Pigs are often colonized by more than one bacterial and/or viral species during respiratory tract infections. This phenomenon is known as the porcine respiratory disease complex (PRDC). Actinobacillus pleuropneumoniae (App) and porcine reproductive and respiratory syndrome virus (PRRSV) are pathogens that are frequently involved in PRDC. The main objective of this project was to study the in vitro interactions between these two pathogens and the host cells in the context of mixed infections. To fulfill this objective, PRRSV permissive cell lines such as MARC-145, SJPL, and porcine alveolar macrophages (PAM) were used. A pre-infection with PRRSV was performed at 0.5 multiplicity of infection (MOI) followed by an infection with App at 10 MOI. Bacterial adherence and cell death were compared. Results showed that PRRSV preinfection did not affect bacterial adherence to the cells. PRRSV and App co-infection produced an additive cytotoxicity effect. Interestingly, a pre-infection of SJPL and PAM cells with App blocked completely PRRSV infection. Incubation of SJPL and PAM cells with an App cell-free culture supernatant is also sufficient to significantly block PRRSV infection. This antiviral activity is not due to LPS but rather by small molecular weight, heat-resistant App metabolites (,1 kDa). The antiviral activity was also observed in SJPL cells infected with swine influenza virus but to a much lower extent compared to PRRSV. More importantly, the PRRSV antiviral activity of App was also seen with PAM, the cells targeted by the virus in vivo during infection in pigs. The antiviral activity might be due, at least in part, to the production of interferon c. The use of in vitro experimental models to study viral and bacterial co-infections will lead to a better understanding of the interactions between pathogens and their host cells, and could allow the development of novel prophylactic and therapeutic tools.

Deoxynivalenol (DON) is a mycotoxin produced by Fusarium spp and is a common contaminant of grains in North America. Among farm animals, swine are the most susceptible to DON because it markedly reduces feed intake and decreases weight gain. Porcine circovirus type 2 (PCV2) is the main causative agent of several syndromes in weaning piglets collectively known as porcine circovirus-associated disease (PCVAD). The objectives of this study were to investigate the impact of DON on PCV2 replication in NPTr permissive cell line, and to determine eventual potentiating effects of DON on PCV2 infection in pigs. Noninfected and infected cells with PCV2 were treated with increasing concentrations of DON (0, 70, 140, 280, 560, 1200 ng/mL) and cell survival and virus titer were evaluated 72 h postinfection. Thirty commercial piglets were randomly divided into 3 experimental groups of 10 animals based on DON content of served diets (0, 2.5 and 3.5 mg/kg DON). All groups were further divided into subgroups of 6 pigs and were inoculated with PCV2b virus. The remaining pigs (control) were sham-inoculated with PBS. In vitro results showed that low concentrations of DON could potentially increase PCV2 replication depending on virus genotype. In vivo results showed that even though viremia and lung viral load tend to be higher in animal ingesting DON contaminated diet at 2.5 mg/kg, DON had no significant effect on clinical manifestation of PCVAD in PCV2b infected animals. DON has neither in vitro nor in vivo clear potentiating effects in the development of porcine circovirus infection despite slight increases in viral replication.

The aim of this pilot project was to investigate association of viruses with bacterial biofilms. Our preliminary data indicate that important viral pathogens of swine, namely, porcine reproductive and respiratory syndrome virus and porcine circovirus type 2, can associate with and persist within bacterial biofilms for several days.

Background Airborne transmitted pathogens, such as porcine reproductive and respiratory syndrome virus (PRRSV), need to interact with host cells of the respiratory tract in order to be able to enter and disseminate in the host organism. Pulmonary alveolar macrophages (PAM) and MA104 derived monkey kidney MARC-145 cells are known to be permissive to PRRSV infection and replication and are the most studied cells in the literature. More recently, new cell lines developed to study PRRSV have been genetically modified to make them permissive to the virus. The SJPL cell line origin was initially reported to be epithelial cells of the respiratory tract of swine. Thus, the goal of this study was to determine if SJPL cells could support PRRSV infection and replication in vitro. Results The SJPL cell growth was significantly slower than MARC-145 cell growth. The SJPL cells were found to express the CD151 protein but not the CD163 and neither the sialoadhesin PRRSV receptors. During the course of the present study, the SJPL cells have been reported to be of monkey origin. Nevertheless, SJPL cells were found to be permissive to PRRSV infection and replication even if the development of the cytopathic effect was delayed compared to PRRSV-infected MARC-145 cells. Following PRRSV replication, the amount of infectious viral particles produced in SJPL and MARC-145 infected cells was similar. The SJPL cells allowed the replication of several PRRSV North American strains and were almost efficient as MARC-145 cells for virus isolation. Interestingly, PRRSV is 8 to 16 times more sensitive to IFNα antiviral effect in SJPL cell in comparison to that in MARC-145 cells. PRRSV induced an increase in IFNβ mRNA and no up regulation of IFNα mRNA in both infected cell types. In addition, PRRSV induced an up regulation of IFNγ and TNF-α mRNAs only in infected MARC-145 cells. Conclusions In conclusion, the SJPL cells are permissive to PRRSV. In addition, they are phenotypically different from MARC-145 cells and are an additional tool that could be used to study PRRSV pathogenesis mechanisms in vitro.

Empirical evidence suggests that ambiguity is prevalent in insurance pricing and underwriting, and that often insurers tend to exhibit more ambiguity than the insured individuals (e.g., [23]). Motivated by these findings, we consider a problem of demand for insurance indemnity schedules, where the insurer has ambiguous beliefs about the realizations of the insurable loss, whereas the insured is an expected-utility maximizer. We show that if the ambiguous beliefs of the insurer satisfy a property of compatibility with the non-ambiguous beliefs of the insured, then there exist optimal monotonic indemnity schedules. By virtue of monotonicity, no ex-post moral hazard issues arise at our solutions (e.g., [25]). In addition, in the case where the insurer is either ambiguity-seeking or ambiguity-averse, we show that the problem of determining the optimal indemnity schedule reduces to that of solving an auxiliary problem that is simpler than the original one in that it does not involve ambiguity. Finally, under additional assumptions, we give an explicit characterization of the optimal indemnity schedule for the insured, and we show how our results naturally extend the classical result of Arrow [5] on the optimality of the deductible indemnity schedule.

Le Staphylococcus aureus résistant à la méthicilline (SARM) est un enjeu majeur en santé publique. Il est responsable d’une grande variété d’infections. Les “Livestock Associated-MRSA” (LA-MRSA) sont des SARM ayant comme origine les animaux de production tels le porc ou la volaille. Ils constituent un risque de transmission à l’humain via la chaîne alimentaire. Les LA-MRSA peuvent former du biofilm ce qui augmente leur tolérance aux stress environnementaux. Le biofilm est partiellement régulé par le système Agr. Il n’existe aucune donnée sur les ‘LA-MRSA’ d’origine aviaire au Québec. Les objectifs de ce projet étaient : (i) de déterminer la prévalence de ces SARM dans la viande de poulet et le poulet à griller de la province de Québec et (ii) de caractériser les isolats retrouvés. La collecte d’échantillons s’est effectuée dans 43 épiceries (309 cuisses et pilons de poulet) et dans deux abattoirs (échantillons nasaux et fécaux de 200 poulets) de la Montérégie. La prévalence de SARM a été évaluée à 1.29% (IC 95%: 0.35-3.28) et 0% dans la viande et les oiseaux respectivement. Les isolats testés se sont révélés résistants aux bêta-lactamines (n=15), à la tétracycline (n=10), à l’oxytétracycline (n=10), à la spectinomycine (n=10) et à la tobramycine (n=1). Le typage a révélé deux clones différents (ST398-V, n=10; et ST8-IVa ’USA300’, n=5). La présence de gènes de résistance aux antibiotiques (blaZ, blaR, blaI, erm(A), lnu(A), aad(D), fosB, tet(K), tet(L) et spc) ainsi que plusieurs gènes codant pour l’évasion du système immunitaire (IEC), la production de toxines ou encore pour la production de biofilm ont aussi été détectés. Une forte production de biofilm a été observée pour la majorité des isolats (n=11) à l’exception de certains isolats ST398. Le taux d’expression du système Agr n’a révélé aucune différence particulière entre les SARM testés. Pour conclure, nos données indiquent une faible prévalence de SARM chez la volaille et la viande de poulet. Les isolats ont été catégorisés en deux génotypes, dont un portant plus de gènes de résistance aux antibiotiques (ST398) et l’autre possédant plus de gènes de virulence (ST8).

Background: Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most important pathogens in the swine industry and causes important economic losses. No effective antiviral drugs against it are commercially available. We recently reported that the culture supernatant of Actinobacillus pleuropneumoniae, the porcine pleuropneumonia causative agent, has an antiviral activity in vitro against PRRSV in SJPL cells. Objectives of this study were (i) to identify the mechanism behind the antiviral activity displayed by A. pleuropneumoniae and (ii) to characterize the active molecules present in the bacterial culture supernatant. Methods: Antibody microarray analysis was used in order to point out cellular pathways modulated by the A. pleuropneumoniae supernatant. Subsequent, flow cytometry analysis and cell cycle inhibitors were used to confirm antibody microarray data and to link them to the antiviral activity of the A. pleuropneumoniae supernatant. Finally, A. pleuropneumoniae supernatant characterization was partially achieved using mass spectrometry. Results: Using antibody microarray, we observed modulations in G2/M-phase cell cycle regulation pathway when SJPL cells were treated with A. pleuropneumoniae culture supernatant. These modulations were confirmed by a cell cycle arrest at the G2/M-phase when cells were treated with the A. pleuropneumoniae culture supernatant. Furthermore, two G2/M-phase cell cycle inhibitors demonstrated the ability to inhibit PRRSV infection, indicating a potential key role for PRRSV infection. Finally, mass spectrometry lead to identify two molecules (m/z 515.2 and m/z 663.6) present only in the culture supernatant. Conclusions: We demonstrated for the first time that A. pleuropneumoniae is able to disrupt SJPL cell cycle resulting in inhibitory activity against PRRSV. Furthermore, two putative molecules were identified from the culture supernatant. This study highlighted the cell cycle importance for PRRSV and will allow the development of new prophylactic or therapeutic approaches against PRRSV.

En Amérique du Nord, les pratiques actuelles de conception de l’industrie de la construction suscitent de nombreux et importants problèmes, tant au niveau environnemental, que social et économique. Certaines tendances conceptuelles, telles que l’industrialisation de la construction, le bâtiment vert, l’approche diachronique et la rationalisation spatiale proposent des moyens afin de répondre à cette problématique. La conception synchronique, qui ne se base encore que sur des besoins immédiats, produit des bâtiments statiques qui ne peuvent accommoder l’évolution des besoins. En plus de la phase d’usage qui se révèle la plus coûteuse en termes de consommation d’énergie, d’eau, de matériaux et de génération de déchets, le bâtiment statique est amené à subir de nombreuses rénovations, profondément délétères pour l’environnement et la situation économique des ménages. Dans une perspective d’opérationnalisation du développement durable, la problématique de recherche concerne la considération de la phase d’usage dans le processus conceptuel de l’habitation. Dans l’optique de favoriser une conception plutôt diachronique, 55 projets de rénovation résidentiels ont été analysés et comparés afin de décrire la nature et l’intensité des interventions effectuées. L’examen des plans avant/après rénovation a permis de quantifier le niveau d’intensité des interventions et ainsi, mettre en relief certaines tendances et récurrences des pratiques actuelles. Dans le but de valider si le bâtiment vert est en mesure de répondre aux enjeux environnementaux contemporains de l’habitation, les contenus de trois programmes de certification utilisés en Amérique du Nord, à savoir BOMA-BESt®, LEED®, et Living Building ChallengeTM, sont analysés. Une classification des stratégies proposées montre la répartition des préoccupations envers le bâtiment, les systèmes et l’usager. Une analyse permet de mettre en relief que les considérations semblent, de façon générale, cohérentes avec les impacts associés à l’habitation. Un examen plus ciblé sur la teneur et les objectifs des stratégies considérant l’usager permet l’émergence de deux ensembles thématiques : les cinq sources potentielles d’impacts (énergie, eau, matériaux, déchets et environnement intérieur), ainsi que les quatre thèmes reliés aux objectifs et aux finalités des stratégies (entretien et surveillance; sensibilisation et formation; modélisation et mesurage; comportement et habitude). La discussion a permis d’émettre des pistes d’écoconception pour permettre à l’habitation d’accommoder l’évolution des besoins des occupants, à savoir la « démontabilité », l’aptitude à l’évolution et l’accessibilité. Des recommandations, telles que la nécessité de mieux prendre en compte l’usager comme facteur d’influence des impacts occasionnés par la phase d’usage, sont ensuite proposées. D’autres suggestions appellent à une concrétisation de la performance en suscitant l’implication, l’engagement, la responsabilisation et l’autonomisation des occupants. Enfin, il semble que certaines modifications des programmes de certification et de la législation en vigueur pourraient favoriser l’émergence d’une vision nouvelle concernant la nécessaire réduction de la superficie habitable. Toutes ces opportunités d’amélioration ont le potentiel de mener ce secteur vers une démarche plus globale et systémique, tout en bonifiant de façon majeure les implications environnementales, économiques et sociales de l’habitation.

De l'ouvrage richement illustré de Lorraine Camerlain et Diane Pavlovic, sur cent ans de théâtre québécois, aux expositions scénographiques de Mario Bouchard et de l'APASQ, en passant par les numéros de revues spécialisées consacrés à la scénographie et jusqu'à la présente publication, on n'arrête pas de redécouvrir l'image au théâtre, dans ce qu'elle est - un ensemble de signes scéniques - comme dans sa conjoncture.

The difficulties arising in the calculation of the nuclear curvature energy are analyzed in detail, especially with reference to relativistic models. It is underlined that the implicit dependence on curvature of the quantal wave functions is directly accessible only in a semiclassical framework. It is shown that also in the relativistic models quantal and semiclassical calculations of the curvature energy are in good agreement.

The recently developed variational Wigner-Kirkwood approach is extended to the relativistic mean field theory for finite nuclei. A numerical application to the calculation of the surface energy coefficient in semi-infinite nuclear matter is presented. The new method is contrasted with the standard density functional theory and the fully quantal approach.
