Objetivou-se no presente trabalho verificar a qualidade do café natural e despolpado durante a secagem em terreiro e secagem com ar aquecido a 40º e 60ºC. O trabalho foi realizado no Departamento de Engenharia e no Pólo de Tecnologia em Pós-Colheita do Café da Universidade Federal de Lavras (UFLA). A colheita do café, cultivar Topázio, foi seletiva. Parte do café foi despolpado e outra parte processado de forma natural. Uma parcela de cada tipo de café foi conduzida para a secagem em terreiro e outra parcela para secagem com temperaturas de 40ºC e 60ºC. Para avaliação da qualidade, foram feitos teste de condutividade elétrica e lixiviação de potássio; determinação de acidez titulável total; teste de acidez graxa; açúcares totais e redutores. Os resultados obtidos permitem concluir que: o tempo para secagem é afetado pelos diferentes tipos de secagem e processamentos; a condutividade elétrica, a lixiviação de potássio, a acidez titulável total e a acidez graxa aumentam com a elevação da temperatura de secagem, independente do tipo de processamento; os açúcares redutores e os açúcares totais diminuem com o aumento da tem peratura de secagem independente do tipo de processamento; a secagem com temperatura de 60ºC afetou negativamente a qualidade do café.

A manutenção da integridade das membranas celulares, entre outros eventos, é um forte indicativo de que a qualidade do café foi preservada na pós-colheita. Objetivou-se neste trabalho, analisar o efeito de diferentes métodos de secagem na manutenção da integridade da parede celular e da membrana plasmática de café natural e café despolpado, buscando determinar as condições e o momento em que ocorrem as rupturas microscópicas. Os cafés foram submetidos a um período de pré-secagem em terreiro. Após este, uma parcela de cada tipo de café foi desidratada no terreiro e, outra, à temperatura de 40ºC e 60ºC em secadores de camada fixa, monitorando-se a temperatura e o teor de água até 11% (bu). Nesse período, grãos foram aleatoriamente amostrados e fragmentos do endosperma preparados para a microscopia eletrônica de varredura, registrando-se diversas eletromicrografias, avaliando-se as alterações na membrana plasmática da célula do endosperma dos grãos de cafés em função do teor de água e tempo de secagem. O citoplasma das células a 11% (bu) de teor de água não foi comprometido na secagem em terreiro e a 40°C; na secagem a 60°C, observou-se comprometimento nas estruturas celulares nos cafés com teor de água de 20% (bu).

O presente experimento foi conduzido em Jaboticabal, SP, e teve como objetivos estudar uma cultura de café (Coffea arabica L., var. Mundo Novo), quanto à biologia floral, a freqüência e comportamento dos insetos na flor, testar o produto Bee-HereR (Hoescht Shering Agrevo do Brasil Ltda) quanto a sua atratividade para as abelhas Apis mellifera e verificar a produção de frutos com e sem a visita dos insetos. Para isso, foram verificados o tempo do desenvolvimento e quantidade de açúcar solúvel do néctar das flores; freqüência das visitações dos insetos, no decorrer do dia, por meio de contagem do número de insetos visitando as flores, a cada 60 minutos, das 8 às 17 horas, 10 minutos em cada horário; tempo (em segundos) e tipo de coleta (néctar e/ou pólen) dos insetos mais freqüentes; perda de botões florais; porcentagem de flores que se transformaram em frutos; tempo de formação e contagem dos grãos de café, observando-se a porcentagem de frutificação em flores visitadas ou não pelos insetos. Também foram realizados testes por pulverização utilizando-se o produto Bee-HereR , diluído em xarope e em água, em diferentes horários. A flor durou, em média, cerca de 3 dias desde sua abertura até o murchamento. A quantidade de açúcares do néctar apresentou diferença significativa entre os horários, sendo maior às 8 horas (em média, 102,18 ± 8,75 mg de carboidratos totais por flor). A abelha A. mellifera foi o inseto mais freqüente nas flores de café, coletando, principalmente, néctar no decorrer do dia. A perda de botões florais causada pelas chuvas foi, em média, 26,50 ± 11,70%. O tempo para a formação do fruto foi 6 meses e o número de frutos decorrentes do tratamento descoberto foi maior (38,79% e 168,38%, em 1993 e 1994, respectivamente) que do tratamento coberto. Apesar da eficiência do produto Bee-HereR ser afetada pelas condições climáticas, ele pode ser usado para atrair as abelhas A. mellifera na cultura.

Two simple methods were developed to determine, 11 pesticides in coconut water, a natural isotonic drink rich in salts, sugars and vitamins consumed by the people and athletes. The first procedure involves solid-phase extraction using Sep-Pak Vac C-18 disposable cartridges with methanol for elution. Isocratic analysis was carried out by means of high-performance liquid chromatography with ultraviolet detection at 254 nm to analyse captan, chlorothalonil, carbendazim, lufenuron and diafenthiuron. The other procedure is based on liquid-liquid extraction with hexane-dichloromethane (1:1, v/v), followed by gas chromatographic analysis with effluent splitting to electron-capture detection for determination of endosulfan, captan, tetradifon and trichlorfon and thermionic specific detection for determination of malathion, parathion-methyl and monocrotophos. The methods were validated with fortified samples at different concentration levels (0.01-12.0 mg/kg). Average recoveries ranged from 75 to 104% with relative standard deviations between 1.4 and 11.5%. Each recovery analysis was repeated at least five times. Limits of detection ranged from 0.002 to 2.0 mg/kg. The analytical procedures were applied to 15 samples and no detectable amounts of the pesticides were found in any samples under the conditions described. (C) 2002 Elsevier B.V. B.V. A population boom of the millipede Plusioporus setigerhas been recorded in coffee crops in the district of Patrocinio-MG, Brazil on the last five years. The millipedes spread over dwelling houses and to vegetable gardens, orchards and small plantations. It was suggested that population increase occurred as a consequence of the organic materials in high scale and also periodic irrigation in the coffee crop.

In this work we describe a versatile and very sensitive way for copper quantification by potentiometric stripping analysis using gold electrodes obtained from recordable compact disks (CDs). This new source of electrodes (CDtrodes) shown similar performance to the commercial gold electrodes with superior versatility and lower cost. Recordable CDs contains a highly pure gold film with thickness between 50 and 100 nm and superficial area of ca. 100 cm(2). The working electrode developed was used successfully in stationary cell and many experimental parameters have been optimized. For copper, the detection limit attained was 30 ng L-1 (600 s deposition time) with remarkable precision (standard deviation of 1.8 % for 20 repetitive measurements using 25 mu gL(-1) of copper with 60 s of deposition time). The gold electrode developed was used for analysis of copper in sugar cane spirits and tap water samples. The results were compared with those obtained by atomic absorption spectroscopy.

Xylella fastidiosa is a xylem-dwelling, insect-transmitted, gamma-proteobacterium that causes diseases in many plants, including grapevine, citrus, periwinkle, almond, oleander, and coffee. X. fastidiosa has an unusually broad host range, has an extensive geographical distribution throughout the American continent, and induces diverse disease phenotypes. Previous molecular analyses indicated three distinct groups of X.fastidiosa isolates that were expected to be genetically divergent. Here we report the genome sequence of X. fastidiosa (Temecula strain), isolated from a naturally infected grapevine with Pierce's disease (PD) in a wine-grape-growing region of California. Comparative analyses with a previously sequenced X.fastidiosa strain responsible for citrus variegated chlorosis (CVC) revealed that 98% of the PD X.fastidiosa Temecula genes are shared with the CVC X. fastidiosa strain 9a5c genes. Furthermore, the average amino acid identity of the open reading frames in the strains is 95.7%. Genomic differences are limited to phage-associated chromosomal rearrangements and deletions that also account for the strain-specific genes present in each genome. Genomic islands, one in each genome, were identified, and their presence in other X.fastidiosa strains was analyzed. We conclude that these two organisms have identical metabolic functions and are likely to use a common set of genes in plant colonization and pathogenesis, permitting convergence of functional genomic strategies.

Alcohol dehydrogenases (ADHs) are oxidoreductases present in animal tissues, plants, and microorganisms. These enzymes attract major scientific interest for the evolutionary perspectives, afforded by their wide occurrence in nature, and for their use in synthesis, thanks to their broad substrate specificity and stereoselectivity. In the present study, the standardization of the activity of the alcohol dehydrogenase from baker's yeast was accomplished, and the pH and temperature stability showed, that the enzyme presented a high stability to pH 6.0-7.0 and the thermal stability were completely maintained up to 50 degrees C during 1 h. The assays of ethanol (detection range 1-5 mM or 4.6 x 10(-2) to 23.0 x 10(-2) g/L) in different samples in alcoholic beverages, presented a maximum deviation of only 7.2%. The standard curve and the analytic curve of this method meet the conditions of precision, sensitivity, simplicity, and low cost, required for a useable analytical method. (c) 2006 Elsevier B.V. Xylella fastidiosa is a xylem-limited, Gram-negative bacterium responsible for citrus variegated chlorosis (CVC) in sweet oranges. In the present study, we present the recombinant expression, purification and characterization of an X. fastidiosa cysteine protease (dubbed Xylellain). The recombinant Xylellain ((HIS)Xylellain) was able to hydrolyze carbobenzoxy-Phe-Arg-7-amido-4-methylcoumarin (Z-FR-MCA) and carbobenzoxy-Arg-Arg-7-amido-4-methylcoumarin (Z-RR-MCA) with similar catalytic efficiencies, suggesting that this enzyme presents substrate specificity requirements similar to cathepsin B. The immunization of mice with (HIS)Xylellain provided us with antibodies, which recognized a protein of c. 31 kDa in the X. fastidiosa pathogenic strains 9a5c, and X. fastidiosa isolated from coffee plants. However, these antibodies recognized no protein in the nonpathogenic X. fastidiosa J1a12, suggesting the absence or low expression of this protein in the strain. These findings enabled us to identify Xylellain as a putative target for combating CVC and other diseases caused by X. fastidiosa strains.

The present research had aimed at studying the auxins and or boron effect on rooting of coffee (Coffea arabica L. cv. Mundo Novo') cuttings. The cuttings were obtained from semi-hardwood orthotropous branches of coffee-tree, containing 2 nodes and with aproximately 10 cm in length. The bases were dipped in treatment solutions composed of IBA or NAA with and without boron during 24 hours. After the treatments, the cuttings were planted in vermiculite. The following could be observed on cuttings taken ninety days after planting - total number of roots formed, average number of roots per cutting and root length. From the results obtained, it can be concluded that cutting treatment with NAA 100 ppm plus boron is responsible for the better rooting of coffee cuttings.

Lymphocytes from beta thalassaemia heterozygote patients, with normal levels of plasma folic acid cultured for 72 h in a folate rich medium, were not found to contain increased rates of sister chromatid exchanges (SCE). A wide intra and interindividual variability was found in both thalassaemic and control groups and methodological and biological factors, such as types of peripheral lymphocytes, sex, age, and smoking, alcohol and coffee drinking, as well as dietary habits, are possibly responsible for these variations.

The construction of a flow-through cell incorporating an array of gold microelectrodes is described and its application to flow injection analysis with amperometric detection is presented, Simple modification of almost any conventional integrated circuit chip, used as an inexpensive source of pre-assembled gold micro-wires, leads to the rapid and successful preparation of arrays of 8-48 elements, the polymeric encapsulation material from the top face of the chip is removed by abrasion until the gold micro-mires (used to interconnect the silicon circuit to the external contact pins of the chip) are disrupted and their transversal (elliptical) sections become exposed. Once polished, the flat and smooth top surface of the gold microelectrode-array chip (MEAC) is provided with a spacer and fitted under pressure against an acrylic block with the reference and auxiliary electrodes, to form the electrochemical (thin-layer) flow cell, while the contact pins are plugged into a standard IC socket, This design ensures autonomous electric contact with each electrode and allows fast dismantling for polishing or substitution, the performance of flow cells with MEACs was investigated utilizing the technique of reverse pulse amperometry without oxygen removal, A method was established for the determination of the copper concentration in sugar cane spirit, regulated by law for beverages, Samples from industrial producers and small-scale (alembic) brewers were compared, With a 24 MEAC, a detection limit of 30 mu g I-l of copper (4.7 x 10(-7) mol l(-1) of Cu-II for 100 mu l injections) was calculated, Routine operation was established at a frequency of 60-90 determinations per hour, Intercomparison with atomic absorption spectrometric determinations resulted in excellent agreement.

Density of binary solutions and combinations of sucrose, glucose, fructose, citric acid, malic acid, pectin, and inorganic salts were measured with an oscillating tube density meter in the temperature range from 10degrees to 60degreesC, at varying concentrations. Density can be predicted with accuracy better than 5 x 10(-5) g cm(-3) using predictive equations obtained by fitting the experimental data. Available literature values agreed well with experimental data. Relations for the excess molar volume of these solutions were derived in terms of mole fraction and temperature. A thermodynamic model for the volumetric analysis of multicomponent aqueous solutions containing electrolyte and non-electrolyte compounds was also proposed. These models can be used for prediction of density of liquid food systems, specially fruit juices and beverages, based on composition and temperature, with high accuracy and without elaborate experimental work.
