Microorganisms distributed in the marine and brackish environments play an important role in the decomposition of organic matter and mineralisation in the system (Seshadri and lgnacimuthu, 2002). Estuary is one of the most productive ecosystems, at the same time one among the least explored ecosystems on earth, which has immense potential as a source of potent microorganisms that produce valuable compounds particularly, enzymes such as proteases. In this scenario, it is very appropriate to embark on finding novel alkaline protease producers from the estuarine system. The area where the present investigation was carried out is a part of the extensive estuarine system of South India viz. Cochin Estuary. There is meagre knowledge regarding the microbial composition, particularly the protease producers of Cochin Estuary. Hence, the present study has been undertaken with the objective of finding novel alkaline protease producing bacteria from Cochin Estuary

The reactions involving fulvenes and its derivatives have received a great deal of attention over the years in synthetic organic chemistry. Functionalizations of fulvenes provide versatile and powerful approaches to various polycyclic systems and natural products. They serve as versatile intermediates in the construction of various ring systems through inter- as well as intramolecular cycloadditions. Compared to the rich literature on the cycloaddition reactions of pentafulvenes, much less attention has been paid to the synthetic utilization of their cycloadducts. Tactical manipulations on the chosen adduct offer the prospects for designing a variety of useful molecular skeletons. Addition of heterodienophiles to fulvenes offers an efficient strategy towards the synthesis of azabicyclic olefins. However, there have been no serious attempts to study the synthetic utility of these substrates. In this context and with the intention of utilizing pentafulvenes towards synthetically important molecules, author decided to explore the reactivity of pentafulvene derived azabicyclic olefins. Our attention was focused on the synthetic potential associated with the ring opening of fulvene derived bicyclic hydrazines under palladium catalysis. It was envisioned that the desymmetrization of these adducts using various soft nucleophiles will provide a novel access to synthetically and biologically important alkylidene cyclopentenes. There exists a need for potential microorganism that could facilitate effective bioremediation of crude oil pollutants in the environment. Hence it was desired to isolate a potential bacterium from marine sediment, which often experiences oil pollution and develop a bioprocess for crude oil biodegradation. In the present study the sediment deposits in the beach of Munakkal, Trichur dist, Kerala, collected immediately after the major event Tsunami in 2004 was collected and analyzed by enrichment culture technique towards isolation of potential strains that could degrade crude oil and its fractions. From the results obtained it was found that the sediment deposits harbor several bacteria with potential for degrading hydrocarbons. However, among the strains obtained, isolate no. BTTS 10 showed capabilities for utilizing both alkanes and aromatic hydrocarbons and hence the same was selected for further studies.

The thesis entitled "Studies on the eco-physiology of heterotrophic and indicator bacteria in the marine environments of Kerala" embodies the results of an investigation carried out by the candidate at the Central Marine Fisheries Research Institute, Cochin. It is presentedd under 4 chapters in two parts (Parts A & B) and includes 6 sections. The material for the study was collected in the Cochin backwater during April 1972 to February. 1973, March 1974 to February 1975, July 1975 to June 1976 and in the ishore area during January to October, 1978 and an account of the heterotropic and indicator bacteria are given with intensity charts and tables. Samples from all the stations contained significant quantities of heterotrophs (Part A, Section I) and faecal pollution indicators (Section II). Maximum number of heterotrophic bacteria was observed during the postmonsoon period. The total counts betwen one station and the other did not vary as much as the counts between months did. The distribution was characterised by overdispersion. During 1972-73 in all the stations except the fourth the minimum heterotrophs (Total counts) were recorded during the monsoon period. Minimum counts were observed during the premonsoon period, with an increasing trend from the premonsoon to postmonsoon seasons. Maximum counts were recorded during monsoon months during 1974-75. No significant difference was noted in the total plate count between stations, months and regions. Seasonal variations in sea water was meagre during 1975-76, whereas in sediments variations were prominent during monsoon in Station I - near the mouth of the sewage effluent of Cochin City and in postmonsoon at Station II in the Mattancherry Channel and Station III near barmouth

The present work "Nature and Ecological Significance of Nutrient Regeneration in different Prawn Culture Fields" was undertaken to understand the seasonal variation of nutrients, nutrient cycling and primary productivity of the prawn culture systems. The main emphasis was to find the qualitative and quantitative estimates of distribution of total phosphorus, inorganic phosphorus, organic phosphorus, total nitrogen and nitrogen fractions in the water. The effect of nutrient cycling on primary productivity and concentration of metals also form one part of the study. The entire thesis comprise of only one major chapter with subchapters such as, Introduction (I), Review of Literature (2), Material and Methods (3), Results (14), Discussion (5), Executive Summary (6) and Biblio~ graphy (7)

Poisoning by pesticides from agricultural fields is a serious water pollution problem and its environmental long-term effect may result in the incidence of poisoning of fish and other aquatic life forms (jyothi and Narayan, 1999). Fishes like Heteropneustesfbssilis and C/arius batrac/nus are especially prone to serious pesticide pollution as their habitat is mostly the agriculture area. Though only few studies are conducted in this area, it can be assessed from the local information that, population of such fish is on the verge of vulnerability due to extensive use of pesticides. The knowledge of sublethal effects of xenobiotic compounds on hematological parameters, enzyme activities and metabolite concentrations is very important to delineate the fish health status and provide a future understanding of ecological impacts. These pesticides act by causing inhibition of cholinesterase enzymes (ChE) by formation of enzyme inhibitor complex (O'Brien, 1976) and damaging the nervous system. These effects may result in metabolic disorders. Associated to cholinesterase activities, a study of other enzymes such as phosphatases and aminotransferases close to intermediary metabolite determination provides a wider view of metabolism. Interest in toxicological aspects has grown in recent years and research is now increasingly focused on mechanistic aspects of oxidative damage and cellular responses in biological system. The term ‘biomarker’ is generally used in a broad sense to include almost any measurement reflecting an interaction between a biological system and a potential hazard, which may be chemical, physical or biological (WHO, 1993). As biomarker stands for immediate responses, they are used as early warning signals of biological effects caused by environmental pollutants. Design and study of molecular receptors capable of mimicking natural processes has found applications in basic research as well as in the development of potentially useful technologies. Of the various receptors reported, the cyclophanes are known to encapsulate guest molecules in their cavity utilizing various non–covalent interactions resulting in significant changes in their optical properties. This unique property of the cyclophanes has been widely exploited for the development of selective and sensitive probes for a variety of guest molecules including complex biomolecules. Further, the incorporation of metal centres into these systems added new possibilities for designing receptors such as the metallocyclophanes and transition metal complexes, which can target a large variety of Lewis basic functional groups that act as selective synthetic receptors. The ligands that form complexes with the metal ions, and are capable of further binding to Lewis-basic substrates through open coordination sites present in various biomolecules are particularly important as biomolecular receptors. In this context, we synthesized a few anthracene and acridine based metal complexes and novel metallocyclophanes and have investigated their photophysical and biomolecular recognition properties.

Emergence of antibiotic resistance among aquaculture pathogens has made it necessary to look into environment friendly, effective and sustainable methods such as probiotic and immunostimulants among others.. In the present study, LAB were isolated from the gut of fish species namely Rastrelliger kanagurta and analyzed for their antibacterial activity against various fish, shrimp and human pathogens. Different LAB species such as Lactobacillus plantarum, L. bulgaricus, L. brevis and L. viridiscens were encountered in the gut of R. kanagurta. Several strains showed good activity against fish, shrimp and human pathogens. LAB from the gut of such marine species may be developed as possible probiont for environment friendly health management of fresh water, estuarine and marine species currently exploited in aquaculture

In the present study we address the issue on gut associated lactic acid bacteria (LAB) isolated from the intestine of estuarine fish Mugil cephalus using de Man Rogossa and Sharpe (MRS) agar. LAB isolates were identified biochemically and screened for their ability to inhibit in vitro growth of various fish, shrimp and human pathogens. Most of the LAB isolates displayed an improved antagonism against fish pathogens compared to shrimp and human pathogens. Selected representative strains displaying high antibacterial activity were identified using 16S rRNA gene sequence analysis. Of the selected strains Lactobacillus brevis was the most predominant. Four other species of Lactobacillus, Enterobacter hormaechei and Enterobacter ludwigii were also identified. It was also observed that even among same species, considerable diversity with respect to substrate utilization persisted. Considering the euryhaline nature of grey mullet (Mugil cephalus), the LAB isolated from the gut possessed good tolerance to varying salt concentrations. This finding merits further investigation to evaluate whether the isolated LAB could be used as probiotics in various fresh and sea water aquaculture

In natural systems phytoplankton interact with planktonic (free living) and attached epiphytic bacteria both synergistically and antagonistically. The specificity of the association with micro algae and bacteria differs in terms of adhesion mechanisms and metabolic cooperation. Present research was carried out to study the effect of bacterial isolates namely Bacillus sp. and Pseudomonas sp. from algal culture systems on the growth of micro algae such as Chaetoceros calcitrans and Nannochloropsis oculata. C. calcitrans (F= 15.34; P<0.05) and N. oculata (F=12.52; P<0.05) showed significantly higher growth, in treatments with Bacillus sp. and Pseudomonas sp when compared to control.

Development of continuous cell lines from shrimp is essential to investigate viral pathogens. Unfortunately, there is no valid cell line developed from crustaceans in general and shrimps in particular to address this issue. Lack of information on the requirements of cells in vitro limits the success of developing a cell line, where the microenvironment of a cell culture, provided by the growthmedium, is of prime importance. Screening and optimization of growth medium components based on statistical experimental designs have been widely used for improving the efficacy of cell culture media. Accordingly, we applied Plackett–Burman design and response surface methodology to study multifactorial interactions between the growth factors in shrimp cell culture medium and to identify the most important ones for growth of lymphoid cell culture from Penaeus monodon. The statistical screening and optimization indicated that insulin like growth factor-I (IGF-I) and insulin like growth factor-II (IGF-II) at concentrations of 100 and 150 ng ml-1, respectively, could significantly influence the metabolic activity and DNA synthesis of the lymphoid cells. An increase of 53 % metabolic activity and 24.8 % DNA synthesis could be obtained, which suggested that IGF-I and IGFII had critical roles in metabolic activity and DNA synthesis of shrimp lymphoid cells

The textile industry is one amongst the rapidly growing industries world wide, which utilizes enormous amounts of synthetic dyes. Consequently, the effluent from these textile industries poses serious threat to the environment which is often very difficult to treat and dispose. This has become a very grave problem in environment conservation and hence natural pigments have drawn the attention of industry as safe alternative. In this context, in the present study an attempt was made to bioprospect marine bacteria towards isolation of a suitable and ideal pigment that could be used as a natural dye. A marine Serratia sp. BTWJ8 was recognized to synthesize enormous amounts of a prodigiosin-like pigment. The pigment was isolated and characterized for various properties. The pigment was evaluated for application as a dye in the textile industry. Results of the studies indicated that this pigment could be used as a natural dye for imparting red-yellow colour to various grades of textile materials. The colour was observed to be stable after wash performance studies

The rising pressure of Population, together with the constantly changing technologies, and development perspectives, contribute to the ever increasing volumes of wastes in different forms. The solid and liquid wastes generated in the urban areas were considered a burden to the society, and hazardous to the environment. The fact is that the growth of consumerist culture and aimless throwing of refuse by the people created the outbreak of environmental pollution. Unhygienic environment and solid waste accumulation coincided with mosquito breeding which causes, the spread of most epidemics. The rationale behind most of the diseases is the unhygienic pattern followed by the people both in rural and urban areas. As an environmental package, the disposal of solid waste from different sources, such as house holds markets, commercial areas, slaughter houses, hospitals and industries, therefore assumed crucial importance. So as a part of the theory and practice, a study on the area, solid waste management of Arppukara Grama Panchayat of Kottayam district is taken into consideration. The study conducted here proposes, to examine the quality and quantity of the solid waste generated in the panchayat and also it's impact on the existing social, economical, environmental and ecological systems

Influence of acute salinity stress on the immunological and physiological response of Penaeus monodon to white spot syndrome virus (WSSV) infection was analysed. P. monodon maintained at 15‰ were subjected to acute salinity changes to 0‰ and 35‰ in 7 h and then challenged orally with WSSV. Immune variables viz., total haemocyte count, phenol oxidase activity (PO), nitroblue tetrazolium salt (NBT) reduction, alkaline phosphatase activity (ALP), acid phosphatase activity (ACP) and metabolic variables viz., total protein, total carbohydrates, total free amino acids (TFAA), total lipids, glucose and cholesterol were determined soon after salinity change and on post challenge days 2 (PCD2) and 5 (PCD5). Acute salinity change induced an increase in metabolic variables in shrimps at 35‰ except TFAA. Immune variables reduced significantly (Pb0.05) in shrimps subjected to salinity stress with the exception of ALP and PO at 35‰ and the reduction was found to be more at 0‰. Better performance of metabolic and immune variables in general could be observed in shrimps maintained at 15‰ that showed significantly higher post challenge survival following infection compared to those under salinity stress. Stress was found to be higher in shrimps subjected to salinity change to lower level (0‰) than to higher level (35‰) as being evidenced by the better immune response and survival at 35‰. THC (Pb0.001), ALP (Pb0.01) and PO (Pb0.05) that together explained a greater percentage of variability in survival rate, could be proposed as the most potential health indicators in shrimp haemolymph. It can be concluded from the study that acute salinity stress induces alterations in the haemolymph metabolic and immune variables of P. monodon affecting the immunocompetence and increasing susceptibility to WSSV, particularly at low salinity stress conditions

mbikulam Tiger Reserve of Western Ghats using Geospatial technology. The major objectives of the study are Land use land cover mapping (LULC) and Phytodiversity analysis. Satellite data was used to map the land use / land cover using supervised classification techniques in Erdas imagine. The change for a period of 32 years was assessed using the multi-temporal satellite datasets from Landsat MSS (1973), Landsat TM (1990), and IRS P6 LISS III (2005). A geospatial approach was used for the land cover analysis. Digital elevation models, Satellite imageries and SOI topo sheets were the data sets used in the analysis. Vegetation sampling plots distributed over the different forest types were enumerated and studied for Phytodiversity analysis.
