A intensa exploração econômica tem causado diminuição do estoque original das espécies madeireiras amazônicas, incluindo aquelas de alta durabilidade natural destinadas a usos em contato com o solo. A escassez dessas madeiras resulta na elevação do preço, inviabilizando economicamente seu uso, sendo uma alternativa a substituição por espécies plantadas de rápido crescimento tratadas com preservativos. Outra vantagem, além da econômica, de utilizar espécies plantadas em substituição às tradicionais é ambiental, pois reduzirá a pressão exploratória sobre aquelas. Este estudo teve por objetivo avaliar o grau de degradação biológica de madeira de eucalipto citriodora (Corymbia citriodora (Hook.) K.D. Hill & L.A.S. Johnson) tratado por processo a vácuo e pressão com arseniato de cobre cromatado (CCA) e exposta em ensaio de campo em Rio Branco-AC. O ensaio foi implantado em maio de 2005 com estacas tratadas ou não com CCA. As variáveis avaliadas foram: 1) degradação por fungos, 2) degradação por cupins, e 3) região ou parte da peça afetada. A partir da terceira avaliação (47 meses de ensaio), 100% dos corpos de prova não tratados foram classificados com o grau de degradação máximo e, ao contrário, 100% dos tratados com CCA, decorridos 60 meses de ensaio, foram classificados com o grau mínimo. As partes não tratadas com maior e menor grau de degradação foram, respectivamente, o topo inferior e a parte aérea acima de 10 cm do solo. A degradação provocada por fungos foi levemente inferior à dos cupins. O ensaio aponta para a viabilidade técnica do uso de madeira tratada de eucalipto em substituição às espécies tradicionais da Amazônia.

Espécies de Mansoa são denominadas de "cipó-d'alho", por exalarem odor de alho das partes vegetativas e reprodutivas. Contudo, dados sobre morfologia e distribuição das estruturas secretoras presentes em Mansoa são escassos e ausentes para M. standleyi. O presente trabalho objetivou caracterizar a ocorrência e morfologia das estruturas secretoras do eixo vegetativo aéreo de M. standleyi. Para tanto, amostras da lâmina foliolar e de regiões nodais foram fixadas e submetidas às técnicas histológicas e de microscopia eletrônica de varredura. Testes histoquímicos, com os respectivos controles foram aplicados nas estruturas secretoras em fase secretora. Indivíduos de formigas e moscas, que visitavam a espécie foram amostrados, preservados e identificados por entomólogo. As estruturas secretoras do eixo vegetativo aéreo de M. standleyi estão representadas por tricomas glandulares dos tipos pateliformes e peltados. Todos com desenvolvimento assincrônico e presentes nas regiões nodais e lâmina foliolar, principalmente nas partes mais jovens. Nas regiões nodais, os tricomas formam um complexo secretor e, na lâmina foliolar, estão dispersos. As análises histoquímicas revelaram que os tricomas pateliformes são de fato nectários extraflorais e que os tricomas peltados, apresentam uma fração de alcaloides. Os visitantes das glândulas nodais correspondem a formigas Crematogaster (Formicidae) e Ectatomma brunea (Vespoidea, Formicidae) e moscas Oxysarcodexia (Sarcophagidae, subfamília Utitidae (Ulidiidae). Mansoa standleyi possui as estruturas secretoras do eixo vegetativo aéreo semelhantes às citadas pela literatura para Bignoniaceae, sendo esta a primeira vez, que um nectário extrafloral é descrito para a espécie.

High-risk human papillomavirus (hrHPV) is an essential cause of cervical carcinoma and is also strongly related to anal cancer development. The hrHPV E6 oncoprotein plays a major role in carcinogenesis. We aimed to evaluate the frequency of hrHPV DNA and E6 oncoprotein in the anuses of women with cervical carcinoma. We analyzed 117 women with cervical cancer and 103 controls for hrHPV and the E6 oncogene. Positive test results for a cervical carcinoma included 66.7 % with hrHPV-16 and 7.7 % with hrHPV-18. One case tested positive for both HPV variants (0.9 %). The samples from the anal canal were positive for HPV-16 in 59.8 % of the cases. Simultaneous presence of HPV in the cervix and anal canal was found in 53.8 % of the cases. Regarding expression of E6 RNA, positivity for HPV-16 in the anal canal was found in 21.2 % of the cases, positivity for HPV-16 in the cervix was found in 75.0 %, and positivity for HPV-18 in the cervix was found in 1.9 %. E6 expression in both the cervix and anal canal was found in 19.2 % of the cases. In the controls, 1 % tested positive for HPV-16 and 0 % for HPV-18. Anal samples from the controls showed a hrHPV frequency of 4.9 % (only HPV16). The presence of hrHPV in the anal canal of women with cervical cancer was detected at a high frequency. We also detected E6 RNA expression in the anal canal of women with cervical cancer, suggesting that these women are at risk for anal hrHPV infection.

Among the various possible embodiements of Advanced Therapies and in particular of Tissue Engineering the use of temporary scaffolds to regenerate tissue defects is one of the key issues. The scaffolds should be specifically designed to create environments that promote tissue development and not merely to support the maintenance of communities of cells. To achieve that goal, highly functional scaffolds may combine specific morphologies and surface chemistry with the local release of bioactive agents. Many biomaterials have been proposed to produce scaffolds aiming the regeneration of a wealth of human tissues. We have a particular interest in developing systems based in nanofibrous biodegradable polymers1,2. Those demanding applications require a combination of mechanical properties, processability, cell-friendly surfaces and tunable biodegradability that need to be tailored for the specific application envisioned. Those biomaterials are usually processed by different routes into devices with wide range of morphologies such as biodegradable fibers and meshes, films or particles and adaptable to different biomedical applications. In our approach, we combine the temporary scaffolds populated with therapeutically relevant communities of cells to generate a hybrid implant. For that we have explored different sources of adult and also embryonic stem cells. We are exploring the use of adult MSCs3, namely obtained from the bone marrow for the development autologous-based therapies. We also develop strategies based in extra-embryonic tissues, such as amniotic fluid (AF) and the perivascular region of the umbilical cord4 (Whartonâ s Jelly, WJ). Those tissues offer many advantages over both embryonic and other adult stem cell sourcess. These tissues are frequently discarded at parturition and its extracorporeal nature facilitates tissue donation by the patients. The comparatively large volume of tissue and ease of physical manipulation facilitates the isolation of larger numbers of stem cells. The fetal stem cells appear to have more pronounced immunomodulatory properties than adult MSCs. This allogeneic escape mechanism may be of therapeutic value, because the transplantation of readily available allogeneic human MSCs would be preferable as opposed to the required expansion stage (involving both time and logistic effort) of autologous cells. Topics to be covered: This talk will review our latest developments of nanostructured-based biomaterials and scaffolds in combination with stem cells for bone and cartilage tissue engineering.

OBJETIVO: Determinar os pontos de melhor sensibilidade e especificidade do Inventário de Depressão de Beck (BDI) e da Escala de Avaliação de Depressão de Hamilton (HAM-D) no diagnóstico de depressão associada à epilepsia. MÃTODOS: Setenta e três pacientes de um centro de referência no tratamento da epilepsia foram submetidos à avaliação neuropsiquiátrica. Foram colhidos dados clínicos e sociodemográficos, sendo utilizados os seguintes instrumentos: entrevista clínica estruturada (MINI-PLUS) para diagnóstico psiquiátrico conforme o DSM-IV, HAM-D e BDI. RESULTADOS: No momento da entrevista, 27,4% dos pacientes estavam deprimidos e 37% preenchiam critérios para diagnóstico de depressão maior ao longo da vida. A análise da curva ROC indicou que o ponto de corte em 16 (> 16) para o BDI (sensibilidade de 94,4%, especificidade de 90,6%) e em 16 (> 16) para a HAM-D (sensibilidade de 95%, especificidade de 75,5%) representou dicotomização ótima entre deprimidos e não deprimidos. Ambos os instrumentos apresentaram um valor preditivo negativo superior a 95%. CONCLUSÃO: A frequência de depressão maior é elevada em pacientes com epilepsia. BDI e a HAM-D podem auxiliar o clínico na identificação da depressão associada à epilepsia, diminuindo seu subdiagnóstico.

The present study aimed to investigate the effect of structure (design and porosity) on the matrix stiffness and osteogenic activity of stem cells cultured on poly(ester-urethane) (PEU) scaffolds. Different three-dimensional (3D) forms of scaffold were prepared from lysine-based PEU using traditional salt-leaching and advanced bioplotting techniques. The resulting scaffolds were characterized by differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), scanning electron microscopy (SEM), mercury porosimetry and mechanical testing. The scaffolds had various pore sizes with different designs, and all were thermally stable up to 300â ðC.àIn vitrotests, carried out using rat bone marrow stem cells (BMSCs) for bone tissue engineering, demonstrated better viability and higher cell proliferation on bioplotted scaffolds compared to salt-leached ones, most probably due to their larger and interconnected pores and stiffer nature, as shown by higher compressive moduli, which were measured by compression testing. Similarly, SEM, von Kossa staining and EDX analyses indicated higher amounts of calcium deposition on bioplotted scaffolds during cell culture. It was concluded that the design with larger interconnected porosity and stiffness has an effect on the osteogenic activity of the stem cells.

Despite the vast investigation and the large amount of products already available in the market to treat the different bone defects there is still a growing need to develop more advanced and complex therapeutic strategies. In this context, a mixture of Marine Hydroxyapatite-Fluorapatite:Collagen (HA-FP:ASC) seems to be a promising solution to overcome these bone defects, specifically, dental defects. HA-FP particles (20â63âμm) were obtained through pyrolysis (950°C, 12âh) of shark teeth (Isurus oxyrinchus, P. glauca), and Type I collagen was isolated from Prionace glauca skin as previously described (1). After the steps of purification, collagen was solubilized in 0.5âM acetic acid and HA-FP added producing three different formulations: were produced, 30:70, 50:50 and 70:30 of HA-FP:ASC, respectively. EDC/NHS and HMDI binding agents were used to stabilize the produced scaffolds. Mechanical properties were evaluated by compression tests. SEM analysis allowed observing the mineral deposition, after immersion in simulated body fluid and also permitted to evaluate how homogenous was the distribution of HA-FP in the different scaffold formulations, also confirmed by μ-CT assay. It was readily visible by Cytotoxicity and life/dead CLSM assays that cells were able to adhere and proliferate in the produced scaffolds. Scaffolds crosslinked with EDC/NHS showed lower cytotoxicity, being the ones chosen for further cellular evaluation.

[Excerpt] Although Acinetobacter baumannii has been the main agent for healthcare infections, recent reports suggest that some Acinetobacter environmental species should be considered as a potential cause of disease. In Angola, there are no previous data on its environmental reservoirs and resistance features. We aimed to unveil the occurrence and diversity of Acinetobacter species and the presence of resistance mechanisms in different non-clinical settings in Angola.
