Triterpane and sterane biomarkers in the YA13-1 condensates from Qiongdongnan Basin, South China Sea

Triterpanes and steranes in condensates from the YA13-1 gas field, Qiongdongnan Basin, were monitored. The YA13-1 condensates have unusual biomarker distributions dominated by terpanes and steranes derived from higher plants. Anomalously abundant 1 got-oleanane and remarkably abundant bicadinanes are present in the YA13-1 condensates, whereas the 17alpha-hopane contents are extremely low. Taraxastane and significantly abundant 17alpha-diahopanes occur in the condensates. In addition, a number of unknown C-29 and C-30 pentacyclic triterpanes including previously unreported compounds were detected in the condensates, some of which are significantly abundant. The unknown compounds may be terrestrial biomarkers. C-29 homologues are relatively predominant among the regular and rearranged steranes. The diasterane concentrations are markedly higher than those of regular steranes. The maturity of the YA13-1 condensates is relatively high, at the peak to late oil generation stage (corresponding to 0.85-1.10% Ro), based on sterane and terpane and including bicadinane maturity parameters (i.e. T/(T-1 + R) and 2T/R bicadinane ratios). The above maturity assessment result is different from that based on diamondoid maturity parameters (%Ro = 1.60-1.70) [Org. Geochem. 25 (1996) 179], which can be explained by a contribution of hydrocarbons from two sources at different depths. The YA13-1 condensates were probably generated from the Yacheng and Lingshui coal-bearing source rocks buried both in the Qiongdongnan Basin (3400-5000 m) and in the Yinggehai Basin (>5000 m). The possible contribution of lower maturity hydrocarbons from the Yacheng and Lingshui Formations (3400-4100 m) in the Qiongdongnan Basin to the YA13-1 gases and condensates should not be neglected. A novel labeling reagent 1-(2-naphthyl)-3-methyl-5-pyrazolone (NMP) coupling to liquid chromatography with electrospray ionization mass spectrometry for the detection of carbohydrates from the derivatized rape bee pollen samples is reported. Carbohydrates are derivatized to their bis-NMP-labeled derivatives. Derivatives showed an intense protonated molecular ion at m/z [M+H](+) in positive-ion detection mode. The mass-to-charge ratios of characteristic fragment ions at m/z 473.0 could be used for the accurately qualitative analysis of carbohydrates. This characteristic fragment ion is from the cleavage of C2-C3 bond in carbohydrate chain giving the specific fragment ions at m/z [MH-CmH2m+1Om-H2O](+) for pentose, hexose and glyceraldehydes and at m/z [MH-CmH2m-1Om+1-H2O](+) for alduronic acids such as galacturonic acid and glucuronic acid (m = n - 2, n is carbon number of carbohydrate). No interferences for all aliphatic and aromatic aldehydes presented in natural environmental samples were observed due to the highly specific parent mass-to-charge ratio and the characteristic fragment ions. The method, in conjunction with a gradient elution, offered a baseline resolution of carbohydrate derivatives on a reversed-phase Hypersil ODS-2 column. The carbohydrates such as mannose, galacturonic acid, glucuronic acid, rhamnose, glucose, galactose, xylose, arabinose and fucose can successfully be detected.

A pre-column derivatization method for the sensitive determination of amines using a labeling reagent 2-(11H-benzo[a]-carbazol-11-yl) ethyl chloroformate (BCEC-Cl) followed by high-performance, liquid chromatography with fluorescence detection has been developed. Identification of derivatives was carried out by LC/APCI/MS in positive-ion mode. The chromophore of 1,2-benzo-3,4-dihydrocarbazole-9-ethyl chloroformate (BCEOC-Cl) reagent was replaced by 2-(11H-benzo[a]-carbazol-11-yl) ethyl functional group, which resulted in a sensitive fluorescence derivatizing reagent BCEC-Cl. BCEC-Cl could easily and quickly label amines. Derivatives were stable enough to be efficiently analyzed by HPLC and showed an intense protonated molecular ion corresponding m/z [M+ H](+) under APCI/MS in positive-ion mode. The collision-induced dissociation of the protonated molecular ion formed characteristic fragment ions at m/z 261.8 and m/z 243.8 corresponding to the cleavages of CH2O-CO and CH2-OCO bonds. Studies on derivatization demonstrated excellent derivative yields over the pH 9.0-10.0. Maximal yields close to 100% were observed with three- to four-fold molar reagent excess. In addition, the detection responses for BCEC-derivatives were compared to those obtained using 1,2-benzo-3,4-dihydrocarbazole-9-ethyl chloroformate (BCEOC-Cl) and 9-fluorenyl methylchloroformate, (FMOC-Cl) as labeling reagents. The ratios I-BCEC/I-BCEOC = 1.94-2.17 and I-BCEC/I-FMOC = 1.04-2.19 for fluorescent (FL) responses (here, I was relative fluorescence intensity). Separation of the derivatized amines had been optimized on reversed-phase Eclipse XDB-C-8 column. Detection limits calculated from 0.50 pmol injection, at a signal-to-noise ratio of 3, were 1.77-14.4 fmol. The relative standard deviations for within-day determination (n = 11) were 1.84-2.89% for the tested amines. The mean intra- and inter-assay precision for all amines levels were < 3.64% and 2.52%, respectively. The mean recoveries ranged from 96.6% to 107.1% with their standard deviations in the range of 0.8-2.7. Excellent linear responses were observed with coefficients of > 0.9996. (C) 2006 Elsevier B.V. A sensitive method for the determination of 30 kinds of free fatty acids (FFAs, C-1-C-30) with 1-[2-(p-toluenesulfonate)-ethyl]-2-phenylimidazole-[4,5-f] 9,10-phenan- threne (TSPP) as labeling reagent and using high performance liquid chromatography with fluorescence detection and identification by online postcolumn mass spectrometry with atmospheric pressure chemical ionization (APCI) source in positive-ion mode (HPLC/MS/APCI) has been developed. TSPP could easily and quickly label FFAs in the presence of K2CO3 catalyst at 90 degrees C for 30 min in N,N-dimethylformamide (DMF) solvent, and maximal labeling yields close to 100% were observed with a 5-fold excess of molar reagent. Derivatives were stable enough to be efficiently analyzed by high performance liquid chromatography. TSPP was introduced into fatty acid molecules and effectively augmented MS ionization of fatty acid derivatives and led to regular MS and MS/MS information. The collision induced cleavage of protonated molecular ions formed specific fragment ions at m/z [MH](+)(molecular ion), m/z [M'+CH2CH2](+)(M' was molecular mass of the corresponding FFA) and m/z 295.0 (the, mass of protonated molecular core structure of TSPP). Fatty acid derivatives were separated on a reversed-phase Eclipse XDB-C-8 column (4.6 x 150 mm, 5 mu m, Agilent) with a good baseline resolution in combination with a gradient elution. Linear ranges of 30 FFAs are 2.441 x 10(-3) to 20 mu mol/L, detection limits are 3.24 similar to 36.97 fmol (injection volume 10 mu L, at a signal-to-noise ratio of 3, S/N 3:1). The mean interday precision ranged from 93.4 to 106.2% with the largest mean coefficients of variation (R.S.D.) < 7,5%. The mean intraday precision for all standards was < 6.4% of the expected concentration. Excellent linear responses were observed with correlation coefficients of > 0.9991. Good compositional data could be obtained from the analysis of extracted fatty acids from as little as 200 mg of bryophyte plant samples.Therefore, the facile TSPP derivatization coupled with HPLC/MS/APCI analysis allowed the development of a highly sensitive method for the quantitation of trace levels of short and long chain fatty acids from biological and natural environmental samples.

A sensitive method for the determination of long-chain fatty acids (LCFAs) (>C20) using 1-[2-(p-toluenesulfonate)-ethyl]-2-phenylimidazole-[4.5-f]-9,10-phenanthrene (TSPP) as tagging reagent with fluorescence detection and identification with post-column APCI/MS has been developed. The LCFAs in bryophyte plant samples were obtained based on distillation extraction with 1: 1 (v/v) chloroform/methanol as extracting solvent. TSPP could easily and quickly label LCFAs at 90 degrees C in the presence of K2CO3 catalyst in DMF. Eleven free LCFAs from the extracts of bryophyte plants were sensitively determined. Maximal labeling yields close to 100% were observed with a five-fold excess of molar reagent. Separation of the derivatized fatty acids exhibited a good baseline resolution in combination with a gradient elution on a reversed-phase Eclipse XDB-C-8 column. Calculated detection limits from 1.0 pmol injection, at a signal-to-noise ratio of 3, were 26.19-76.67 fmol. Excellent linear responses were observed with coefficients of >0.9996. Good compositional data were obtained from the analysis of the extracted LCFAs containing as little as 0.2 g of bryophyte plant samples. Therefore, the facile TSPP derivatization coupled with HPLC/APCI/MS analysis allowed the development of a highly sensitive method for the quantitation of trace levels of LCFAs from biological and natural environmental samples. A simple and sensitive high-performance liquid chromatographic (HPLC) method with fluorescence detection and mass spectrometric identification has been developed for analysis of 30 long-chain and short-chain free Fatty acids (FFAs). The fatty acids were derivatized to their esters with 1-[2-(p-toluenesulfonate)ethyl]-2-phenylimidazole-[4,5-f]-9,10-phenanthrene (TSPP) in N,N-dimethylformamide (DMF) at 90 degrees C with anhydrous K2CO3 as catalyst. A mixture Of C-1-C-30 fatty acids was completely separated within 60 min by gradient elution on a reversed-phase C-8 column. Qualitative identification of the acids was performed by atmospheric-pressure chemical ionization mass spectrometry (APCI-MS) in positive-ion mode. The fluorescence excitation and emission wavelengths were 260 and 380 nm, respectively. Quantitative determination of the 30 acids in two Tibetan medicines Gentiana straminea and G. dahurica was performed. The results indicated that the medicines contained many FFAs. Linear correlation coefficients for the FFA derivatives were > 0.9991. Relative standard deviations (RSDs, n = 6) for the fatty acid derivatives were < 3%. Detection limits (at a signal-to-noise ratio of 3:1) were 3.1-38 fmol. When the fatty acid derivatives were determined in the two real samples results were satisfactory and the sensitivity and reproducibility of the method were good.

The collision-induced dissociation of protonated molecular ion formed a product at m/z 260 corresponding to the cleavage of CH2-OCO bond. Studies on derivatization demonstrate excellent derivative yields over the pH 9.0-10.0. Maximal yields close to 100% are observed with a 3 to 4-fold molar reagent excess. In addition, the detection responses for BCIC derivatives are compared with those obtained using CEOC and FMOC as derivatization reagents. The ratios of l(BCIC)/l(CEOC) and l(BCIC)/l(FMOC) are, respectively, 1.23-3.14 and 1.25-3.08 for fluorescent (FL) responses (here, l is relative fluorescence intensity). Separation of the derivatized amines had been optimized on reversed-phase Eclipse XDB-C-8 column. Detection limits are calculated from 1.0 pmol injection, at a signal-to-noise ratio of 3, are 10.6-37.8 fmol. The mean interday accuracy ranges from 94 to 105% for fluorescence detection with the largest mean %CV < 7.5. The mean interday precision for all standards is < 6.0% of the expected concentration. Excellent linear responses are observed with coefficients of > 0.9997.

A simple, sensitive, and mild method for the determination of amino compounds based on a condensation reaction with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride (EDC-HCI) as the dehydrant with fluorescence detection has been developed. Amines were derivatized to their acidamides with labeling reagent 2-(2-phenyl-1H-phenanthro-[9,10-d]imidazole-1-yl)-acetic acid (PPIA). Studies on derivatization conditions indicated that the coupling reaction proceeded rapidly and smoothly in the presence of a base catalyst in acetonitrile to give the corresponding sensitively fluorescent derivatives with an excitation maximum at lambda(ex) 260nm and an emission maximum at lambda(em) 380nm. The labeled derivatives exhibited high stability and were enough to be efficiently analyzed by high-performance liquid chromatography. Identification of derivatives was carried out by online post-column mass spectrometry (LC/APCI-MS/MS) and showed an intense protonated molecular ion corresponding m/z [MH](+) under APCI in positive-ion mode. At the same time, the fluorescence properties of derivatives in various solvents or at different temperature were investigated. The method, in conjunction with a gradient elution, offered a baseline resolution of the common amine derivatives on a reversed-phase Eclipse XDB-C-8 column. LC separation for the derivatized amines showed good reproducibility with acetonitrile-water as mobile phase. Detection limits calculated from 0.78 pmol injection, at a signal-to-noise ratio of 3, were 3.1-18.2 fmol. The mean intra- and inter-assay precision for all amine levels were < 3.85% and 2.11%, respectively. Excellent linear responses were observed with coefficients of > 0.9996. A rapid and sensitive liquid chromatography-atmospheric pressure chemical ionization mass spectrometry (HPLC-APCI-MS) assay for the determination of five pharmacologically active compounds (PAC) extracted from the traditional Chinese medicine, Rhodiola , namely salidroside, tyrosol, rhodionin, gallic acid, and ethyl gallate has been developed. In this method, PAC could be baseline separated and detected with DAD at 275 nm. The validation of the method, including sensitivity, linearity, repeatability, and recovery, was examined. The linear calibration curves were acquired with correlation coefficient >0.999 and the limits of detection LOD (at a signal-to-noise ratio=3:1) were between 0.058 and 1.500 mu mol/L. It was found, that the amounts of PAC varied with different species of Rhodiola . The established method is rapid and reproducible for the separation of five natural pharmacologically active compounds from extracts of Rhodiola with satisfactory results.

The chromophore of 2-(9-carbazole)-ethyl chloroformate (CEOC) reagent was replaced by 1,2-benzo-3,4-dihydrocarbazole functional group, which resulted in a sensitive fluorescence derivatizing reagent BCEOC. BCEOC can easily and quickly label peptides and amino acids. Derivatives are stable enough to be efficiently analyzed by high-performance liquid chromatography. The derivatives showed an intense protonated molecular ion corresponding m/z (M + H)(+) under electrospray ionization (ESI) positive-ion mode with an exception being Tyr detected at negative mode. The collision-induced dissociation of protonated molecular ion formed a product at m/z 246.2 corresponding to the cleavage of C-O bond of BCEOC molecule. Studies on derivatization demonstrate excellent derivative yields over the pH 9.0-10.0. Maximal yields close to 100% are observed with a 3-4-fold molar reagent excess. Derivatives exhibit strong fluorescence and extracted detzvatization solution with n-hexane/ethyl acetate (10:1, v/v) allows for the direct injection with no significant interference from the major fluorescent reagent degradation by-products, such as 1,2-benzo-3,4-dihydrocarbazole-9-ethanol (BDC-OH) (a major by-product), mono- 1,2-benzo-3,4-dihydrocarbazole-9-ethyl carbonate (BCEOC-OH) and bis-(1,2-benzo-3,4-dihydrocarbazole-9-ethyl) carbonate (BCEOC)(2). In addition, the detection responses for BCEOC derivatives are compared to those obtained with previously synthesized 2-(9-carbazole)-ethyl chloroformate (CEOC) in our laboratory. The ratios AC(BCEOC)/AC(CEOC) = 2.05-6.51 for fluorescence responses are observed (here, AC is relative fluorescence response). Separation of the derivatized peptides and amino acids had been optimized on Hypersil BDS C-18 column. Detection limits were calculated from 1.0 pmol injection at a signal-to-noise ratio of 3, and were 6.3 (Lys)-177.6 (His) fmol. The mean interday accuracy ranged from 92 to 106% for fluorescence detection with mean %CV < 7.5. The mean interday precision for all standards was < 10% of the expected concentration. Excellent linear responses were observed with coefficients of > 0.9999. Good compositional data could be obtained from the analysis of derivatized protein hydrolysates containing as little as 50.5 ng of sample. Therefore, the facile BCEOC derivatization coupled with mass spectrometry allowed the development of a highly sensitive and specific method for the quantitative analysis of trace levels of amino acids and peptides from biological and natural environmental samples. Using three different types of yaks, 30 head of 1-yr-old calves (liveweight 60.7 +/- 6.3 kg), 30 head of 2-yr-old calves (97.7 +/- 10.2 kg) and 30 head of yak cows (160.2 +/- 15.1 kg) were randomly selected from the same yak herds. Each type of yak herd was divided into control (C) and trial (T) groups using a completely randomized design, with 10 and 20 animals in the C and T groups, respectively. The animals in the C group were grazed on natural rangeland, and the animals in the T group were supplemented with urea multinutritional molasses blocks (UMMB), together with grazing on natural rangeland from January to May of 1998. The objective was to determine the effect of UMMB on productive performance of yak calves and yak cows in the cold season. Live weight loss of 1-yr-old calves, 2-yr-old calves and yak cows was reduced by 1.2, 8.3 and 7.9 kg after UMMB supplementation (P < 0.01). The 1-yr-old calves gained the most in the first month of supplementation, but the 2-yr-old calves and yak cows gained the most both in the first and last supplementation months. Daily milk yield of yak cows increased by 0.21 kg d(-1) when the lactating animals were supplemented with UMMB (P < 0.01), although there was no effect (P < 0.01) of UMMB supplementation on hair and downy hair production. Supplementation with UMMB also improved reproductive performance of yak cows, with 8.8 and 30.9% increments in pregnancy rate and newborn weight, respectively. We conclude that the benefit of UMMB supplementation the 1-yr-old calves was not economical, with only 0.3:1 output to input ratio, but supplementation of the 2-yr-old calves and yak cows may be economical, with 1.8:1 and 1.4:1 output to input ratios, respectively.

In this paper, in view of characteristics and appraisal difficulty of complicate fault block reservoir, based on the theory of reservoir geology, tectonic geology, sequence stratigraphy, logging geology and sedimentology, according to related multidisciplinary data, such as geological, geophysical, logging and testing, taking 3D seismic acquisition processing, delicate structure interpretation, fine reservoir prediction, reservoir engineering and logging evaluation as research methods, on the basis of comprehensive geologic research, innovative appraisal thinking of complicate fault block reservoir is probed into, reservoir evaluation technology series of different reservoirs were created, and appraisal circuit of complicate fault block reservoir was proposed. According to the research on main controlling factors of hydrocarbon and concentration and concealing laws of Dagang exploration area, family assemble patterns of complicate fault block reservoir were proposed firstly, such as twinning, juxtaposed, overlying, concurrent, connected in series and so on, so theory basement was provided to reservoir evaluation. Taking into account of difficulties of low signal to noise ratio and border identification of lithologic trap of complicate fault block reservoir, in the middle of seismic data acquisition, double focusing layout and optimization technology were adopted, geophysical model was set up, and best observation system was ascertained. During object processing, matching processing technology of removing time difference, phase difference, energy difference and frequency spectrum difference generated by different blazing and receiving elements was studied. Imagery, low amplitude structure and identification of subtle lithologic reservoir was interpreted reasonably. On the basis of characteristics and technical description analysis of structural, structural--lithologic and lithologic reservoirs, innovative appraisal thinking of these reservoirs was formatted, appraisal circuit of complicate fault block reservoir was proposed, and this method could be used in other similar oilfields

In this paper we base on the anisotropic theory and Zoeppritz function of the transmission theory and the law of amplitude versus offset simplify seismic reflection coefficient of different media, analyze the characteristic of the gas or oil saturated stratum or the VTI and HTI models. Discuss the P wave reflection relationship and the meanings of the different parameters. We use measured parameters of a reservoir to simulate the characteristic of the reservoir, study the different effects of stratum saturated with gas or oil and analyze the characteristic of the seismic response of different models which change with different incident angles and different azimuths. Using the field data of logs ,analyze the rock property parameters, build the relationship of logs and parameters by Gassmann theory or empirical function. Calculate the density and the shear modulus and bulk modulus, reconstruct the log curves, calculate shear wave logs and correlate the logs affected by mud and other environmental factors. Finally perform the relationship of the seismic data log of saturated stratum and enhance the ability and reliability in reservoir prediction. Our aim is by the prestack seismic processing to get high solution and amplitude preserved seismic data. Because in incident angle gathers or azimuthal gathers, the low signal to noise ratio and low different covers affect the result of the prestack reservoir prediction. We apply prestack noise erase, cell regularization process and relatively amplitude preservation in the high solution seismic process routine to preserve the characteristic of stratum response, and erase the effects of the noise. In this paper we finished prestack invertion in the BYT survey and fractured reservoir depiction in MB survey. By the invertion and multiple attributes crossplot. we can get the stratum profiles and oil indicator profiles which can predict the distribution of the reservoir and oil. In the MB survey, we get orientation and density of fractured reservoir by the azimuthal seismic amplitude and depict the potential oil and gas reservoir. Prestak invertion works better in distinguishing oil and reservoir.

Maichen Depression lie between Leizhou Peninsula and Qiongzhou Strait. Oil and gas have been discovered in Weixinan Depression, Wushi Depression and Fushan Depression, which pertain to a same basin — North Sea Basin along with Maichen Depression.Jiangsu Oil started exploration at 2002. The first well began to drill at November, 2004 after gravity survey, electric method prospecting and 2D seismic exploration had been finished. Generating rock and hydrocarbon shows have been verified by the drilling. Low yield oil stream has been tested. And we started 3D seismic exploration at November, 2005. My thesis topic came from the actual needs of our exploration in the Maichen Depression. In the thesis, I give emphasis to analyse the own seismic geologic conditions of Maichen Depression. By real tests, we choosed the means to overcome or weaken the unfavorably impress owing to the own coditions in Maichen Depression. Finally, we obtained the usable seismic data. 1. Owing to the multiphase eruptive rock during the Quaternary Period, the near surface layers are very inhomogeneous. By simultaneous testing at same point with short refraction, uphole surveys of radial source and of surface source, the most appropriate method had been sorted out. Radial source uphole survey has been regarding the best practicable means in the complex area. Accurate surficial geology was very helpful to choosing of acquirement means and parameters. Basically the appropriate method of seismic acquirement has been built at Maichen area. 2. The seismic primary data has many, very strong and complex noise. By noise characteristic analysis in different domain, many means of denoising had been paralleled individual and joint application researched. As a result, the pre-stack multidomain joint denoise flow was the appropriate method. It can improve the seismic signal-to-noise ratio. 3. The problem of seismic static correction at Maichen Depression is very conspicuous. Many static correction methods had been tested individual and joint researched. The seismic data quality has been improved after choosing the appropriate combination of static correction flows. 4. Although the above-mentioned process are resultful, the seismic profile quality is just passable. Some reflector continuity and fault zone imagery are ambiguity.
