Lactic acid bacteria (LAB) are Gram-positive bacteria and are generally regarded as safe (GRAS) organisms. Therefore, LAB could be used for heterologous protein secretion and they are good potential candidates as antigen delivery vehicles. To develop such live vaccines, a better control of protein secretion is required. We developed an efficient secretion system in the model LAB, Lactococcus lactis. Staphylococcal nuclease (Nuc) was used as the reporter protein. We first observed that the quantity of secreted Nuc correlated with the copy number of the cloning vector. The nuc gene was cloned on a high-copy number cloning vector and no perturbation of the metabolism of the secreting strain was observed. Replacement of nuc native promoter by a strong lactococcal one led to a significant increase of nuc expression. Secretion efficiency (SE) of Nuc in L. lactis was low, i.e., only 60% of the synthesized Nuc was secreted. Insertion of a synthetic propeptide between the signal peptide and the mature moiety of Nuc increased the SE of Nuc. On the basis of these results, we developed a secretion system and we applied it to the construction of an L. lactis strain which secretes a bovine coronavirus (BCV) epitope-protein fusion (BCV-Nuc). BCV-Nuc was recognized by both anti-BCV and anti-Nuc antibodies. Secretion of this antigenic fusion is the first step towards the development of a novel antigen delivery system based on LAB-secreting strains.

The present study was carried out to evaluate the antioxidant and antimicrobial activities of a methanol extract of Bauhinia racemosa (MEBR) (Caesalpiniaceae) stem bark in various systems. 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) radical, superoxide anion radical, nitric oxide radical, and hydroxyl radical scavenging assays were carried out to evaluate the antioxidant potential of the extract. The antioxidant activity of the methanol extract increased in a concentration-dependent manner. About 50, 100, 250, and 500 µg MEBR inhibited the peroxidation of a linoleic acid emulsion by 62.43, 67.21, 71.04, and 76.83%, respectively. Similarly, the effect of MEBR on reducing power increased in a concentration-dependent manner. In DPPH radical scavenging assays the IC50 value of the extract was 152.29 µg/ml. MEBR inhibited the nitric oxide radicals generated from sodium nitroprusside with an IC50 of 78.34 µg/ml, as opposed to 20.4 µg/ml for curcumin. Moreover, MEBR scavenged the superoxide generated by the PMS/NADH-NBT system. MEBR also inhibited the hydroxyl radical generated by Fenton's reaction, with an IC50 value of more than 1000 µg/ml, as compared to 5 µg/ml for catechin. The amounts of total phenolic compounds were also determined and 64.7 µg pyrocatechol phenol equivalents were detected in MEBR (1 mg). The antimicrobial activities of MEBR were determined by disc diffusion with five Gram-positive, four Gram-negative and four fungal species. MEBR showed broad-spectrum antimicrobial activity against all tested microorganisms. The results obtained in the present study indicate that MEBR can be a potential source of natural antioxidant and antimicrobial agents.

We developed a forced non-electric-shock running wheel (FNESRW) system that provides rats with high-intensity exercise training using automatic exercise training patterns that are controlled by a microcontroller. The proposed system successfully makes a breakthrough in the traditional motorized running wheel to allow rats to perform high-intensity training and to enable comparisons with the treadmill at the same exercise intensity without any electric shock. A polyvinyl chloride runway with a rough rubber surface was coated on the periphery of the wheel so as to permit automatic acceleration training, and which allowed the rats to run consistently at high speeds (30 m/min for 1 h). An animal ischemic stroke model was used to validate the proposed system. FNESRW, treadmill, control, and sham groups were studied. The FNESRW and treadmill groups underwent 3 weeks of endurance running training. After 3 weeks, the experiments of middle cerebral artery occlusion, the modified neurological severity score (mNSS), an inclined plane test, and triphenyltetrazolium chloride were performed to evaluate the effectiveness of the proposed platform. The proposed platform showed that enhancement of motor function, mNSS, and infarct volumes was significantly stronger in the FNESRW group than the control group (P<0.05) and similar to the treadmill group. The experimental data demonstrated that the proposed platform can be applied to test the benefit of exercise-preconditioning-induced neuroprotection using the animal stroke model. Additional advantages of the FNESRW system include stand-alone capability, independence of subjective human adjustment, and ease of use.

Se realizó el estudio comparativo de distintas técnicas extractivas de naringina utilizando cáscaras de pomelo (Citrus paradisi). Las mejores condiciones de preparación previa del material son el fragmentado de cáscara fresca y el secado en estufa con posterior fragmentado. Para cáscara fresca resulta más conveniente la extracción con etanol puro por Soxhlet (obteniéndose 26g/kg de peso fresco), mientras que para cáscara seca los mejores resultados se obtienen en la extracción con la mezcla etanol-agua en las proporciones 70:30 por maceración (20g/kg de peso fresco) y reflujo (22g/kg de peso fresco). Se observaron evidencias de la importancia del agua en el proceso de extracción. Se determinaron los valores de concentración de naringina en cáscaras de distintas variedades de Citrus paradisi de la región del Noroeste Argentino.

Las papayas obtenidas en el raleo (papayas menos desarrolladas entresacadas de los árboles para beneficiar el crecimiento de las otras) generalmente se descartan. Alternativamente, estas papayas pueden secarse por un proceso de deshidratación osmótica y secado térmico convencional para usarlas como snacks o como ingrediente para otros productos. Se comparó la cinética de transferencia de masa en la deshidratación osmótica de papayas de raleo frente a papayas desarrolladas con diferente grado de madurez, considerando el efecto de la geometría de la muestra y de la presión del proceso. La deshidratación osmótica se efectuó en una solución agitada de sacarosa a 50 °Brix, a 25 °C. Se consideraron tres niveles de madurez: raleo, verde y pintona, tres geometrías: lámina, cilindro y anillo y dos niveles de presión: atmosférica y vacío. Se estudiaron como variables cinéticas la variación de peso (WR), pérdida de agua (WL) y ganancia de sólidos (SG) entre 10 y 180 min. La madurez tuvo efecto significativo sobre las tres respuestas de la cinética a 30 min y sobre SG a 180 min; la geometría tuvo efecto sobre WR y SG a 30 min y sobre los tres parámetros cinéticos a 180 min y la presión solamente tuvo efecto sobre WL y SG a 30 min. La mayor pérdida de agua (65%) se obtuvo a 30 min para la combinación raleo/cilindro/vacío; mientras que la mayor ganancia de sólidos fue 31% para el tratamiento verde/lámina/vacío a 180 min.

Anthocyanins are the pigments responsible for the color of most red grapes and are easily degraded following various reaction mechanisms affected by oxygen, enzymes, pH, and temperature among other variables. In this study, a jam model system was developed using Merlot and Bordô grape extracts and polysaccharides (xanthan and locust bean gums) and different temperatures (45, 55 and 65 °C). The stability of the anthocyanin pigments and the rheological behavior of the jam model system were studied. For the determination of the stability, the half-life time and first-order reaction rate constants for the anthocyanin pigments were calculated. The rheological behavior was determined through the Power law model. The jam model system produced using a temperature of 45 °C showed the best results for the anthocyanin half-life time. The first-order reaction rate constants for the 45, 55, and 65 °C treatments were not significantly different among each other (p > 0.05). It was observed that with an increase in the jam model system temperature there was an increase in the index of consistency.

Los objetivos de este trabajo fueron identificar los hongos asociados a las semillas de zanahoria de procedencia local y determinar la eficiencia del control químico sobre la flora fúngica transportada, en relación con la calidad fisiológica. Se analizó la calidad fisiológica y sanitaria de lotes de semillas de zanahoria provenientes de Córdoba, Argentina y se evaluó el efecto de tratamientos combinados de fungicidas en diferentes dosis y tiempos de inmersión. Se observó alta incidencia de hongos transportados con predominio de A. alternata, Fusarium sp. y Epicoccum sp los cuales afectaron la germinación y vigor de las semillas. Los tratamientos fungicidas redujeron el porcentaje de semillas infectadas y su efecto dependió principalmente del tiempo de inmersión. Los tratamientos más eficientes fueron Tebuconazole, Tiram y Carboxin + Tiram en dosis de 1.000 y 500 ppm de p.a. aplicados por inmersión durante una hora. Estos tratamientos indujeron además un incremento de la germinación de semillas de zanahoria.
