Candida albicans is classified into different serotypes according to cell wall mannan composition and cell surface hydrophobicity. Since the effectiveness of photodynamic therapy (PDT) depends on the cell wall structure of microorganisms, the objective of this study was to compare the sensitivity of in vitro biofilms of C. albicans serotypes A and B to antimicrobial PDT. Reference strains of C. albicans serotype A (ATCC 36801) and serotype B (ATCC 36802) were used for the assays. A gallium-aluminum-arsenide laser (660 nm) was used as the light source and methylene blue (300 mu M) as the photosensitizer. After biofilm formation on the bottom of a 96-well microplate for 48 h, each Candida strain was submitted to assays: PDT consisting of laser and photosensitizer application (L + P+), laser application alone (L + P-), photosensitizer application alone (L-P+), and application of saline as control (L-P-). After treatment, biofilm cells were scraped off and transferred to tubes containing PBS. The content of the tubes was homogenized, diluted, and seeded onto Sabouraud agar plates to determine the number of colony-forming units (CFU/mL). The results were compared by analysis of variance and Tukey test (p < 0.05). The two strains studied were sensitive to PDT (L + P+), with a log reduction of 0.49 for serotype A and of 2.34 for serotype B. Laser application alone only reduced serotype B cells (0.53 log), and the use of the photosensitizer alone had no effect on the strains tested. It can be concluded that in vitro biofilms of C. albicans serotype B were more sensitive to PDT.

Natural products have been studied aiming to understand their biological properties. Thus, this study aimed to investigate the antimicrobial activity of twenty-seven essential oils (EOs) used in aromatherapy procedures, a natural therapy with great emphasis currently used against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa strains. The agar dilution method was carried out and minimal inhibitory concentration against 50% and 90% of strains (MIC50% and MIC90% values) were reported. The S.aureus strains were highly susceptible with MIC90% from 0.21mg/mL to black pepper (Piper nigrum) and tea tree (Melaleuca alternifolia) to 26.52mg/mL with copaiba (Copaifera officinalis) EO. Cinnamon (Cinnamomum cassia) and clove (Syzygium aromaticum) EOs were effective against E.coli (2.0mg/mL) while the S.aromaticum EO was against P.aeruginosa (8.29mg/mL). Thus, the higher susceptibility of Gram-positive bacteria when compared with Gram-negative strains was found, and a large variability in the potential antibacterial has also been observed.

This article presents details of fabrication, biological activity (i.e., anti-matrix metalloproteinase [anti-MMP] inhibition), cytocompatibility, and bonding characteristics to dentin of a unique doxycycline (DOX)-encapsulated halloysite nanotube (HNT)-modified adhesive. We tested the hypothesis that the release of DOX from the DOX-encapsulated nanotube-modified adhesive can effectively inhibit MMP activity. We incorporated nanotubes, encapsulated or not with DOX, into the adhesive resin of a commercially available bonding system (Scotchbond Multi-Purpose [SBMP]). The following groups were tested: unmodified SBMP (control), SBMP with nanotubes (HNT), and DOX-encapsulated nanotube-modified adhesive (HNT+DOX). Changes in degree of conversion (DC) and microtensile bond strength were evaluated. Cytotoxicity was examined on human dental pulp stem cells (hDPSCs). To prove the successful encapsulation of DOX within the adhesivesbut, more important, to support the hypothesis that the HNT+DOX adhesive would release DOX at subantimicrobial levelswe tested the antimicrobial activity of synthesized adhesives and the DOX-containing eluates against Streptococcus mutans through agar diffusion assays. Anti-MMP properties were assessed via -casein cleavage assays. Increasing curing times (10, 20, 40 sec) led to increased DC values. There were no statistically significant differences (p > .05) in DC within each increasing curing time between the modified adhesives compared to SBMP. No statistically significant differences in microtensile bond strength were noted. None of the adhesives eluates were cytotoxic to the human dental pulp stem cells. A significant growth inhibition of S. mutans by direct contact illustrates successful encapsulation of DOX into the experimental adhesive. More important, DOX-containing eluates promoted inhibition of MMP-1 activity when compared to the control. Collectively, our findings provide a solid background for further testing of encapsulated MMP inhibitors into the synthesis of therapeutic adhesives that may enhance the longevity of hybrid layers and the overall clinical performance of adhesively bonded resin composite restorations.

The objective of this study was to compare the efficiency of washing and trimming broiler carcasses to reduce bacterial contamination. At the poste-visceration site, 100 broiler carcasses were collected during 4 visits to a slaughterhouse in Santa Catarina State, Brazil. Birds were from the same flock, age, and approximately 2.4 kg of weight. Groups were as follows: group 1, with fecal contamination; group 2, without fecal contamination; group 3, with fecal contamination and trimmed; group 4, with fecal contamination and washed; group 5, with fecal contamination, and washed and trimmed. Carcass washings were performed with at least 1.5 L/bird of potable water (0.5 to 1 mg/kg of residual chlorine) at room temperature (20-25 degrees C) using spray cabinets with 44 spray nozzles distributed into 2 chambers (pressure of 2 kgf/cm(2) and 4 kgf/cm(2)). Washed carcasses (trimmed or not) showed significantly (P < 0.05) lower counts of aerobic mesophiles (plate count agar) on the third evaluation, and even lower (P < 0.01) counts for total coliforms (CT) and fecal coliforms (Escherichia coli). Trimmed carcasses showed significantly lower counts (P < 0.05) for plate count agar; however, we observed higher counts for E. coli (P < 0.05). The association of both treatments (washing and trimming) showed significantly higher (P < 0.05) counts for coliforms (CT and E. coli). We can conclude that the washing method is overall more efficient than the trimming method to decontaminate chicken carcasses at the postevisceration site. Hopefully, our findings can help poultry companies to minimize production costs by applying the washing method for carcass decontamination.

Uma coleção de 50 amostras de Staphylococcus aureus e 50 de estafilococos coagulase-negativa (ECN) isoladas de recém-nascidos (RN) da unidade neonatal do Hospital das Clínicas da Faculdade de Medicina de Botucatu foram estudadas quanto à produção de biofilme. Para isto foi utilizada a técnica de PCR na detecção dos genes icaA, icaC e icaD e os métodos fenotípicos de aderência em placa de poliestireno, aderência em tubo de borossilicato e método do Agar Congo Vermelho (CRA). Dos 50 S. aureus estudados, 100% foram positivos para a produção de biofilme pela PCR, 98% pelo método do tubo, 100% pelo método da placa de poliestireno e 98% pelo CRA. Já das 50 amostras de ECN, 94% foram positivas pela PCR, 76% pelo método do tubo, 82% pelo teste da placa e 74% pelo CRA. Feita a comparação dos métodos utilizados, tendo por referência o padrão-ouro (PCR), foi possível observar que o método que melhor se correlacionou com o padrão-ouro, foi o método da aderência em placa de poliestireno, apresentando melhor sensibilidade e especificidade para ambas as espécies

Coleus blumei Benth (Lamiaceae), popularly known in Brazil as heart-hurt or coleus-of-Java is an ornamental plant widely used due to the color of its leaves. Several species of the genus Coleus present compounds with antioxidant, antimicrobial and allopathic activities due to the presence of chemicals such coleonol, forskolin and rosmarinic acid. Therefore, this study aimed to carry out phytochemical study and biological tests to evaluate the antibacterial, antioxidant and allopathic activities of the 70% ethanol extract and its fractions ethyl acetate, dichloromethane and end aqueous fraction leaf of Coleus blumei Benth. The plant extract was prepared by turbolisis from the powder of dried leaves, as a solvent for extraction using 70% ethanol. The ethanol extract was fractionated with solvents dichloromethane and ethyl acetate. The phytochemical study identified the presence of saponins, free anthraquinones, flavonoids and anthocyanidins. In evaluating the antibacterial dichloromethane fraction showed activity against all microorganisms tested both in agar diffusion test and microdilution test. The 70% ethanol extract showed activity against the micro-organisms S. aureus and S. epidermidis in agar diffusion test and against all microorganisms tested in microdilution test. The ethyl acetate fraction showed activity against the micro-organism S. aureus in the agar diffusion test and against all microorganisms tested in microdilution test. The end aqueous fraction showed no activity against any micro-organism tested. In the test of evaluation activity allopathic dichloromethane fraction showed greater inhibition of seed germination and growth of tomato (Lycopersicon esculentum Mill), followed by 70% ethanolic extract, the ethyl acetate fraction and the end aqueous fraction. A utilização de novas tecnologias empregando a biologia molecular busca superar os principais problemas encontrados nas vacinas atualmente comercializadas no combate à brucelose bovina. Desta forma, a identificação de proteínas imunogênicas e a posterior transformação dos genes correspondentes em micro-organismos competentes tem sido um dos principais alvos para o desenvolvimento de novas formas de controle da infecção. O presente trabalho tem como objetivo propor, em base teórica, uma vacina de eficácia e segurança superiores às encontradas atualmente no mercado contra a brucelose bovina, antropozoonose contagiosa provocada principalmente pela espécie Brucella abortus. Essa enfermidade produz infecção característica em bovinos e bubalinos e é infecciosa ao homem, causando doença crônica que leva à incapacidade parcial ou total para o trabalho. Por ter distribuição universal, acarreta problemas sanitário e de saúde pública importantes e grandes prejuízos econômicos. A vacina proposta visa o desenvolvimento de microorganismos transformados contendo genes de proteínas de potencial imunológico que, após purificação, são usadas para o combate da doença. As proteínas recombinantes abordadas no presente estudo são as proteína ribossomal L7/L12 e a lumazina sintetase que, ao serem utilizadas em uma vacina subcelular, diferente das comercializadas atualmente, induzem resposta de longa duração, não induzem a produção de anticorpos que interfiram no diagnóstico e não são patogênicas ao homem.

The studies related to research on new antimicrobial products have received special attention from researchers, especially given the emergence of microbial strains resistant to conventional antimicrobials. Thus, the present study was aimed to test the antimicrobial action of hydro-alcoholic extracts of plants collected in Cerrado region of Botucatu, following the species: Achyrocline satureioides (Lam) DC (macela), Stryphnodendron adstringens (Mart) Coville (barbatimão), Miconia rubiginosa (Bonpl.) DC (quaresma-branca), Davilla elliptica A. St-Hil (lixinha), Siparuna guianensis (negramina) e Solanum lycocarpum A.St-Hil (lobeira). The plants were always collected in the morning, in areas near the town of Botucatu, and extracts were prepared using a solvent such as methanol 70% from materials dried (50°C) and ground into mill knives. The extraction was performed for 48 hours at refrigerator temperature, followed by filtration, removal of methanol solvent in a rotary evaporator, determination of the dry weight of the extracts (mg / mL) and phytochemical analysis of the same. The sensitivity tests for 10 S. aureus, 11 E. coli and 11 P. aeruginosa, isolated from human clinical cases were performed by diluting volumes of the extracts in Mueller Hinton Agar (MHA) and determination of minimum inhibitory concentration (MIC) (mg / mL). According to the results and statistical analysis, it was found that depending on the bacteria tested, and in descending order of antibacterial activity for S. aureus: Lixinha sheet > Barbatimão sheet > Quaresma-Branca > Macela > Lixinha fruit > Barbatimão shell > Lobeira > Negramina; E. coli: Lixinha sheet > Barbatimão sheet > Lixinha fruit = barbatimão peel > Quaresma-Branca > Macela = Lobeira > Negramina and P. aeruginosa: Lixinha leaf > Barbatimão bark > Barbatimão leaf > Lixinha fruit > Macela > Lobeira > Quaresma - Branca = Negramina...
