The C-phycocyanin and the R-phycoerythrin were purified from the blue-green alga Spirulina platensis and red alga Polysiphonia urceolata respectively. Both sodium periodate and glutaraldehyde are effective coupling agents being capable of constructing the R-phycoerythrin-C-phycocyanin conjugate, which was also called phycobiliproteins energy transfer model. The two artificial conjugates constructed with different methods were purified by Sephadex G-200 chromatography respectively. Spectra analysis indicated that energy transfer occurred in the two conjugates. The conjugate with sodium periodate had the higher efficiency of energy transfer than that with glutaraldehyde conjugate.

B-phycoerythrin (BPE) and R-phycocyanin (RPC) were purified from Porphyridium cruentum by Sephadex G-200 chromatography, then the BPE was attached covalently to the RPC by reacting their amino groups to form the artificially covalent BPE-RPC conjugate in which the excitation energy can transfer from the BPE to the RPC with low efficiency. Meanwhile, the intact phycobilisome (PBS) consisting of BPE, RPC, APC and L-CM was isolated and purified from Porphyridium cruentum, and the purified PBS was found to keep intact if the solution contains sucrose. Comparison of spectroscopic properties between the purified PBS and the BPE-RPC conjugate suggests that the BPE-RPC conjugate is much more stable than the purified PBS. The construction of BPE-RPC conjugate with low efficiency of the excitation energy transfer may be useful for preparing phycobiliprotein probes. (C) 2002 Elsevier Science Ireland Ltd. Interpretation of high-resolution two-dimensional (2D) and three-dimensional (3D) seismic data collected in the Qiongdongnan Basin, South China Sea reveals the presence of polygonal faults, pockmarks, gas chimneys and slope failure in strata of Pliocene and younger age. The gas chimneys are characterized by low-amplitude reflections, acoustic turbidity and low P-wave velocity indicating fluid expulsion pathways. Coherence time slices show that the polygonal faults are restricted to sediments with moderate-amplitude, continuous reflections. Gas hydrates are identified in seismic data by the presence of bottom simulating reflectors (BSRs), which have high amplitude, reverse polarity and are subparallel to seafloor. Mud diapirism and mounded structures have variable geometry and a great diversity regarding the origin of the fluid and the parent beds. The gas chimneys, mud diapirism, polygonal faults and a seismic facies-change facilitate the upward migration of thermogenic fluids from underlying sediments. Fluids can be temporarily trapped below the gas hydrate stability zone, but fluid advection may cause gas hydrate dissociation and affect the thickness of gas hydrate zone. The fluid accumulation leads to the generation of excess pore fluids that release along faults, forming pockmarks and mud volcanoes on the seafloor. Our analysis of approximately 40,000 km of multichannel 2-D seismic data, reef oil-field seismic data, and data from several boreholes led to the identification of two areas of reef carbonate reservoirs in deepwater areas (water depth >= 500 in) of the Qiongdongnan Basin (QDNB), northern South China Sea. High-resolution sequence stratigraphic analysis revealed that the transgressive and highstand system tracts of the mid-Miocene Meishan Formation in the Beijiao and Ledong-Lingshui Depressions developed reef carbonates. The seismic features of the reef carbonates in these two areas include chaotic bedding, intermittent internal reflections, chaotic or blank reflections, mounded reflections, and apparent amplitude anomalies, similar to the seismic characteristics of the LH11-1 reef reservoir in the Dongsha Uplift and Island Reef of the Salawati Basin, Indonesia, which house large oil fields. The impedance values of reefs in the Beijiao and Ledong-Lingshui Depressions are 8000-9000 g/cc x m/s. Impedance sections reveal that the impedance of the LH11-1 reef reservoir in the northern South China Sea is 800010000 g/cc x m/s, whereas that of pure limestone in BD23-1-1 is > 10000 g/cc x m/s. The mid-Miocene paleogeography of the Beijiao Depression was dominated by offshore and neritic environments, with only part of the southern Beijiao uplift emergent at that time. The input of terrigenous sediments was relatively minor in this area, meaning that terrigenous source areas were insignificant in terms of the Beijiao Depression: reef carbonates were probably widely distributed throughout the depression, as with the Ledong-Lingshui Depression. The combined geological and geophysical data indicate that shelf margin atolls were well developed in the Beijiao Depression, as in the Ledong-Lingshui Depression where small-scale patch or pinnacle reefs developed. The equatorial Pacific upwelling zone has been suspected of playing an important role in the global atmospheric CO2 changes associated with glacial-interglacial cycles. In order to assess the influencing scope of the surface water deglacial delta(13)C minimum in the tropical low-latitude Pacific, the core DGKS9603, collected from the middle Okinawa Trough, was examined for 4513 C records of planktonic foraminifera N. dutertrei and G. ruber. The planktonic foraminiferal delta(13)C records show a clear decreasing event from 20 to 6 cal. kaBP., which is characterized by long duration of about 14 ka and amplitude shift of 0.4 x 10(-3). Its minimum value occurred at 15.7 cal kaBP. The event shows fairly synchrony with the surface water deglacial delta(13)C minimum identified in the tropical Pacific and its marginal seas. Because there is no evidence in planktonic foraminiferal fauna and 45180 records for upwelling and river runoff enhancement, the broad deglacial delta(13)C minimum event in planktonic foraminiferal records revealed in core DGKS9603 might have been the direct influencing result of the deglacial surface water of the tropical Pacific. The identification for the event in the Okinawa Trough provides new evidence that the water evolution in the tropical low-latitude Pacific plays a key role in large regional, even global carbon cycle.

Eolian flux in the Chinese Loess Plateau was reconstructed by measuring the dry bulk density and CaCO3 content of the late Cenozoic loess-paleosol-red clay sequences in the Lingtai profile. Comparison of eolian flux variation between the Lingtai profile and the ODP sites 885/886 in the North Pacific shows a significant wet-dry variability in addition to a gradual drying trend in the dust source regions in interior Asia. Especially, the increase of eolian fluxes from both continental and pelagic eolian sediments indicates a sharp drying of the dust source regions between 3.6 and 2.6 MaBP, which might be attributed to the tectonic uplift of the Tibetan Plateau, which cut down the moisture input to the interior Asia. The average value and variability of eolian flux are higher after 2.6 MaBP than before, which may be related to the Quaternary climatic fluctuations on the glacial-interglacial timescale after the commencement of major Northern Hemisphere Glaciations. The eolian fluxes of the Lingtai profile and Core V21-146 in northwest Pacific show a synchronous variation on the 10(4)-10(5) a timescale, indicating that the flux variations from both continental and marine records are closely correlated to the Quaternary climatic fluctuation forced by the ice volume changes on a global scale.

We here reconstruct the past change of the East Asian monsoon since 20 Ma using samples from Ocean Drilling Program (ODP) Site 1146 in the northern South China Sea based On a multi-proxy approach including a monomineralic quartz isolation procedure, identification of clay minerals by X-ray Diffraction (XRD) and grain-size analysis of isolated terrigenous materials. Terrigenous supply to ODP Site 1146 was dominated by changes in the strength of multiple sources and transport processes. Grain-size data modeled by an end-member modeling algorithm indicate that eolian dust from the and Asian inland and fluvial input have contributed on average 20% and 80% of total terrigenous material to ODP Site 1146, respectively. Specifically, about 40-53% of the total (quartz+feldspar) and only 6-11% of the total clay is related to eolian supply at the study site. Detailed analysis of the sedimentary environment, and clay minerals combined with previous studies shows that smectite originates mainly from Luzon, kaolinite from the Pearl River and illite and chlorite from the Pearl River, Taiwan and/or the Yangtze River. The proportion and mass accumulation rate (MAR) of the coarsest end-member EM1 (interpreted as eolian dust), ratios of (illite+chlorite)/smectite, (quartz+feldspar)% and mean grain-size of terrigenous materials at ODP Site 1146 were adopted as proxies for East Asian monsoon evolution. The consistent variation of these independent proxies since 20 Ma shows three profound shifts in the intensity of East Asian winter monsoon relative to summer monsoon, as well as aridity of the Asian continent, occurred at similar to 15 Ma, similar to 8 Ma and the youngest at about 3 Ma. In comparison, the summer monsoon intensified contemporaneously with the winter monsoon at 3 Ma. The phased uplift of the Himalaya-Tibetan plateau may have played a significant role in strengthening the Asian monsoon at similar to 15 Ma, 8 Ma and 3 Ma. (C) 2007 Elsevier B.V. 273 samples from Ocean Drilling Program (ODP) Site 1146 in the northern South China Sea (SCS) were analyzed for grain-size distributions using grain-size class vs. standard deviation method and end-member modeling algorithm (EMMA) in order to investigate the evolution of the East Asian mon-soon since about 20 Ma. 10-19 mu m/1.3-2.4 mu m, the ratio of two grain-size populations with the highest variability through time was used to indicate East Asian winter monsoon intensity relative to summer monsoon. The mass accumulation rate of the coarsest end member EM1 (eolian), resulting from EMMA, can be used as a proxy of winter monsoon strength and Asian inland aridity, and the ratio of EM1/(EM2+EM3) as a proxy of winter monsoon intensity relative to summer monsoon. The combined proxies show that a profound enhancement of East Asian winter monsoon strength and winter monsoon intensity relative to summer monsoon occurred at about 8 Ma, and it is possible that the summer monsoon simultaneously intensified with winter monsoon at 3 Ma. Our results are well consistent with the previous studies in loess, eolian deposion in the Pacifc, radiolarians and planktonic foraminifera in the SCS. The phased uplift of the Himalaya-Tibetan Plateau may have played a significant role in strengthening the Asian monsoon at 8 Ma and 3 Ma.

128 samples from Ocean Drilling Program (ODP) Site 1143 in the southern South China Sea were analyzed for grain size, clay minerals, biogenic opal content and quartz in order to reconstruct changes in East Asian monsoon climate since 8.5 Ma. An abrupt change of terrigenous mass accumulation rate (MAR), clay mineral assemblage, median grain size and biogenic opal MAR about 5.2 Ma suggests that between 8.5-5.2 Ma the source of terrigenous sediment was mainly in the region of surface uplift and basaltic volcanism in southern Vietnam. A simple model of East Asian summer monsoon evolution was based on the clay/feldspar ratio, kaolinite/chlorite ratio and biogenic opal MAR. The summer monsoon has two periods of maximum strength at 8.5-7.6 Ma and 7.1-6.2 Ma. Subsequently, there was a relatively stable period at 6.2-3.5 Ma, continued intensification about 3.5-2.5 Ma, and gradually weakening after 2.5 Ma. Since I Ma the monsoon has intensified, with remarkable high-frequency and amplitude variability. Simultaneous increase in sedimentation rates at ODP Sites 1143, 1146 and 1148, as well as in MAR of terrigenous materials, quartz, feldspar and clay minerals at ODP Site 1143 at 3.5-2.5 Ma, may be the erosional response to both global climatic deterioration and the strengthening of the East Asian summer monsoon after about 3-4 Ma. Sectional velocity distribution of the East China Sea Kuroshio is one of the basic points in the study of the Kuroshio. Hydrographic temperature and salinity data at G-PN section in the East China Sea from June 1955 to November 2001 are collected and properly processed to calculate the geostrophic current using dynamic height method at the transect of the Kuroshio. After analysis of calculation results, the basic current structure of the Kuroshio in its main part is examined together with scalar estimate and characters of multi- core structure, and spacial-temporal variations of current cores' position. Main result shows that (1) single-core structure, double-core structure and multi-core structure are basic forms in axial part of the Kuroshio; (2) abvious temporal variations exist in current structure of the Kuroshio; (3) the current of structure of the Kuroshio has distinctly seasonal association. The number of current cores is on the high side of core numbers in average and multi-core stucture appears in fall mostly.

扇贝养殖是我国传统的海水养殖产业,但自1997 年以来,养殖扇贝陆续爆发的大规模死亡,不但造成了巨大的经济损失,而且严重影响了该产业的健康发展。扇贝病害的不断爆发以及病因的多样性迫切要求制定新的疾病防治措施和开发新型的抗菌物质。 从扇贝自身的免疫防御因子入手,筛选和克隆参与免疫防御的功能基因,一方面可以研究抗病功能基因在病原感染或环境胁迫条件下的表达规律,深入探讨扇贝的免疫防御机制,并可作为抗病良种选育的分子标记,指导扇贝的遗传改良和抗病品系的培育;另一方面,可对抗菌效应物实现重组表达,开发新型的病害预防治疗制剂,取代目前普遍使用的抗生素和化学药物。抗菌效应物是机体在免疫应答过程中产生的多肽类物质,对侵入生物体内的细菌、病毒具有很强的免疫杀灭作用,对抗菌效应物的研究有助于深入了解机体先天性免疫防御的机制。 本研究采用大规模EST测序方法,结合cDNA末端快速扩增(RACE)技术,从海湾扇贝血淋巴中克隆到了大防御素基因(big defensin, AiBD)的全长cDNA序列,该cDNA全长为531 bp,其中5' 非编码区(UTR)为24 bp,开放阅读框(Open Reading Frame, ORF)含有369 bp,编码122 个氨基酸残基;随后为138-bp 的3' UTR,包括一个多聚腺苷酸信号序列(AATAAA)和ploy A尾巴。分析表明,海湾扇贝大防御素是以前体的形式合成,前体分子包括信号肽、前域和成熟肽三部分。采用Northern blot方法,以DIG标记的DNA探针检测了 AiBD mRNA在不同组织中的表达。结果发现,AiBD 基因的转录本主要在血淋巴中表达,在鳃中也有微量的表达,而在外套膜、闭壳肌、性腺及肝胰腺中检测不到杂交信号。采用QRT-PCR(quantitative real time PCR)对鳗弧菌感染后海湾扇贝血淋巴中AiBD mRNA 的表达量进行了检测,结果发现在感染后8 h 内, AiBD mRNA 的相对表达量平缓升高;随着刺激时间的增长,AiBD基因的mRNA表达量急剧增加,在刺激后16 h 和32 h 分别达到了空白组的72.3 倍和131.1 倍。为了研究海湾扇贝大防御素的抗菌活性,将其成熟肽编码区克隆到毕赤酵母表达载体pPIC9K并实现了重组表达。抑菌实验表明,重组AiBD具有广谱的抗菌活性,其对供试的三株革兰氏阳性菌(藤黄微球菌、溶壁微球菌、金黄色葡萄球菌)都表现出显著的抗菌活性,而对革兰氏阴性菌(鳗弧菌、亮弧菌)的抑菌活性则相对较弱;此外,重组AiBD对表达宿主也表现出杀菌活性,证明其具有抗真菌活性。 根据栉孔扇贝G 型溶菌酶基因的cDNA序列,利用构建的Genome Walking 文库获得了栉孔扇贝G 型溶菌酶基因的全长序列,该基因序列全长为8131 bp,由六个外显子和五个内含子组成。六个外显子长度分别为55 bp,60 bp,90 bp,113 bp,145 bp 和140 bp;五个内含子的长度分别为1126 bp,2161 bp,2744 bp,750 bp和592 bp;内含子的两侧都具有RNA正确剪接所必需的识别位点(GT/AG)。利用TRANSFAC 软件对栉孔扇贝G 型溶菌酶基因的5' 侧翼序列分析发现,该基因的5' 侧翼具有 TATA box 和 CAAT box 的共有序列;此外,在该基因的5' 侧翼发现了C/EBP、NF-κB、OCT-1 和 NF-IL6 等参与免疫基因激活的转录因子潜在结合位点。采用Northern blot方法,以生物素标记的RNA 探针检测了栉孔扇贝G 型溶菌酶基因在不同组织中的表达。结果发现,该基因的转录本主要在鳃、性腺及肝胰腺中表达,在血细胞和外套膜中也有微量的的表达,而在闭壳肌中检测不到杂交信号,这表明栉孔扇贝G 型溶菌酶可能兼备参与机体免疫防御和消化的功能。为了研究栉孔扇贝G 型溶菌酶的抗菌活性,将其成熟肽编码区克隆到毕赤酵母表达载体pPIC9K并实现了重组表达。抑菌实验表明,重组产物具有显著的抗阳性菌活性,其对供试的藤黄微球菌、溶壁微球菌表现出明显的抑制作用,对金黄色葡萄球菌未检测到抑制活性;而对革兰氏阴性菌仅表现出微弱的抑菌活性(亮弧菌和鳗弧菌),对大肠杆菌则基本无抑制活性。

Accumulation and distributions of aliphatic and polyaromatic hydrocarbons (PAHs) and heavy metals were measured in tissues of the clam Ruditapes philippinarum collected from 5 sites in Jiaozhou Bay, Qingdao, China. The concentrations of total aliphatic hydrocarbon and PAHs ranged from 570 to 2 574 ng/gdw (gram dry weight) and from 276 to 939 ng/gdw, in the most and least polluted sites, respectively. The bio-accumulation of hydrocarbons and PAHs in the clams appeared to be selective. Aliphatic hydrocarbons were predominantly represented by short chain (< nC(23)) n-alkanes, suggesting that petroleum hydrocarbons were likely the major contamination source. The selective uptake of 3 and 4 ring PAHs, such as naphthalene, fluorene, phenanthrene, fluoranthene and pyrene, by the clams was probably related to the physiological and bio-kinetic processes that were energetically favorable for uptake of compounds with fewer rings. Accumulation of the metals Cd, Cu, Zn, Pb, Cr, Hg, and As in the clam tissues also showed high variability, ranging from 0.043 to 87 A mu g/gdw. Among the 7 detected metals, Zn, Cd, Cu, and As had a particularly high potential of accumulation in R. philippinarum. In general, a positive correlation was found between the tissue concentrations and sediment concentrations of hydrocarbons and of some metals. Our study suggests that moderate contamination with polyaromatic hydrocarbons, and low to moderate contamination with metals, currently exists for clam R. philippinarum in Jiaozhou Bay, in comparison with other regional studies. A long-term monitoring program is certainly needed for assessment of the potential ecological influence and toxicity of these contaminants of R. philippinarum in Jiaozhou Bay.

Elemental (TOC, TN, C/N) and stable carbon isotopic (delta(13)C) compositions and n-alkane (nC(16-38)) concentrations were measured for Spartina alterniflora, a C-4 marsh grass, Typha latifolia, a C-3 marsh grass, and three sediment cores collected from middle and upper estuarine sites from the Plum Island salt marshes. Our results indicated that the organic matter preserved in the sediments was highly affected by the marsh plants that dominated the sampling sites. delta(13)C values of organic matter preserved in the upper fresh water site sediment were more negative (-23.0+/-0.3) as affected by the C-3 plants than the values of organic matter preserved in the sediments of middle (-18.9+/-0.8) and mud flat sites (-19.4+/-0.1) as influenced mainly by the C4 marsh plants. The distribution of n-alkanes measured in all sediments showed similar patterns as those determined in the marsh grasses S. alterniflora and T. latifolia, and nC(21) to nC(33) long-chain n-alkanes were the major compounds determined in all sediment samples. The strong odd-to-even carbon numbered n-alkane predominance was found in all three sediments and nC(29) was the most abundant homologue in all samples measured. Both delta(13)C compositions of organic matter and n-alkane distributions in these sediments indicate that the marsh plants could contribute significant amount of organic matter preserved in Plum Island salt marsh sediments. This suggests that salt marshes play an important role in the cycling of nutrients and organic carbon in the estuary and adjacent coastal waters. (C) 2003 Elsevier Ltd. All rights reserved.

The composition and distribution of aliphatic (n-alkanes) and polyaromatic hydrocarbons (PAHs) were measured for the surface sediments collected at 25 sites from Jiaozhou Bay, Qingdao, China. Total n-alkanes and PAH concentrations ranged from 0.5 to 8.2 mu g/gdw and 0.02 to 2.2 mu g/gdw, respectively, and the distribution of both n-alkanes and PAHs showed large spatial variations in the bay. The distribution of PAHs in the sediments was predominated by the three or more ring compounds. High hydrocarbon levels were generally found in the areas associated with high anthropogenic impact and port activities in the bay. The calculated hydrocarbon indexes suggest that petroleum contamination was the main source of n-alkanes, while both pyrolytic and petrogenic sources contributed PAHs to the surface sediments of Jiaozhou Bay. In comparison to other polluted coastal sediments, the level of contamination from both aliphatic hydrocarbons and PAHs in Jiaozhou Bay sediments is relatively low at the present time.
