus plantarum have the greatest potential for use as a probiotic for marine shrimp.

O objetivo deste trabalho foi avaliar o efeito de diferentes comprimentos de onda no desenvolvimento in vitro de mudas de cana-de-açúcar. Explantes foram submetidos a quatro tratamentos de diodos emissores de luz (LED): 100% azul; 70% azul + 30% vermelha; 30% azul + 70% vermelha; 100% vermelha, além do controle com lâmpada fluorescente branca. As plântulas foram avaliadas quanto a: número de brotações; altura; massa de matéria fresca e seca; e conteúdo de carotenoides e das clorofilas a e b. Observou-se desmanche dos tilacoides nos cloroplastos, proporcional ao aumento na incidência de luz vermelha. O porte das mudas diminui com o aumento na incidência de luz vermelha.

Reconstruction of defects in the craniomaxillofacial (CMF) area has mainly been based on bone grafts or metallic fixing plates and screws. Particularly in the case of large calvarial and/or craniofacial defects caused by trauma, tumours or congenital malformations, there is a need for reliable reconstruction biomaterials, because bone grafts or metallic fixing systems do not completely fulfill the criteria for the best possible reconstruction methods in these complicated cases. In this series of studies, the usability of fibre-reinforced composite (FRC) was studied as a biostable, nonmetallic alternative material for reconstructing artificially created bone defects in frontal and calvarial areas of rabbits. The experimental part of this work describes the different stages of the product development process from the first in vitro tests with resin-impregnated fibrereinforced composites to the in vivo animal studies, in which this FRC was tested as an implant material for reconstructing different size bone defects in rabbit frontal and calvarial areas. In the first in vitro study, the FRC was polymerised in contact with bone or blood in the laboratory. The polymerised FRC samples were then incubated in water, which was analysed for residual monomer content by using high performance liquid chromatography (HPLC). It was found that this in vitro polymerisation in contact with bone and blood did not markedly increase the residual monomer leaching from the FRC. In the second in vitro study, different adhesive systems were tested in fixing the implant to bone surface. This was done to find an alternative implant fixing system to screws and pins. On the basis of this study, it was found that the surface of the calvarial bone needed both mechanical and chemical treatments before the resinimpregnated FRC could be properly fixed onto it. In three animal studies performed with rabbit frontal bone defects and critical size calvarial bone defect models, biological responses to the FRC implants were evaluated. On the basis of theseevaluations, it can be concluded that the FRC, based on E-glass (electrical glass) fibres forming a porous fibre veil enables the ingrowth of connective tissues to the inner structures of the material, as well as the bone formation and mineralization inside the fibre veil. Bone formation could be enhanced by using bioactive glass granules fixed to the FRC implants. FRC-implanted bone defects healed partly; no total healing of defects was achieved. Biological responses during the follow-up time, at a maximum of 12 weeks, to resin-impregnated composite implant seemed to depend on the polymerization time of the resin matrix of the FRC. Both of the studied resin systems used in the FRC were photopolymerised and the heat-induced postpolymerisation was used additionally.

Cell therapy for nucleus pulposus (NP) regeneration is an attractive treatment for early disc degeneration as shown by studies using autologous NP cells or stem cells. Another potential source of cells is foetal cells. We investigated the feasibility of isolating foetal cells from human foetal spine tissues and assessed their chondrogenic potential in alginate bead cultures. Histology and immunohistochemistry of foetal tissues showed that the structure and the matrix composition (aggrecan, type I and II collagen) of foetal intervertebral disc (IVD) were similar to adult IVD. Isolated foetal cells were cultured in monolayer in basic media supplemented with 10% Fetal Bovine Serum (FBS) and from each foetal tissue donation, a cell bank of foetal spine cells at passage 2 was established and was composed of around 2000 vials of 5 million cells. Gene expression and immunohistochemistry of foetal spine cells cultured in alginate beads during 28 days showed that cells were able to produce aggrecan and type II collagen and very low level of type I and type X collagen, indicating chondrogenic differentiation. However variability in matrix synthesis was observed between donors. In conclusion, foetal cells could be isolated from human foetal spine tissues and since these cells showed chondrogenic potential, they could be a potential cell source for IVD regeneration.

The objective of this work was to evaluate the effects of hormonal synchronization protocols, associated or not with follicular development stimulation, on the recovery of oocytes and on in vitro production of Bos indicus and B. taurus embryos, in different seasons. Ultrasound-guided follicular aspirations (n=237) were performed without pre-treatment (G1, control group) and after follicular wave synchronization (G2), or after follicular wave synchronization and follicle growth induction (G3). Bos indicus produced more oocytes and embryos than B. taurus (18.7±0.9 vs. 11.9±0.6 oocytes and 4.8±0.3 vs. 2.1±0.2 embryos). On average, oocyte and embryo yields were higher in G3 than in G2, and both were greater than in G1, which lead to a higher conversion of oocytes to embryos in these treatments. The hot or the cold season did not affect the B. indicus outcomes, whereas, in B. taurus, both oocyte recovery and embryo production were higher in the cold season. Follicular wave synchronization improves ovum pick-up and in vitro production of embryos in both cattle subspecies evaluated.

Children conceived by assisted reproductive technologies (ART) display a level of vascular dysfunction similar to that seen in children of mothers with preeclamspia. The long-term consequences of ART-associated vascular disorders are unknown and difficult to investigate in healthy children. Here, we found that vasculature from mice generated by ART display endothelial dysfunction and increased stiffness, which translated into arterial hypertension in vivo. Progeny of male ART mice also exhibited vascular dysfunction, suggesting underlying epigenetic modifications. ART mice had altered methylation at the promoter of the gene encoding eNOS in the aorta, which correlated with decreased vascular eNOS expression and NO synthesis. Administration of a deacetylase inhibitor to ART mice normalized vascular gene methylation and function and resulted in progeny without vascular dysfunction. The induction of ART-associated vascular and epigenetic alterations appeared to be related to the embryo environment; these alterations were possibly facilitated by the hormonally stimulated ovulation accompanying ART. Finally, ART mice challenged with a high-fat diet had roughly a 25% shorter life span compared with control animals. This study highlights the potential of ART to induce vascular dysfunction and shorten life span and suggests that epigenetic alterations contribute to these problems.

Hyperglycosylated human chorionic gonadotropin (H-hCG) is secreted by the placenta in early pregnancy. Decreased H-hCG levels have been associated with abortion in spontaneous pregnancy. We retrospectively measured H-hCG and dimeric hCG in the sera of 87 in vitro fertilization patients obtained in the 3 weeks following embryo transfer and set the results in relation to pregnancy outcome. H-hCG and dimeric hCG were correlated (r(2) = 0.89), and were significantly decreased in biochemical pregnancy (2 microg/l and 18 IU/l, respectively) compared to early pregnancy loss (22 microg/l and 331 IU/l) and ongoing pregnancy (32 microg/l and 353 IU/l). Only H-hCG tended to discriminate between these last two groups.

The objective of this work was to evaluate the in vitro maintenance of oil palm (Elaeis guineensis and E. oleifera) accessions under slow-growth conditions. Plants produced by embryo rescue were subject to 1/2MS culture medium supplemented with the carbohydrates sucrose, mannitol, and sorbitol at 1, 2, and 3% under 20 and 25±2ºC. After 12 months, the temperature of 20°C reduced plant growth. Sucrose is the most appropriate carbohydrate for maintaining the quality of the plants, whereas mannitol and sorbitol result in a reduced plant survival.

Contexte : L'insuffisance cardiaque touche environ 150 personnes sur 100'000 habitants en Suisse, avec une¦prévalence évaluée à 1.45 %, et cause 42.3 décès par 100'000 habitants. Globalement, la prévalence de¦l'insuffisance cardiaque augmente, d'une part à cause du vieillissement de la population, d'autre part par¦l'amélioration de la prise en charge de pathologies cardiaques. La transplantation reste actuellement le gold¦standard pour l'insuffisance cardiaque réfractaire au traitement pharmacologique, mais les organes sont¦rares. Une alternative a donc été développée, celle des systèmes d'assistance ventriculaire (ventricular assist¦device, VAD). Les appareils existants actuellement sur le marché fonctionnent en déviant le sang du¦ventricule vers un système de projection à flux pulsatile ou continu placé dans la cage thoracique, avant de le¦renvoyer vers l'artère. Ils comportent certains défauts, en particulier la nécessité de léser le coeur pour les¦implanter et les risques hémorragique et thrombo-embolique importants. Pour remédier à ces défauts, des¦VAD externes sont en cours de développement. Fixés autour du coeur, ils permettent de l'assister dans la¦contraction, sans contact direct avec le sang ni lésion du coeur. Dans cette étude, nous avons créé deux¦prototypes de VAD externes basés sur la technique du muscle artificiel. Ils sont faits de fils de Nitinol, un¦alliage à mémoire de forme qui raccourcit lorsqu'il est chauffé. Placés autour du coeur, ils lui impriment un¦mouvement de contraction, tel un muscle artificiel.¦Méthode : deux VAD externes ont été créés en utilisant du Nitinol. Les fibres de Nitinol du VAD N°1¦passent à travers des charnières qui augmentent son pouvoir de contraction. Celles du VAD N°2 sont¦orientées dans un maillage de fibres de Kevlar de manière à reproduire la direction des fibres musculaires du¦ventricule humain. Ils ont été testés sur un banc d'essai avec un coeur en silicone. Nous avons mesuré la¦fraction d'éjection, le débit et la pression générée, à différentes valeurs de précharge et post-charge. Les¦VAD étaient alimentés par une génératrice ou par une unité de contrôle, qui permettait de fournir l'énergie¦précisément dans chaque fil de Nitinol et d'imposer une certaine fréquence cardiaque.¦Résultats : Tant avec la génératrice que l'unité de contrôle, le ventricule gauche du VAD N°1 fournit une¦fraction d'éjection maximale de 16.09 %. Le débit maximal est de 191.42 ml/min. La génératrice permet au¦VAD N°2 de fournir une fraction d'éjection de 6.18 %, contre 2.48 % avec l'unité de contrôle. Le débit¦maximal est de 27.37 ml/min. La pression générée atteint 75 mmHg pour le VAD N°1 et 6 mmHg pour le¦VAD N°2.¦Discussion/conclusion : Le VAD N°1 est le plus performant, il permet une augmentation significative de la¦fraction d'éjection et pourrait avoir un impact sur la qualité de vie des patients. L'unité de contrôle apporte¦un avantage sur la génératrice pour le VAD N°1, en dirigeant plus précisément l'énergie dans les fils de¦Nitinol et en limitant les pertes. Le VAD N°2, lui, est peu performant et l'unité de contrôle n'améliore pas¦ses performances. Cela est probablement dû à sa configuration initiale, la taille du VAD n'étant pas adaptée¦au coeur en silicone. Cette étude prouve qu'il est possible d'assister un coeur depuis l'extérieur, sans l'altérer,¦et que la position des fibres de Nitinol a plus d'importance que leur nombre.

Beta-hemolytic Streptococcus agalactiae is the leading cause of bacteremia and invasive infections. These diseases are treated with β-lactams or macrolides, but the emergence of less susceptible and even fully resistant strains is a cause for concern. New bacteriophage lysins could be promising alternatives against such organisms. They hydrolyze the bacterial peptidoglycan at the end of the phage cycle, in order to release the phage progeny. By using a bioinformatic approach to screen several beta-hemolytic streptococci, a gene coding for a lysin was identified on a prophage carried by Streptococcus dysgalactiae subsp. equisimilis SK1249. The gene product, named PlySK1249, harbored an original three-domain structure with a central cell wall-binding domain surrounded by an N-terminal amidase and a C-terminal CHAP domain. Purified PlySK1249 was highly lytic and bactericidal for S. dysgalactiae (2-log10 CFU/ml decrease within 15 min). Moreover, it also efficiently killed S. agalactiae (1.5-log10 CFU/ml decrease within 15 min) but not several streptococcal commensal species. We further investigated the activity of PlySK1249 in a mouse model of S. agalactiae bacteremia. Eighty percent of the animals (n = 10) challenged intraperitoneally with 10(6) CFU of S. agalactiae died within 72 h, whereas repeated injections of PlySK1249 (45 mg/kg 3 times within 24 h) significantly protected the mice (P < 0.01). Thus, PlySK1249, which was isolated from S. dysgalactiae, demonstrated high cross-lytic activity against S. agalactiae both in vitro and in vivo. These encouraging results indicated that PlySK1249 might represent a good candidate to be developed as a new enzybiotic for the treatment of systemic S. agalactiae infections.

Major advances in the understanding of the molecular biology of hepatitis C virus (HCV) have been made recently. While the chimpanzee is the only established animal model of HCV infection, several in vivo and in vitro models have been established that allow us to study various aspects of the viral life cycle. In particular, the replicon system and the production of recombinant infectious virions revolutionized the investigation of HCV-RNA replication and rendered all steps of the viral life cycle, including entry and release of viral particles, amenable to systematic analysis. In the following we will review the different in vivo and in vitro models of HCV infection.

Most hematopoietic stem cells (HSC) in the bone marrow reside in a quiescent state and occasionally enter the cell cycle upon cytokine-induced activation. Although the mechanisms regulating HSC quiescence and activation remain poorly defined, recent studies have revealed a role of lipid raft clustering (LRC) in HSC activation. Here, we tested the hypothesis that changes in lipid raft distribution could serve as an indicator of the quiescent and activated state of HSCs in response to putative niche signals. A semi-automated image analysis tool was developed to map the presence or absence of lipid raft clusters in live HSCs cultured for just one hour in serum-free medium supplemented with stem cell factor (SCF). By screening the ability of 19 protein candidates to alter lipid raft dynamics, we identified six factors that induced either a marked decrease (Wnt5a, Wnt3a and Osteopontin) or increase (IL3, IL6 and VEGF) in LRC. Cell cycle kinetics of single HSCs exposed to these factors revealed a correlation of LRC dynamics and proliferation kinetics: factors that decreased LRC slowed down cell cycle kinetics, while factors that increased LRC led to faster and more synchronous cycling. The possibility of identifying, by LRC analysis at very early time points, whether a stem cell is activated and possibly committed upon exposure to a signaling cue of interest could open up new avenues for large-scale screening efforts.

Explantes de bananeira cv. Caipiras (AAA) foram cultivados in vitro. Nas subculturas de estabelecimento e multiplicação, o meio utilizado foi MS + 5 mg/l de BAP e, na de enraizamento, 50%MS sem BAP. Na avaliação das subculturas, a maior contaminação ocorreu no estabelecimento (74,7%), seguindo decrescente até o enraizamento, com 5,6%. O número de brotos por frasco diminui ao longo das subculturas, atingindo 3,7 brotos na subculturas 1 e 1,7 na subcultura 4. O tamanho dos brotos permaneceu entre 24 e 26 mm, nas subculturas de 1 a 3, diminuindo na subcultura 4, com 22 mm. Como conseqüência, a classe de brotos menores (x £ 20 mm) predominou, variando de 56% na subcultura 1 para 65% na subcultura 4. No enraizamento, a predominância (62%) foi de brotos na faixa entre 30 e 60 mm, com média de 4,6 brotos por frasco. A avaliação geral do processo indicou um rendimento real de 70,9 mudas/rizoma.

Este experimento teve como objetivo avaliar a influência de diferentes concentrações de ferro, cobre e zinco do meio MS (MURASHIGE & SKOOG, 1962) no controle da oxidação de explantes iniciais de bananeira-Prata (Musa AAB). Foram utilizadas três concentrações (100, 50 e 0 miM) de FeEDTA, duas concentrações (0,1 e 0miM) de (CuSO4).5H2O e duas concentrações (30 e 0miM) de (ZnSO4).7H2O, num delineamento inteiramente casualizado, arranjado em um fatorial completo 3 x 2 x 2, utilizando-se de 15 repetições. Ápices caulinares foram inoculados em meio MS modificado e, decorridos 28 dias após a inoculação, avaliaram-se a massa de matéria fresca, altura e grau de oxidação. Observou-se que esses micronutrientes são essenciais para o crescimento dos explantes e que a concentração de ferro influencia na oxidação de explantes, sendo que maiores graus de escurecimento foram observados nas concentrações mais elevadas. A redução ou retirada destes elementos do meio MS, isoladamente ou em combinações, não foi suficiente para eliminar a oxidação dos explantes.
