O objetivo deste trabalho foi avaliar o efeito de subdoses de 2,4-D no desenvolvimento e produtividade da cultura do algodoeiro. Dois experimentos foram conduzidos utilizando-se da variedade IAPAR-95 e o sistema de avaliação por meio de testemunhas duplas. No primeiro experimento, as aplicações foram realizadas no estádio F1 (início do florescimento), aplicando-se dosagens de 0,84; 1,68; 3,36; 6,72; 13,44 e 26,88 g de equivalente ácido (e.a.) por hectare, equivalentes a derivas de 0,125; 0,25; 0,5; 1,0; 2,0 e 4,0% da dose de 670 g e.a. ha-1. No segundo experimento, os tratamentos foram constituídos pela combinação em esquema fatorial de duas doses (6,72 e 13,44 g e.a. ha-1) e três épocas de aplicação (C1, C3/C4 e C6), visando a avaliar a variação da sensibilidade do algodão ao 2,4-D em função de seu estádio de desenvolvimento. Os resultados evidenciaram que doses maiores que 3,36 g e.a. ha-1 (0,50%) aplicadas na fase de florescimento afetaram de forma significativa a produtividade, que a queda dos botões florais foi o sintoma mais importante para a redução da produtividade. No segundo experimento, observou-se que a sensibilidade do algodão caiu drasticamente em função do estádio de desenvolvimento. O único tratamento que provocou queda significativa de produtividade foi a dose de 13,44 g e.a. ha-1 (2,0%) aplicada no estádio C1. Dessa forma, a partir do momento em que as maçãs começam a se formar, a sensibilidade da cultura cai substancialmente.

O presente trabalho teve como objetivo determinar os efeitos de subdoses do herbicida 2,4-D na cultura do fumo. Os experimentos foram conduzidos a campo durante o ano de 2003, no município de Santa Isabel do Ivaí - PR, utilizando-se de fumo tipo Virgínia. No primeiro experimento, foram aplicadas, no estádio de pré-florescimento, doses de 0,84; 1,68; 3,36; 6,72; 13,44 e 26,88 g e.a. ha-1 de 2,4-D, equivalentes à deriva de 0,125; 0,25; 0,5; 1,0; 2,0 e 4,0% de produtos comerciais aplicados a 1,0 L ha-1 (670 g e.a. ha-1). Para o segundo experimento, os tratamentos foram constituídos pela combinação em esquema fatorial de duas doses (6,72 e 13,44 g e.a. ha-1, equivalentes a 1,0 e 2,0% de deriva) e três épocas de aplicação após o transplante para o campo. Nenhum sintoma visual marcante de fitotoxicidade foi observado dentro da faixa de doses avaliada. Visualmente, alguns dos possíveis efeitos atribuídos à fitotoxicidade ocasionada pela deriva de 2,4-D na cultura do fumo podem estar associados à utilização de antibrotantes. Nenhuma das doses aplicadas no primeiro experimento afetou a produtividade da cultura, nem o aspecto e qualidade das folhas, tendo o fumo tolerado até 4,0% de deriva avaliada. Resultados obtidos no segundo experimento confirmaram esses dados, constatando-se que níveis de deriva de até 2,0% (13,44 g e.a. ha-1), aplicados em três fases após o transplante das mudas para o campo, não causaram qualquer efeito negativo sobre a produtividade da cultura.

Durante os anos agrícolas de 2002-2003 e 2003-2004 foram conduzidos trabalhos no município de Maringá - PR, com o objetivo de avaliar o dano potencial de subdoses de 2,4-D sobre plantas de uva, imitando depósitos decorrentes de deriva. No primeiro experimento, a aplicação foi realizada cerca de 30 dias após a poda de inverno, num pomar de uva Itália. As doses utilizadas foram de 6,72; 13,44; 26,88; 53,76 e 107,52 g de equivalente ácido (e.a.) por hectare de 2,4-D, equivalentes a depósitos de 1,0%; 2,0%; 4,0%; 8,0% e 16,0%, assumindo-se uma aplicação de 1 L ha-1 (670 g e.a. ha-1). Nessa data, as plantas encontravam-se na fase de emissão de cachos e florescimento (estádio 15). O surgimento de sintomas visuais de fitointoxicação foi imediato e proporcional às doses aplicadas. A produtividade da cultura foi afetada por todas as doses aplicadas nesse estádio de crescimento. No entanto, mesmo com as injúrias severas registradas na dose mais alta, as plantas afetadas se recuperaram após duas podas para as condições de manejo regionais (duas safras por ano). No segundo experimento, foram aplicadas doses equivalentes a derivas de 1,0 e 2,0% (6,72 e 13,44 g e.a. ha-1) em três estádios do ciclo de desenvolvimento. A aplicação de doses < 13,44 g e.a. ha-1 (2,0% de deriva simulada) a partir do estádio de "meia-baga", não causou repercussões negativas em termos de injúrias visuais e produtividade.

Social media has become a part of many people’s everyday lives. In the library field the adoption of social media has been widespread and discussions of the development of “Library 2.0” began at an early stage. The aim with this thesis is to study the interface between public libraries, social media, and users, focusing on information activities. The main research question is: How is the interface between public libraries and social media perceived and acted upon by its main stakeholders (library professionals and users)? The background of Library 2.0 is strongly associated with the development of the Web and social media, as well as with the public libraries and their user-centered and information technological development. The theoretical framework builds on the research within the area of Library and Information Science concerning information behavior, information practice, and information activities. Earlier research on social media and public libraries is also highlighted in this thesis. The methods survey and content analysis were applied to map the interface between social media and public libraries. A questionnaire was handed out to the users and another questionnaire was sent out to the library professionals. The results were statistically analyzed. In the content analysis public library Facebook pages were studied. All the empirical investigations were conducted in the area of Finland Proper. An integrated analysis of the results deepens the understanding of the key elements of the social media and public library context. These elements are interactivity, information activities, perceptions, and stakeholders. In this context seven information activities were distinguished: reading, seeking, creating, communicating, informing, mediating, and contributing. This thesis contributes to develop the research concerning information activities and draws a realistic picture of the challenges and opportunities in the social media and public library context. It also contributes with knowledge on library professionals and library users, and the existing differences in their perceptions of the interface between libraries and social media.

In the beginning of its 10th year of existence Facebook has engaged and connected 1.2 billion monthly active users. This article-based dissertation Disconnect.Me – User Engagement and Facebook approaches this engagement from the opposite direction: disconnection. The research articles focus on social media specific phenomena including leaving Facebook, tactical media works such as Web 2.0 SuicideMachine, memorializing dead Facebook users and Facebook trolling. The media theoretical framework for this study is built around affect theory, software studies, biopolitics as well as different critical studies of new media. The argument is that disconnection is a necessary condition of social media connectivity and exploring social media through disconnection – as an empirical phenomenon, future potential and theoretical notion – helps us to understand how users are engaged with social media, its uses and subsequent business models. The results of the study indicate that engagement is a relation that precedes user participation, a notion often used to conceptualize social media. Furthermore, this engagement turns the focus from users’ actions towards the platform and how the platform actively controls users and their behavior. Facebook aims to engage new users and maintain the old ones by renewing its platform and user interface. User engagement with the platform is thus social but also technical and affective. When engaged, the user is positioned to algorithmic connectivity where machinc processes mine user data. This data is but sold also used to affect and engage other users. In the heart of this study is the notion that our networked engagements matter and disconnection can bring us to the current limits of network culture.

This thesis studies crowdfunding with qualitive methods to introduce the phenomenon as well as provide guidance to those interested in its utilization. Knowledge and ideas were gathered form several sources, from academic literature to commercial media and expert interviews. Crowdfunding has already demonstrated its ability to impact the startup scene but is still far from being utilized to its full extent, especially in Finland, where even its legality has been questioned. Crowd financing can provide capital to entrepreneurs who might not otherwise be able to obtain funding as well as enable crowdsourcing the funders in several ways. A successful campaign, however, requires a wealth of knowledge on the subject, careful planning and hard work on the implementation. The thesis will provide most benefit to entrepreneurs who are considering the use of this new form of finance, but should also be of value for investors, academics, politicians and everyone else interested in the subject.

This study discusses the procedures of value co-creation that persist in gaming industry. The purpose of this study was to identify the procedures that persist in current video gaming industry which answers the main research problem how value is co-created in video gaming industry followed by three sub questions: (i) What is value co-creation in gaming industry? (ii) Who participates in value co-creation in gaming industry? (iii) What are the procedures that are involved in value co-creation in gaming industry? The theoretical background of the study consists of literature relating to the theory of marketing i.e., notion of value, conventional understanding of value creation, value chain, co-creation approach, co-production approach. The research adopted qualitative research approach. As a platform of relationship researcher used web 2.0 tool interface. Data were collected from the social networks and netnography method was applied for analyzing them. Findings show that customer and company both co-create optimum level of value while they interact with each other and within the customers as well. However mostly the C2C interaction, discussions and dialogues threads that emerged around the main discussion facilitated to co-create value. In this manner, companies require exploiting and further motivating, developing and supporting the interactions between customers participating in value creation. Hierarchy of value co-creation processes is the result derived from the identified challenges of value co-creation approach and discussion forums data analysis. Overall three general sets and seven topics were found that explored the phenomenon of customer to customer (C2C) and business to customer (B2C) interaction/debating for value co-creation through user generated contents. These topics describe how gamer contributes and interacts in co-creating value along with companies. A methodical quest in current research literature acknowledged numerous evolving flows of value in this study. These are general management perspective, new product development and innovation, virtual customer environment, service science and service dominant logic. Overall the topics deliver various realistic and conceptual implications for using and handling gamers in social networks for augmenting customers’ value co-creation process.

To determine the effects of combined therapy of gliclazide and bedtime insulin on glycemic control and C-peptide secretion, we studied 25 patients with type 2 diabetes and sulfonylurea secondary failure, aged 56.8 ± 8.3 years, with a duration of diabetes of 10.6 ± 6.6 years, fasting plasma glucose of 277.3 ± 64.6 mg/dl and a body mass index of 27.4 ± 4.8 kg/m². Patients were submitted to three therapeutic regimens lasting 2 months each: 320 mg gliclazide (phase 1), 320 mg gliclazide and bedtime NPH insulin (phase 2), and insulin (phase 3). At the end of each period, glycemic and C-peptide curves in response to a mixed meal were determined. During combined therapy, there was a decrease in all glycemic curve values (P<0.01). Twelve patients (48%) reached fasting plasma glucose <140 mg/dl with a significant weight gain of 64.8 kg (43.1-98.8) vs 66.7 kg (42.8-101.4) (P<0.05), with no increase in C-peptide secretion or decrease in HbA1. C-Peptide glucose score (C-peptide/glucose x 100) increased from 0.9 (0.2-2.1) to 1.3 (0.2-4.7) during combined therapy (P<0.01). Despite a 50% increase in insulin doses in phase 3 (12 U (9-30) vs 18 U (11-60); P<0.01) only 3 patients who responded to combined therapy maintained fasting plasma glucose <140 mg/dl (P<0.02). A tendency to a higher absolute increase in C-peptide (0.99 (0.15-2.5) vs 0.6 (0-2.15); P = 0.08) and C-peptide incremental area (2.47 (0.22-6.2) vs 1.2 (0-3.35); P = 0.07) was observed among responders. We conclude that combined therapy resulted in a better glucose response to a mixed meal than insulin alone and should be tried in type 2 diabetic patients before starting insulin monotherapy, despite difficulties in predicting the response.

Analysoin tutkielmassani suomalaisten kansanedustajien poliitikko-sivujen soveltumista kansanedustajien viestintävälineeksi. Pyrin selvittämään, valjastavatko perinteiset poliittiset toimijat samoja sosiaalisen median toimintoja, joita erityisesti epähierarkkiset, uusyhteisölliset ja spontaanit kansalaisliikkeet ovat pystyneet käyttämään poliittiseen mobilisaatioon 2000-luvulla, ja miten he ottavat huomioon käyttämänsä Web 2.0 –ympäristön yleiset ominaisuudet. Apunani aineiston erittelyssä käytän Jay David Bolterin ja Richard Grusinin jo 1990-luvulla lanseeraamaa remediaation teoriaa sekä Axel Brunsin esittämiä käyttäjätuotannon perusperiaatteita. Bruns haastaa perinteisestä tuotantomallista hyötyvät sisällöntuottajat analyysillään käyttäjätuotannosta ja sen aiheuttamista muutoksista tulevaisuuden poliittisessa toimintakulttuurissa. Hän kategorisoi Web 2.0 –ajan teknologioiden myötä syntyneiden julkaisumahdollisuuksien seurauksia perinteiselle hierarkkiselle sisällöntuotannolle käyttäjätuotannon neljän perusperiaatteen avulla. Bolter ja Grusin taas analysoivat mediaympäristöä remediaation kautta esittäen, että jokainen mediamuoto lainaa ja remedioi aikaisempien medioiden sisältöjä ja ominaisuuksia pyrkiessään kohti välittömämpää ja hypermedioituneempaa ilmaisua. Teorioiden avulla sekä kuvaan sosiaalisen median toiminnallisia ominaisuuksia että kategorisoin poliittisen viestinnän tuotteita sekä niiden takana piileviä motiiveja. Tutkielman materiaalina käytän kahdeksantoista suosituimman kansanedustajan poliitikko-sivujen sisältöjä. Perehdyn sekä sivuille julkaistuun alkuperäissisältöön että niiden aikaansaamaan keskusteluun kansanedustajien osalta. Kohtaan tutkielman aikana historiografisia ongelmia dynaamisten ja muuttuvien sisältöjen kanssa työskennellessäni, mistä johtuen erittelen myös digitaalisen historiantutkimuksen erityispiirteitä. Bolterin ja Grusinin sekä Brunsin teorioihin sisältyvien teemojen lisäksi otannasta erottuu toistuvia aihepiirejä, joista merkittävimmät ovat poliitikon julkisuuden ja yksityisyyden välinen jännite, perinteisen vahvan poliitikon roolin kohtaamat haasteet puoluepoliittisten toimijoiden piirin ulkopuolelta, jatkuvan kampanjoinnin tarve sosiaalisen median aikakautena sekä rajattujen yleisöjen katoaminen verkkoviestinnän avoimuuden ja esteettömyyden takia. Tutkielma osoittaa, että vaikka aktiivisimmat poliitikko-sivut imitoivat joissain määrin Brunsin esittämää käyttäjätuotantoa kommunikoidessaan palvelun käyttäjien kanssa, ne eivät tunnusta kaikkia Brunsin käyttäjätuotantoprojekteille asettamia perusperiaatteita. Eduskuntalaitoksen hierarkkisuuteen ja kansanedustajakauden aikaiseen työrauhaan ei pystytä vaikuttamaan viestinnän kautta. Tutkielma osoittaa myös, että poliitikko-sivut hyväksikäyttävät remediaatiota välittääkseen Facebookin käyttäjille haluamiansa sisältöjä muista digitaalisista lähteistä ja luodakseen niiden ympärille oman viestintänsä kannalta heitä hyödyttävän näkökulman. Molemmat teoriat soveltuvat hyvin tutkittujen poliitikkojen viestinnän erittelyyn. Tutkielman aikana havaitsen kolme erilaista suhtautumistapaa Facebookiin. Autioiden sivujen omistajat eivät käytä sivujaan viestintävälineenä. Passiiviset sisällön tuottajat välittävät sivujensa kautta sisältöä vertikaalisesti kuitenkaan ottamatta kontaktia käyttäjiin. Aktiiviset sisältöihin osallistujat käyttävät Facebookia monipuolisesti keskustelemalla ja argumentoimalla sisällöissään käyttäjien kanssa sekä antamalla näille tunnustusta sen ollessa perusteltua.

Hypoxia activates endothelial cells by the action of reactive oxygen species generated in part by cyclooxygenases (COX) production enhancing leukocyte transmigration. We investigated the effect of specific COX inhibition on the function of endothelial cells exposed to hypoxia. Mouse immortalized endothelial cells were subjected to 30 min of oxygen deprivation by gas exchange. Acridine orange/ethidium bromide dyes and lactate dehydrogenase activity were used to monitor cell viability. The mRNA of COX-1 and -2 was amplified and semi-quantified before and after hypoxia in cells treated or not with indomethacin, a non-selective COX inhibitor. Expression of RANTES (regulated upon activation, normal T cell expressed and secreted) protein and the protective role of heme oxygenase-1 (HO-1) were also investigated by PCR. Gas exchange decreased partial oxygen pressure (PaO2) by 45.12 ± 5.85% (from 162 ± 10 to 73 ± 7.4 mmHg). Thirty minutes of hypoxia decreased cell viability and enhanced lactate dehydrogenase levels compared to control (73.1 ± 2.7 vs 91.2 ± 0.9%, P < 0.02; 35.96 ± 11.64 vs 22.19 ± 9.65%, P = 0.002, respectively). COX-2 and HO-1 mRNA were up-regulated after hypoxia. Indomethacin (300 µM) decreased COX-2, HO-1, hypoxia-inducible factor-1alpha and RANTES mRNA and increased cell viability after hypoxia. We conclude that blockade of COX up-regulation can ameliorate endothelial injury, resulting in reduced production of chemokines.

(E)-2-nonenal is considered an important off-flavor of beer, related to the flavor of beer staling. In this study, a new method for determination of (E)-2-nonenal in beer using headspace solid-phase microextraction and gas chromatographic coupled mass spectrometry (HS-SPME-GC-MS) was developed and applied in Brazilian beer samples. The extractions were carried out in CAR-PDMS (carboxen-polydimethylsiloxane) fiber and the best results were found with 15 minutes of equilibrium and 90 minutes of extraction at 50 °C. The method was linear in the range from 0.02 to 4.0 μg.L-1 with correlation coefficient of 0.9994. The limits of detection and quantification were 0.01 and 0.02 μg.L-1, respectively. 96.5% of recovery and 4% precision (RSD) were obtained in the fortification of beer samples with 2.0 μg.L-1 of (E)-2-nonenal. The developed method proved to be simple, efficient and highly sensitive to the determination of this analyte being easily applied in the quality control of the brewery. (E)-2-nonenal was found in all beer samples analyzed with levels between 0.17 and 0.42 μg.L-1.

Low levels of ionizing radiation induce two translocation responses in soybean: a reduction in photoassimilate export from leaves and a change in the distribution pattern of exported photoassimilate within the plant. In this investigation these responses have been further studied specifically to ascertain the site of radiation damage and to better understand the physiological responses observed. Experimentally the primary data was obtained from studies in which a mature trifoliate leaf of a young soybean plant (Glycine ~ L. cultivar Harosoy '63) is isolated in a closed transparent chamber and allowed to photoassimilate 14C02 for 15 minutes. This is followed by an additional 45 ~_il'1;ute period before the plant is sectl.o ne d an d 14 C-ra dl' oactl.v.l ty d eterml. ne d'l n a 11 parts. Such 14c data provides one with the magnitude and distribution pattern of translocation. Further analyses were conducted to determine the relative levels of the major photosynthetic products using the techniques of paper chromatography and autoradiography. Since differences between control and irradiated P 1 ants were not 0 b serve d l' n t h e par tl't"lo nlng 0 f 14 C between the 80% ethanol-soluble and -insoluble fractions 14 or in the relative amounts of C-products of photosynthesis, the reduction in export in irradiated plants is not likely due to reduced availability of translocatable materials. Data presented in this thesis shows that photoassimilate export was not affected by gamma radiation until a threshold dose between 2.0 and 3.0 krads was reached. It was also observed that radiation-induced damage to the export process was capable of recovery in a period of 1 to 2 hours provided high light intensity was supplied. In contrast, the distribution pattern was shown to be extremely radiosensitive with a low threshold dose between .25 and .49 krads. Although this process was also capable of recovery,lt" occurred much earlier and was followed by a secondary effect which lasted at least for the duration of the experiments. The data presented in this thesis is interpreted to suggest that the sites of radiation action for the two translocation responses are different. In regards to photoassimilate export, the site of action of ionizing radiation is the leaf, quite possibly the process of photophosphorylation which may provide energy directly for phloem loading and for membrane integrity of the phloem tissue* In regards to the pattern of distribution of exported photoassimilate, the site is likely the apical sink, possibly the result of changes of levels of endogenous hormones. By the selection of radiation exposure dose and time post-irradiation, it is possible to affect independently these two processes suggesting that each may be regulated independent of the other and involves a distinct site.

Les institutions documentaires doivent, de leur côté, renouveler leur positionnement et repenser leurs savoir-faire.
