The FOB-3, anew type fiber optic biosensor, is designed to rapidly detect a variety of biological agents or analytes with better stability, sensitivity and specificity. In order to detect Y. Pestis, a sandwich immunoassay was developed by using the purified antibody against antigen FI immobilized on polystyrene probes as the capture antibody and the monoclonal antibody-Cy5 conjugate as the detector. After a series of optimization for the stability, sensitivity and specificity of the FOB-3, 50-1000 ng/ml of antigen FI and 6 x 10(1)-6 x 10(7) CFU/ml Y. pestis could be detected constantly in about 20 min, and Y pestis could be detected specifically from Y. pseudotuberculosis, Y. enterocolitica, B. anthracis and E. coli. Then, 39 blind samples, including 27 tissues of mice infected with Y pestis and 12 tissues of healthy mice as negative control, were detected with the FOB-3. 92.6% infected tissues were identified from the tissues of healthy mice and the tissues containing more than 100 CFU/ml bacteria could be detected by the biosensor. The results demonstrated the feasibility of the FOB-3 as an effective method to detect Y. pestis rapidly and directly from the infected animal specimens with the advantage of portability, simple-operation as well as high sensitivity and specificity. The aim of the study was to evaluate the resistance of white spot syndrome virus (WSSV) in shrimps (Penaeus monodon) to the process of cooking. The cooking was carried out at 1000C six different durations 5, 10, 15, 20, 25 and 30 min. The presence of WSSV was tested by single step and nested polymerase chain reaction (PCR). In the single step PCR, the primers 1s5 & 1a16 and IK1 & IK2 were used. While in the nested PCR, primers IK1 &IK2 – IK3 & IK4 were used for the detection of WSSV. WSSV was detected in the single step PCR with the primers 1s5 and 1a16 and the nested PCR with the primers IK1 and IK2 – IK3 & IK4 from the cooked shrimp samples. The cooked shrimps, which gave positive results for WSSV by PCR, were further confirmed for the viability of WSSV by conducting the bio-inoculation studies. Mortality (100%) was observed within 123 h of intra-muscular post injection (P.I) into the live healthy WSSV-free shrimps (P. monodon). These results show that the WSSV survive the cooking process and even infected cooked shrimp products may pose a transmission risk for WSSV to the native shrimp farming systems.

The toxicity of the herbicide glyphosate was tested on water hyacinth (Eichhornia crassipes) samples cultivated in glass aquariums. The lowest dose (0.09 g.m-2) leads to an increasing plant growth rate. This growth rate decreases with intermediate doses (0.18 and 0.36 g.m-2), the consequence of which is to increase stolons (vegetative reproduction). On the other hand, the dose of 0.72 g.m-2 leads to a total and irreversible destruction of plants.

Ichthyoplankton surveys have been used to provide an independent estimate of adult spawning biomass of commercially exploited species and to further our understanding of the recruitment processes in the early life stages. However, predicting recruitment has been difficult because of the complex interaction of physical and biological processes operating at different spatial and temporal scales that can occur at the different life stages. A model of first-year life-stage recruitment was applied to Georges Bank Atlantic cod (Gadus morhua) and haddock (Melanogrammus aeglefinus) stocks over the years 1977–2004 by using environmental and densitydependent relationships. The best lifestage mortality relationships for eggs, larvae, pelagic juveniles, and demersal juveniles were first determined by hindcasting recruitment estimates based on egg and larval abundance and mortality rates derived from two intensive sampling periods, 1977–87 and 1995–99. A wind-driven egg mortality relationship was used to estimate losses due to transport off the bank, and a wind-stress larval mortality relationship was derived from feeding and survival studies. A simple metric for the density-dependent effects of Atlantic cod was used for both Atlantic cod and haddock. These life stage proxies were then applied to the virtual population analysis (VPA) derived annual egg abundances to predict age-1 recruitment. Best models were determined from the correlation of predicted and VPA-derived age-1 abundance. The larval stage was the most quantifiable of any stage from surveys, whereas abundance estimates of the demersal juvenile stage were not available because of undersampling. Attempts to forecast recruitment from spawning stock biomass or egg abundance, however, will always be poor because of variable egg survival.

A case study of the reproductive biology of the endemic Hawaiian grouper or hapu’upu’u (Hyporthodus quernus) is presented as a model for comprehensive future studies of economically important epinephelid groupers. Specimens were collected throughout multiple years (1978–81, 1992–93, and 2005–08) from most reefs and banks of the Northwestern Hawaiian Islands. The absence of small males, presence of atretic oocytes and brown bodies in testes of mature males, and both developed ovarian and testicular tissues in the gonads of five transitional fish provided evidence of protogynous hermaphroditism. No small mature males were collected, indicating that Hawaiian grouper are monandrous (all males are sex-changed females). Complementary microscopic criteria also were used to assign reproductive stage and estimate median body sizes (L50) at female sexual maturity and at adult sex change from female to male. The L50 at maturation and at sex change was 580 ±8 (95% confidence interval [CI]) mm total length (TL) and 895 ±20 mm TL, respectively. The adult sex ratio was strongly female biased (6:1). Spawning seasonality was described by using gonadosomatic indices. Females began ripening in the fall and remained ripe through April. A February–June main spawning period that followed peak ripening was deduced from the proportion of females whose ovaries contained hydrated oocytes, postovulatory follicles, or both. Testes weights were not affected by season; average testes weight was only about 0.2% of body weight—an order of magnitude smaller than that for ovaries that peaked at 1–3% of body weight. The species’ reproductive life history is discussed in relation to its management.

We evaluated the conservation benefits of the use of circle hooks compared with standard J hooks in the recreational fishery for Atlantic istiophorid billfishes, noting hooking location and the presence of trauma (bleeding) for 123 blue marlin (Makaira nigricans), 272 white marlin (Kajikia albida), and 132 sailfish (Istiophorus platypterus) caught on natural baits rigged with one of the two hook types. In addition, we used pop-up satellite archival tags (PSATs) to follow the fate of 61 blue marlin caught on natural baits rigged with circle hooks or on a combination of artificial lure and natural bait rigged with J hooks. The frequencies of internal hooking locations and bleeding were significantly lower with circle hooks than with J hooks for each of the three species and were significantly reduced for blue marlin caught on J hooks than for white marlin and sailfish taken on the same hook type. Analysis of the data received from 59 PSATs (two tags released prematurely) indicated no mortalities among the 29 blue marlin caught on circle hooks and two mortalities among the 30 blue marlin caught on J hooks (6.7%). Collectively, the hook location and PSAT data revealed that blue marlin, like white marlin and sailfish, derive substantial conservation benefits from the use of circle hooks, and the negative impacts of J hooks are significantly reduced for blue marlin relative to the other two species.
