The authors have studied the chromatographic behavior of parenteral preparations for pediatric use containing inorganic cations. After separation and identification by thin-layer chromatography, Mn2+, Zn2+, and Cu2+ were analyzed by a method based on reaction with an appropriate reagent and extraction with an organic solvent which yielded elution and preconcentration, resulting in an appropriate solution for colorimetric quantitation. Cr3+ cation was determined by atomic absorption spectrophotometry after appropriate chromatographic separation, using microcrystalline cellulose (adsorbent) and an acetone:water:hydrochloric acid mixture (80:5:8) as the mobile phase.

A simple, precise, fast and low-cost potentiometric method for alendronate determination in pharmaceutical formulations has been proposed. Alendronate present in tablets at the known concentration was potentiometrically titrated in an aqueous solution using a combined glass electrode coupled with an autotitrator. Determination limit of alendronate was 20.0 mug mL(-1). In the presence of common excipients, such as lactose, microcrystalline cellulose, glucose, croscarmellose sodium, starch, talc, magnesium stearate and titanium dioxide no interferences were observed. The method proposed was validated by determining alendronate in various dosage forms. The average RSD was 0.75%.

Rheological characteristics of gels were studied, with the focus on their use as a cosmetic base. Some ideal characteristics can be predicted by the rheological characterization, such as the performance, with easy application and without dripping or forming lumps and bubbles. Moreover, it is possible to detect signs of physical instability. The gels were prepared with sodium carboxymethyl cellulose 3% and 5%, with Carbopol 940 (INCI: Carbomer) and with Carbopol Ultrez (INCI: Acrylates/C10-30 alkyl acrylate crosspolymer). The tests performed were yield stress, stress sweep and creep and recovery. The gel with 3% of sodium carboxymethyl cellulose presented the most appropriated behavior and can be indicated as the most suitable cosmetic base.

Many microorganisms that decompose lignocellulosic material are being studied as producers of enzymes to perform enzymatic hydrolysis of the lignocellulosic material present in residues from the agroindustries. Although the cellulose and hemicellulose present in these materials have their value for feeding cattle, their bioavailability requires breakdown of the bonds with indigestible lignin. Predigestion of such materials with ligninases, xylanases and pectinases (cellulase free) may transform the lignocellulosic substrate into a feed with greater digestibility and higher quality for ruminants.. This review provides an overview of variables to be considered in the utilization of fungal plantdepolymerizing enzymes produced by solid-state fermentation from agricultural production residues in Brazil. The activation of pre-kininogenin to kininogenin (pre-kallikrein to kallikrein) is one of the steps in the series of reactions of a complex system, linked also to fibrinolysis and coagulation, that leads to kinin release in plasma (See Cochrane et al., 1976; Wuepper, 1976; Kaplan et al., 1976; Kaplan et al., 1976). For human plasma, a test using kaolin as activator and measuring kallikrein activity with the chromogenic substrate Chromozym PK (Nα-benzoyl-prolyl-phenylalanyl-arginyl-nitroanilide, Pentapharm, Basle) is routinely employed. The purpose of this paper is to further study the mechanism of this activation, by means of different activators and using as inhibitor hexadimethrine bromide (Polybrene). Besides kaolin, sulfated polysaccharides, such as heparin and cellulose sulfate are able to activate pre-kininogenin to kininogenin. Hexadimethrine as expected, inhibited the activation by heparin and also that by cellulose sulfate. The activation by kaolin however followed a different pattern suggesting, at least partially, a different mode of action of this activator. © 1979.

Soluble and bound peroxidases were isolated from the pulp of ripening papaya fruit. During papaya ripening, soluble and bound peroxidase activities increased 2.5- and 4.2-fold, respectively. Soluble peroxidase was purified 59-fold by ammonium sulphate precipitation and chromatography on Sephadex G-25, DEAE-cellulose and Sephadex G-100. Bound peroxidase was purified 140-fold by ammonium sulphate precipitation and chromatography on Sephadex G-100 and DEAE-cellulose. Polyacrylamide gel electrophoresis of the purified preparations revealed that both enzymes were highly purified by the procedures adopted. The soluble and bound forms had a Mr of 41 000 and 54 000, respectively. Soluble and bound peroxidases showed optimum activity at pH 6.0 and 5.5, respectively, and were inhibited by p-chloromercuribenzoate, iodoacetamide, N-ethylmaleimide, potassium cyanide and Fe2+. Soluble peroxidase was activated by ammonium sulphate and this activation was prevented by cyanide. © 1990.
