Background: [NiFe] hydrogenases are enzymes that catalyze the oxidation of hydrogen into protons and electrons, to use H2 as energy source, or the production of hydrogen through proton reduction, as an escape valve for the excess of reduction equivalents in anaerobic metabolism. Biosynthesis of [NiFe] hydrogenases is a complex process that occurs in the cytoplasm, where a number of auxiliary proteins are required to synthesize and insert the metal cofactors into the enzyme structural units. The endosymbiotic bacterium Rhizobium leguminosarum requires the products of eighteen genes (hupSLCDEFGHIJKhypABFCDEX) to synthesize an active hydrogenase. hupF and hupK genes are found only in hydrogenase clusters from bacteria expressing hydrogenase in the presence of oxygen. Results: HupF is a HypC paralogue with a similar predicted structure, except for the C-terminal domain present only in HupF. Deletion of hupF results in the inability to process the hydrogenase large subunit HupL, and also in reduced stability of this subunit when cells are exposed to high oxygen tensions. A ?hupF mutant was fully complemented for hydrogenase activity by a C-terminal deletion derivative under symbiotic, ultra low-oxygen tensions, but only partial complementation was observed in free living cells under higher oxygen tensions (1% or 3%). Co-purification experiments using StrepTag-labelled HupF derivatives and mass spectrometry analysis indicate the existence of a major complex involving HupL and HupF, and a less abundant HupF-HupK complex. Conclusions: The results indicate that HupF has a dual role during hydrogenase biosynthesis: it is required for hydrogenase large subunit processing and it also acts as a chaperone to stabilize HupL when hydrogenase is synthesized in the presence of oxygen.

Dos tipos diferentes de pollinaza mezclados con el material de cama (paja o serrín) añadidas en tasas de 0 y 10% más dos substratos comerciales de turba (turba negra y turba rubia) se usaron para estudiar el crecimiento de crisantemo en maceta. En todos los casos, la calidad y el tamaño de las plantas fue mejor con las mezclas de substratos de turba negra que con las mezclas de turba rubia. Con las mezclas de turba negra con pollinaza hubo un aumento significativo del número de flores sin que hubiese un aumento en el tamaño de las plantas. También se pudo observar que con la adición de pollinaza, el substrato de turba rubia mejoró significativamente los parámetros de crecimiento estudiados, esto fue debido a que el pH del substrato fue más adecuado para el cultivo. Por último, no hubo mortalidad de plantas con las mezclas experimentadas, pero cuando la cantidad de gallinaza aumentó en la mezcla fue más notable la mortalidad de plantas.

The intermediate band solar cell (IBSC) is a solar cell that, in order to increase its efficiency over that of single gap solar cells, takes advantage of the absorption of below-bandgap energy photons by means of an intermediate band (IB) located in the semiconductor bandgap. For this process to improve the solar cell performance, the belowbandgap photon absorption has to be effective and the IB cannot limit the open-circuit voltage of the cell. In this paper we provide a guide to the new researcher interested in the idea in order he can quickly become familiar with the concept and updated with the most relevant experimental results.

IBPOWER is a Project awarded under the 7th European Framework Programme that aims to advance research on intermediate band solar cells (IBSCs). These are solar cells conceived to absorb below bandgap energy photons by means of an electronic energy band that is located within the semiconductor bandgap, whilst producing photocurrent with output voltage still limited by the total semiconductor bandgap. IBPOWER employs two basic strategies for implementing the IBSC concept. The first is based on the use of quantum dots, the IB arising from the confined energy levels of the electrons in the dots. Quantum dots have led to devices that demonstrate the physical operation principles of the IB concept and have allowed identification of the problems to be solved to achieve actual high efficiencies. The second approach is based on the creation of bulk intermediate band materials by the insertion of an appropriate impurity into a bulk semiconductor. Under this approach it is expected that, when inserted at high densities, these impurities will find it difficult to capture electrons by producing a breathing mode and will cease behaving as non-radiative recombination centres. Towards this end the following systems are being investigated: a) Mn: In1-xGax N; b) transition metals in GaAs and c) thin films.

Today's motivation for autonomous systems research stems out of the fact that networked environments have reached a level of complexity and heterogeneity that make their control and management by solely human administrators more and more difficult. The optimisation of performance metrics for the air traffic management system, like in other networked system, has become more complex with increasing number of flights, capacity constraints, environmental factors and safety regulations. It is anticipated that a new structure of planning layers and the introduction of higher levels of automation will reduce complexity and will optimise the performance metrics of the air traffic management system. This paper discusses the complexity of optimising air traffic management performance metrics and proposes a way forward based on higher levels of automation.

A gene encoding a homolog to the cation diffusion facilitator protein DmeF from Cupriavidus metallidurans has been identified in the genome of Rhizobium leguminosarum UPM791. The R. leguminosarum dmeF gene is located downstream of an open reading frame (designated dmeR) encoding a protein homologous to the nickel- and cobalt-responsive transcriptional regulator RcnR from Escherichia coli. Analysis of gene expression showed that the R. leguminosarum dmeRF genes are organized as a transcriptional unit whose expression is strongly induced by nickel and cobalt ions, likely by alleviating the repressor activity of DmeR on dmeRF transcription. An R. leguminosarum dmeRF mutant strain displayed increased sensitivity to Co(II) and Ni(II), whereas no alterations of its resistance to Cd(II), Cu(II), or Zn(II) were observed. A decrease of symbiotic performance was observed when pea plants inoculated with an R. leguminosarum dmeRF deletion mutant strain were grown in the presence of high concentrations of nickel and cobalt. The same mutant induced significantly lower activity levels of NiFe hydrogenase in microaerobic cultures. These results indicate that the R. leguminosarum DmeRF system is a metal-responsive efflux mechanism acting as a key element for metal homeostasis in R. leguminosarum under free-living and symbiotic conditions. The presence of similar dmeRF gene clusters in other Rhizobiaceae suggests that the dmeRF system is a conserved mechanism for metal tolerance in legume endosymbiotic bacteria.

La presente tesis es el resultado de un trabajo de investigación sobre diferentes métodos constructivos y estructurales desarrollado en el ámbito de las Estructuras y de la Historia de la Construcción y la Arquitectura. Esta disciplina tiene por objeto el conocimiento de la eficacia constructiva y tipológica, así como aspectos mecánicos en las obras arquitectónicas que se han sucedido a lo largo de la Historia, concretamente en el periodo estudiado en la presente investigación, en un recorrido aproximado de diez siglos. Su interés radica en la aplicación de las distintas estrategias metodológicas utilizadas para abordar un mismo problema; una constante; cubrir grandes espacios sin apoyos intermedios. Estamos hablando de la resolución de cúpulas en el ámbito de la mecánica de cúpulas. La investigación se ha desarrollado en el campo de la teoría de las estructuras y la construcción en general, y particularmente en la teoría de las bóvedas de fábrica aplicada al proyecto de edificios cupulados. Edificios cupulados significativamente importantes en la Historia de la Arquitectura, obras que han sido y son referencia en el campo mencionado; hitos arquitectónicos. El objetivo es el de, en base a un recorrido cronológico, partiendo de la arquitectura paleocristiana y bizantina (tomando y citando referencias de edificios de la Roma Republicana y de la Roma Imperial) llegar a relacionar métodos y soluciones con la arquitectura otomana de mitad del siglo XVI. Relacionar la iglesia de Santa Sofía de Constantinopla, obra cumbre del mundo bizantino con las mezquitas otomanas que diez siglos más tarde levantaría el maestro Sinán al servicio de los sultanes otomanos. Se realizará un análisis profundo y comparativo de métodos constructivos y estructurales, esto es, aspectos mecánicos. No se trata de enfrentar métodos orientales y occidentales, se trata cuanto de relacionarlos. Las preguntas fundamentales a las que tratará de responder esta tesis son: 1. Validez de los métodos orientales y occidentales en el desarrollo de cúpulas y su comportamiento mecánico. 2. Qué hay del “espíritu” de Santa Sofía de Constantinopla en las obras de Mimar Sinán mil años después. 3. Eficacia constructiva y tipológica de las soluciones de los edificios cupulados del estudio.

El ser humano nunca ha tenido una capacidad semejante a la actual para alterar el medio en el que vive. A través fundamentalmente de cambios en el uso del hábitat, alteración de los ciclos biogeoquímicos, cambio climático y presencia de especies invasoras, la tasa de extinción de especies se ha acelerado enormemente. Al crecer más rápido la población humana dentro de las áreas en las que se concentra una mayor biodiversidad (biodiversity hotspots), las tasas de transformación son mayores, por lo que dicha biodiversidad se encuentra más amenazada. Entre los principales 25 biodiversity hotspots globales se encuentra la cuenca mediterránea. Las afecciones a la biodiversidad hacen por tanto que en dicha cuenca mediterránea sea necesario poner en práctica iniciativas innovadoras para su conservación. Pero además de una cuidadosa selección, es necesario el seguimiento y la evaluación de la eficacia y la eficienca de dichas medidas, para comprobar su idoneidad y mejorarlas cuando sea posible. Esta es la tarea de la ingeniería de la conservación. Esta Memoria busca analizar y proponer mejoras en cuatro acciones experimentales de conservación de la biodiversidad que tienen un marco común, la conservación de especies subrogadas del monte mediterráneo. En concreto de cinco especies amenazadas: conejo de monte (Oryctolagus cuniculus, especie llave), águila imperial ibérica (Aquila adalberti), águila perdicera (Aquila fasciata), cigüeña negra (Ciconia nigra) y lince ibérico (Lynx pardinus), todas ellas especies paraguas y bandera. Casi todas estas especies se enfrentan a amenazas comunes, como son la pérdida de la calidad del hábitat, la mortalidad de origen humano, la escasez de alimento o la falta de eficiencia en las técnicas de seguimiento. Las acciones destinadas a revertir o minimizar estas amenazas han tenido un marco geográfico y ecológico común, 49 fincas privadas de monte mediterráneo situadas en 9 espacios de la Red Natura 2000 del centro-occidental peninsular, en las que se ha podido trabajar merced a acuerdos de custodia del territorio. Dichas actividades han sido financiadas casi por completo con cargo a fondos LIFE Naturaleza de la Comisión Europea. El presente documento contribuye a aportar el conocimiento científico imprescindible para mejorar la eficacia y eficiencia de las medidas técnicas de gestión y conservación: la ingeniería de la conservación. Como resultados más relevantes se pueden citar que la selección del conejo de las zonas sembradas aumenta con la presencia tanto de isletas de matorral como de refugios y resulta inferior en las zonas de matorral más cerrado; que la electrocución es una de las principales causas de mortalidad de las rapaces amenazadas, así como que se ha detectado que la mortalidad se concentra en determinadas zonas que deben ser prioritarias para su corrección y que éstas deben pasar por las modificaciones estructurales; que la selección de zonas de alimentación por parte de la cigüeña negra se ve afectada por numerosos factores y que una adecuada ubicación y gestión de las charcas para ganadería y caza mayor puede favorecer no sólo la alimentación de esta especie, sino suponer un lugar de cría para diversas especies de anfibios y peces autóctonos, en franco declive; finalmente, hay que destacar que la mejora en el seguimiento de las poblaciones de lince ibérico pasa por la incorporación de métodos de seguimiento novedosos, como las cámaras infrarrojas en letrinas, dada la capacidad de aprendizaje de la especie. Abstract Nowadays the human being has reached its top ability to exchange its environment. Mainly through habitat change, altering biogeochemical cycles, climatic change and alien species, global extinction rate has increased dramatically. Population increases faster in biodiversity hotspots, thus change rates are higher. Mediterranean basin is included within the 25 most valuable biodiversity hotspots. Affections on biodiversity turns necessary to develop innovative actions to maintain the most threatened species in this hotspot. But further than a careful selection and implementarion, survey and evaluation of those practices is necessary to check its suitability. Those are actions for the conservation engineering. This Memory looks for to analyze and to put forward improvements in four experimental biodiversity conservation actions with a common framework: conservation of surrogate Mediterranean species. Those actions were specifically directed to five threatened species: the European wild rabbit (Oryctolagus cuniculus, key species), Spanish imperial eagle (Aquila adalberti) and Bonelli’s eagle (Aquila fasciata, both of the umbrella and flagship species), black stork (Ciconia nigra, umbrella and flagship species) and the Iberian lynx (Lynx pardinus, umbrella and flagship species). Those species have common threats: habitat loss, maninduced mortality, food shortening and lack of surveying efficiency. Actions aimed to minimize those threats also had a common geographical frame: 49 privately owned states within 9 Natura 2000 areas in Southwestern Spain. Actions were developed through lands tewardship agreements, and were even wholly undertaken through European Commision LIFE’s fund. This document contributes to develop cientific knowledge necessary for increasing efficacy and efficiency for implementing technical measures devoted to conservation, the conservation engineering. One of the most outstanding result might be the necessity of including bushy islets or rabbit shelter within sowings when enhacing rabbit populations is the goal. Electrocution is one of the commonest man-induced death causes for raptors, especially for the threatened ones. Areas of concentration of this mortality have been detected, and when mitigation measures have been evaluated, only structural modifications had results. The black stork’s selection of feeding areas is influenced by several factors. An adequate location and management of livestock’s ponds can favour not only this species, but also creating new breeding places for amphibians and indigenous fish species, most of them in a sharp decline. Finally, improving the survey of the Iberian lynx might be achieved through implementing newly developed surveying systems, such as the infrared cameras in latrines.

In the intricate maturation process of [NiFe]-hydrogenases, the Fe(CN)2CO cofactor is first assembled in a HypCD complex with iron coordinated by cysteines from both proteins and CO is added after ligation of cyanides. The small accessory protein HypC is known to play a role in delivering the cofactor needed for assembling the hydrogenase active site. However, the chemical nature of the Fe(CN)2CO moiety and the stability of the cofactor–HypC complex are open questions. In this work, we address geometries, properties, and the nature of bonding of all chemical species involved in formation and binding of the cofactor by means of quantum calculations. We also study the influence of environmental effects and binding to cysteines on vibrational frequencies of stretching modes of CO and CN used to detect the presence of Fe(CN)2CO. Carbon monoxide is found to be much more sensitive to sulfur binding and the polarity of the medium than cyanides. The stability of the HypC–cofactor complex is analyzed by means of molecular dynamics simulation of cofactor-free and cofactor-bound forms of HypC. The results show that HypC is stable enough to carry the cofactor, but since its binding cysteine is located at the N-terminal unstructured tail, it presents large motions in solution, which suggests the need for a guiding interaction to achieve delivery of the cofactor.

The concept of "intermediate band solar cell" (IBSC) is, apparently, simple to grasp. However, since the idea was proposed, our understanding has improved and we feel now that we can explain better some concepts than we initially introduced. Clarifying these concepts is important, even if they are well-known for the advanced researcher, so that efforts can be driven in the right direction from start. The six pieces of this work are: Does a miniband need to be formed when the IBSC is implemented with quantum dots?; What are the problems of each of the main practical approaches that exist today? What are the simplest experimental techniques to demonstrate whether an IBSC is working as such or not? What is the issue with the absorption coefficient overlap? and Mott's transition? What the best system would be, if any?

The Europe-Japan Collaborative Research Project on Concentrator Photovoltaics (CPV) has been initiated under support by the EC (European Commission) and NEDO (New Energy and Industrial Technology Development Organization) since June 2011. This is project (NGCPV Project; a New Generation of Concentrator PhotoVoltaic cells, modules and systems) is aiming to accelerate the move to very high efficiency and lower cost CPV technologies and to enhance widespread deployment of CPV systems. 7 organizations such as UPM, FhG-ISE Imperial College, BSQ, CEA-INES, ENEA, and PSE in Europe and 9 organizations such as TTI, Univ. Tokyo, AIST, Sharp Co. Daido Steel Co., Kobe Univ., Miyazaki Univ., Asahi Kasei Co., and Takano Co. participate in this project. The targets of this project are 1) to develop world-record efficiency CPV cells of more than 45%, 2) to develop world-record efficiency CPV modules of 35%, 3) to establish standard measurements of CPV cells and modules, 4) to install 50kW CPV system in Spain, to carry out field test of CPV system and to manage power generation of CPV systems, and 5) to develop high-efficiency and low-cost new materials and structure cells such as III-V-N, III-V-on-Si tandem, quantum dots and wells. This paper presents outline of this project and most recent results such as world record efficiency (37.9% under 1-sun) cell and high-efficiency (43.5% under 240-306 suns) concentrator cell with inverted epitaxial grown InGaP/GaAs/InGaAs 3-junction solar cells.

Las células solares de banda intermedia se inventaron en España. Constituyen una propuesta para aumentar la eficiencia de las células solares por encima del límite de Shockley y Queisser para células de un solo gap. El número de ideas para llevarlas a la práctica (desde la utilización de puntos cuánticos a la implantación selectiva de impurezas) se ha ido expandiendo a medida que nuestra comprensión ha ido aumentando. En este artículo repasamos el concepto de célula solar de banda intermedia y algunas de estas ideas para llevarla a la práctica.
