In standard cylindrical gradient coils consisting of a single layer of wires, a limiting factor in achieving very large magnetic field gradients is the rapid increase in coil resistance with efficiency. This is a particular problem in small-bore scanners, such as those used for MR microscopy. By adopting a multi-layer design in which the coil wires are allowed to spread out into multiple layers wound at increasing radii, a more favourable scaling of resistance with efficiency is achieved, thus allowing the design of more powerful gradient coils with acceptable resistance values. Previously this approach has been applied to the design of unshielded, longitudinal, and transverse gradient coils. Here, the multi-layer approach has been extended to allow the design of actively shielded multi-layer gradient coils, and also to produce coils exhibiting enhanced cooling characteristics. An iterative approach to modelling the steady-state temperature distribution within the coil has also been developed. Results indicate that a good level of screening can be achieved in multi-layer coils, that small versions of such coils can yield higher efficiencies at fixed resistance than conventional two-layer (primary and screen) coils, and that performance improves as the number of layers of increases. Simulations show that by optimising multi-layer coils for cooling it is possible to achieve significantly higher gradient strengths at a fixed maximum operating temperature. A four-layer coil of 8 mm inner diameter has been constructed and used to test the steady-state temperature model. (C) 2003 Elsevier Inc. All rights reserved.

The National Health and Medical Research Council, Research Agenda Working Group (RAWG), and the literature on Indigenous health have identified the need to fill gaps in descriptive data on Aboriginal and Torres Strait Islander health and noted both the lack of research with urban populations and the need for longitudinal studies. This paper presents some of the broad ethical and methodological challenges associated with longitudinal research in Indigenous health and focuses particularly on national studies and studies in urban areas. Our goal is to advance debate in the public health arena about the application of ethical guidelines and the conduct of longitudinal studies in Aboriginal and Torres Strait Islander communities. We encourage others to offer their experiences in this field.

The BRN2 transcription factor (POU3F2, N-Oct-3) has been implicated in development of the melanocytic lineage and in melanoma. Using a low calcium medium supplemented with stem cell factor, fibroblast growth factor-2, endothelin-3 and cholera toxin, we have established and partially characterised human melanocyte precursor cells, which are unpigmented, contain immature melanosomes and lack L-dihydroxyphenylalanine reactivity. Melanoblast cultures expressed high levels of BRN2 compared to melanocytes, which decreased to a level similar to that of melanocytes when cultured in medium that contained phorbol ester but lacked endothelin-3, stem cell factor and fibroblast growth factor-2. This decrease in BRN2 accompanied a positive L-dihydroxyphenylalanine reaction and induction of melanosome maturation consistent with melanoblast differentiation seen during development. Culture of primary melanocytes in low calcium medium supplemented with stem cell factor, fibroblast growth factor-2 and endothelin-3 caused an increase in BRN2 protein levels with a concomitant change to a melanoblast-like morphology. Synergism between any two of these growth factors was required for BRN2 protein induction, whereas all three factors were required to alter melanocyte morphology and for maximal BRN2 protein expression. These finding implicate BRN2 as an early marker of melanoblasts that may contribute to the hierarchy of melanocytic gene control.

Objective: To explore relationships between physical activity and mental health cross-sectionally and longitudinally in a large cohort of older Australian women. Method: Women in their 70s participating in the Australian Longitudinal Study on Women's Health responded in 1996 (aged 70-75) and in 1999 (aged 73-78). Cross-sectional data were analyzed for 10,063 women and longitudinal data for 6472. Self-reports were used to categorize women into four categories of physical activity at each time point as well as to define four physical activity transition categories across the 3-year period. Outcome variables for the cross-sectional analyses were the mental health component score (MCS) and mental health subscales of the Medical Outcomes Study Short Form (SF-36). The longitudinal analyses focused on changes in these variables. Confounders included the physical health component scale (PCS) of the SF-36, marital status, body mass index (BMI) and life events. Adjustment for baseline scores was included for the longitudinal analyses. Results: Cross-sectionally, higher levels of physical activity were associated with higher scores on all dependent variables, both with and without adjustment for confounders. Longitudinally, the effects were weaker, but women who had made a transition from some physical activity to none generally showed more negative changes in emotional well-being than those who had always been sedentary, while those who maintained or adopted physical activity had better outcomes. Conclusion: Physical activity is associated with emotional well-being among a population cohort of older women both cross-sectionally and longitudinally, supporting the need for the promotion of appropriate physical activity in this age group. Background Latent left ventricular (LV) dysfunction in patients with valvular or myocardial disease may be identified by loss of contractile reserve (CR) at exercise echocardiography. Contraction in the LV longitudinal axis may be more sensitive than radial contraction to minor disturbances of LV function. We sought to determine whether tissue Doppler measurement of longitudinal function could be used to identify CR. Methods Exercise echocardiography was performed in 86 patients (20 women, age 53 +/- 18 years), 72 with asymptomatic or minimally symptomatic mitral regurgitation, and 14 normal controls. Pulsed-wave tissue Doppler imaging (DTI) was used to measure maximum annular systolic velocity at rest and stress. Inducible ischemia was excluded by analysis of wall motion by an experienced observer. CR was defined by greater than or equal to5% improvement of stress compared with rest ejection fraction (EF). Exercise capacity was assessed from expired gas analysis. As survival of patients with CF increases,glucose intolerance and cystic fibrosisrelated diabetes (CFRD),ar e increasingly recognised common complications. CFRD may be preceded by a pre-diabetic state. Using markers identified as being associated with CFRD may improve targeted screening. Aim: To identify features consistently predicting CFRD in paediatric patients. Patients diagnosed with CFRD between January 1997–January 2002 were compared with age and sex matched controls. Clinical,micr obiological, and hospitalisation data was collected at time of CFRD diagnosis,and at six monthly intervals for 3 yr prior to diagnosis. Eight patients with CFRD were identified,mean age 13.7 yr (S.D. 3.49) at time of diagnosis. Control patients underwent OGTT to ensure normal glucose tolerance. Patients with CFRD had a lower FEV1 up to 12 months prior to diagnosis however, this was only significant at diagnosis. There was no difference in weight and height z scores between the 2 groups; however,the decrease in weight and height z scores in the CFRD group over 3 yr prior to diagnosis was significant. Mean number of days in hospital and admissions per patient significantly increased in the CFRD group,6 months prior to diagnosis. No other significant differences were observed between the 2 groups. Conclusions: This study has shown a difference in lung function,gr owth parameters and frequency of hospital admissions between patients with CFRD and controls. These differences may be utilised as tools for targeted screening in the paediatricyadolescent population. Further larger scale studies are required to improve guidelines for targeted screening in this population.

In order to develop a method for use in investigations of spatial biomass distribution in solid-state fermentation systems, confocal scanning laser microscopy was used to determine the concentrations of aerial and penetrative biomass against height and depth above and below the substrate surface, during growth of Rhizopus oligosporus on potato dextrose agar. Penetrative hyphae had penetrated to a depth of 0.445 cm by 64 h and showed rhizoid morphology, in which the maximum biomass concentration, of 4.45 mg dry wt cm(-3), occurred at a depth of 0.075 cm. For aerial biomass the maximum density of 39.54 mg dry wt(-3) occurred at the substrate surface. For both aerial and penetrative biomass, there were two distinct regions in which the biomass concentration decayed exponentially with distance from the surface. For aerial biomass, the first exponential decay region was up to 0.1 cm height. The second region above the height of 0.1 cm corresponded to that in which sporangiophores dominated. This work lays the foundation for deeper studies into what controls the growth of fungal hyphae above and below the surfaces of solid substrates. (C) Wiley Periodicals, Inc.

Two methods were compared for determining the concentration of penetrative biomass during growth of Rhizopus oligosporus on an artificial solid substrate consisting of an inert gel and starch as the sole source of carbon and energy. The first method was based on the use of a hand microtome to make sections of approximately 0.2- to 0.4-mm thickness parallel to the substrate surface and the determination of the glucosamine content in each slice. Use of glucosamine measurements to estimate biomass concentrations was shown to be problematic due to the large variations in glucosamine content with mycelial age. The second method was a novel method based on the use of confocal scanning laser microscopy to estimate the fractional volume occupied by the biomass. Although it is not simple to translate fractional volumes into dry weights of hyphae due to the lack of experimentally determined conversion factors, measurement of the fractional volumes in themselves is useful for characterizing fungal penetration into the substrate. Growth of penetrative biomass in the artificial model substrate showed two forms of growth with an indistinct mass in the region close to the substrate surface and a few hyphae penetrating perpendicularly to the surface in regions further away from the substrate surface. The biomass profiles against depth obtained from the confocal microscopy showed two linear regions on log-linear plots, which are possibly related to different oxygen availability at different depths within the substrate. Confocal microscopy has the potential to be a powerful tool in the investigation of fungal growth mechanisms in solid-state fermentation. (C) 2003 Wiley Periodicals, Inc.

This paper describes a process-based metapopulation dynamics and phenology model of prickly acacia, Acacia nilotica, an invasive alien species in Australia. The model, SPAnDX, describes the interactions between riparian and upland sub-populations of A. nilotica within livestock paddocks, including the effects of extrinsic factors such as temperature, soil moisture availability and atmospheric concentrations of carbon dioxide. The model includes the effects of management events such as changing the livestock species or stocking rate, applying fire, and herbicide application. The predicted population behaviour of A. nilotica was sensitive to climate. Using 35 years daily weather datasets for five representative sites spanning the range of conditions that A. nilotica is found in Australia, the model predicted biomass levels that closely accord with expected values at each site. SPAnDX can be used as a decision-support tool in integrated weed management, and to explore the sensitivity of cultural management practices to climate change throughout the range of A. nilotica. The cohort-based DYMEX modelling package used to build and run SPAnDX provided several advantages over more traditional population modelling approaches (e.g. an appropriate specific formalism (discrete time, cohort-based, process-oriented), user-friendly graphical environment, extensible library of reusable components, and useful and flexible input/output support framework). Nodal shoot cultures of 'Clone 003', a selected Australian papaya cultivar, were cultured on modified De Fossard medium supplemented with chemicals that either promote ethylene evolution or inhibit action while in culture. Nodal shoot cultures grown in the presence of 1-aminocyclopropane carboxylic acid (ACC, 1.0 mM) resulted in a significant reduction in percent fresh and dry weights, shoot length, leaf area, petiole length and chlorophyll content, but leaf development was significantly increased. In contrast, nodal cultures grown in the presence of silver thiosulphate (STS, 0.5 mM) significantly produced the highest percentage of fresh and dry weights, shoot length, leaf production, leaf area expansion, petiole length and leaf chlorophyll content. Nodal cultures and rooted whole plantlets placed in medium-sized (125 mL) culture vessels had significantly better growth than those cultures placed in small (70 mL) or in large (250 mL) vessels. Cultures grown in medium-sized vessels had higher fresh and dry weights, longer shoots, more leaves and larger leaf area than those cultures placed in smaller or larger vessels. Similarly, values for said growth parameters and for chlorophyll content of the nodal and rooted whole plantlets were higher when they were incubated under high light intensity of 120 mumol m(-2)s(-1) at a prevailing temperature of either 20+/-1 C or 25+/-1 C.

O objetivo deste estudo foi estimar a cronologia e sequência de erupção da dentição decídua e seus fatores relacionados em amostra de crianças de duas regiões do município de Vitória, ES. Os dados utilizados no estudo são provenientes de um estudo longitudinal realizado entre 2003 e 2006 com 86 recém-nascidos que foram acompanhados até a idade de 36 meses de vida, cuja coleta de dados foi obtida por meio da aplicação de um formulário as mães e da realização de um exame clínico nas crianças. Um total de 67 crianças permaneceram até o final do estudo. Calculou-se a idade média de erupção dos dentes decíduos de cada criança e foram aplicados o teste de kappa, McNemar e kappa ajustado pela prevalência. Em seguida realizou-se a Análise de Sobrevivência. Os resultados mostraram que a média de erupção dos dentes decíduos variou de oito a 29 meses de vida no arco inferior, e de 11 a 30 meses no arco superior e que os maiores níveis de concordância foram para os tempos de erupção dos incisivos e caninos decíduos (71/81, kappa = 0,82; IC95% = 0,72-0,93; 53/63, kappa = 0,76; IC95% = 0,62-0,88) do que para os molares decíduos. Dos fatores relacionados a cronologia de erupção da dentição decídua, foi identificado na Regressão de Cox que os hábitos alimentares infantis podem influenciar, acelerando e retardando esse processo eruptivo. Recomenda-se o conhecimento do perfil de erupção decídua de cada população para que tais evidências sirvam de base para a implementação de medidas de prevenção e controle da saúde dessa população e auxilie na elaboração de estratégias, com ações de proteção e promoção da saúde. As ações tem como finalidade a prevenção de possíveis alterações bucais e gerais durante o crescimento e desenvolvimento infantil e melhoria da qualidade de vida dessa população.

A acumulação de tecido adiposo abdominal apresenta associação positiva com eventos cardiovasculares, pressão arterial e alterações metabólicas. Dentre os fatores de risco para o aumento da obesidade abdominal está o alto consumo de bebidas alcoólicas, particularmente a cerveja. O objetivo deste estudo foi identificar associação entre consumo de bebidas alcoólicas (CBA) e adiposidade abdominal. Trata-se de uma investigação de corte transversal conduzida a partir da linha de base do Estudo Longitudinal de Saúde do Adulto – ELSA-Brasil, composta por 15.105 indivíduos (35 a 74 anos). Foram analisadas variáveis antropométricas, socioeconômicas e consumo de bebidas alcoólicas e utilizados, para diagnóstico de obesidade abdominal, os pontos de corte da circunferência da cintura (CC) e relação cintura/quadril (RCQ) preconizados pela Organização Mundial de Saúde. O CBA foi categorizado em quintis. Teste de Kolmogorov-Smirnov foi utilizado para avaliar a normalidade das variáveis. A associação entre variáveis antropométricas e o CBA foi avaliada utilizando-se teste Mann-Whitney, Kruskal-Wallis e teste qui-quadrado. Foram testados modelos de regressão linear e Poisson, ajustados por idade, sexo, IMC, tabagismo, atividade física, renda e escolaridade. A CC inadequada foi associada a maior CBA em toda amostra (1,03, IC95% 1,01-1,05) e em homens (1,05, IC95% 1,03-1,08). A RCQ inadequada foi associada a maior CBA tanto para o total da amostra (1,04, IC95% 1,01-1,06) como para mulheres (1,07, IC95% 1,03-1,12). Homens no quinto quintil de consumo de cerveja apresentaram chance 1,05 maior (IC95% 1,02-1,08) de ter a CC inadequada quando comparados aos que se encontravam no primeiro quintil. Já entre as mulheres a chance foi 1,16 (IC95% 1,13-1,20). Homens e mulheres no quinto quintil de consumo de cerveja tinham, respectivamente, 1,03 (IC95% 1,00-1,07) e 1,10 (IC95%1,04-1,15) vezes mais chance de apresentar RCQ inadequada. O consumo de vinho só foi associado a maior chance de ter CC aumentada entre mulheres (β=0,026, p<0,027). Neste estudo, o consumo de álcool foi associado positivamente com obesidade abdominal, sendo mais importante a contribuição da cerveja para aumento da CC e da RCQ.

Em crianças usuárias de Implante Coclear (IC), o registro do potencial de ação composto do nervo auditivo evocado eletricamente (ECAP) representa uma maneira de avaliar as modificações nas respostas neurais e entre o feixe de eletrodos e o tecido neural ao longo do tempo. OBJETIVO: Estudar o ECAP em crianças ao longo do primeiro ano de uso do IC. MATERIAL E MÉTODO: As características do ECAP foram analisadas em 13 crianças, implantadas com idades inferiores a três anos de idade. Estudo de série. RESULTADOS: Houve aumento estatisticamente significante na amplitude do pico N1, nos eletrodos basais, entre o segundo e o terceiro retorno. Não foram obtidas diferenças significantes para: a latência do pico N1, o slope, o p-NRT e o período de recuperação entre os retornos. Nos três retornos, a maioria dos sujeitos apresentou tempo de recuperação entre 1000 e 2000µs. CONCLUSÃO: Ao longo do primeiro ano de uso do IC, a estimulação elétrica liberada pelos eletrodos intracocleares não causou alterações significativas às características do ECAP, exceto pelo aumento da amplitude do pico N1.

This study was carried out to investigate the effects of light spectra, additional UV-A, and different growth regulators on the in vitro germination of Senecio cineraria DC. Seeds were surface-sterilized and inoculated in MS medium to evaluate the following light spectra: white, white plus UV-A, blue, green, red or darkness. The maximum germinability was obtained using MS0 medium under white light (30%) and MS + 0.3 mg L-1 GA3 in the absence of light (30.5%). S. cineraria seeds were indifferent to light. Blue and green lights inhibited germination. Different concentrations of gibberellic acid (GA3) (0.1; 0.4; 0.6; 0.8; 1.0 and 2.0 mg L-1) and indole-3-acetic acid IAA (0.1; 0.3 and 1.0 mg L-1) were evaluated under white light and darkness. No concentration of GA3 enhanced seed germination percentage under white light. However, when the seeds were maintained in darkness, GA3 improved germination responses in all tested concentrations, except at 1.0 mg L-1. Under white light, these concentrations also increased the germination time and reduced germination rate. Germination rate, under light or darkness, was lower using IAA compared with GA3.
