Drug development is being continuously scrutinised for its lack of productivity. Novel drug development is associated with high costs, high failure rates and lengthy development process. These downfalls combined with a huge demand in blood cancer for new therapeutic treatments have led many to consider the method of drug repurposing. Finding new therapeutic indications for already established drug substances is known as redirecting, repositioning, reprofiling, or repurposing of drugs. Off-patent and on-patent drugs can be screened for additional targets and new indications thus bringing them to clinical trials at a faster pace. This approach offers smaller research groups, such as those that are academic based, into the drug development industry. Drug repurposing can make use of previously published data concerning dosage, toxicology and mechanism of activity.

Electroless nickel composite coatings with silicon carbide, SiC, as reinforcing particles deposited with Ni–P onto aluminium alloy, LM24, having zincating as under layer were subjected to heat treatment using air furnace. The changes at the interface were investigated using scanning electron microscope (SEM) and energy dispersive X-ray (EDX) to probe the chemistry changes upon heat treatment. Microhardness tester with various loads using both Knoop and Vickers indenters was used to study the load effect clubbed with the influence of second phase particles on the coating at the vicinity of the interface. It was observed that zinc was absent at the interface after elevated temperature heat treatment at 400–500 °C. Precipitation of copper and nickel with a distinct demarcation (copper rich belt) along the coating interface was seen with irregular thickness of the order of 1 μm. Migration of copper from the bulk aluminium alloy could have been the factor. Brittleness of the coating was confirmed on heat treatment when indented with Vickers. However, in composite coating the propagation of the microcrack was stopped by the embedded particles but the microcracks continue in the matrix when not interrupted by second phase particles (SiC).

The microhardness is increased on incorporation of SiC in Ni–P matrix. The hardening mechanism is the formation of intermetallic phase Ni3P on annealing at elevated temperature.

Electroless nickel (EN) and electroless nickel composite (ENC) coatings were deposited on aluminium alloy substrate, LM24. The micro abrasion test was conducted to study the wear behaviour of the coatings with the effect of SiC concentration. Microhardness of the coatings was tested also. The wear scars were analysed using optical microscope and scanning electron microscope (SEM). The wear resistance was found to be improved in composite coating that has higher microhardness as compared to particles free and the bare aluminium substrate. In as-deposited condition for the composite coating, the wear volume increases on increase in SiC percentage in the coating but is found to be minimum for lower SiC percentage. The increase in hardness on heat treatment at 400°C is due to the hardening or grain coarsening with the formation Ni3P.

Carbon-flow from plant roots to the rhizosphere provides a major source of nutrients for the soil microbial population. However, quantification of carbon-flow is problematic due to its complex composition. This study investigated the potential of lux-marked Pseudomonas fluorescens to discriminate between forms of carbon present in the rhizosphere by measuring the light response to a range of carbon compounds. Results indicate that bioluminescence of short-term carbon-starved P. fluorescens is dependent upon the source and concentration of carbon. This system, therefore, has the potential to both quantify and qualify organic acids present in rhizodeposits.

Prominent theories of plant defence have predicted that plants growing on nutrient-poor soils produce more phenolic defence compounds than those on richer soils. Only recently has the Protein Competition Model (PCM) of phenolic allocation suggested that N and P limitation could have different effects because the nutrients are involved in different cellular metabolic processes. 2. We extend the prediction of the PCM and hypothesize that N will have a greater influence on the production of phenolic defensive compounds than P availability, because N limitation reduces protein production and thus competition for phenylalanine, a precursor of many phenolic compounds. In contrast, P acts as a recyclable cofactor in these reactions, allowing protein and hence phenolic production to continue under low P conditions. 3. We test this hypothesis by comparing the foliar concentrations of phenolic compounds in (i) phenotypes of 21 species growing on P-rich alluvial terraces and P-depleted marine terraces in southern New Zealand, and (ii) 87 species growing under similar climates on comparatively P-rich soils in New Zealand vs. P-depleted soils in Tasmania. 4. Foliar P concentrations of plants from the marine terraces were about half those of plants from alluvial soils, and much lower in Tasmania than in New Zealand. However, foliar concentrations of N and phenolic compounds were similar across sites in both comparisons, supporting the hypothesis that N availability is a more important determinant of plant investment in phenolic defensive compounds than P availability. We found no indication that reduced soil P levels influenced plant concentrations of phenolic compounds. There was wide variation in the foliar N and P concentrations among species, and those with low foliar nutrient concentrations produced more phenolics (including condensed tannins). 5. Our study is the first trait comparison extending beyond standard leaf economics to include secondary metabolites related to defence in forest plants, and emphasizes that N and P have different influences on the production of phenolic defence compounds. © 2009 British Ecological Society.

Persistent organic pollutants (POPs) are toxic substances, highly resistant to environmental degradation, which can bio-accumulate and have long-range atmospheric transport potential (UNEP 2001). The majority of studies on endocrine disruption have focused on interferences on the sexual steroid hormones and so have overlooked disruption to glucocorticoid hormones. Here the endocrine disrupting potential of individual POPs and their mixtures has been investigated in vitro to identify any disruption to glucocorticoid nuclear receptor transcriptional activity. POP mixtures were screened for glucocorticoid receptor (GR) translocation using a GR redistribution assay (RA) on a CellInsight(TM) NXT High Content Screening (HCS) platform. A mammalian reporter gene assay (RGA) was then used to assess the individual POPs, and their mixtures, for effects on glucocorticoid nuclear receptor transactivation. POP mixtures did not induce GR translocation in the GR RA or produce an agonist response in the GR RGA. However, in the antagonist test, in the presence of cortisol, an individual POP, p,p'-dichlorodiphenyldichloroethylene (DDE), was found to decrease glucocorticoid nuclear receptor transcriptional activity to 72.5% (in comparison to the positive cortisol control). Enhanced nuclear transcriptional activity, in the presence of cortisol, was evident for the two lowest concentrations of perfluorodecanoic acid (PFOS) potassium salt (0.0147mg/ml and 0.0294mg/ml), the two highest concentrations of perfluorodecanoic acid (PFDA) (0.0025mg/ml and 0.005mg/ml) and the highest concentration of 2,2',4,4'-tetrabromodiphenyl ether (BDE 47) (0.0000858mg/ml). It is important to gain a better understanding of how POPs can interact with GRs as the disruption of glucocorticoid action is thought to contribute to complex diseases.

Fermi-level pinning of aluminium on n-type germanium (n-Ge) was reduced by insertion of a thin interfacial dielectric by atomic layer deposition. The barrier height for aluminium contacts on n-Ge was reduced from 0.7 eV to a value of 0.28 eV for a thin Al2O3 interfacial layer (∼2.8 nm). For diodes with an Al2O3 interfacial layer, the contact resistance started to increase for layer thicknesses above 2.8 nm. For diodes with a HfO2 interfacial layer, the barrier height was also reduced but the contact resistance increased dramatically for layer thicknesses above 1.5 nm.

Recently, new lines of yellow-seeded (CS-Y) and black-seeded canola (CS-B) have been developed with chemical and structural alteration through modern breeding technology. However, no systematic study was found on the bioactive compounds, chemical functional groups, fatty acid profiles, inherent structure, nutrient degradation and absorption, or metabolic characteristics between the newly developed yellow- and black-seeded canola lines. This study aimed to systematically characterize chemical, structural, and nutritional features in these canola lines. The parameters accessed include bioactive compounds and antinutrition factors, chemical functional groups, detailed chemical and nutrient profiles, energy value, nutrient fractions, protein structure, degradation kinetics, intestinal digestion, true intestinal protein supply, and feed milk value. The results showed that the CS-Y line was lower (P ≤ 0.05) in neutral detergent fiber (122 vs 154 g/kg DM), acid detergent fiber (61 vs 99 g/kg DM), lignin (58 vs 77 g/kg DM), nonprotein nitrogen (56 vs 68 g/kg DM), and acid detergent insoluble protein (11 vs 35 g/kg DM) than the CS-B line. There was no difference in fatty acid profiles except C20:1 eicosenoic acid content (omega-9) which was in lower in the CS-Y line (P < 0.05) compared to the CS-B line. The glucosinolate compounds differed (P < 0.05) in terms of 4-pentenyl, phenylethyl, 3-CH3-indolyl, and 3-butenyl glucosinolates (2.9 vs 1.0 μmol/g) between the CS-Y and CS-B lines. For bioactive compounds, total polyphenols tended to be different (6.3 vs 7.2 g/kg DM), but there were no differences in erucic acid and condensed tannins with averages of 0.3 and 3.1 g/kg DM, respectively. When protein was portioned into five subfractions, significant differences were found in PA, PB1 (65 vs 79 g/kg CP), PB2, and PC fractions (10 vs 33 g/kg CP), indicating protein degradation and supply to small intestine differed between two new lines. In terms of protein structure spectral profile, there were no significant differences in functional groups of amides I and II, α helix, and β-sheet structure as well as their ratio between the two new lines, indicating no difference in protein structure makeup and conformation between the two lines. In terms of energy values, there were significant differences in total digestible nutrient (TDN; 149 vs 133 g/kg DM), metabolizable energy (ME; 58 vs 52 MJ/kg DM), and net energy for lactation (NEL; 42 vs 37 MJ/kg DM) between CS-Y and CS-B lines. For in situ rumen degradation kinetics, the two lines differed in soluble fraction (S; 284 vs 341 g/kg CP), potential degradation fraction (D; 672 vs 590 g/kg CP), and effective degraded organic matter (EDOM; 710 vs 684 g/kg OM), but no difference in degradation rate. CS-Y had higher digestibility of rumen bypass protein in the intestine than CS-B (566 vs 446 g/kg of RUP, P < 0.05). Modeling nutrient supply results showed that microbial protein synthesis (MCP; 148 vs 171 g/kg DM) and rumen protein degraded balance (DPB; 108 vs 127 g/kg DM) were lower in the CS-Y line, but there were no differences in total truly digested protein in small intestine (DVE) and feed milk value (FMV) between the two lines. In conclusion, the new yellow line had different nutritional, chemical, and structural features compared to the black line. CS-Y provided better nutrient utilization and availability.

This paper builds on previous work to show how using holistic and iterative design optimisation tools can be used to produce a commercially viable product that reduces a costly assembly into a single moulded structure. An assembly consisting of a structural metallic support and compression moulding outer shell undergo design optimisation and analysis to remove the support from the assembly process in favour of a structural moulding. The support is analysed and a sheet moulded compound (SMC) alternative is presented, this is then combined into a manufacturable shell design which is then assessed on viability as an alternative to the original.
Alongside this a robust material selection system is implemented that removes user bias towards materials for designs. This system builds on work set out by the Cambridge Material Selector and Boothroyd and Dewhurst, while using a selection of applicable materials currently available for the compression moulding process. This material selection process has been linked into the design and analysis stage, via scripts for use in the finite element environment. This builds towards an analysis toolkit that is suggested to develop and enhance manufacturability of design studies.

Sweet cherries (Prunus avium L.) ‘Sweetheart’ were harvested at different production regions from Portugal (Cova da Beira and Portalegre) and Spain (Valle de Jerte). Cherries were harvested at their commercial maturation according to the empirical knowledge of external color corresponding to good quality. Fruits were stored and evaluated in order to study their quality on the harvest day and during a period of 21 days, at cold storage (1 ºC, 95% RH). The sweet cherry ‘Sweetheart’ is a well-known variety and a highly appreciated one but fruits present a short shelf life. On the other hand the effect of different “terroir” on cherry characteristics should be known and clarified. Fruits from day 0, considered without storage, were kept at 20ºC and analyzed. Every weak, 3 replicas were randomly picked up and 10 fruits from each one were submitted to several analyses after fruit temperature stabilized at 20ºC. Several quality parameters were evaluated: external colour (L*, a*, b*), texture, soluble solids content (SSC), titratable acidity (TA) and the ratio between soluble solid contents (SSC) and tritratable acidity (TA). Fruits from different orchards and locations were significantly different according to these parameters. Fruits from Cova da Beira were less firm comparing with other two regions, Valle de Jerte and Portalegre, which may indicate a higher maturation rate at harvest in those fruits. This is in accordance with SSC/titratable acidity rate suggesting a late harvest in Cova da Beira comparing with other two orchards, however fruits from Cova da Beira exhibit a poor color at harvest. These results clearly showed a lower correlation between SSC and firmness considering fruits origin.

Com o presente trabalho pretendeu-se determinar e compreender melhor quais os alvos do Alumínio (Al) nas plantas, e contribuir para um melhor entendimento dos mecanismos de tolerância presentes em genótipos com elevado grau de tolerância ao Al. O Al é um dos maiores constituintes do solo e torna-se biodisponível em solos com baixo pH. Nesses casos, a exposição ao Al afecta negativamente o crescimento das plantas conduzindo a uma diminuição da produção. Estes factos são especialmente visíveis nos cereais, sendo a exposição ao Al uma das principais causas das quebras de produção nestas espécies. O Capítulo I consiste numa revisão geral sobre a toxicidade do Al nas plantas, apontando os seus principais alvos. Apresenta também os mecanismos de resistência, que inclui Al-destoxificação externa e interna, em diferentes espécies. O Capítulo II aborda os estudos sobre a exposição de curto prazo ao Al em duas espécies de cereais: Triticum aestivum L. e Secale cereale L., tendo-se sempre utilizado um genótipo Al-tolerante e um Al-sensível para cada espécie. Este capítulo está dividido em três estudos: no Capítulo II.1 realça-se o efeito da exposição a 185 μM de Al no equilíbrio nutricional em trigo. Verificou-se que em ambos os genótipos (sensível e tolerante) o perfil de macro e micro nutrientes se alterou, tendo uma interferência negativa, sobretudo no nível de P, Mg e K. Além disso, registaram-se diferenças na diferenciação da endoderme consoante o grau de tolerância/sensibilidade do genótipo. No Capítulo II.2 apresenta-se uma visão mais abrangente dos efeitos da exposição a 185 μM de Al em trigo, incluindo parâmetros fisiológicos, estruturais, citológicos e genotóxicos. Demonstra-se, pela primeira vez, que a progressão do ciclo celular é diferentemente regulada, dependendo da tolerância/sensibilidade do genótipo e que, mesmo em zonas já diferenciadas da raiz a exposição ao Al leva à deposição de calose. O Capítulo II.3 aborda os efeitos da exposição de 1.1 mM de Al em centeio, numa perspectiva bastante alargada. Apresenta-se o desequilíbrio nutricional, sobretudo no genótipo sensível, assim como a translocação de Al para a parte aérea nesse mesmo genótipo. Analisa-se também o comportamento de ambos os genótipos no que se refere ao ciclo celular, diferenciação da endoderme, crescimento radicular, reservas de hidratos de carbono, entre outros. Os resultados apontam para estratégias bem definidas adoptadas pelo genótipo tolerante de forma a minimizar a acção do Al no sistema radicular. O Capítulo III compreende a exposição longa ao Al. Dois genótipos de centeio com diferentes graus de tolerância ao Al foram expostos a 1.11 mM e 1.85 mM de Al durante 21 dias, tendo sido usados dois pontos de amostragem (15 e 21 dias). Este capítulo está dividido em dois estudos: No Capítulo III. 1 analisamse os mecanismos antioxidantes (folhas e raízes) como resposta à exposição ao Al, dando-se especial atenção ao ciclo do ascorbato-glutationas. A exposição ao Al levou a stress oxidativo e a alterações na actividade de enzimas antioxidantes e no conteúdo de antioxidantes não-enzimáticos. Demonstra-se que os dois órgãos apresentam respostas diferentes à exposição ao Al e que a capacidade de sobreviver em ambientes ricos em Al depende da eficácia da resposta antioxidante. Para além disso, a resposta do ciclo ascorbato-glutationas parece estar dependente do tipo de órgão, grau de tolerância e do tempo de exposição ao Al. No Capítulo III. 2 analisam-se os efeitos da exposição ao Al na fotossíntese. Verificou-se que o Al afecta negativamente a taxa fotossintética em ambos os genótipos, embora as alterações que o Al provoca nas trocas gasosas e no Ciclo de Calvin sejam dependentes do genótipo. Verificou-se também que os danos no genótipo sensível surgem mais cedo do que no genótipo tolerante, mas que ambos apresentam susceptibilidade ao Al após exposição de longo termo. Por fim, no Capítulo IV são apresentadas as conclusões da Tese de Doutoramento.
