The present investigation was undertaken to identify and characterize trophozoite proteases of five axenic strains of Giardia duodenalis isolated in Brazil and the reference strain Portland 1 isolated in the United States. Trophozoite cell lysates of each strain were analysed for the pattern of proteins and for proteolytic activity. Samples were tested in SDS-polyacrylamide gel electrophoresis for the protein profiles, and the detection of proteases in cell lysates was performed using substrate gel electrophoresis [gelatin, collagen, bovine serum albumin (BSA) and haemoglobin] and azocasein assays. Indeed, synthetic inhibitors were included in the assays to characterize the protease classes. Differences on the hydrolysis patterns of protein substrates were observed in relation to the substrate composition as much as the Giardia trophozoite strain. The substrate-containing gels revealed hydrolysis bands with molecular masses ranging from > 97 to 20-15 kDa, and most zones were common to the five strains. However, some pronounced differences could be detected in the BTU-11 pattern. Azocasein was also degraded; however, depending on the lysate assayed, the degree of substrate degradation was variable. It was observed that inhibitory effects are substrate-dependent since the activity was predominantly due to cysteine proteases against gelatin, collagen, BSA and azocasein substrates and due to serine against haemoglobin. The presence of aspartic protease and aminopeptidase activity in the lysates was also indicated.

Human purine nucleoside phosphorylase (PNP) is a ubiquitous enzyme which plays a key role in the purine salvage pathway, and PNP deficiency in humans leads to an impairment of T-cell function, usually with no apparent effect on B-cell function. PNP is highly specific for 6-oxopurine nucleosides and exhibits negligible activity for 6-aminopurine nucleosides. The catalytic efficiency for inosine is 350,000-fold greater than for adenosine. Adenine nucleosides and nucleotides are deaminated by adenosine deaminase and AMP deaminase to their corresponding inosine derivatives which, in turn, may be further degraded. Here we report the crystal structures of human PNP in complex with inosine and 2',3'-dideoxymosine, refined to 2.8 Angstrom resolution using synchrotron radiation. The present structures provide explanation for ligand binding, refine the purine-binding site, and can be used for future inhibitor design. (C) 2003 Elsevier B.V. All rights reserved.

Diplopods feed organic matter in decomposition; however, some environmental factors can promote changes in tissues of these animals. Sewage sludge has been applied for recuperation of physical structure of degraded soil. This work analyzed the influence of the sludge from a city of So Paulo in the midgut of the diplopod Rhinocricus padbergi. After the exposition to sludge, the midgut was prepared for histological and ultra-structural analyses. After 1 week of exposition, there were various glycoprotein globules in the fat body, which appeared, ultrastructurally, little electron dense. In the animals exposed for 2 weeks, there was an intensive renovation of the epithelium with the invasion of regenerative cells, which was observed in the histological and ultra-structural analyses. These data showed that the sludge present various substances that were very hazardous for these animals; more studies were necessary before the application of this in agriculture.

The keratin is not degraded by common enzyme, keratinases have the ability to degrade native keratin and others insoluble enzymes. In the present work was Studied keratinase produced by Streptomyces sp LMI-1 isolated from industrial plant of poultry processing. The enzyme degraded 87% of feathers after 120 h, it was stimulated by Ba(2+) and inhibited by Ca(2+), Mn(2+), EDTA and Hg(+). The optimum pH and temperature for the enzyme was 8.5 and 60 degrees C, respectively. The enzyme was stable after 2 hours at 50 degrees C. The culture broth analysis by thin layer chromatography showed presence of amino acids serine, methionine, proline, tyrosine and leucine after 72 hours of incubation. The microorganism showed potential for use in industrial process because of higher enzyme production and feathers degradation.

Poly(hydroxybutyrate-co-valerate) (PHBV) and poly(epsilon-caprolactone) (PCL) PCL/PHBV (4:1) blend films were prepared by melt-pressing. The biodegradation of the films in response to burial in soil for 30 days was investigated by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), differential scanning calorimetry (DSC) and thermogravimetry (TG). The PHBV film was the most susceptible to microbial attack, since it was rapidly biodegraded via surface erosion in 15 days and completely degraded in 30 days. The PCL film also degraded but more slowly than PHBV. The degradation of the PCL/PHBV blend occurred in the PHBV phase, inducing changes in the PCL phases (interphase) and resulting in an increase of its crystalline fraction.

To investigate the movement of seeds transported by fruit-eating birds in an agricultural, fragmented landscape of the Atlantic forest of southeast Brazil, I asked which bird species are the main seed dispersers in such environment, and how they use the available habitats (small forest fragments, forest thickets, live fences, isolated trees, and active pastures) where they are most likely to drop the seeds they swallow the relative importance of fruit-eating birds as seed vectors was evaluated based on the number of fruit species eaten, the number of visits, and visitation rate to fruiting plants. Habitat use was accessed by recording the habitats where birds were seen or heard during walks conducted throughout the study area. Sixteen plant species were observed during 308.3 plant-hours. Forty-one bird species were observed eating fruits in a total of 830 visits to fruiting plants. Sayaca Tanagers (Thraupis sayaca) and Pale-breasted Thrushes (Turdus leucomelas) ate the greatest number of fruit species, were the most frequent plant visitors in terms of number and rate of visits, and had a broad range of habitat use. These two species and the Rusty-margined Guan (Penelope superciliaris), which is able to swallow large fruits with large seeds that smaller bird species cannot cat, likely have a great contribution to the movement of seeds throughout this highly degraded landscape.

This study was conducted in an area of restinga in the municipality of Silo Vicente (SP). The study aimed to evaluate the seed rain in three different conditions of regeneration. Was selected the following conditions: High Forest of Restinga in the middle stage of regeneration, an area of clearing in the same condition mentioned above and a third in early stages of regeneration. hi each condition were installed 10 seed collectors made of wood and screen with 80% of shade, supported to a height of 20cm of soil. They were evaluated monthly for one yew; the densities of seed deposited in the collector these seedlings are identified and categorized based on their dispersion syndrome and successional classes. The Area of High Forest area was carried out a phytosociological survey to identify which seedlings present in the rain could be from these areas. It was found that the densities of seedlings are relatively low compared to other formations of the Atlantic rain forest, but consistent with other studies of the same type of vegetation used in the study. Regarding dispersal syndromes and successional classes, there was prevalence of the zoocoric syndrome and species of secondary successional classes. The results indicate that the area has good ability to maintain their succession dynamics.
