This study aimed to verify that chicks from eggs injected with ascorbic acid and subjected to heat stress would have changes in acid-base balance, compared to chicks incubated at thermoneutral without injection of ascorbic acid. The parameters evaluated were blood pressure of carbon dioxide and oxygen, base excess, total carbon dioxide, concentration of sodium, potassium, ionized calcium, bicarbonate and pH of newly hatched male chicks, hatched from eggs injected with acid ascorbic acid (AA) and subjected to heat stress during incubation. The experimental design was completely randomized in factorial scheme 5 (application levels of ascorbic acid) x 2 (incubation temperatures). The data were subjected to analysis of variance using the General Linear Model procedure (GLM) of SAS ®. For the blood pH was observed significant interaction (p <0.05) between treatments with application in eggs and incubation temperatures. For the other parameters were not significant effects (p< 0.05) of AA level and neither temperature of incubation. Analyzing the unfolding of the interaction to pH was observed that chicks from eggs injected with 6% ascorbic acid and subjected to heat stress during incubation had a higher pH value compared with the thermoneutral temperature incubated (p <0.05). Therefore, it is suggested that the incubation of eggs in high temperature (39°C) can alter the metabolic rate of these embryos.

The red gum lerp psyllid Glycaspis brimblecombei Moore (Hemiptera: Psyllidae) was first recorded in Brazil in 2003 and since then has caused serious damage in Eucalyptus forests. The damage is mainly due to defoliation, sooty mold and dieback, thus requiring the development of efficient management strategies. The settling and ovipositional preference of G. brimblecombei by species and clones of Eucalyptus were evaluated in free- and no-choice tests. Preliminary tests were done to determine the density of psyllid couples that produced the greatest densities of eggs, leaf position selection for oviposition, and whether oviposition was affected by preimaginal conditioning. Apical leaves were used more frequently for oviposition by the psyllid, and the number of eggs decreased from the apex to the base. The densities of 10 and 15 psyllid couples per seedling produced the highest oviposition. There was no evidence of preimaginal conditioning. In the free-choice test, E. grandis, E. urophylla, VM-1, I-144, C-219 and H-13 were less attractive to G. brimblecombei adults, with H-13, E. grandis and E. urophylla having the least oviposition. In the no-choice test, E. urophylla, GG-100 and E. grandis also demonstrated a lower oviposition, but E. camaldulensis and 3025 were highly susceptible. The low preference for the genotypes E. grandis and E. urophylla suggests the occurrence of a non-preference type resistance against the red gum lerp psyllid. Our results can be utilized for the development of management programs for G. brimblecombei in Eucalyptus forests. © 2012 Springer Science + Business Media B.V.

This work evaluated the effect of triiodothyronine (T3) on larviculture of matrinxã, Brycon amazonicus. Oocytes of three females were pooled, fertilized with pooled semen of two males and separated in four batches that were immersed in triiodothyronine solutions as follows: M1 (control - water); M2 (0.01mg/L T3); M3 (0.05mg/L T3); and M4 (0.1mg/L T3). Triiodothyronine did not affect fertilization rate and number of hatched larvae. Weight of hatched larvae was significantly higher in treatments M3 and M4, as well as among larvae sampled at Day 12 in all treatments. After 12d of rearing, biomass gain was higher in the hormone treatments (M1 688±569mg; M2 2436±562mg; M3 3572±569mg; and M4 4129±770mg). In general, coefficients of variation of weight (CVw) and length (CVl) did not differ among treatments and cannibalism was registered between 36 and 72hours post-hatching (h.p.h.) without differences among treatments. Larval survival increased in the hormone treatments (M1 26.5%; M2 37.6%; M3 40.6%; and M4 40.8%). The results indicate that the immersion of matrinxã eggs in triiodothyronine can promote beneficial effects to its larviculture and indicate promising perspectives for culture of this tropical species. © by the World Aquaculture Society 2013.

Background:Hepatitis C is a disease spread throughout the world. Hepatitis C virus (HCV), the etiological agent of this disease, is a single-stranded positive RNA virus. Its genome encodes a single precursor protein that yields ten proteins after processing. NS5A, one of the non-structural viral proteins, is most associated with interferon-based therapy response, the approved treatment for hepatitis C in Brazil. HCV has a high mutation rate and therefore high variability, which may be important for evading the immune system and response to therapy. The aim of this study was to analyze the evolution of NS5A quasispecies before, during, and after treatment in patients infected with HCV genotype 3a who presented different therapy responses.Methods:Viral RNA was extracted, cDNA was synthesized, the NS5A region was amplified and cloned, and 15 clones from each time-point were sequenced. The sequences were analyzed for evolutionary history, genetic diversity and selection.Results:This analysis shows that the viral population that persists after treatment for most non-responder patients is present in before-treatment samples, suggesting it is adapted to evade treatment. In contrast, the population found in before treatment samples from most end-of-treatment responder patients either are selected out or appears in low frequency after relapse, therefore changing the population structure. The exceptions illustrate the uniqueness of the evolutionary process, and therefore the treatment resistance process, in each patient.Conclusion:Although evolutionary behavior throughout treatment showed that each patient presented different population dynamics unrelated to therapy outcome, it seems that the viral population from non-responders that resists the treatment already had strains that could evade therapy before it started. © 2013 Bittar et al.

Specimens (n= 41) of the amphisbaenid Amphisbaena wuchereri taken from a population in Minas Gerais state, south-eastern Brazil, were examined for gastrointestinal parasites. A single nematode species was found, Paradollfusnema amphisbaenia. This was a new host record for this nematode species. This parasite was encountered in the large intestine (prevalence of 100%), in the stomach (prevalence of 2%) and in the small intestine (prevalence of 7.3%). The intensity of infection ranged from 1 to 457 individual parasites per host and was positively correlated with body size of both male and female amphisbaenians. The discrepancy index (D) indicated that P. amphisbaenia tended to an even distribution in this host population. The nematode, which did not affect fat body mass, induced inflammatory infiltrations in the small intestine, indicating that the parasites might injure the host's organs. Free amino acids (FAA) are used principally as substrate in protein synthesis and the source of energy in aerobic catabolism. In marine fish, embryo and larvae FAA are used to maintain body fluid osmolality during fish early development. However, there is essentially no information about FAA concentrations in early ontogeny of freshwater neotropical species in comparison to marine fishes. Therefore, the aim of this study was to evaluate the FAA concentrations in pacu, Piaractus mesopotamicus, eggs and larvae. Broodstock fish were induced to spawn and ovulated females were stripped of their eggs and immediately sampled for analysis. Larvae were sampled right after hatching (HL) and after the completion of the yolk-sac absorption (YSA). The wet weight of the HL and YSA larvae amounted to 0.5±0.1mg and 1.1±0.3mg, respectively. HL larvae showed higher levels of most of the indispensable amino acids (IAA) in comparison to eggs and YSA larvae. Exceptions were observed with His and Trp that showed higher or similar levels, respectively, in YSA larvae. The FAA Orn, Tau, Glu, Gln, Gly, and Tyr increased concentrations in both larval stages while that of Tau was found in higher concentration in all analyzed stages. Also, the concentrations of Asn, Ala, Pro, Ser, and Asp were higher in HL larvae. Both larval stages displayed a rise in total free IAA/total free DAA (dispensable amino acids) ratio. The authors conclude that the highest level of FAA in HL pacu larvae is indicative of active proteolysis of yolk reserves and a probable catabolism regulation of some FAA through spare-effect. In addition, Tau is one of the major FAA occurring during pacu ontogeny and may be performing regulation on body fluid osmolality regulation. © Copyright by the World Aquaculture Society 2013.

Telenomus remus Nixon Egg Parasitization of Three Species of Spodoptera Under Different Temperatures

Telenomus remus Nixon is a promising biocontrol agent as an egg parasitoid of Spodoptera spp., but the lack of information on the host-parasitoid interactions in this system precludes its applied use in agriculture. Therefore, we studied the parasitism capacity of T. remus on eggs of Spodoptera cosmioides (Walker), Spodoptera eridania (Cramer), and Spodoptera frugiperda (Smith) in a range of temperatures (19, 22, 25, 28, 31, and 34 ± 1°C) under controlled conditions (70 ± 10% RH and 12 h photophase). Egg masses of Spodoptera spp. were offered to a single-mated T. remus female on a daily basis. More than 80% lifetime parasitism on eggs of S. cosmioides, S. frugiperda, and S. eridania was reached from 1 to 5, 1 to 7, and 1 to 9 days, respectively, at temperatures from 19 to 34°C. More than 80% parasitization was obtained at extreme temperatures for all hosts studied. Lifetime parasitization of S. frugiperda, S. cosmioides, and S. eridania was affected by temperature, with the lowest values for S. frugiperda (34°C) and S. cosmioides (19 and 34°C). Parasitization of S. eridania eggs was reduced around 18% at 28 and 31°C, but dropped more severely at 34°C. Parasitoid longevity was reduced as temperature increased. Thus, our data indicated that T. remus might be suitable as a biocontrol agent against S. eridania, S. cosmioides, and S. frugiperda in geographical areas that fit the temperature range studied here, even though T. remus parasitism was reduced at 34°C. © 2013 Sociedade Entomológica do Brasil.

Feline immunodeficiency virus (FIV) infection has been the focus of several studies because this virus exhibits genetic and pathogenic characteristics that are similar to those of the human immunodeficiency virus (HIV). FIV causes acquired immunodeficiency syndrome (AIDS) in cats, nevertheless, a large fraction of infected cats remain asymptomatic throughout life despite of persistent chronic infection. This slow disease progression may be due to the presence of factors that are involved in the natural resistance to infection and the immune response that is mounted by the animals, as well as due to the adaptation of the virus to the host. Therefore, the study of virus-host interaction is essential to the understanding of the different patterns of disease course and the virus persistence in the host, and to help with the development of effective vaccines and perhaps the cure of FIV and HIV infections. © 2013 Elsevier Ltd. Currently, schistosomiasis mansoni is treated clinically with praziquantel (PZQ). Nevertheless, cases of tolerance and resistance to this drug have been reported, creating the need to develop new drugs or to improve existing drugs. Considering the small number of new drugs against Schistosoma mansoni, the design of nanotechnology-based drug delivery systems is an important strategy in combating this disease. The aim of this study was to evaluate the activity of PZQ containing liposome (lip.PZQ) on S. mansoni, BH strain. Mice were treated orally with different concentrations of PZQ and lip.PZQ 30 and 45 days following infection. The number of worms, recovered by perfusion of the hepatic portal system, and the number of eggs found in the intestine and liver were analysed. Parasite egg counts were also performed. The most active formulation for all parameters was 300. mg/kg of lip.PZQ, since as it decreased the total number of worms by 68.8%, the number of eggs in the intestine by 79%, and the number of hepatic granulomas by 98.4% compared to untreated controls. In addition, this concentration decreased egg counts by 55.5%. The improved efficacy of the treatment with lip.PZQ, especially when administered 45 days following infection, compared with the positive-control group (untreated) and the groups that received free PZQ, can be explained by greater bioavailability in the host organism; the preferred target of lip.PZQ is the liver, and lip.PZQ is better absorbed by the tegument of S. mansoni, which has an affinity for phospholipids. © 2013 Elsevier B.V.

The aim of the present study was to analyse the genetic and pathogenic variability of Colletotrichum spp. isolates from various organs and cultivars of mango with anthracnose symptoms, collected from different municipalities of São Paulo State, Brazil. Colletotrichum gloeosporioides isolates from symptomless citrus leaves and C. acutatum isolates from citrus flowers with post-bloom fruit drop symptoms were included as controls. Sequencing of the ITS region allowed the identification of 183 C. gloeosporioides isolates from mango; only one isolate was identified as C. acutatum. amova analysis of ITS sequences showed larger genetic variability among isolates from the same municipality than among those from different populations. fAFLP markers indicated high levels of genetic variability among the C. gloeosporioides isolates from mango and no correlation between genetic variability and isolate source. Only one C. gloeosporioides mango isolate had the same genotype as the C. gloeosporioides isolates from citrus leaves, as determined by ITS sequencing and fAFLP analysis. Pathogenicity tests revealed that C. gloeosporioides and C. acutatum isolates from either mango or citrus can cause anthracnose symptoms on leaves of mango cvs Palmer and Tommy Atkins and blossom blight symptoms in citrus flowers. These outcomes indicate a lack of host specificity of the Colletotrichum species and suggest the possibility of host migration. © 2012 British Society for Plant Pathology.
