This paper first examines splits and mergers among Kenya’s political parties (and inner-party factions) from the restoration of a multi-party system in 1991 until 2007, before the turbulent 10th general elections were conducted. It then considers what functions “political parties” have in Kenya with special reference to the period since 2002, the year in which President Moi announced his intention to retire. A look back at NARC’s five years of rule reveals that, although it succeeded in changing the government, NARC, as a “political party,” remained throughout an organization without any real substance. The paper looks at (1) NARC’s de facto split after its overwhelming win in the ninth general election, (2) malfunctions of the anti-defection laws that were introduced in the 1960s, and (3) Kenya’s election rules that require candidates to be nominated by registered political parties in general elections. The paper proceeds to argue that as a result of the operation of these three elements, Kenya’s political parties, and especially the victorious coalition sides, tend to end up being nothing more than temporary vehicles for political elites angling for post-election posts.

This paper deals with the assessment of the contribution of the second flexural mode to the dynamic behaviour of simply supported railway bridges. Alluding to the works of other authors, it is suggested in some references that the dynamic behaviour of simply supported bridges could be adequately represented taking into account only the contribution of the fundamental flexural mode. On the other hand, the European Rail Research Institute (ERRI) proposes that the second mode should also be included whenever the associated natural frequency is lower than 30 Hz]. This investigation endeavours to clarify the question as much as possible by establishing whether the maximum response of the bridge, in terms of displacements, accelerations and bending moments, can be computed accurately not taking account of the contribution of the second mode. To this end, a dimensionless formulation of the equations of motion of a simply supported beam traversed by a series of equally spaced moving loads is presented. This formulation brings to light the fundamental parameters governing the behaviour of the beam: damping ratio, dimensionless speed $ \alpha$=VT/L, and L/d ratio (L stands for the span of the beam, V for the speed of the train, T represents the fundamental period of the bridge and d symbolises the distance between consecutive loads). Assuming a damping ratio equal to 1%, which is a usual value for prestressed high-speed bridges, a parametric analysis is conducted over realistic ranges of values of $ \alpha$ and L/d. The results can be extended to any simply supported bridge subjected to a train of equally spaced loads in virtue of the so-called Similarity Formulae. The validity of these formulae can be derived from the dimensionless formulation mentioned above. In the parametric analysis the maximum response of the bridge is obtained for one thousand values of speed that cover the range from the fourth resonance of the first mode to the first resonance of the second mode. The response at twenty-one different locations along the span of the beam is compared in order to decide if the maximum can be accurately computed with the sole contribution of the fundamental mode.

The paper shows the results of the new steps that have been done in the development of the tidal energy converter GESMEY. These are the design, construction and trials into the sea of a 1/10 scale prototype and also the construction with the same scale of the buoy BOSCEM, that anchors the device and lets it in the correct work position and depth, along the two directions of the flow that the daily tidal cycle have. Inside the paper is described the objectives and the methodology of the experimental trials that were ca rry out the last summer with the scale prototype. GESMEY is a new type of tidal energy converter (TEC) that has the capability to exploit currents in waters over forty meters by itself and it gets only using its internal ballast system the necessary equilibrium between hy drostatics and hydrodynamics forces to make the emersion and the immersion procedures without any other help. Finally the paper shows the description of the results obtained over the performance of the devices along the immersion, emersion and floating transport manoeuvres and afterwards the results, that were obtained along the generation power tests that were carried out, are shown.

Speech is the major function, emergence and which development radically changes all course of formation of the identity of the child already in the early childhood. If language and speech development in solitary born children is investigated today quite well, at twin children this process practically is not studied. Our research was carried out for the purpose of studying of an originality of mastering by speech by heterosexual children of pair of twins within communicative and pragmatist approach (T.N. Ushakov,G. V. Chirkina). Application of this approach to the analysis of process of communication at twin children allowed us to allocate those peculiar receptions and means of communication which they functionally develop in a situation of pair of twins, as allows them to show the phenomena of the speech which are not meeting at solitary born contemporaries. In this work results of supervision and research of pair of heterosexual twins of the second year of the life, carried out by a technique developed by us under the scientific guide of G. V. Chirkina

This paper deals with the assessment of the contribution of the second bending mode to the dynamic behavior of simply supported railway bridges. Traditionally the contributions of modes higher than the fundamental have been considered of little importance for the computation of the magnitudes of interest to structural engineers (vertical deflections, bending moments, etc.). Starting from the dimensionless equations of motion of a simply supported beam subjected to moving loads, the key parameters governing the dynamic behavior are identified. Then, a parametric study over realistic ranges of values of those parameters is conducted, and the influence of the second mode examined in detail. The main purpose is to decide whether the second mode should be taken into account for the determination of the maximum displacement and acceleration in high-speed bridges. In addition, the reasons that cause the contribution of the second bending mode to be relevant in some situations are highlighted, particularly with regard to the computation of the maximum acceleration.

ABSTRACT: The surveys carried out in the municipality of Pinto (Madrid) have enabled us to locate various structural remains linked to the military operations that took place around the capital during the Spanish Civil War (1936-1939). In order to identify and record them, surveys were complemented with the use of GPS and air photographs from different time periods. Afterwards, and in collaboration with researchers from various universities, further methods aimed at generating a complete special representation of the area were applied directly to one of the sites which produced the best results, known as "los Yesares". These methods include topographic mapping that resulted in cartographic material at different scales, the photographic recording with flying Unmanned Aerial Vehicles, and the use of land scanners and GPS-corrected photogrammetrics with which to obtain 3D models.

The AG dinucleotide at the 3′ splice sites of metazoan nuclear pre-mRNAs plays a critical role in catalytic step II of the splicing reaction. Previous studies have shown that replacement of the guanine by adenine in the AG (AG → GG) inhibits this step. We find that the second step was even more severely inhibited by cytosine (AG → CG) or uracil (AG → UG) substitutions at this position. By contrast, a relatively moderate inhibition was observed with a hypoxanthine substitution (AG → HG). When adenine was replaced by a purine base (AG → PG) or by 7-deazaadenine (AG → c7AG), little effect on the second step was observed, suggesting that the 6-NH2 and N7 groups do not play a critical role in adenine recognition. Finally, replacement of adenine by 2-aminopurine (AG → 2-APG) had no effect on the second step. Taken together, our results suggest that the N1 group of adenine functions as an essential determinant in adenine recognition during the second step of pre-mRNA splicing.

Persistent infection with hepatitis C virus (HCV) is among the leading causes of chronic liver disease. Previous studies suggested that genetic variation in hypervariable region 1 (HVR1) of the second envelope protein, possibly in response to host immune pressure, influences the outcome of HCV infection. In the present study, a chimpanzee transfected intrahepatically with RNA transcripts of an infectious HCV clone (pCV-H77C) from which HVR1 was deleted became infected; the ΔHVR1 virus was subsequently transmitted to a second chimpanzee. Infection with ΔHVR1 virus resulted in persistent infection in the former chimpanzee and in acute resolving infection in the latter chimpanzee. Both chimpanzees developed hepatitis. The ΔHVR1 virus initially replicated to low titers, but virus titer increased significantly after mutations appeared in the viral genome. Thus, wild-type HCV without HVR1 was apparently attenuated, suggesting a functional role of HVR1. However, our data indicate that HVR1 is not essential for the viability of HCV, the resolution of infection, or the progression to chronicity.

Cryptococcus neoformans STE12α, a homologue of Saccharomyces cerevisiae STE12, exists only in MATα strains. We identified another STE12 homologue, STE12a, which is MATa specific. As in the case with Δste12α, the mating efficiency for Δste12a was reduced significantly. The Δste12a strains surprisingly still mated with Δste12α strains. In MATα strains, STE12a functionally complemented STE12α for mating efficacy, haploid fruiting, and regulation of capsule size in the mouse brain. Furthermore, when STE12a was replaced with two copies of STE12α, the resulting MATa strain produced hyphae on filament agar. STE12a regulates mRNA levels of several genes that are important for virulence including CNLAC1 and CAP genes. STE12a also modulates enzyme activities of phospholipase and superoxide dismutase. Importantly, deletion of STE12a markedly reduced the virulence in mice, as is the case with STE12α. Brain smears of mice infected with the Δste12a strain showed yeast cells with a considerable reduction in capsule size compared with those infected with STE12a strains. When the disrupted locus of ste12a was replaced with a wild-type STE12a gene, both in vivo and in vitro mutant phenotypes were reversed. These results suggest that STE12a and STE12α have similar functions, and that the mating type of the cells influences the alleles to exert their biological effects. C. neoformans, thus, is the first fungal species that contains a mating-type-specific STE12 homologue in each mating type. Our results demonstrate that mating-type-specific genes are not only important for saprobic reproduction but also play an important role for survival of the organism in host tissue.

We had earlier identified the pcnB locus as the gene for the major Escherichia coli poly(A) polymerase (PAP I). In this report, we describe the disruption and identification of a candidate gene for a second poly(A) polymerase (PAP II) by an experimental strategy which was based on the assumption that the viability of E. coli depends on the presence of either PAP I or PAP II. The coding region thus identified is the open reading frame f310, located at about 87 min on the E. coli chromosome. The following lines of evidence support f310 as the gene for PAP II: (i) the deduced peptide encoded by f310 has a molecular weight of 36,300, similar to the molecular weight of 35,000 estimated by gel filtration of PAP II; (ii) the deduced f310 product is a relatively hydrophobic polypeptide with a pI of 9.4, consistent with the properties of partially purified PAP II; (iii) overexpression of f310 leads to the formation of inclusion bodies whose solubilization and renaturation yields poly(A) polymerase activity that corresponds to a 35-kDa protein as shown by enzyme blotting; and (iv) expression of a f310 fusion construct with hexahistidine at the N-terminus of the coding region allowed purification of a poly(A) polymerase fraction whose major component is a 36-kDa protein. E. coli PAP II has no significant sequence homology either to PAP I or to the viral and eukaryotic poly(A) polymerases, suggesting that the bacterial poly(A) polymerases have evolved independently. An interesting feature of the PAP II sequence is the presence of sets of two paired cysteine and histidine residues that resemble the RNA binding motifs seen in some other proteins.
