The Pliocene and Early Pleistocene, between 5.3 and 0.8 million years ago, span a transition from a global climate state that was 2-3 °C warmer than present with limited ice sheets in the Northern Hemisphere to one that was characterized by continental-scale glaciations at both poles. Growth and decay of these ice sheets was paced by variations in the Earth's orbit around the Sun. However, the nature of the influence of orbital forcing on the ice sheets is unclear, particularly in light of the absence of a strong 20,000-year precession signal in geologic records of global ice volume and sea level. Here we present a record of the rate of accumulation of iceberg-rafted debris offshore from the East Antarctic ice sheet, adjacent to the Wilkes Subglacial Basin, between 4.3 and 2.2 million years ago. We infer that maximum iceberg debris accumulation is associated with the enhanced calving of icebergs during ice-sheet margin retreat. In the warmer part of the record, between 4.3 and 3.5 million years ago, spectral analyses show a dominant periodicity of about 40,000 years. Subsequently, the powers of the 100,000-year and 20,000-year signals strengthen. We suggest that, as the Southern Ocean cooled between 3.5 and 2.5 million years ago, the development of a perennial sea-ice field limited the oceanic forcing of the ice sheet. After this threshold was crossed, substantial retreat of the East Antarctic ice sheet occurred only during austral summer insolation maxima, as controlled by the precession cycle.

The Saemaul Undong of the Republic of Korea has been world-widely recognized as a successful model of rural community development. The Saemaul Undong was a pure Korean way of community development program which was initiated by the political will of the top national leadership in order to escape from poverty. There are several key factors to the success of the Saemaul Undong. First, the national government's guidance and support for the movement played a very important role in the whole period of the movement. Second, there was a wide range of people's participation in the implementation process. Third, the Saemaul Undong could make a big success by nurturing community leadership which was selected by rural residents themselves. Finally, as a movement for the spiritual reform, the Saemaul Undong imbued the people with the spirits of diligence, the self-reliance, and cooperation.

The aim of this paper Is lo discuss the influence of the selection of the interpolation kernel in the accuracy of the modeling of the internal viscous dissipation in Tree surface Hows, Simulations corresponding to a standing wave* for which an analytic solution available, are presented. Wendland and renormalized Gaussian kernels are considered. The differences in the flow pattern* and Internal dissipation mechanisms are documented for a range of Reynolds numbers. It is shown that the simulations with Wendland kernels replicate the dissipation mechanisms more accurately than those with a renormalized Gaussian kernel. Although some explanations are hinted we have Tailed to clarify which the core structural reasons for Mich differences are*

El presente proyecto de fin de carrera describe y analiza el estudio integral del efecto de las vibraciones producidas por voladuras superficiales realizadas en el proyecto del “Tercer Juego de Esclusas” ejecutado para la Expansión del Canal de Panamá. Se recopilan un total de 53 registros, data generada por el monitoreo de 7 sismógrafos en 10 voladuras de producción realizadas en el año 2010. El fenómeno vibratorio tiene dos parámetros fundamentales, la velocidad pico-partícula (PPV) y la frecuencia dominante, los cuales caracterizan cuan dañino puede ser éste frente a su influencia sobre las estructuras civiles; por ello, se pretende caracterizarlas y fundamentalmente predecirlas, lo que permitirá su debido control. En función a lo expuesto, el estudio realizado consta de dos partes, la primera describe el comportamiento del terreno mediante la estimación de la ley de atenuación de la velocidad pico-partícula a través del uso de la regresión lineal por mínimos cuadrados; la segunda detalla un procedimiento validable para la predicción de la frecuencia dominante y del pseudo-espectro de respuesta de velocidad (PVRS) basada en la teoría de Newmark & Hall. Se ha obtenido: (i) la ley de atenuación del terreno para distintos grados de fiabilidad, (ii) herramientas de diseño de voladuras basadas en la relación de carga – distancia, (iii) la demostración que los valores de PPV se ajustan a una distribución log-normal, (iv) el mapa de isolíneas de PPV para el área de estudio, (v) una técnica detallada y válida para la predicción de la frecuencia dominante y del espectro de respuesta, (vi) formulaciones matemáticas de los factores de amplificación para el desplazamiento, velocidad y aceleración, (vii) mapa de isolíneas de amplificación para el área de estudio. A partir de los resultados obtenidos se proporciona información útil para su uso en el diseño y control de las voladuras posteriores del proyecto. ABSTRACT This project work describes and analyzes the comprehensive study of the effect of the vibrations produced by surface blasting carried out in the "Third Set of Locks" project executed for the expansion of the Panama Canal. A total of 53 records were collected, with the data generated by the monitoring of 7 seismographs in 10 production blasts carried out in 2010. The vibratory phenomenon has two fundamental parameters, the peak-particle velocity (PPV) and the dominant frequency, which characterize how damaging this can be compared to their influence on structures, which is why this is intended to characterize and predict fundamentally, that which allows proper control. Based on the above, the study consists of two parts; the first describes the behavior of the terrain by estimating the attenuation law for peak-particle velocity by using the ordinary least squares regression analysis, the second details a validable procedure for the prediction of the dominant frequency and pseudo-velocity response spectrum (PVRS) based on the theory of Newmark & Hall. The following have been obtained: (i) the attenuation law of the terrain for different degrees of reliability, (ii) blast design tools based on charge-distance ratio, (iii) the demonstration that the values of PPV conform to a log-normal distribution, (iv) the map of isolines of PPV for the area of study (v) detailed and valid technique for predicting the dominant frequency response spectrum, (vi) mathematical formulations of the amplification factors for displacement, velocity and acceleration, (vii) amplification of isolines map for the study area. From the results obtained, the study provides useful information for use in the design and control of blasting for subsequent projects.

Abstract Potential consequences of climate change on crop production can be studied using mechanistic crop simulation models. While a broad variety of maize simulation models exist, it is not known whether different models diverge on grain yield responses to changes in climatic factors, or whether they agree in their general trends related to phenology, growth, and yield. With the goal of analyzing the sensitivity of simulated yields to changes in temperature and atmospheric carbon dioxide concentrations [CO2], we present the largest maize crop model intercomparison to date, including 23 different models. These models were evaluated for four locations representing a wide range of maize production conditions in the world: Lusignan (France), Ames (USA), Rio Verde (Brazil) and Morogoro (Tanzania). While individual models differed considerably in absolute yield simulation at the four sites, an ensemble of a minimum number of models was able to simulate absolute yields accurately at the four sites even with low data for calibration, thus suggesting that using an ensemble of models has merit. Temperature increase had strong negative influence on modeled yield response of roughly 0.5 Mg ha 1 per °C. Doubling [CO2] from 360 to 720 lmol mol 1 increased grain yield by 7.5% on average across models and the sites. That would therefore make temperature the main factor altering maize yields at the end of this century. Furthermore, there was a large uncertainty in the yield response to [CO2] among models. Model responses to temperature and [CO2] did not differ whether models were simulated with low calibration information or, simulated with high level of calibration information.

Chromatin remodeling complexes such as the SWI/SNF complex make DNA accessible to transcription factors by disrupting nucleosomes. However, it is not known how such complexes are targeted to the promoter. For example, a SWI/SNF1-like chromatin remodeling complex erythroid Krüppel-like factor (EKLF) coactivator-remodeling complex 1 (E-RC1) disrupts the nucleosomes over the human β-globin promoter in an EKLF-dependent manner. However, it is not known whether E-RC1 is targeted specifically to the β-globin promoter or whether E-RC1 is randomly targeted, but its activity is evident only at the β-globin promoter. Because E-RC1 cannot remodel chromatin over the β-globin promoter without EKLF in vitro, it has been proposed that SWI/SNF1-like complexes such as E-RC1 are targeted specifically to the promoter by selectively interacting with promoter-associated transcription factors such as EKLF. In this report, we test this hypothesis in the cellular context by using the ProteIN POsition Identification with Nuclease Tail (PIN*POINT) assay. We find that the Brahma-related gene (BRG) 1 and BRG1-associated factor (BAF) 170 subunits of E-RC1 are both recruited near the transcription initiation site of the β-globin promoter. On transiently transfected templates, both the locus control region and the EKLF-binding site are important for their recruitment to the β-globin promoter in mouse erythroleukemia cells. When the β-globin promoter was linked to the cytomegalovirus enhancer, the E-RC1 complex was not recruited, suggesting that recruitment of the E-RC1 complex is not a general property of enhancers.

An essential component of regulated steroidogenesis is the translocation of cholesterol from the cytoplasm to the inner mitochondrial membrane where the cholesterol side-chain cleavage enzyme carries out the first committed step in steroidogenesis. Recent studies showed that a 30-kDa mitochondrial phosphoprotein, designated steroidogenic acute regulatory protein (StAR), is essential for this translocation. To allow us to explore the roles of StAR in a system amenable to experimental manipulation and to develop an animal model for the human disorder lipoid congenital adrenal hyperplasia (lipoid CAH), we used targeted gene disruption to produce StAR knockout mice. These StAR knockout mice were indistinguishable initially from wild-type littermates, except that males and females had female external genitalia. After birth, they failed to grow normally and died from adrenocortical insufficiency. Hormone assays confirmed severe defects in adrenal steroids—with loss of negative feedback regulation at hypothalamic–pituitary levels—whereas hormones constituting the gonadal axis did not differ significantly from levels in wild-type littermates. Histologically, the adrenal cortex of StAR knockout mice contained florid lipid deposits, with lesser deposits in the steroidogenic compartment of the testis and none in the ovary. The sex-specific differences in gonadal involvement support a two-stage model of the pathogenesis of StAR deficiency, with trophic hormone stimulation inducing progressive accumulation of lipids within the steroidogenic cells and ultimately causing their death. These StAR knockout mice provide a useful model system in which to determine the mechanisms of StAR’s essential roles in adrenocortical and gonadal steroidogenesis.

The signal transducer and activator of transcription, STAT5b, has been implicated in signal transduction pathways for a number of cytokines and growth factors, including growth hormone (GH). Pulsatile but not continuous GH exposure activates liver STAT5b by tyrosine phosphorylation, leading to dimerization, nuclear translocation, and transcriptional activation of the STAT, which is proposed to play a key role in regulating the sexual dimorphism of liver gene expression induced by pulsatile plasma GH. We have evaluated the importance of STAT5b for the physiological effects of GH pulses using a mouse gene knockout model. STAT5b gene disruption led to a major loss of multiple, sexually differentiated responses associated with the sexually dimorphic pattern of pituitary GH secretion. Male-characteristic body growth rates and male-specific liver gene expression were decreased to wild-type female levels in STAT5b−/− males, while female-predominant liver gene products were increased to a level intermediate between wild-type male and female levels. Although these responses are similar to those observed in GH-deficient Little mice, STAT5b−/− mice are not GH-deficient, suggesting that they may be GH pulse-resistant. Indeed, the dwarfism, elevated plasma GH, low plasma insulin-like growth factor I, and development of obesity seen in STAT5b−/− mice are all characteristics of Laron-type dwarfism, a human GH-resistance disease generally associated with a defective GH receptor. The requirement of STAT5b to maintain sexual dimorphism of body growth rates and liver gene expression suggests that STAT5b may be the major, if not the sole, STAT protein that mediates the sexually dimorphic effects of GH pulses in liver and perhaps other target tissues. STAT5b thus has unique physiological functions for which, surprisingly, the highly homologous STAT5a is unable to substitute.
