Los recientes desarrollos tecnológicos permiten la transición de la oceanografía observacional desde un concepto basado en buques a uno basado en sistemas autónomos en red. Este último, propone que la forma más eficiente y efectiva de observar el océano es con una red de plataformas autónomas distribuidas espacialmente y complementadas con sistemas de medición remota. Debido a su maniobrabilidad y autonomía, los planeadores submarinos están jugando un papel relevante en este concepto de observaciones en red. Los planeadores submarinos fueron específicamente diseñados para muestrear vastas zonas del océano. Estos son robots con forma de torpedo que hacen uso de su forma hidrodinámica, alas y cambios de flotabilidad para generar movimientos horizontales y verticales en la columna de agua. Un sensor que mide conductividad, temperatura y profundidad (CTD) constituye un equipamiento estándar en la plataforma. Esto se debe a que ciertas variables dinámicas del Océano se pueden derivar de la temperatura, profundidad y salinidad. Esta última se puede estimar a partir de las medidas de temperatura y conductividad. La integración de sensores CTD en planeadores submarinos no esta exenta de desafíos. Uno de ellos está relacionado con la precisión de los valores de salinidad derivados de las muestras de temperatura y conductividad. Específicamente, las estimaciones de salinidad están significativamente degradadas por el retardo térmico existente, entre la temperatura medida y la temperatura real dentro de la celda de conductividad del sensor. Esta deficiencia depende de las particularidades del flujo de entrada al sensor, su geometría y, también se ha postulado, del calor acumulado en las capas de aislamiento externo del sensor. Los efectos del retardo térmico se suelen mitigar mediante el control del flujo de entrada al sensor. Esto se obtiene generalmente mediante el bombeo de agua a través del sensor o manteniendo constante y conocida su velocidad. Aunque recientemente se han incorporado sistemas de bombeo en los CTDs a bordo de los planeadores submarinos, todavía existen plataformas equipadas con CTDs sin dichos sistemas. En estos casos, la estimación de la salinidad supone condiciones de flujo de entrada al sensor, razonablemente controladas e imperturbadas. Esta Tesis investiga el impacto, si existe, que la hidrodinámica de los planeadores submarinos pudiera tener en la eficiencia de los sensores CTD. Específicamente, se investiga primero la localización del sensor CTD (externo al fuselaje) relativa a la capa límite desarrollada a lo largo del cuerpo del planeador. Esto se lleva a cabo mediante la utilización de un modelo acoplado de fluido no viscoso con un modelo de capa límite implementado por el autor, así como mediante un programa comercial de dinámica de fluidos computacional (CFD). Los resultados indican, en ambos casos, que el sensor CTD se encuentra fuera de la capa límite, siendo las condiciones del flujo de entrada las mismas que las del flujo sin perturbar. Todavía, la velocidad del flujo de entrada al sensor CTD es la velocidad de la plataforma, la cual depende de su hidrodinámica. Por tal motivo, la investigación se ha extendido para averiguar el efecto que la velocidad de la plataforma tiene en la eficiencia del sensor CTD. Con este propósito, se ha desarrollado un modelo en elementos finitos del comportamiento hidrodinámico y térmico del flujo dentro del CTD. Los resultados numéricos indican que el retardo térmico, atribuidos originalmente a la acumulación de calor en la estructura del sensor, se debe fundamentalmente a la interacción del flujo que atraviesa la celda de conductividad con la geometría interna de la misma. Esta interacción es distinta a distintas velocidades del planeador submarino. Específicamente, a velocidades bajas del planeador (0.2 m/s), la mezcla del flujo entrante con las masas de agua remanentes en el interior de la celda, se ralentiza debido a la generación de remolinos. Se obtienen entonces desviaciones significantes entre la salinidad real y aquella estimada. En cambio, a velocidades más altas del planeador (0.4 m/s) los procesos de mezcla se incrementan debido a la turbulencia e inestabilidades. En consecuencia, la respuesta del sensor CTD es mas rápida y las estimaciones de la salinidad mas precisas que en el caso anterior. Para completar el trabajo, los resultados numéricos se han validado con pruebas experimentales. Específicamente, se ha construido un modelo a escala del sensor CTD para obtener la confirmación experimental de los modelos numéricos. Haciendo uso del principio de similaridad de la dinámica que gobierna los fluidos incompresibles, los experimentos se han realizado con flujos de aire. Esto simplifica significativamente la puesta experimental y facilita su realización en condiciones con medios limitados. Las pruebas experimentales han confirmado cualitativamente los resultados numéricos. Más aun, se sugiere en esta Tesis que la respuesta del sensor CTD mejoraría significativamente añadiendo un generador de turbulencia en localizaciones adecuadas al interno de la celda de conductividad. ABSTRACT Recent technological developments allow the transition of observational oceanography from a ship-based to a networking concept. The latter suggests that the most efficient and effective way to observe the Ocean is through a fleet of spatially distributed autonomous platforms complemented by remote sensing. Due to their maneuverability, autonomy and endurance at sea, underwater gliders are already playing a significant role in this networking observational approach. Underwater gliders were specifically designed to sample vast areas of the Ocean. These are robots with a torpedo shape that make use of their hydrodynamic shape, wings and buoyancy changes to induce horizontal and vertical motions through the water column. A sensor to measure the conductivity, temperature and depth (CTD) is a standard payload of this platform. This is because certain ocean dynamic variables can be derived from temperature, depth and salinity. The latter can be inferred from measurements of temperature and conductivity. Integrating CTD sensors in glider platforms is not exempted of challenges. One of them, concerns to the accuracy of the salinity values derived from the sampled conductivity and temperature. Specifically, salinity estimates are significantly degraded by the thermal lag response existing between the measured temperature and the real temperature inside the conductivity cell of the sensor. This deficiency depends on the particularities of the inflow to the sensor, its geometry and, it has also been hypothesized, on the heat accumulated by the sensor coating layers. The effects of thermal lag are usually mitigated by controlling the inflow conditions through the sensor. Controlling inflow conditions is usually achieved by pumping the water through the sensor or by keeping constant and known its diving speed. Although pumping systems have been recently implemented in CTD sensors on board gliders, there are still platforms with unpumped CTDs. In the latter case, salinity estimates rely on assuming reasonable controlled and unperturbed flow conditions at the CTD sensor. This Thesis investigates the impact, if any, that glider hydrodynamics may have on the performance of onboard CTDs. Specifically, the location of the CTD sensor (external to the hull) relative to the boundary layer developed along the glider fuselage, is first investigated. This is done, initially, by applying a coupled inviscid-boundary layer model developed by the author, and later by using a commercial software for computational fluid dynamics (CFD). Results indicate, in both cases, that the CTD sensor is out of the boundary layer, being its inflow conditions those of the free stream. Still, the inflow speed to the CTD sensor is the speed of the platform, which largely depends on its hydrodynamic setup. For this reason, the research has been further extended to investigate the effect of the platform speed on the performance of the CTD sensor. A finite element model of the hydrodynamic and thermal behavior of the flow inside the CTD sensor, is developed for this purpose. Numerical results suggest that the thermal lag effect is mostly due to the interaction of the flow through the conductivity cell and its geometry. This interaction is different at different speeds of the glider. Specifically, at low glider speeds (0.2 m/s), the mixing of recent and old waters inside the conductivity cell is slowed down by the generation of coherent eddy structures. Significant departures between real and estimated values of the salinity are found. Instead, mixing is enhanced by turbulence and instabilities for high glider speeds (0.4 m/s). As a result, the thermal response of the CTD sensor is faster and the salinity estimates more accurate than for the low speed case. For completeness, numerical results have been validated against model tests. Specifically, a scaled model of the CTD sensor was built to obtain experimental confirmation of the numerical results. Making use of the similarity principle of the dynamics governing incompressible fluids, experiments are carried out with air flows. This significantly simplifies the experimental setup and facilitates its realization in a limited resource condition. Model tests qualitatively confirm the numerical findings. Moreover, it is suggested in this Thesis that the response of the CTD sensor would be significantly improved by adding small turbulators at adequate locations inside the conductivity cell.

Habitualmente en los ensayos de barcos amarrados se reproduce la acción del oleaje. Se ha diseñado un sistema para la reproducción del efecto del viento en estos ensayos, utilizando motores, poleas, hilos de conexión, muelles, galgas y controladoras, de forma que la fuerza ejercida por el viento, previamente calculada considerando las características del barco y del viento, se transmite al barco mediante los elementos que componen el sistema. Este sistema consiste en un circuito cerrado de transmisión de fuerza con un motor que simultáneamente actúa sobre dos muelles deformándolos en sentidos opuestos, los cuales transmiten las fuerzas en los costados del buque, siendo la diferencia del alargamiento de los muelles la fuerza resultante sobre el barco. El sistema se aplicó a un buque crucero, obteniendo unos resultados que muestran la importancia de tener en cuenta el viento racheado como un agente adicional al oleaje que provoca movimientos y esfuerzos en las amarras y defensas elevados, dependiendo de su intensidad y dirección. Se ensayaron tres condiciones: modelo alimentado únicamente con olas, solo con viento y con ambos. En cada ensayo realizado se registraron los movimientos del buque, las fuerzas en las amarras y las reacciones en las defensas. La aportación de la Tesis es un sistema mecánico para excitar el modelo del buque amarrado con viento racheado superpuesto a la acción del oleaje. Evidentemente los resultados que se obtengan se ajustarán más a la situación real que reproduciendo sólo el oleaje. ABSTRACT Traditionally, moored ship tests with small-scale models only take into account the disturbance effect of waves. In this thesis, the design and testing of a system also implementing the effect of wind in moored ships is analyzed. The system is based on rotatory actuators acting on linear springs. This solution has a swift enough response to reproduce the fluctuating component of the wind. Three scenarios have been tested: waves, wind and combination of both. In order to assess the results, different sensors are connected to a computer for data acquisition, allowing the recording and subsequent analysis of the measured variables (forces in ropes, reactions in fenders and ship motions). The results obtained from the experiments show a great impact when wind effect is considered. A superposition effect is observed when waves and wind act together on the ship, emphasizing therefore the importance of taking the wind into account in berthed vessel tests, achieving safer and more realistic results.

The French CEA, together with EDF and the IAEA, recently organised an international benchmark to evaluate the ability to model the mechanical behaviour of a typical nuclear reinforced concrete structure subjected to seismic demands. The participants were provided with descriptions of the structure and the testing campaign; they had to propose the numerical model and the material laws for the concrete (stage #1). A mesh of beam and shell elements was generated; for modelling the concrete a damaged plasticity model was used, but a smeared crack model was also investigated. Some of the initial experimental results, with the mock-up remaining in the elastic range, were provided to the participants for calibrating their models (stage #2). Predictions had to be produced in terms of eigen-frequencies and motion time histories. The calculated frequencies reproduced reasonably the experimental ones; the time histories, calculated by modal response analysis, also reproduced adequately the observed amplifications. The participants were then expected to predict the structural response under strong ground motions (stage #3), which increased progressively up to a history recorded during the 1994 Northridge earthquake, followed by an aftershock. These results were produced using an explicit solver and a damaged plasticity model for the concrete, although an implicit solver with a smeared crack model was also investigated. The paper presents the conclusions of the pre-test exercise, as well as some observations from additional simulations conducted after the experimental results were made available.

Estudo sobre a história política do primeiro presidente do Brasil, Manoel Deodoro da Fonseca, na ótica da propaganda política. Os objetivos foram resgatar os materiais de propaganda utilizados pelo movimento republicano que culminou com a Proclamação da República chefiada por Deodoro em 15 de novembro de 1889. Posteriormente, compreender o processo eleitoral ao qual o generalíssimo foi escolhido presidente pelos congressistas, por meio do voto indireto, no dia 25 de fevereiro de 1891. A metodologia adotada foi a Pesquisa Histórica enfatizando o estudo bibliográfico e documental. Também foram descritas as charges publicadas na Revista Illustrada no Governo Provisório, a cobertura do jornal O País na eleição presidencial e o cenário de censura na imprensa da época. Na conclusão consta que Deodoro não fez campanha eleitoral. Nem precisou, pois caso não fosse eleito, o Exército o proclamaria ditador. No entanto, os deodoristas convenceram os deputados e senadores a votarem no marechal. Já, a oposição fez campanha política lançando edição extra de jornal, espalhando cartazes pela Capital Federal e aprovando moções de repúdio à candidatura da situação. Essa eleição foi simplesmente uma formalidade para manter Deodoro no cargo mais importante do país. (AU)

Este estudo pretendeu abordar o assunto de forma a concentrar-se na investigação, com um estudo de caso em uma empresa pública de saneamento ambiental da região do grande ABC paulista, o SEMASA, com o intuito de avaliar as mudanças nas rotinas operacionais dos diversos departamentos da empresa e o impacto causado pela TI na satisfação das pessoas envolvidas. Com base na literatura sobre trabalho e sociedade na era da globalização e empresas públicas, foram analisados inicialmente os motivos que podem contribuir para as pessoas buscarem ingressar na carreira pública. Esse estudo verificou a importância da adoção de ferramentas de tecnologia nos processos produtivos e na prestação de serviços das organizações. Verificou também, que estimular os funcionários de empresas públicas a criarem mecanismos para facilitarem seus trabalhos, através de ferramentas de TI atualizadas, pode ser benéfico, ao mesmo tempo em que pode proporcionar condições destes adquirirem novas capacitações profissionais com a aquisição de novos conhecimentos tecnológicos. Os resultados obtidos apontaram para um significativo aumento na satisfação das pessoas envolvidas sendo estas estimuladas a estarem em constante aperfeiçoamento, através da realização de novos treinamentos e cursos de tecnologia.(AU)

Normal mammalian hearing is refined by amplification of the motion of the cochlear partition. This partition, comprising the organ of Corti sandwiched between the basilar and tectorial membranes, contains the outer hair cells that are thought to drive this amplification process. Force generation by outer hair cells has been studied extensively in vitro and in situ, but, to understand cochlear amplification fully, it is necessary to characterize the role played by each of the components of the cochlear partition in vivo. Observations of cochlear partition motion in vivo are severely restricted by its inaccessibility and sensitivity to surgical trauma, so, for the present study, a computer model has been used to simulate the operation of the cochlea under different experimental conditions. In this model, which uniquely retains much of the three-dimensional complexity of the real cochlea, the motions of the basilar and tectorial membranes are fundamentally different during in situ- and in vivo-like conditions. Furthermore, enhanced outer hair cell force generation in vitro leads paradoxically to a decrease in the gain of the cochlear amplifier during sound stimulation to the model in vivo. These results suggest that it is not possible to extrapolate directly from experimental observations made in vitro and in situ to the normal operation of the intact organ in vivo.

As a step toward understanding their functional role, the low frequency vibrational motions (<300 cm−1) that are coupled to optical excitation of the primary donor bacteriochlorophyll cofactors in the reaction center from Rhodobacter sphaeroides were investigated. The pattern of hydrogen-bonding interaction between these bacteriochlorophylls and the surrounding protein was altered in several ways by mutation of single amino acids. The spectrum of low frequency vibrational modes identified by femtosecond coherence spectroscopy varied strongly between the different reaction center complexes, including between different mutants where the pattern of hydrogen bonds was the same. It is argued that these variations are primarily due to changes in the nature of the individual modes, rather than to changes in the charge distribution in the electronic states involved in the optical excitation. Pronounced effects of point mutations on the low frequency vibrational modes active in a protein-cofactor system have not been reported previously. The changes in frequency observed indicate a strong involvement of the protein in these nuclear motions and demonstrate that the protein matrix can increase or decrease the fluctuations of the cofactor along specific directions.

The use of molecular genetics for introducing fluorescent molecules enables the use of donor–donor energy migration to determine intramolecular distances in a variety of proteins. This approach can be applied to examine the overall molecular dimensions of proteins and to investigate structural changes upon interactions with specific target molecules. In this report, the donor–donor energy migration method is demonstrated by experiments with the latent form of plasminogen activator inhibitor type 1. Based on the known x-ray structure of plasminogen activator inhibitor type 1, three positions forming the corners of a triangle were chosen. Double Cys substitution mutants (V106C-H185C, H185C-M266C, and M266C-V106C) and corresponding single substitution mutants (V106C, H185C, and M266C) were created and labeled with a sulfhydryl specific derivative of BODIPY (=the D molecule). The side lengths of this triangle were obtained from analyses of the experimental data. The analyses account for the local anisotropic order and rotational motions of the D molecules, as well as for the influence of a partial DD-labeling. The distances, as determined from x-ray diffraction, between the Cα-atoms of the positions V106C–H185C, H185C–M266C, and M266C–V106C were 60.9, 30.8, and 55.1 Å, respectively. These are in good agreement with the distances of 54 ± 4, 38 ± 3, and 55 ± 3 Å, as determined between the BODIPY groups attached via linkers to the same residues. Although the positions of the D-molecules and the Cα-atoms physically cannot coincide, there is a reasonable agreement between the methods.

The “3-color, 46-bead” model of a folding polypeptide is the vehicle for adapting to proteins a mode of analysis used heretofore for atomic clusters, to relate the topography of the potential surface to the dynamics that lead to formation of selected structures. The analysis is based on sequences of stationary points—successive minima, joined by saddles—that rise monotonically in energy from basin bottoms. Like structure-seeking clusters, the potential surface of the model studied here is staircase-like, rather than sawtooth-like, with highly collective motions required for passage from one minimum to the next. The surface has several deep basins whose minima correspond to very similar structures, but which are separated by high energy barriers.

The bacterial aspartate receptor was reconstructed to eliminate the transmembrane domain, thus connecting the recognition domain directly to the effector domain. The resulting soluble receptor folded correctly and was no longer an integral membrane protein. Upon aspartate binding, this soluble receptor was stabilized to a similar extent as that of the native receptor. Of interest, this soluble receptor retained the ability to signal from the recognition to the effector domain. This result defines more clearly the role of the membrane and transmembrane domains in signal transduction and suggests that some ligand-induced motions in receptor proteins do not require the membrane or transmembrane domain for information transmission.

The EPR spectra of spin-labeled lipid chains in fully hydrated bilayer membranes of dimyristoyl phosphatidylcholine containing 40 mol % of cholesterol have been studied in the liquid-ordered phase at a microwave radiation frequency of 94 GHz. At such high field strengths, the spectra should be optimally sensitive to lateral chain ordering that is expected in the formation of in-plane domains. The high-field EPR spectra from random dispersions of the cholesterol-containing membranes display very little axial averaging of the nitroxide g-tensor anisotropy for lipids spin labeled toward the carboxyl end of the sn-2 chain (down to the 8-C atom). For these positions of labeling, anisotropic 14N-hyperfine splittings are resolved in the gzz and gyy regions of the nonaxial EPR spectra. For positions of labeling further down the lipid chain, toward the terminal methyl group, the axial averaging of the spectral features systematically increases and is complete at the 14-C atom position. Concomitantly, the time-averaged 〈Azz〉 element of the 14N-hyperfine tensor decreases, indicating that the axial rotation at the terminal methyl end of the chains arises from correlated torsional motions about the bonds of the chain backbone, the dynamics of which also give rise to a differential line broadening of the 14N-hyperfine manifolds in the gzz region of the spectrum. These results provide an indication of the way in which lateral ordering of lipid chains in membranes is induced by cholesterol.

The electronic excitations of naphthalene and a family of bridged naphthalene dimers are calculated and analyzed by using the Collective Electronic Oscillator method combined with the oblique Lanczos algorithm. All experimentally observed trends in absorption profiles and radiative lifetimes are reproduced. Each electronic excitation is linked to the corresponding real-space transition density matrix, which represents the motions of electrons and holes created in the molecule by photon absorption. Two-dimensional plots of these matrices help visualize the degree of exciton localization and explain the dependence of the electronic interaction between chromophores on their separation.

The propagation of inhomogeneous, weakly nonlinear waves is considered in a cochlear model having two degrees of freedom that represent the transverse motions of the tectorial and basilar membranes within the organ of Corti. It is assumed that nonlinearity arises from the saturation of outer hair cell active force generation. I use multiple scale asymptotics and treat nonlinearity as a correction to a linear hydroelastic wave. The resulting theory is used to explain experimentally observed features of the response of the cochlear partition to a pure tone, including: the amplification of the response in a healthy cochlea vs a dead one; the less than linear growth rate of the response to increasing sound pressure level; and the amount of distortion to be expected at high and low frequencies at basal and apical locations, respectively. I also show that the outer hair cell nonlinearity generates retrograde waves.
