The phytopathogen Xylella fastidiosa produces long type IV pili and short type I pili involved in motility and adhesion. In this work, we have investigated the role of sigma factor sigma(54) (RpoN) in the regulation of fimbrial biogenesis in X. fastidiosa. An rpoN null mutant was constructed from the non-pathogenic citrus strain J1a12, and microarray analyses of global gene expression comparing the wild type and rpoN mutant strains showed few genes exhibiting differential expression. In particular, gene pilA1 (XF2542), which encodes the structural pilin protein of type IV pili, showed decreased expression in the rpoN mutant, whereas two-fold higher expression of an operon encoding proteins of type I pili was detected, as confirmed by quantitative RT-PCR (qRT-PCR) analysis. The transcriptional start site of pilA1 was determined by primer extension, downstream of a sigma(54)-dependent promoter. Microarray and qRT-PCR data demonstrated that expression of only one of the five pilA paralogues, pilA1, was significantly reduced in the rpoN mutant. The rpoN mutant made more biofilm than the wild type strain and presented a cell-cell aggregative phenotype. These results indicate that sigma(54) differentially regulates genes involved in type IV and type I fimbrial biogenesis, and is involved in biofilm formation in X. fastidiosa.

The PilZ protein was originally identified as necessary for type IV pilus (T4P) biogenesis. Since then, a large and diverse family of bacterial PilZ homology domains have been identified, some of which have been implicated in signaling pathways that control important processes, including motility, virulence and biofilm formation. Furthermore, many PilZ homology domains, though not PilZ itself, have been shown to bind the important bacterial second messenger bis(3`-> 5`)cyclic diGMP (c-diGMP). The crystal structures of the PilZ orthologs from Xanthomonas axonopodis pv Citri (PilZ(XAC1133), this work) and from Xanthomonas campestris pv campestris (XC1028) present significant structural differences to other PilZ homologs that explain its failure to bind c-diGMP. NMR analysis of PilZ(XAC1133) shows that these structural differences are maintained in solution. In spite of their emerging importance in bacterial signaling, the means by which NZ proteins regulate specific processes is not clear. In this study, we show that PilZ(XAC1133) binds to PilB, an ATPase required for TV polymerization, and to the EAL domain of FiMX(XAC2398), which regulates TV biogenesis and localization in other bacterial species. These interactions were confirmed in NMR, two-hybrid and far-Western blot assays and are the first interactions observed between any PilZ domain and a target protein. While we were unable to detect phosphodiesterase activity for FimXX(AC2398) in vitro, we show that it binds c-diGMP both in the presence and in the absence of PilZ(XAC1133). Site-directed mutagenesis studies for conserved and exposed residues suggest that PilZ(XAC1133) interactions with FimX(XAC2398) and PilB(XAC3239) are mediated through a hydrophobic surface and an unstructured C-terminal extension conserved only in PilZ orthologs. The FimX-PilZ-PilB interactions involve a full set of ""degenerate"" GGDEF, EAL and PilZ domains and provide the first evidence of the means by which PilZ orthologs and FimX interact directly with the TP4 machinery. Organotellurium(]V) compounds have been reported to have multiple biological activities including cysteine protease-inhibitory activity, mainly cathepsin B. As cathepsin B is a highly predictive indicator for prognosis and diagnosis of cancer, a possible antitumor potential for these new compounds is expected. In this work, it was investigated the effectiveness of organotellurium(IV) RT-04 to produce lethal effects in the human promyelocytic leukaemia cell line HL60. Using the MTT tetrazolium reduction test, and trypan blue exclusion assay, the IC50 for the compound after 24 h incubation was 6.8 and 0.35 mu M, respectively. Moreover, the compound was found to trigger apoptosis in HL60 cells, inducing DNA fragmentation and caspase-3, -6, and -9 activations. The apoptsosis-induced by RT-04 is probably related to the diminished Bcl-2 expression, observed by RT-PCR, in HL60-treated cells. In vivo studies demonstrated that the RT-04 treatment (2.76 mg/kg given for three consecutive days) produces no significant toxic effects for bone marrow and spleen CFU-GM. However, higher doses (5.0 and 10 mg/kg) produced a dose-dependent reduction in the number of CFU-GM of RT-04-treated mice. These results suggest that RT-04 is able to induce apoptosis in HL60 cells by Bcl-2 expression down-modulation. Further studies are necessary to better clarify the effects of this compound on bone marrow normal cells. (C) 2008 Elsevier Ltd. All rights reserved.

Four new diorganotin(IV) complexes have been prepared from R(2)SnCl(2) (R = Me, Ph) with the ligands 5-hydroxy-3-metyl-5-phenyl-1-(S-benzildithiocarbazate)-pyrazoline (H(2)L(1)) and 5-hydroxy-3-methyl-5-phenyl-1-(2-thiophenecarboxylic)-pyrazoline (H(2)L(2)). The complexes were characterized by elemental analysis, IR. (1)H (13)C, (119)Sn NMR and Mossbauer spectroscopes The complexes [Me(2)SnL(1)], [Ph(2)SnL(1)] and [Me(2)SnL(2)] were also studied by single crystal X-ray diffraction and the results showed that the Sn(IV) central atom of the complexes adopts a distorted trigonal bipyramidal (TBP) geometry with the N atom of the ONX-tridentate (X = O and S) ligand and two organic groups occupying equatorial sites. The C-Sn-C angles for [Me(2)Sn(L(1))] and [Ph(2)Sn(L(1))] were calculated using a correlation between (119)Sn Mossbauer and X-ray crystallographic data based on the point-charge model Theoretical calculations were performed with the B3LYP density functional employing 3-21G(*) and DZVP all electron basis sets showing good agreement with experimental findings General and Sn(IV) specific IR harmonic frequency scale factors for both basis sets were obtained from comparison with selected experimental frequencies (C) 2010 Elsevier B V All rights reserved

Os autores objetivam, com este trabalho preliminar, bem como com aqueles que lhe darão continuidade, na sequência de composição de um livro de matemática para economistas, registrar as suas experiências ao longo dos últimos anos ministrando cadeiras de matemática nos cursos de pós-graduação em economia da Fundação Getúlio Vargas, da UFF (Universidade Federal Fluminense) e da PUC-RJ. Reveste-se de constante repetição em tais cursos a discussão sobre que pontos abordar, bem como com qual grau de profundidade, e em que ordem. É neste sentido que os autores esperam, com a sequência didática que aqui se inicia, trazer alguma contribuição para o assunto. A parte teórica relativa à demonstração do Teorema de Kuhn Tucker aqui apresentada transcreve, com a aquiescência do autor, textos selecionados de "Análise Convexa do Rn." de Mario Henrique Simonsen.

This paper considers the general problem of Feasible Generalized Least Squares Instrumental Variables (FG LS IV) estimation using optimal instruments. First we summarize the sufficient conditions for the FG LS IV estimator to be asymptotic ally equivalent to an optimal G LS IV estimator. Then we specialize to stationary dynamic systems with stationary VAR errors, and use the sufficient conditions to derive new moment conditions for these models. These moment conditions produce useful IVs from the lagged endogenous variables, despite the correlation between errors and endogenous variables. This use of the information contained in the lagged endogenous variables expands the class of IV estimators under consideration and there by potentially improves both asymptotic and small-sample efficiency of the optimal IV estimator in the class. Some Monte Carlo experiments compare the new methods with those of Hatanaka [1976]. For the DG P used in the Monte Carlo experiments, asymptotic efficiency is strictly improved by the new IVs, and experimental small-sample efficiency is improved as well.

The complex of Brookhart Ni(α-diimine)Cl2 (1) (α-diimine = 1,4-bis(2,6- diisopropylphenyl)-acenaphthenediimine) has been characterized after impregnation on silica (S1) and MAO-modified silicas (4.0, 8.0 and 23.0 wts.% Al/SiO2 called S2, S3 and S4, respectively). The treatment of these heterogeneous systems with MAO produces some active catalysts for the polymerization of the ethylene. A high catalytic activity has been gotten while using the system supported 1/S3 (196 kg of PE/mol[Ni].h.atm; toluene, Al/Ni = 1000, 30ºC, 60 min and atmospheric pressure of ethylene). The effects of polymerization conditions have been tested with the catalyst supported in S2 and the best catalytic activity has been gotten with solvent hexane, MAO as cocatalyst, molar ratio Al/Ni of 1000 and to the temperature of 30°C (285 kg of PE/mol[Ni].h.atm). When the reaction has been driven according to the in situ methodology, the activity practically doubled and polymers showed some similar properties. Polymers products by the supported catalysts showed the absence of melting fusion, results similar to those gotten with the homogeneous systems by DSC analysis. But then, polymers gotten with the transplanted system present according to the GPC’s curves the polydispersity (MwD) varies between 1.7 and 7.0. A polyethylene blend (BPE/LPE) was prepared using the complex Ni(α-diimine)Cl2 (1) (α-diimine = 1,4-bis(2,6-diisopropylphenyl)-acenaphthenediimine) and {TpMs*}TiCl3 (2) (TpMs* = hydridobis(3-mesitylpyrazol-1-yl)(5-mesitylpyrazol-1-yl)) supported in situ on MAO-modified silica (4.0 wts. -% Al/SiO2, S2). Reactions of polymerization of ethylene have been executed in the toluene in two different temperatures (0 and 30°C), varying the molars fraction of nickel (xNi), and using MAO as external cocatalyst. To all temperatures, the activities show a linear variation tendency with xNi and indicate the absence of the effect synergic between the species of nickel and the titanium. The maximum of activity have been found at 0°C. The melting temperature for the blends of polyethylene produced at 0 °C decrease whereas xNi increases indicating a good compatibility between phases of the polyethylene gotten with the two catalysts. The melting temperature for the blends of polyethylene showed be depend on the order according to which catalysts have been supported on the MAO-modified silica. The initial immobilization of 1 on the support (2/1/S2) product of polymers with a melting temperature (Tm) lower to the one of the polymer gotten when the titanium has been supported inicially (1/2/S2). The observation of polyethylenes gotten with the two systems (2/1/S2 and 1/2/S2) by scanning electron microscopy (SEM) showed the spherical polymer formation showing that the spherical morphology of the support to been reproduced. Are described the synthesis, the characterization and the catalytic properties for the oligomerization of the ethylene of four organometallics compounds of CrIII with ligands ([bis[2-(3,5-dimethyl-1-pyrazolyl)ethyl]amine] chromium (III) chloride (3a), [bis[2-(3,5- dimethyl-l-pyrazolyl)ethyl]benzylamine] chromium (III) chloride (3b), [bis[2-(3,5-dimethyl-lpyrazolyl) ethyl]ether] chromiun(III)chloride (3c), [bis[2-(3-phenyl-lpyrazolyl) ethyl]ether]chromiun(III)chloride (3d)). In relation of the oligomerization, at exception made of the compounds 3a, all complex of the chromium showed be active after activation with MAO and the TOF gotten have one effect differentiated to those formed with CrCl3(thf)3. The coordination of a tridentate ligand on the metallic center doesn't provoke any considerable changes on the formation of the C4 and C6, but the amount of C8 are decrease and the C10 and C12+ have increased. The Polymers produced by the catalyst 3a to 3 and 20 bar of ethylene have, according to analyses by DSC, the temperatures of fusion of 133,8 and 136ºC respectively. It indicates that in the two cases the production of high density polyethylene. The molar mass, gotten by GPC, is 46647 g/mols with MwD = 2,4 (3 bar). The system 3c/MAO showed values of TOF, activity and selectivity to different α-olefins according to the pressure of ethylene uses. Himself that shown a big sensibility to the concentration of ethylene solubilized.

A acidemia metilmalônica é uma desordem metabólica hereditária caracterizada bioquimicamente pelo acúmulo tecidual de ácido metilmalônico (MMA) e clinicamente por deterioração neurológica progressiva e falência renal. Os avanços no tratamento dessa doença alcançados nos últimos anos possibilitaram uma diminuição significativa na mortalidade dos mesmos. Entretanto, a morbidade continua alta, pois a maioria dos pacientes afetados por acidemia metilmalônica, mesmo recebendo o melhor tratamento disponível no momento, apresenta graus variáveis de comprometimento do sistema nervoso central refletido em retardo mental e atraso no desenvolvimento psicomotor. No presente estudo investigamos os efeitos in vivo da administração crônica do MMA em ratos Wistar durante o seu desenvolvimento (do 5º ao 28º dia de vida pós-natal) sobre o comportamento dos mesmos na tarefa do labirinto aquático de Morris. A tarefa foi realizada após um período de recuperação dos animais de 30 dias com o intuito de verificar dano neurológico permanente ou de longa duração nos animais. O labirinto aquático de Morris é uma tarefa bastante útil para a avaliação de aprendizado e memória espaciais. O protocolo da tarefa foi ligeiramente modificado para servir ao propósito de nosso trabalho, ou seja, o de avaliar o efeito da administração crônica de drogas sobre o comportamento de ratos. Verificamos que a administração crônica de MMA não provocou efeito no peso corporal, velocidade de natação e na fase de aquisição da tarefa. Porém, o tratamento prejudicou o desempenho dos animais no treino reverso, o que é condizente com comportamento perseverativo. Também avaliamos o efeito do ácido ascórbico, que foi administrado isoladamente ou em combinação com o MMA para testarmos se o estresse oxidativo poderia estar relacionado com as alterações comportamentais observadas no grupo tratado com MMA. Observamos que este antioxidante preveniu as alterações comportamentais provocadas pelo MMA, indicando que o estresse oxidativo pode estar envolvido com o efeito encontrado. Passamos então a avaliar o efeito in vitro do MMA sobre parâmetros de estresse oxidativo, mais especificamente na técnica de dosagem de substâncias reativas ao ácido tiobarbitúrico (TBA-RS), que é um parâmetro de lipoperoxidação, e sobre o potencial antioxidante total do tecido (TRAP) e a reatividade antioxidante do tecido (TAR), que são parâmetros de defesas antioxidantes teciduais. O MMA na concentração de 2,5 mM aumentou a lipoperoxidação in vitro em homogeneizado de estriado e hipocampo de ratos e diminuiu o TRAP e o TAR em homogeneizado de estriado de ratos. Tais resultados indicam fortemente que o MMA induz estresse oxidativo. Finalmente investigamos o efeito in vitro do MMA sobre a atividade enzimática dos complexos da cadeia respiratória em várias estruturas cerebrais e em órgãos periféricos em ratos de 30 dias de vida no sentido de melhor esclarecer os mecanismos fisiopatológicos dos danos teciduais desta doença. Verificamos que o MMA causou uma inibição significativa da atividade do complexo II da cadeia respiratória em estriado e hipocampo quando baixas concentrações de sucinato foram utilizadas no meio de incubação. Além disso, verificamos que este efeito inibitório do MMA sobre o complexo II ocorreu somente após exposição do homogeneizado ao ácido por pelo menos 10 minutos, além do que esta inibição não foi prevenida pela co-incubação com o inibidor da óxido nítrico sintetase Nω- nitro-L-argininametilester (L-NAME) ou por uma associação de catalase e superóxido dismutase. Estes resultados sugerem que as espécies reativas de oxigênio e nitrogênio mais comuns não estão envolvidas neste efeito, tornando improvável que a inibição do complexo II da cadeia respiratória seja mediada por estresse oxidativo. O MMA também causou uma inibição do complexo II-III em estriado, hipocampo, rim, fígado e coração; e inibiu o complexo I-III em fígado e rim. O complexo IV não foi afetado pela incubação com o ácido em nenhuma das estruturas testadas. Portanto, tomados em seu conjunto, estes resultados indicam que o MMA bloqueia a cadeia respiratória. Os resultados de nosso trabalho indicam que a administração crônica de MMA em ratos em desenvolvimento provocou alterações comportamentais de longa duração provavelmente mediadas por radicais livres, pois tais alterações foram prevenidas pelo antioxidante ácido ascórbico. O MMA também induziu estresse oxidativo in vitro em estriado e hipocampo e inibiu de forma diferenciada os complexos da cadeia respiratória nos tecidos estudados, sendo que as estruturas mais vulneráveis a esta ação foram o estriado e o hipocampo. Finalmente, nossos resultados sugerem que antioxidantes podem ajudar a prevenir, ou pelo menos atenuar, os danos teciduais provocados pelo MMA na acidemia metilmalônica.
