Net photosynthesis (A) and transpiration rates (E), stomatal conductance (g), water use efficiency (WUE), intrinsic water use efficiency (IWUE) and internal leaf CO2 concentration (C) in response to different vapor pressure deficit (1.2 and 2.5 kPa) were investigated in 'Pera' sweet orange plants affected by citrus variegated chlorosis (CVC), a disease caused by Xylella fastidiosa. All plants were well watered and leaf water potential (Pw) was also measured by the psychrometric technique. Results showed that healthy plants responded to higher vapor pressure deficit (VPD), lowering its net photosynthesis and transpiration rates, and stomatal conductance. However, diseased plants presented no clear response to VPD, showing lower A, E and g for both VPDs studied and very similar values to these variables in healthy plants at the highest VPD. Internal leaf CO2 concentration also decreased for healthy plants when under the highest VPD, and surprisingly, the same pattern of response was found in plants with CVC. These results, the lower Psi(w) and higher WUE values for diseased plants, indicated that this disease may cause stomatal dysfunction and affect the water resistance through xylem vessels, which ultimately may play some role in photosynthetic metabolism. Cordyline terminalis is a tropical ornamental with high decorative value due its variety of colors and foliar patterns. This ornamental is new in the Brazilian market and has large potential to be exported. The lack of studies on postharvest aspects is a problem for new crops, and the development of good post-production techniques can reduce costs due to loss of unsalable, damaged plants. The present work aimed to characterize the vase life of red and green cultivars of C. terminalis. Conservation techniques, such as pulsing with gibberellic acid (GA(3)), wax spray, and cold storage for 10 and 20 days at 5, 10, and 13 degrees C were tested. Also, macro-and micronutrients concentration was tracked during the senescence process. In the water-based treatment (control), both cultivars had a vase life of 21 days. Vase life was not influenced by wax spray, GA pulsing, or cold storage for 10 days at 5, 10 or 13 degrees C. In both cultivars, the postharvest techniques used did not improve the vase life on this species. on the other hand, by cold storing the foliage for 20 days in any of the tested temperatures, the vase life was shortened one week, with a total of 14 days longevity compared with all other treatments. Color changes were not correlated with senescence of the foliage. Nutrients did not change during the evaluation period. C. terminalis had a vase life long enough for export, supported by cold storage up to 10 days, which did not shorten the vase life.

Casearia sylvestris Sw. is a widespread neotropical tree utilized in popular medicine. Recent research ranked Casearia as one of the most promising genus in the search of drugs against cancer. Despite its wide distribution and pharmacological importance, no microsatellite markers have yet been developed for this genus. In this study, we provide 10 polymorphic microsatellite loci specifically designed for C. sylvestris, used to analyse 90 individuals distributed in two populations from São Paulo state, Brazil. on average, 12.3 alleles per locus were identified, showing the ability of the markers to detect microsatellite polymorphism in this species.

Polygalacturonases are part of the group of enzymes involved in pectin degradation. The aim of this work was to investigate some of the factors affecting polygalacturonase production by an Aspergillus giganteus strain and to characterize this pectinolytic activity. Several carbon sources, both pure substances and natural substrates, were tested in standing cultures, and the best results were obtained with orange bagasse and purified citrus pectin. on citrus pectin as sole carbon source, the highest extracellular activity (9.5 U/ml and 40.6 U/mg protein) was obtained in 4.5-day-old cultures shaken at 120 rpm, pH 3.5 and 30 degrees C, while on orange bagasse, the highest extracellular activity (48.5 U/ml and 78.3 U/mg protein) was obtained in 3.5-day-old cultures shaken at 120 rpm, pH 6.0 and 30 degrees C. Optimal polygalacturonase activity was observed in assays conducted at pH 5.5-6.5 and 55-60 degrees C. The activity showed good thermal stability, with half-lives of 90 and 30 min when incubated at 55 and 60 degrees C, respectively. High stability was observed from pH 4.5 to 8.5; more than 90% of the activity remained after 24 h in this pH range.

The influence of a population of the understorey woody bamboo Merostachys riedeliana and different flooding regimes on tree community dynamics in a section of tropical semideciduous forest in South-Eastern Brazil was examined. A forest section with an area of 1.6 ha composed of 71 adjacent plots was located on a slope ending at the river margin. The section was divided into five topographical sectors according to the mean duration of river floods. In 1991 and 1998 all trees with a diameter at the base of the trunk greater than or equal to 5 cm were measured, identified and tagged, and all live bamboo culms were counted. Annualised estimates of the rates of tree mortality and recruitment, gain and loss of tree basal area, and change in bamboo density were calculated for each of the 71 plots and five topographical sectors as well as for diameter classes and tree species. To segregate patterns arising from spatially autocorrelated events, geostatistical analyses were used prior to statistical comparisons and correlations. In general, mortality rates were not compensated by recruitment rates but there was a net increase in basal area in all sectors, suggesting that the tree community as a whole was in a building phase. Tree community dynamics of the point bar forest (Depression and Levee sectors) differed from that of the upland forest (Ridgetop, Middle Slope and Lower Slope sectors) in the extremely high rates of gain in basal area. The predominant and specialised species, Inga vera and Salix humboldtiana, are probably favoured by relaxed competition in an environment stressed by long-lasting floods. In the upland forest, mortality rates were highest at the Middle Slope, particularly for smaller trees, while recruitment rates were lowest. As bamboo clumps were concentrated in this sector, the locally higher instability in the tree community probably resulted from the direct interference of bamboos. The density of bamboo culms in the upland forest was negatively correlated with the rates of tree recruitment and gain in basal area, and positively correlated with tree mortality rates. Bamboos therefore seemed to restrict the recruitment, growth and survival of trees.

A estrutura horizontal e vertical do componente arbóreo foi investigada em um trecho de Floresta Atlântica baixo-montana através de um levantamento fitossociológico em dois blocos amostrais de 0,99 ha cada no Parque Estadual Intervales. Todos os indivíduos com DAP > 5 cm foram registrados. Foram amostrados 3.078 indivíduos distribuídos em 172 espécies. O índice de diversidade de Shannon foi de H' = 3,85 nat.ind.-1. A família Myrtaceae se destacou tanto em número de espécies (38) quanto em número de indivíduos (745) no levantamento. Euterpe edulis Mart. teve o maior valor de importância (33,98%), abrangendo 21,8% do total de indivíduos registrados. O índice de similaridade quantitativo foi maior do que o qualitativo, mostrando pouca variação estrutural entre os blocos amostrais, mas a grande quantidade de espécies pouco abundantes, resultou em pronunciadas diferenças florísticas entre eles. Uma análise de correspondência retificada (DCA) gerou três estratos verticais arbitrários. O estrato A (> 26 m) teve a menor densidade e foi bem representado pelas espécies Sloanea guianensis (Aubl.) Benth. e Virola bicuhyba (Schott. ex A.DC.) Warb. O estrato B (8 m < h < 26 m) mostrou a maior riqueza e diversidade florística, e o estrato C (< 8 m) a maior densidade. Euterpe edulis, Guapira opposita (Vell.) Reitz, Garcinia gardneriana (Planch. & Triana) Zappi e Eugenia mosenii (Kausel) Sobral foram bem representadas nos estratos B e C da floresta. A existência de estratos verticais em florestas tropicais é discutida, recomendando-se o uso da DCA para estudos da estratificação vertical em outras florestas tropicais.

Seed dispersal by animals is a complex process involving several distinct stages: fruit removal by frugivores, seed delivery in different microhabitats, seed germination, seedling establishment, and adult recruitment. Nevertheless, studies conducted until now have provided scarce information concerning the sequence of stages in a plant's life cycle in its entirety. The main objective of this study was to evaluate the immediate consequences of frugivore activity for Eugenia umbelliflora ( Myrtaceae) early recruitment by measuring the relative importance of each fruit-eating bird species on the establishment of new seedlings in scrub and low restinga vegetation in the Atlantic rainforest, Brazil. We conducted focal tree observations on E. umbelliflora trees recording birds' feeding behaviour and post-feeding movements. We also recorded the fate of dispersed seeds in scrub and low restinga vegetation. We recorded 17 bird species interacting with fruits in 55 h of observation. Only 30% of the handled fruits were successfully removed. From 108 post flight movements of exit from the fruiting trees, 30.6% were to scrub and 69.4% to low restinga forest. Proportion of seed germination was higher in low restinga than in the scrub vegetation. Incorporating the probabilities of seeds' removal, deposition, and germination in both sites, we found that the relative importance of each frugivorous bird as seed dispersers varies largely among species. Turdus amaurochalinus and Turdus rufiventris were the best dispersers, together representing almost 12% probability of seed germination following removal. Our results show the importance of assessing the overall consequence of seed dispersal within the framework of disperser effectiveness, providing a more comprehensive and realistic evaluation of the relative importance of different seed dispersers on plant population dynamics.

Lorryia formosa Cooreman has been collected on many different host plants around the world. The biological cycle of this species was studied at 28 degrees C with mites collected from a rubber tree crop. The life table was constructed based on sixty individuals. The average lifetime was 37.43 +/- 20.23 (SE) days, and the r(m) was 0.08. The sex ratio was totally female biased, and unfertilized eggs developed into females. This is the first record of thelytoky in Tydeoidea proved by rearing.

A comparative cytogenetic analysis was carried out on four Hylinae tree frogs from Brazil (Aparasphenodon brunoi, Corthomantis greeningi, Osteocephalus langsdorffi and Scinax fuscovarius) using Giemsa staining, BrdU replication banding, Ag-NOR staining, C-banding, DAPI and CMA(3) fluorochrome staining, and fluorescence in situ hybridization (FISH) with an rDNA probe. All the species share closely similar 2n = 24 karyotypes, almost indistinguishable by standard staining. The technique of BrdU incorporation allowed the identification of each pair of homologs and the establishment of extensive homeology for the great majority of the chromosomes, mainly of A. brunoi, C greeningi, and O. langsdorffii. Despite highly conserved replication banding patterns, the use of the other banding techniques disclosed some minor differsences, which reinforces the importance of extensive cytogenctic analyses for the karyotypic characterization of Anuran species. The present cytogenctic data confirm the closer proximity of A. brunoi, C greeningi, and O. langsdorfjii, whereas S. fuscovarius is phylogenetically more distant. Copyright (C) 2003 S.
