A tentativa merleau-pontyana de aproximação do marxismo, empreendida nos idos do pós-guerra, é perpassada por constante ambigüidade. Não obstante o propósito do filósofo de se filiar à teoria de Marx, suas análises políticas revelam-se distantes de suas intenções. Concebendo a história como uma aventura que escapa a qualquer esquema racional, Merleau-Ponty questiona, desde seus primeiros escritos, a dialética marxista entre lógica e contingência na história. A tensão interna que dilacera os textos do autor nos anos 40, anunciando (e preparando) a recusa da teoria da revolução estampada mais tarde nas Aventuras da Dialética, permite indagar se esse desfecho dos anos 50 não teria sido, ao invés de um corte no interior da obra, o resultado necessário dessa tentativa problemática de aproximação do marxismo a partir de categorias que lhe são estranhas (próprias às filosofias da existência e à fenomenologia).

Por meio da utilização de corpus de textos técnicos, jornalísticos e literários escritos originalmente em inglês e traduzidos para o português, pode-se efetuar uma análise das soluções propostas pelos tradutores ao lidarem com semelhanças e diferenças lingüísticas e culturais dos textos de partida e de chegada. Com esse intuito, aplicou-se o modelo descritivo-comparativo sugerido por Aubert (1984, 1998), o qual se origina das categorias de Vinay e Darbelnet ([1958, 1977] 1995), a fim de, primeiramente, identificar e, depois, classificar os procedimentos ou modalidades empregadas na tradução desses gêneros textuais. Registraram-se como modalidades de maior incidência nos três corpora: a tradução literal, a transposição e a modulação. em virtude de a alta freqüência da tradução literal ocorrer em textos técnicos e, contrariamente ao esperado, incidir com proporção ainda maior em textos jornalísticos, poder-se-ia inferir uma tendência para automatismos na tradução desses dois tipos de textos. Já a necessidade de um emprego acentuado da modulação e da transposição com modulação em textos literários poderia ser decorrente de uma participação mais ativa do tradutor na tentativa de escapar da literalidade e proceder a uma maior reelaboração, a fim de passar características normalmente associadas à linguagem do romance.

A pair of primers directed to 16S-23S rDNA interspacer (ITS) was designed directed to Brucella genetic sequences in order to develop a polymerase chain reaction (PCR) putatively capable of amplifying DNA from any Brucella species. Nucleic acid extracts from whole-blood from naive dogs were spiked with decreasing amounts of Brucella canis RM6/66 DNA and the resulting solutions were tested by PCR. In addition, the ability of PCR to amplify Brucella spp. genetic sequences from naturally infected dogs was evaluated using 210 whole-blood samples of dogs from 19 kennels. The whole-blood samples collected were subjected to blood culture and PCR. Serodiagnosis was performed using the rapid slide agglutination test with and without 2-mercaptoethanol. The DNA from whole blood was extracted using proteinase-K, sodium dodecyl sulphate and cetyl trimethyl ammonium bromide followed by phenol-chloroform purification. The PCR was capable of detecting as little as 3.8 fg of Brucella DNA mixed with 450 ng of host DNA. Theoretically, 3.8 fg of Brucella DNA represents the total genomic mass of fewer than two bacterial cells. The PCR diagnostic sensitivity and specificity were 100%. From the results observed in the present study, we conclude that PCR could be used as confirmatory test for diagnosis of B. canis infection.

A bovine male-specific marker was identified in our laboratory through random amplified polymorphic DNA (RAPD) analysis. This fragment of 3216 bp was cloned, sequenced and mapped by fluorescent in situ hybridization (FISH) on the taurine Yq. Primers derived from this sequence were initially screened by polymerase chain reaction (PCR) for their ability to detect Y-specific segments in zebu and taurine genomic DNA. Two of these primers amplified a 655 bp Y-specific sequence present in taurine and zebu male genomic DNA. These primers were then used for detecting the 655 bp male sequence in DNA from 173 zebu and 30 taurine embryos, which had been previously sexed using primers for the sequence BC 1.2. From a 0.72-kb fragment universally generated in Paracoccidioides brasiliensis strains, primers were designed and tested on genomic DNA of this and other pathogenic fungi. They were specific and highly sensitive for P. brasiliensis DNA. Positive results were obtained when these were tested in clinical samples.

Background: The increasing number of genomic sequences of bacteria makes it possible to select unique SNPs of a particular strain/species at the whole genome level and thus design specific primers based on the SNPs. The high similarity of genomic sequences among phylogenetically-related bacteria requires the identification of the few loci in the genome that can serve as unique markers for strain differentiation. PrimerSNP attempts to identify reliable strain-specific markers, on which specific primers are designed for pathogen detection purpose.Results: PrimerSNP is an online tool to design primers based on strain specific SNPs for multiple strains/species of microorganisms at the whole genome level. The allele-specific primers could distinguish query sequences of one strain from other homologous sequences by standard PCR reaction. Additionally, PrimerSNP provides a feature for designing common primers that can amplify all the homologous sequences of multiple strains/species of microorganisms. PrimerSNP is freely available at http://cropdisease.ars.usda.gov/similar to primer.Conclusion: PrimerSNP is a high-throughput specific primer generation tool for the differentiation of phylogenetically-related strains/species. Experimental validation showed that this software had a successful prediction rate of 80.4 - 100% for strain specific primer design.

This article discusses some methodological possibilities for analyzing textbooks in Mathematics Education. Its starting point was a review - which allowed us to build a general map - of the production developed by consolidated Research Groups that deal with this theme in Brazil. This map allowed us to propose a complementary/alternative guide to analyse textbooks according to a hermeneutical approach.

Randomly amplified polymorphic DNA (RAPD) analysis of 35 Paracoccidioides brasiliensis isolates was carried out to evaluate the correlation of RAPD profiles with the virulence degree or the type of the clinical manifestations of human paracoccidioidomycosis. The dendrogram presented two main groups sharing 64% genetic similarity. Group A included two isolates from patients with chronic paracoccidioidomycosis; group B comprised the following isolates showing 65% similarity: two non-virulent, six attenuated, five virulent, eight from patients with chronic paracoccidioidomycosis and two from patients with acute paracoccidioidomycosis. The virulent Pb18 isolate and six attenuated or non-virulent samples derived from it were genetically indistinguishable (100% of similarity). Thus, in our study, RAPD patterns could not discriminate among 35 P. brasiliensis isolates according to their differences either in the degree of virulence or in the type of the clinical manifestation of this fungal infection. © 2002 Federation of European Microbiological Societies. Since the use of air abrasion has grown in pediatric dentistry, the aim of this study was to evaluate, by means of shear bond strength testing, the need to use the total etching technique or self-etching primers on dentin of primary teeth after air abrasion. Twenty-five exfoliated primary molars had their occlusal dentin exposed by trimming and polishing. Specimens were treated by: Air abrasion + Scotchbond MultiPurpose adhesive (G1); 37% phosphoric acid + Scotchbond MP adhesive (G2); Clearfil SE (G3); Air abrasion ( 37% phosphoric acid + Scotchbond MP adhesive (G4); Air abrasion + Clearfil SE (G5). On the treated surface, a cylinder of 2 mm by 6 mm was made using a composite resin (Z100). Duncan's test showed that: (G2 = G3 = G5) > (G1 = G4). The use of a selfetching primer on air abraded dentin is recommended to obtain higher bond strengths.
