In a system in which fertilization is recommended, diagnosis of soil K availability and the establishment of critical levels are made difficult by the possibility of a contribution of non-exchangeable forms of K for plant nutrition. Due to its magnitude, this contribution is well diagnosed in long term experiments and in those which compare fertilization systems with positive and negative balances in terms of replacement of the K extracted by plants. The objective of this study was to evaluate K availability in a Hapludalf under fertilization for sixteen years with the addition of K doses. The study was undertaken in an experiment set up in 1991 and carried out until 2007 in the experimental area of the Soil Department of the Federal University of Santa Maria (Universidade Federal de Santa Maria - UFSM), in Santa Maria (RS), Brazil. The soil was a Typic Hapludalf submitted to four doses of K (0, 60, 120 and 180 kg ha-1 K2O) and subdivided in the second year, when 60 kg ha-1 of K2O were reapplied in the subplots in 0, 1, 2 and 3 times. As of the fifth year, the procedure was repeated. Grain yield above ground dry matter and total K content contained in the plant tissue were evaluated. Soil samples were collected, oven dried, ground, passed through a sieve and submitted to exchangeable K analysis by the Mehlich-1 extractor; non-exchangeable K by boiling HNO3 1 mol L-1 and total K by HF digestion. Potassium fertilization guidelines should foresee the establishment of a critical level as of which the recommended dose should accompany crop needs, which coincides with the quantity exported by the grain, without there being the need for the creation of broad ranges of K availability to predict K fertilization. In adopting the K fertilization recommendations proposed in this manner, there will not be K translocation in the soil profile.

Phytoremediation strategies utilize plants to decontaminate or immobilize soil pollutants. Among soil pollutants, metalloid As is considered a primary concern as a toxic element to organisms. Arsenic concentrations in the soil result from anthropogenic activities such as: the use of pesticides (herbicides and fungicides); some fertilizers; Au, Pb, Cu and Ni mining; Fe and steel production; coal combustion; and as a bi-product during natural gas extraction. This study evaluated the potential of pigeon pea (Cajanus cajan), wand riverhemp (Sesbania virgata), and lead tree (Leucaena leucocephala) as phytoremediators of soils polluted by As. Soil samples were placed in plastic pots, incubated with different As doses (0; 50; 100 and 200 mg dm-3) and then sown with seeds of the three species. Thirty (pigeon pea) and 90 days after sowing, the plants were evaluated for height, collar diameter and dry matter of young, intermediate and basal leaves, stems and roots. Arsenic concentration was determined in different aged leaves, stems and roots to establish the translocation index (TI) between the plant root system and aerial plant components and the bioconcentration factors (BF). The evaluated species showed distinct characteristics regarding As tolerance, since the lead tree and wand riverhemp were significantly more tolerant than pigeon pea. The high As levels found in wand riverhemp roots suggest the existence of an efficient accumulation and compartmentalization mechanism in order to reduce As translocation to shoot tissues. Pigeon pea is a sensitive species and could serve as a potential bioindicator plant, whereas the other two species have potential for phytoremediation programs in As polluted areas. However, further studies are needed with longer exposure times in actual field conditions to reach definite conclusions on relative phytoremediation potentials.

The contribution of humic substances of different composts to the synthesis of humin in a tropical soil was evaluated. Increasing doses (0, 13, 26, 52, and 104 Mg ha-1) of five different composts consisting of agroinpowderrial residues were applied to a Red-Yellow Latosol. These composts were chemically characterized and 13C NMR determined and the quantity of the functional alkyl groups of humic acids applied to the soil as compost was estimated. Thirty days after application of the treatments, organic matter samples were collected for fractionation of humic acids (HA), fulvic acids (FA) and humin (HU), from which the ratios HA/FA and (HA + FA)/HU were calculated. The application of the composts based on castor cake resulted in the highest HU levels in the soil; alkyl groups of the HA fraction of the composts were predominant in the organic components added to the HU soil fraction.

In the region of the Serra do Espinhaço Meridional, peat bog is formed in hydromorphic environments developed in sunken areas on the plain surfaces with vegetation adapted to hydromorphic conditions, favoring the accumulation and preservation of organic matter. This pedoenvironment is developed on the regionally predominant quartzite rocks. Peat bog in the Environmental Protection Area - APA Pau-de-Fruta, located in the watershed of Córrego das Pedras, Diamantina,Brazil, was mapped and three representative profiles were morphologically characterized and sampled for physical, chemical and microbiological analyses. The organic matter was fractionated into fulvic acid (FA), humic acids (HA) and humin (H). Two profiles were sampled to determine the radiocarbon age and δ13C. The structural organization of the three profiles is homogeneous. The first two layers consist of fibric, the two subsequent of hemic and the four deepest of sapric peat, showing that organic matter decomposition advances with depth and that the influence of mineral materials in deeper layers is greater. Physical properties were homogeneous in the profiles, but varied in the sampled layers. Chemical properties were similar in the layers, but the Ca content, sum of bases and base saturation differed between profiles. Contents of H predominated in the more soluble organic matter fractions and were accumulated at a higher rate in the surface and deeper layers, while HA levels were higher in the intermediate and FA in the deeper layers. Microbial activity did not vary among profiles and was highest in the surface layers, decreasing with depth. From the results of radiocarbon dating and isotope analysis, it was inferred that bog formation began about 20 thousand years ago and that the vegetation of the area had not changed significantly since then.

Purpose:We previously observed that anti- and pro-apoptotic genes of the Bcl-2 family were differentially expressed during the development of LCA in the Rpe65-/- mouse model (Cottet et al. 2006). Moreover, we reported that activation and translocation of pro-apoptotic Bax to the mitochondria was associated with apoptosis of rod photoreceptors as the disease progressed (Cottet et al. 2008). In this study we challenged whether disruption of the pro-apoptotic pro-apoptotic Bax protein is sufficient to protect photoreceptor cells against apoptosis. Methods:Apoptosis of photoreceptor cells was addressed by TUNEL assay on flatmounted retinas. Counting of the rod nuclei within the ONL was performed following hematoxylin/eosin histological staining of retina sections. Expression level and localization of photoreceptor gene markers were assessed by quantitative PCR and immunohistological analyses. Results:While expression of rod photoreceptor genes was decreased in Rpe65-deficient retina, expression level remained unchanged in Rpe65-/- / Bax-/- mice. Moreover, OS dysorganization and shortening as well as decrease in ONL thickness observed in diseased retina were prevented in mice lacking functional Bax protein. TUNEL assay confirmed that Bax-dependent rod photoreceptor apoptosis was abolished in Rpe65-/- / Bax-/- mice. However, early and fast degeneration of cone cells was not prevented in Rpe65-/- / Bax-/- mice, indicating that Bax-induced apoptotic pathway was not involved in the degenerating process of cones in Rpe65-deficient retina. Conclusions:Altogether, these data show for the first time that a single genetic mutation can trigger two independent apoptotic pathways in rod and cone photoreceptors in LCA disease. While pro-apoptotic Bax is essential to trigger rod photoreceptor apoptosis, early degeneration of cones is not dependent on Bax-mediated apoptotic pathway in Rpe65-deficientmice.

Hypoxia is an essential component of tumor microenvironment. In this study, we investigated the influence of hypoxia (1% PO(2)) on CTL-mediated tumor cell lysis. We demonstrate that exposure of target tumor cells to hypoxia has an inhibitory effect on the CTL clone (Heu171)-induced autologous target cell lysis. Such inhibition correlates with hypoxia-inducible factor-1alpha (HIF-1alpha) induction but is not associated with an alteration of CTL reactivity as revealed by granzyme B polarization or morphological change. Western blot analysis indicates that although hypoxia had no effect on p53 accumulation, it induced the phosphorylation of STAT3 in tumor cells by a mechanism at least in part involving vascular endothelial growth factor secretion. We additionally show that a simultaneous nuclear translocation of HIF-1alpha and phospho-STAT3 was observed. Interestingly, gene silencing of STAT3 by small interfering RNA resulted in HIF-1alpha inhibition and a significant restoration of target cell susceptibility to CTL-induced killing under hypoxic conditions by a mechanism involving at least in part down-regulation of AKT phosphorylation. Moreover, knockdown of HIF-1alpha resulted in the restoration of target cell lysis under hypoxic conditions. This was further supported by DNA microarray analysis where STAT3 inhibition resulted in a partly reversal of the hypoxia-induced gene expression profile. The present study demonstrates that the concomitant hypoxic induction of phospho-STAT3 and HIF-1alpha are functionally linked to the alteration of non-small cell lung carcinoma target susceptibility to CTL-mediated killing. Considering the eminent functions of STAT3 and HIF-1alpha in the tumor microenvironment, their targeting may represent novel strategies for immunotherapeutic intervention.

O sistema plantio direto (SPD), em função de seu tempo de estabelecimento, pode promover aumento na quantidade de resíduos vegetais adicionados à superfície do solo e, consequentemente, modificações nos seus atributos químicos e físicos. O trabalho teve por objetivo quantificar a deposição de resíduos vegetais na superfície do solo (RVS) e as modificações nos teores de matéria orgânica leve (MOL), nos estoques de carbono e nitrogênio, nos teores de fósforo remanescente (Prem) e nos atributos físicos do solo - densidade de partículas (Dp), densidade do solo (Ds) e volume total de poros (VTP) -, bem como avaliar a origem do carbono por meio de técnicas isotópicas (13C). Para isso, em Montividiu (GO) foram selecionadas áreas sob SPD com diferentes tempos de implantação: SPD com três anos de implantação (SPD3), SPD com 15 anos de implantação (SPD15) e SPD com 20 anos de implantação (SPD20), as quais foram comparadas a uma área de Cerrado nativo stricto sensu (CE) e a uma área de pastagem plantada de Brachiaria decumbens (PA). Em cada uma das áreas, foram coletadas amostras nas profundidades de 0-5, 5-10 e 10-20 cm. O solo das áreas de estudo foi classificado como Latossolo Vermelho distroférrico. O delineamento utilizado foi inteiramente casualizado. Foi observado aumento nos teores de RVS, MOL, VTP, Prem, C e N em função do tempo de implantação do SPD. A área de SPD com 20 anos apresentou maiores valores de C e N e valores semelhantes de Ds e MOL, em relação ao CE. As análises de 13C demonstraram que as leguminosas estão contribuindo de forma significativa para a composição da matéria orgância nas áreas sob SPD. Nas áreas sob SPD, verificou-se aumento dos valores de estoque de C e de N em função do tempo de implantação em todas as profundidades analisadas; as áreas SPD15 e SPD20 apresentaram nas camadas superficiais valores semelhantes e, ou, superiores aos da área de CE. A área de PA apresentou os piores valores dos atributos analisados, demonstrando estar em estádio mais avançado de degradação.

No Brasil, boa parte dos plantios florestais tem sido realizada em solos que apresentam algum grau de degradação física, química ou biológica. A proteção ao solo fornecida pelas árvores, em função da deposição de serapilheira, constitui um dos processos de recuperação de áreas degradadas, e, como a matéria orgânica está relacionada com muitos dos atributos físicos, químicos e biológicos do solo, ela se caracteriza como um fator-chave na manutenção da qualidade ambiental de sistemas florestais. Assim, objetivou-se neste trabalho caracterizar frações da matéria orgânica em solos sob cultivo mínimo de eucalipto com diferentes históricos de uso e posições na paisagem. O estudo foi realizado em três hortos florestais (HFs): Terra Dura, Canafístula e Sanga das Pedras, cultivados com eucalipto no Estado do Rio Grande do Sul. Os solos foram amostrados nas profundidades de 0-5, 5-20 e 20-40 cm. Na profundidade de 0-5 cm, a maioria dos Argissolos dos HFs estudados apresentou teores de carbono orgânico (CO) superiores aos de matas nativas adjacentes. Nas áreas cultivadas, há maior armazenamento de CO no HF Canafístula, sendo notados, em geral, menores teores de CO no terço inferior da encosta em relação às outras posições das topossequências avaliadas. O índice de humificação, em geral, é maior nas áreas de mata em comparação às áreas cultivadas; contudo, ele varia em função da profundidade do solo, da posição amostrada na topossequência e do HF avaliado. Nas profundidades de 0-5 e 5-20 cm, a abundância natural de 13C dos HFs é similar à das áreas de mata nativa, porém há maior teor de 13C na camada mais profunda dos solos cultivados, o que sinaliza para a contribuição à matéria orgânica do solo (MOS) de C oriundo das pastagens que antecederam o eucalipto.

Systemic mastocytoses represent neoplastic proliferations of mast cells. In about 20% of cases systemic mastocytoses are accompanied by clonal haematopoietic non-mast cell-lineage disorders, most commonly myeloid neoplasms. A case of systemic mastocytosis carrying the characteristic mutation at codon 816 (D816V) in the KIT gene of mast cells, with two concurrent accompanying clonal haematopoietic non-mast cell-lineage disorders, chronic myeloproliferative disease, unclassifiable and precursor B lymphoblastic leukaemia is documented. Both accompanying clonal haematopoietic non-mast cell-lineage disorders carried the wild-type KIT gene, but had a novel t(13;13)(q12;q22) involving the FLT3 locus at 13q12. The chronic myeloproliferative disease, unclassifiable and the precursor B lymphoblastic leukaemia were cured by syngenous stem cell transplantation, but the systemic mastocytosis persisted for more than 10 years. The additional impact of molecular techniques on the correct diagnosis in haematological malignancies is highlighted, and evidence is provided that, apart from internal tandem duplications and mutations, FLT3 can be activated by translocations.

A matéria orgânica do solo (MOS) é um dos grandes reservatórios de carbono (C) da Terra e constitui um dos principais componentes do ciclo do C. Turfeiras, ambientes acumuladores de MOS, são produto da decomposição de vegetais, que se desenvolvem e se acumulam em ambientes saturados com água, sendo o estádio inicial da sequência de carbonificação. A fitomassa participa de forma marcante no ciclo global do C, armazenando em torno de 85 % de todo o C terrestre acima do solo. O tecido vegetal é composto principalmente por lignina, celulose e hemicelulose, constituindo até 85 % da biomassa seca. As plantas discriminam C de forma diferenciada, em razão de seu ciclo fotossintético (C3, C4 e CAM). As turfeiras da Serra do Espinhaço Meridional (SdEM-MG) são colonizadas por vegetação de Campo Limpo Úmido (CLU) e de Floresta Estacional Semidecidual (FES), onde ocorrem espécies dos ciclos fotossintéticos C3 e C4. Este trabalho objetivou avaliar a contribuição dessas duas fitofisionomias para o acúmulo de MOS, por meio da avaliação da fitomassa e da composição lignocelulósica e isotópica da vegetação e da MOS. A turfeira estudada localiza-se na SdEM e ocupa 81,75 ha. Para a estimativa da fitomassa do CLU e da FES, foram marcadas três parcelas de 0,5 x 0,5 m em cada fitofisionomia, onde todos os indivíduos da parcela foram cortados e armazenados. Para as análises isotópicas e lignocelulósicas da vegetação, identificaram-se as espécies dominantes em cada fitofisionomia. Amostras de solo foram coletadas em três locais representativos sob cada fitofisionomia, a cada 5 cm de profundidade, até 50 cm. Foram extraídas a celulose e a lignina das folhas das 15 espécies dominantes e das 60 amostras de turfeira para quantificação e determinação dos valores de δ13C e δ15N. Para datação da MOS, o 14C foi determinado em três profundidades, sob o CLU e a FES. A produção da fitomassa da FES foi muito superior à produção da do CLU. Os sinais isotópicos e a composição lignocelulósica da vegetação e da matéria orgânica do solo evidenciaram que a turfeira foi formada pela deposição de matéria orgânica da vegetação que a coloniza. O crescimento vertical e a taxa de acúmulo de C foram muito mais elevados sob a FES do que sob o CLU.

Grande parte da matéria orgânica de Organossolos das turfeiras é composta por substâncias húmicas, formadas pela transformação de resíduos orgânicos pelos microrganismos do solo e pela polimerização dos compostos orgânicos em macromoléculas resistentes à degradação biológica. Os processos de humificação da matéria orgânica do solo (MOS) ainda são pouco compreendidos e o conhecimento sobre os precursores das substâncias húmicas é limitado, sendo apresentadas rotas diferentes para a formação dessas substâncias. Contudo, em todas as rotas, destaca-se a participação da lignina. Isótopos estáveis (13C, 15N) podem ser utilizados para rastrear processos de humificação da MOS, por meio da identificação de seus precursores. Este trabalho teve como objetivo avaliar comparativamente a composição isotópica da vegetação das fitofisionomias que colonizam uma turfeira tropical de altitude composta de Campo Limpo Úmido (CLU) e de Floresta Estacional Semidecidual (FES), em relação à composição isotópica das substâncias húmicas da MOS. A turfeira estudada ocupa 81,75 ha. Para as análises isotópicas e lignocelulósicas da vegetação, foram identificadas as espécies dominantes em cada fitofisionomia. Amostras de solo foram coletadas em três locais representativos sob cada fitofisionomia, a cada 5 cm de profundidade, até 50 cm. As substâncias húmicas dessas amostras foram fracionadas, assim como calculados os valores de δ13C e δ15N nas frações húmicas, respectivamente a partir da determinação dos isótopos estáveis 12C e 13C e 14N e 15N. Os teores de lignina e seus valores de δ13C são mais elevados na vegetação e MOS sob FES em relação à vegetação e MOS sob CLU. Os teores de humina são mais elevados entre as substâncias húmicas na MOS, sob as duas fitofisionomias; os de ácidos húmicos são mais elevados na MOS sob CLU, em relação à FES; e os de ácidos fúlvicos são mais elevados na MOS sob a FES, em relação ao CLU. O δ13C da lignina apresenta similaridade elevada em relação ao δ13C da humina, dos ácidos húmicos e dos ácidos fúlvicos. As variações na composição lignocelulósica das espécies que colonizam o CLU e a FES promovem diferenças nas taxas e nos produtos da humificação da MOS.

Six healthy human subjects were studied during three 75-g oral, [13C]glucose tolerance tests to assess the kinetics of dexamethasone-induced impairment of glucose tolerance. On one occasion, they received dexamethasone (4 x 0.5 mg/day) during the previous 2 days. On another occasion, they received a single dose (0. 5 mg) of dexamethasone 150 min before ingestion of the glucose load. On the third occasion, they received a placebo. Postload plasma glucose was significantly increased after both 2 days dexamethasone and single dose dexamethasone compared with control (P < 0.05). This corresponded to a 20-23% decrease in the metabolic clearance rate of glucose, whereas total glucose turnover ([6,6-2H]glucose), total (indirect calorimetry) and exogenous glucose oxidation (13CO2 production), and suppression of endogenous glucose production were unaffected by dexamethasone. Plasma insulin concentrations were increased after 2 days of dexamethasone but not after a single dose of dexamethasone. In a second set of experiments, the effect of a single dose of dexamethasone on insulin sensitivity was assessed in six healthy humans during a 2-h euglycemic hyperinsulinemic clamp. Dexamethasone did not significantly alter insulin sensitivity. It is concluded that acute administration of dexamethasone impairs oral glucose tolerance without significantly decreasing insulin sensitivity.

We examined the role of heterotrimeric G protein signaling components in insulin and insulin-like growth factor I (IGF-I) action. In HIRcB cells and in 3T3L1 adipocytes, treatment with the Galpha(i) inhibitor (pertussis toxin) or microinjection of the Gbetagamma inhibitor (glutathione S-transferase-betaARK) inhibited IGF-I and lysophosphatidic acid-stimulated mitogenesis but had no effect on epidermal growth factor (EGF) or insulin action. In basal state, Galpha(i) and Gbeta were associated with the IGF-I receptor (IGF-IR), and after ligand stimulation the association of IGF-IR with Galpha(i) increased concomitantly with a decrease in Gbeta association. No association of Galpha(i) was found with either the insulin or EGF receptor. Microinjection of anti-beta-arrestin-1 antibody specifically inhibited IGF-I mitogenic action but had no effect on EGF or insulin action. beta-Arrestin-1 was associated with the receptors for IGF-I, insulin, and EGF in a ligand-dependent manner. We demonstrated that Galpha(i), betagamma subunits, and beta-arrestin-1 all play a critical role in IGF-I mitogenic signaling. In contrast, neither metabolic, such as GLUT4 translocation, nor mitogenic signaling by insulin is dependent on these protein components. These results suggest that insulin receptors and IGF-IRs can function as G protein-coupled receptors and engage different G protein partners for downstream signaling.

The precise mechanisms underlying the interaction between intestinal bacteria and the host epithelium lead to multiple consequences that remain poorly understood at the molecular level. Deciphering such events can provide valuable information as to the mode of action of commensal and probiotic microorganisms in the gastrointestinal environment. Potential roles of such microorganisms along the privileged target represented by the mucosal immune system include maturation prior, during and after weaning, and the reduction of inflammatory reactions in pathogenic conditions. Using human intestinal epithelial Caco-2 cell grown as polarized monolayers, we found that association of a Lactobacillus or a Bifidobacterium with nonspecific secretory IgA (SIgA) enhanced probiotic adhesion by a factor of 3.4-fold or more. Bacteria alone or in complex with SIgA reinforced transepithelial electrical resistance, a phenomenon coupled with increased phosphorylation of tight junction proteins zonula occludens-1 and occludin. In contrast, association with SIgA resulted in both enhanced level of nuclear translocation of NF-κB and production of epithelial polymeric Ig receptor as compared with bacteria alone. Moreover, thymic stromal lymphopoietin production was increased upon exposure to bacteria and further enhanced with SIgA-based complexes, whereas the level of pro-inflammatory epithelial cell mediators remained unaffected. Interestingly, SIgA-mediated potentiation of the Caco-2 cell responsiveness to the two probiotics tested involved Fab-independent interaction with the bacteria. These findings add to the multiple functions of SIgA and underscore a novel role of the antibody in interaction with intestinal bacteria.

5-aminosalicylic acid (5-ASA) is an antiinflammatory drug widely used in the treatment of inflammatory bowel diseases. It is known to inhibit the production of cytokines and inflammatory mediators, but the mechanism underlying the intestinal effects of 5-ASA remains unknown. Based on the common activities of peroxisome proliferator-activated receptor-gamma (PPAR-gamma) ligands and 5-ASA, we hypothesized that this nuclear receptor mediates 5-ASA therapeutic action. To test this possibility, colitis was induced in heterozygous PPAR-gamma(+/-) mice and their wild-type littermates, which were then treated with 5-ASA. 5-ASA treatment had a beneficial effect on colitis only in wild-type and not in heterozygous mice. In epithelial cells, 5-ASA increased PPAR-gamma expression, promoted its translocation from the cytoplasm to the nucleus, and induced a modification of its conformation permitting the recruitment of coactivators and the activation of a peroxisome-proliferator response element-driven gene. Validation of these results was obtained with organ cultures of human colonic biopsies. These data identify PPAR-gamma as a target of 5-ASA underlying antiinflammatory effects in the colon.
