The aim of the study was to compare the efficacy and safety of transvaginal trocar-guided polypropylene mesh insertion with traditional colporrhaphy for treatment of anterior vaginal wall prolapse.This is a randomized controlled trial in which women with advanced anterior vaginal wall prolapse, at least stage II with Ba a parts per thousand yenaEuro parts per thousand+1 cm according to the Pelvic Organ Prolapse Quantification (POP-Q) classification, were randomly assigned to have either anterior colporrhaphy (n = 39) or repair using trocar-guided transvaginal mesh (n = 40). the primary outcome was objective cure rate of the anterior compartment (point Ba) assessed at the 12-month follow-up visit, with stages 0 and I defined as anatomical success. Secondary outcomes included quantification of other vaginal compartments (POP-Q points), comparison of quality of life by the prolapse quality of life (P-QOL) questionnaire, and complication rate between the groups after 1 year. Study power was fixed as 80 % with 5 % cutoff point (p < 0.05) for statistical significance.The groups were similar regarding demographic and clinical preoperative parameters. Anatomical success rates for colporrhaphy and repair with mesh placement groups were 56.4 vs 82.5 % (95 % confidence interval 0.068-0.54), respectively, and the difference between the groups was statistically significant (p = 0.018). Similar total complication rates were observed in both groups, with tape exposure observed in 5 % of the patients. There was a significant improvement in all P-QOL domains as a result of both procedures (p < 0.001), but they were not distinct between groups (p > 0.05).Trocar-guided transvaginal synthetic mesh for advanced anterior POP repair is associated with a higher anatomical success rate for the anterior compartment compared with traditional colporrhaphy. Quality of life equally improved after both techniques. However, the trial failed to detect differences in P-QOL scores and complication rates between the groups.

RESUMO: Este trabalho teve como objetivo comparar a severidade da mancha de Alternaria de genótipos de girassol em ambientes do Cerrado do Distrito Federal. Três experimentos foram avaliados, dois na Embrapa Cerrados (Planaltina, DF) e um localizado na Embrapa Produtos e Mercado (Recanto das Emas, DF). Foram feitas análises ao longo do ciclo da cultura com um intervalo de 15 dias, sendo a primeira aos 35 dias após emergência. Cada planta foi analisada em três partes: terço inferior, terço médio e terço superior. Ao final de cada experimento, a área abaixo da curva de progresso da doença (AACPD) foi calculada para cada genótipo. De acordo com os resultados observados, o ambiente de sequeiro na Embrapa Cerrados, que deteve a maior média da severidade da doença, entre os ambientes avaliados. Dentre os genótipos avaliados, MG 360 mostrou menor severidade da doença no sequeiro e AGUARA 06 no irrigado da Embrapa Cerrados. O genótipo BSG 42 na Embrapa Produtos e Mercado, se destacou pela menor severidade do fungo. Houve influência significativa do ambiente nos resultados de severidade da doença, que está estreitamente relacionada com os fatores climáticos como temperatura e umidade relativa do ar. A compreensão dessas condições favoráveis ao desenvolvimento do fungo é essencial quando se avalia a resistência em genótipos de girassol. ABSTRACT: This study had the purpose of comparing the severity of Alternaria leaf spot on genotypes of sunflower in the Brazilian Cerrado. Three field trials were established, two at Embrapa Cerrados (Planaltina, DF) and one at Embrapa Produtos e Mercados (Recanto das Emas, DF). Evaluations were made during the crop cycle every 15 days, the first one happening 35 days after crop emergency. Each plant was analyzed in three parts: lower, middle and upper thirds. At the end of each trial the average area under the disease progress curve (AUDPC), for each third, was calculated for all genotypes. According to the observed results, the dryland environment at Embrapa Cerrados had the highest mean disease severity, among the studied environments. Between the evaluated genotypes, MG360 showed less severity in dryland and AGUARA 06 in the irrigated environment at Embrapa Cerrados. At Embrapa Produtos e Mercado the genotype that presented the best performance was BRS G42. Environmental conditions deeply affected the trials, mainly temperature and relative humidity. Their influence on Alternaria leaf spot epidemics in the Cerrado region must be studied in more detail, to avoid misinterpreting data when evaluating sunflower genotypes for resistance to this important fungal disease.

RESUMO: O presente trabalho tem por objetivo caracterizar o comportamento de 16 genótipos de girassol em ambiente de sequeiro e ambiente irrigado do Cerrado do Distrito Federal visando aumentar disponibilidade de cultivares mais produtivas e adaptadas. Os ensaios foram conduzidos na área experimental da Embrapa Cerrados, Planaltina, DF. Os ensaios foram arranjados experimentalmente em blocos ao acaso, com quatro repetições. Os genótipos avaliados foram: CF 101, ADV 5504, BRS G42, BRS 323, HELIO 250, HELIO 251, SYN 045, SYN 3950HO, MG 305, MG 360, AGUARÁ 04, AGUARÁ 06, PARAÍSO 20, GNZ NEON, HLA 2012 e M734. Os caracteres avaliados foram rendimento estimado de grãos, tamanho do capítulo, peso de mil aquênios, altura de plantas, teor de óleo e dias para floração inicial. Houve diferenças significativas entre os genótipos para todas as características avaliadas. Dos 16 genótipos avaliados, os híbridos SYN 045 (3.786,2 kg ha-1) e MG 305 (4.987,2 kg ha-1) se sobressaíram quanto ao rendimento estimado de grãos. Quanto ao teor de óleo, os híbridos MG 306 (53,95%) e SYN 3950HO (49,49%) se destacaram. Foram identificados os genótipos mais promissores dentre os avaliados, podendo ser explorados em programas de melhoramento que visam o desenvolvimento de cultivares mais adaptadas.

RESUMO: O objetivo deste trabalho foi estimar caracteres genéticos, fenotípicos e ambientais relacionados aos componentes de produção e caracteres agromorfológicos de girassol em três núcleos rurais do Distrito Federal. Os experimentos foram conduzidos nas áreas experimentais da Embrapa Cerrados, Planaltina, DF, Embrapa Produtos e Mercado, Recanto das Emas, DF e na Fazenda Agua Limpa, da Universidade de Brasília. Através dos resultados obtidos, foram verificadas diferenças significativas entre os genótipos de girassol nos três núcleos rurais do Cerrado para todas as características agronômicas avaliadas. Ainda, baixos coeficientes de variação ambiental para quase todas as características, exceto para o tamanho de capitulo, indicaram boa precisão experimental e altos valores de herdabilidade, coeficientes de variação genéticos e acurácia evidenciaram condições favoráveis à seleção dos materiais para as características agronômicas avaliadas. ABSTRACT: The purpose of this study was to assess genetic, phenotypic and environmental characteristics related to agro-morphological traits of sunflower in three rural centers of Distrito Federal. The experiments were conducted at the experimental areas of Embrapa Cerrados, Planaltina, DF, Embrapa Produtos e Mercado, Recanto das Emas, DF e na Fazenda Agua Limpa, da Universidade de Brasília. Through the obtained results, were verified significate genotypic differences of sunflower at the three rural centers of Brazilian Savannah for all traits evaluated. In addition, low coefficients of environmental variation for almost all trait, except the head size, indicate good experimental precision and high values of heritability, genetic variation and accuracy showed favorable conditions to selecting materials for the agronomic traits evaluated.

RESUMO:Com o objetivo de avaliar o comportamento agronômico de genótipos de girassol no Cerrado do Distrito Federal, foram conduzidos ensaios na safrinha dos anos de 2014 e 2015, na estação experimental da Embrapa Cerrados, Planaltina, DF. O delineamento experimental foi de blocos ao acaso com quatro repetições, e foram avaliados 12 genótipos de girassol: HLA 2015, NTC 90, SYN 065, M734, BRS G44, HLA 2014, BRS G45, BRS G43, HLA 2013, HLA 2017, BRS G46, HLA 2016. As características avaliadas foram rendimento de grãos, tamanho do capitulo, peso de mil aquênios, altura de plantas e dias de floração inicial. Diferenças significativas foram encontradas para as características avaliadas. Os genótipos que se destacaram em relação ao rendimento de grãos foram HLA 2014 (3.161 kg ha-1) e a testemunha M743 (3.212 kg ha-1). Além disso, o ensaio do ano 2014 apresentou uma média de rendimento maior (2.829 kg ha-1) e mais precoces (63,10 dias) em relação a 2015. O trabalho permitiu a identificação de materiais promissores para exploração em programas de melhoramento genético. ABSTRACT: Aiming the evaluation on agronomic behavior of sunflower genotypes in the Brazilian savannah, experiments were carried on in the second crop of 2014 and 2015 at Centro de Pesquisa Agropecuária dos Cerrados (Embrapa), Planaltina, DF. A complete randomized block design was used with four replications and 12 genotypes of sunflower were analyzed: HLA 2015, NTC 90, SYN 065, M734, BRS G44, HLA 2014, BRS G45, BRS G43, HLA 2013, HLA 2017, BRS G46, HLA 2016.The evaluated characteristics were grain yield, head, weight thousand achenes, plant height, and flowering time. Significant differences were found in all evaluated characteristics. The genotypes that stood out in seed yield were HLA 2014 (3161 kg ha-1) and M743 (3212 kg ha-1). Besides, the 2014 experiment presented a seed yield average higher (2829 kg ha-1), and earlier flowering (63,10 days) when compared to 2015 experiment. This study allowed the identification of promising materials to explore in breeding programs.

El presente artículo es un documento conjunto de la Sociedad Española de Geriatría y Gerontología, la Sociedad Española de Cuidados Paliativos y la Sección de Cardiología Geriátrica de la Sociedad Española de Cardiología. Su objetivo es paliar la laguna que existe en España en lo que respecta al manejo de los desfibriladores automáticos implantables (DAI) en las fases finales de la vida. Cada vez es más frecuente encontrarse enfermos portadores de DAI en fase terminal de una enfermedad avanzada, como insuficiencia cardíaca refractaria, enfermedades oncológicas, otras insuficiencias orgánicas o enfermedades neurodegenerativas con mal pronóstico a corto plazo. La enorme mayoría de estos pacientes tiene más de 65 años, por ello el documento se enfoca de forma particular a los ancianos que se encuentran en esta situación, aunque el proceso de toma de decisiones es similar en portadores de DAI más jóvenes que están en la fase final de su vida.

McArdle disease, caused by inherited deficiency of the enzyme muscle glycogen phosphorylase (GP-MM), is arguably the paradigm of exercise intolerance. The recent knock-in (p.R50X/p.R50X) mouse disease model allows an investigation of the phenotypic consequences of muscle glycogen unavailability and the physiopathology of exercise intolerance. We analysed, in 2-month-old mice [wild-type (wt/wt), heterozygous (p.R50X/wt) and p.R50X/p.R50X)], maximal endurance exercise capacity and the molecular consequences of an absence of GP-MM in the main glycogen metabolism regulatory enzymes: glycogen synthase, glycogen branching enzyme and glycogen debranching enzyme, as well as glycogen content in slow-twitch (soleus), intermediate (gastrocnemius) and glycolytic/fast-twitch (extensor digitorum longus; EDL) muscles.

McArdle disease is arguably the paradigm of exercise intolerance in humans. This disorder is caused by inherited deficiency of myophosphorylase, the enzyme isoform that initiates glycogen breakdown in skeletal muscles. Because patients are unable to obtain energy from their muscle glycogen stores, this disease provides an interesting model of study for exercise physiologists, allowing insight to be gained into the understanding of glycogen-dependent muscle functions. Of special interest in the field of muscle physiology and sports medicine are also some specific (if not unique) characteristics of this disorder, such as the so-called 'second wind' phenomenon, the frequent exercise-induced rhabdomyolysis and myoglobinuria episodes suffered by patients (with muscle damage also occurring under basal conditions), or the early appearance of fatigue and contractures, among others. In this article we review the main pathophysiological features of this disorder leading to exercise intolerance as well as the currently available therapeutic possibilities.

McArdle disease is a metabolic disorder caused by pathogenic mutations in the PYGM gene. Timely diagnosis can sometimes be difficult with direct genomic analysis, which requires additional studies of cDNA from muscle transcripts. Although the "nonsense-mediated mRNA decay" (NMD) eliminates tissue-specific aberrant transcripts, there is some residual transcription of tissue-specific genes in virtually all cells, such as peripheral blood mononuclear cells (PBMCs).We studied a subset of the main types of PYGM mutations (deletions, missense, nonsense, silent, or splicing mutations) in cDNA from easily accessible cells (PBMCs) in 12 McArdle patients.Analysis of cDNA from PBMCs allowed detection of all mutations. Importantly, the effects of mutations with unknown pathogenicity (silent and splicing mutations) were characterized in PBMCs. Because the NMD mechanism does not seem to operate in nonspecific cells, PBMCs were more suitable than muscle biopsies for detecting the pathogenicity of some PYGM mutations, notably the silent mutation c.645G>A (p.K215=), whose effect in the splicing of intron 6 was unnoticed in previous muscle transcriptomic studies.We propose considering the use of PBMCs for detecting mutations that are thought to cause McArdle disease, particularly for studying their actual pathogenicity.

We recently generated a knock-in mouse model (PYGM p.R50X/p.R50X) of McArdle disease (myophosphorylase deficiency). One mechanistic approach to unveil the molecular alterations caused by myophosphorylase deficiency, which is arguably the paradigm of 'exercise intolerance', is to compare the skeletal-muscle tissue of McArdle, heterozygous, and healthy (wild type (wt)) mice. We analyzed in quadriceps muscle of p.R50X/p.R50X (n=4), p.R50X/wt (n=6) and wt/wt mice (n=5) (all male, 8 wk-old) molecular markers of energy-sensing pathways, oxidative phosphorylation (OXPHOS) and autophagy/proteasome systems, oxidative damage and sarcoplamic reticulum (SR) Ca handling. We found a significant group effect for total AMPK (tAMPK) and ratio of phosphorylated (pAMPK)/tAMPK (P=0.012 and 0.033), with higher mean values in p.R50X/p.R50X mice vs. the other two groups. The absence of massive accumulation of ubiquitinated proteins, autophagosomes or lysosomes in p.R50X/p.R50X mice suggested no major alterations in autophagy/proteasome systems. Citrate synthase activity was lower in p.R50X/p.R50X mice vs. the other two groups (P=0.036) but no statistical effect existed for respiratory chain complexes. We found higher levels of 4-hydroxy-2-nonenal-modified proteins in p.R50X/p.R50X and p.R50X/wt mice compared with the wt/wt group (P=0.011). Sarco(endo)plasmic reticulum ATPase 1 (SERCA1) levels detected at 110kDa tended to be higher in p.R50X/p.R50X and p.R50X/wt mice compared with wt/wt animals (P=0.076), but their enzyme activity was normal. We also found an accumulation of phosphorylated SERCA1 in p.R50X/p.R50X animals. Myophosphorylase deficiency causes alterations in sensory energetic pathways together with some evidence of oxidative damage and alterations in Ca handling but with no major alterations in OXPHOS capacity or autophagy/ubiquitination pathways, which suggests that the muscle tissue of patients is likely to adapt overall favorably to exercise training interventions.

This thesis creates a multi-faceted archaeological context for early Irish monasticism, so as to ‘rematerialise’ a phenomenon that has been neglected by recent archaeological scholarship. Following revision of earlier models of the early Irish Church, archaeologists are now faced with redefining monasticism and distinguishing it from other diverse forms of Christian lifestyle. This research addresses this challenge, exploring the ways in which material limits can be set on the monastic phenomenon. The evidence for early Irish monasticism does not always conform to modern expectations of its character, and monastic space must be examined as culturally unique in its own right - though this thesis demonstrates that early Irish monasticism was by no means as unorthodox in its contemporary European setting as has previously been suggested. The research is informed by theories of the body, habitus and space, drawing on a wide body of archaeological, religious, sociological and anthropological thought. The data-set comprises evidences gathered through field-survey, reassessment of archaeological scholarship, historical research and cartographic research, enabling consideration of the ways in which early Irish monastics engaged with their environments. A sample of thirty-one early Irish ecclesiastical sites plus Iona forms the basis for discussion of the location and layout of monastic space, the ways in which monastics used buildings and space in their daily lives, the relationship of monasticism and material culture, the setting of mental and physical limits on monastic space and monastic bodies, and the variety of monastic lifestyles that pertained in early medieval Ireland. The study then examines the Christian landscapes of two case-studies in mid-Western Ireland in order to illustrate how monasticism functioned on the ground in these areas. As this research shows, the material complexities of early Irish monastic life are capable of archaeological definition in terms of both communal and personal lived experience.

This thesis seeks to explore the development of sport in Munster in the late nineteenth and early twentieth centuries by comparing developments in three counties: Cork, Tipperary and Waterford. In particular this thesis considers the development of rugby and soccer in comparative perspective across these three counties, asking what local factors impacted on their uneven development in the region and considering the extent to which the traditional model of diffusion applies to the reception of these sports in the three counties. By giving consideration to these two particular non-indigenous sports, the thesis will, through answering that question, explore ideas of cultural reception, national identity and class as expressed at local level. These themes will be explored by placing the comparative analysis of these two sports into a wider context of sporting development regionally and nationally in the period, in particular the emerging commercialisation of sport, and also the diverse sporting culture of which these two sports were a part. Utilising a wide range of archival sources from local, national and sporting newspapers, to club records, official publications and ephemera this thesis builds a picture of sport in Munster that is deeply rooted in the community, and that forms an important facet of the social world of Cork, Tipperary and Waterford from 1880-1930.

This thesis explores the impact of Christianity on the landscape in Ireland from the conversion period to the coming of the Anglo-Normans. The premise is that ecclesiastical and secular settlement formed a cohesive whole which characterised the societal organisation of early medieval Ireland. The matter of the thesis is to isolate some of the agents of cohesion to see was this homogenous or did it vary in different areas. One of these agents was the ownership of land and the thesis undertakes to identify ecclesiastical landholding and examine the manner of settlement on it. A corollary is to explore the contribution of the genealogical link between kin-group, founding saint and territory to the construction of local identities. This necessitated a narrow focus; thus small study areas were chosen, which approximated to early medieval kingdoms in North Louth, Rathdown, Co Dublin and Ross, Co Cork. A multidisciplinary approach was taken using both archaeological and documentary evidence. The thesis found ecclesiastical sites were at the same density through the study areas, but there were considerable regional variations in related secular settlement. Ecclesiastical estates were identified in the three study areas and common settlement patterns were found in two of them. Settlement in all areas indicated the foundation of minor churches by local groups. Ecclesiastical sites were found to be integral to kin-group identity and status, but the manner in which each group negotiated this, was very different. Finally the thesis examined material evidence for a change from diffused to concentrated power in the political organisation of Irish society, a process entwined with developments of the Viking Age. This centralisation of power and associated re-formation of identity was still often mediated through the ecclesiastical sphere but the thesis demonstrates diversity in the materialising of the mediation.
