The aim of the thesis is to investigate the hybrid LC filter behavior in modern power drives; to analyze the influence of such a du/dt filter on the control system stability. With the implementation of the inverter output RLC filter the motor control becomes more complicated. And during the design process the influence of the filter on the motor should be considered and the filter RLC parameters should be constrained.

The precise nature of the reaction between nitric acid and sodium ortho-vanadate solutions has been studied by means of electrometric techniques involving potentiometric and conductometric titrations. The well defined inflections and breaks in the titration curves confirm the existence of the anions, pyro-V2O7(4-), meta-VO3- and poly-H2V10O28(4-) corresponding to the ratios of VO4(3-):H+ as 1:1, 1:2 and 1:2.6 in the neighborhood of pH 10.5, 7.4 and 3.6, respectively. The interaction of cerium(III) nitrate with sodium vanadate solutions, at specific pH levels 12.4, 10.5, 7.4 and 3.6 was also studied by potentiometric and conductometric titrations between the reactants. The end-points obtained from the sharp inflections in the titration curves provide definite evidence for the formation and precipitation of cerium ortho-Ce2O3.V2O5, pyro-2Ce2O3.3V2O5 and meta-Ce2O3.3V2O5 vanadates in the neighborhood of pH 7.4, 6.2 and 4.8, respectively. Analytical investigations on the precipitates formed confirm the results of the electrometric study.

The human genome comprises roughly 20 000 protein coding genes. Proteins are the building material for cells and tissues, and proteins are functional compounds having an important role in many cellular responses, such as cell signalling. In multicellular organisms such as humans, cells need to communicate with each other in order to maintain a normal function of the tissues within the body. This complex signalling between and within cells is transferred by proteins and their post-translational modifications, one of the most important being phosphorylation. The work presented here concerns the development and use of tools for phosphorylation analysis. Mass spectrometers have become essential tools to study proteins and proteomes. In mass spectrometry oriented proteomics, proteins can be identified and their post-translational modifications can be studied. In this Ph.D. thesis the objectives were to improve the robustness of sample handling methods prior to mass spectrometry analysis for peptides and their phosphorylation status. The focus was to develop strategies that enable acquisition of more MS measurements per sample, higher quality MS spectra and simplified and rapid enrichment procedures for phosphopeptides. Furthermore, an objective was to apply these methods to characterize phosphorylation sites of phosphopeptides. In these studies a new MALDI matrix was developed which allowed more homogenous, intense and durable signals to be acquired when compared to traditional CHCA matrix. This new matrix along with other matrices was subsequently used to develop a new method that combines multiple spectra from different matrises from identical peptides. With this approach it was possible to identify more phosphopeptides than with conventional LC/ESI-MS/MS methods, and to use 5 times less sample. Also, phosphopeptide affinity MALDI target was prepared to capture and immobilise phosphopeptides from a standard peptide mixture while maintaining their spatial orientation. In addition a new protocol utilizing commercially available conductive glass slides was developed that enabled fast and sensitive phosphopeptide purification. This protocol was applied to characterize the in vivo phosphorylation of a signalling protein, NFATc1. Evidence for 12 phosphorylation sites were found, and many of those were found in multiply phosphorylated peptides

In recent years, the network vulnerability to natural hazards has been noticed. Moreover, operating on the limits of the network transmission capabilities have resulted in major outages during the past decade. One of the reasons for operating on these limits is that the network has become outdated. Therefore, new technical solutions are studied that could provide more reliable and more energy efficient power distributionand also a better profitability for the network owner. It is the development and price of power electronics that have made the DC distribution an attractive alternative again. In this doctoral thesis, one type of a low-voltage DC distribution system is investigated. Morespecifically, it is studied which current technological solutions, used at the customer-end, could provide better power quality for the customer when compared with the current system. To study the effect of a DC network on the customer-end power quality, a bipolar DC network model is derived. The model can also be used to identify the supply parameters when the V/kW ratio is approximately known. Although the model provides knowledge of the average behavior, it is shown that the instantaneous DC voltage ripple should be limited. The guidelines to choose an appropriate capacitance value for the capacitor located at the input DC terminals of the customer-end are given. Also the structure of the customer-end is considered. A comparison between the most common solutions is made based on their cost, energy efficiency, and reliability. In the comparison, special attention is paid to the passive filtering solutions since the filter is considered a crucial element when the lifetime expenses are determined. It is found out that the filter topology most commonly used today, namely the LC filter, does not provide economical advantage over the hybrid filter structure. Finally, some of the typical control system solutions are introduced and their shortcomings are presented. As a solution to the customer-end voltage regulation problem, an observer-based control scheme is proposed. It is shown how different control system structures affect the performance. The performance meeting the requirements is achieved by using only one output measurement, when operating in a rigid network. Similar performance can be achieved in a weak grid by DC voltage measurement. An additional improvement can be achieved when an adaptive gain scheduling-based control is introduced. As a conclusion, the final power quality is determined by a sum of various factors, and the thesis provides the guidelines for designing the system that improves the power quality experienced by the customer.

O fungo Cylindrocladium spathiphylli causa podridão no colo e nas raízes de espatifilo, induzindo amarelecimento e murcha nas folhas da parte aérea. Há poucos estudos com o fungo quanto às enzimas extracelulares. Assim, delineou-se ensaio inteiramente casualizado in vitro, com três tratamentos (dois isolados normais: LFEEI016 - EPAGRI/SC e MMBF 01/01 - IB/SP; mais testemunha) e seis repetições, visando a detecção de enzimas. A amilase (AM), lipase (LP), carboximetilcelulase (CMC) e lacase (LC) foram mensuradas pelo cálculo da área da coroa circular, local de atividade das enzimas. A catalase (CT) e gelatinase (GL) foram mensuradas por símbolos, depois transformados em notas (1 a 4, de ausência à intensa produção). O isolado MMBF 01/01 produziu as maiores áreas e com mais intensidade as enzimas. Das enzimas, a maior área foi a da LC e depois a LP. LFEEI016 e MMBF 01/01 não apresentaram área para AM e CMC e exibiram intensidade fraca e igual para CT. A intensidade de GT foi moderada no MMBF 01/01 (nota média 3,0) e ausente para o LFEEI016 (nota média 1,0). Após a produção das enzimas extracelulares, os isolados foram alterados vegetativamente por temperatura. Instalou-se ensaio inteiramente ao acaso, em fatorial contendo disco de micélio dos isolados do fungo versus três temperaturas [23ºC (T1); 33ºC (T2) e de 33ºC para 23ºC (T3)] versus quatro períodos (3; 6; 9 e 12 dias), com oito repetições, em placa de Petri com BDA mantida em BOD. O diâmetro das colônias foi medido diariamente (mm) em dois sentidos. As alterações de temperatura vistas foram: inócuo, fungistático (FS) e fungicida (FC). Aplicou-se estatística nas temperaturas T1 e T3 dos isolados para checar efeito FS e escolher aqueles com maior e menor efeito. O isolado LFEEIO16 mostrou efeito FS com três dias às temperaturas 33ºC e de 33ºC para 23ºC. Nos demais períodos, o isolado sofreu efeito FC. O MMBF 01/01 mostrou efeito FS em todos os períodos. Ambos isolados sofreram, estatisticamente, efeito FS. O MMBF 01/01 mostrou maior efeito FS aos nove dias, nas temperaturas referidas e menor com três dias. Após a constatação de alteração na taxa de crescimento micelial dos isolados, procurou-se verificar, novamente, a produção de enzimas extracelulares. Os isolados alterados e utilizados foram o MMBF 01/01 com três e nove dias e o LFEEIO16 com três dias. A produção e a avaliação das enzimas foram realizadas da mesma maneira para os isolados normais. O isolado MMBF 01/01 - três dias produziu as maiores áreas e com mais intensidade as enzimas. Das enzimas, a maior área foi a da LC. Após LC, a área da LP foi maior que a da CMC nos isolados. Não se detectou área de AM nos isolados. A intensidade de GL foi maior que a da CT nos isolados MMBF 01/01 com três e nove dias. Para LFEEI016 - três dias, a intensidade média de CT foi nota 2,0 e ausência de GL (nota média 1,3). Sugere-se investigar a severidade da doença na planta entre os isolados e em condições naturais investigar a evolução da LC em comparação as demais enzimas na patogênese do fungo na cultura.

This study aimed at quantifying total organic carbon stocks and its pools in Acrisol under agroforestry systems with six (AFS6) and thirteen years old (AFS13), slash-and-burn agriculture (SBA) and savanna native forest (SNF) in northeastern Brazil. Soil samples were collected at 0-0.05 m, 0.05-0.10 m, 0.10-0.20 m and 0.20-0.40 m depths in the dry and rainy seasons to evaluate total organic carbon (TOC) stocks and labile carbon (LC), fulvic acid fraction (C-FAF), humic acid fraction (C-HAF), humin (C-HF) and microbial biomass carbon (Cmic) contents. Additionally, carbon management index (CMI) was determined. Higher TOC stocks (97.7 and 81.8 Mg ha-1 for the 0-0.40 m depth in the dry and rainy seasons, respectively) and LC, humic substances and Cmic contents were observed in the AFS13 in all the depths. CMI also was higher in the AFS13 (0-0. 05 m: 158 and 86; 0.05-0.10 m: 171 and 67, respectively for the dry and rainy seasons) especially when compared to the SBA (0-0.05 m: 5.6 and 5.4; 0.05-0.10 m: 5.3 and 5.8, respectively for dry and rainy seasons). The agroforestry systems increased soil quality through the conservation of organic matter and can be considered an excellent strategy to assurance sustainability in tropical soil of Northeastern Brazil

The aim of this study was to identify, by multivariate statistical technique, the physic, chemical and biological variables that best characterize the quality of surface waters in two small rural catchments with different land uses (eucalyptus silviculture (SC) vs. pasture and extensive livestock (LC)) located in Rosário do Sul, RS - Brazil. Monitoring was conducted during the months of August 2011 to August 2012 and the following parameters were analyzed: Ca2+, Mg2+, K+, SO42-, Cl-, pH, electrical conductivity, turbidity, alkalinity, suspended and dissolved solids, biochemical oxygen demand , total coliforms, Escherichia coli and temperature, flow and rainfall. Through the use of FA/PCA, it was found that the model best fit to express water quality of in LC that was composed of five factors which account for 83.5% of the total variance, while for SC, four factors accounted for 85.12% of the variance. In LC, the five main factors were, respectively, soluble salts, diffuse pollution, solid, and both anthropogenic and organic factors. In SC, the four factors were namely: soluble salts, mineral, nutritional and diffuse pollution factors. The results of this study showed that by replacing the traditional soil usage (pasture and livestock) with planted forest, diffuse pollution was attenuated but, however, it did not result in major changes in the physical-chemical and biological characteristics of the water. Another point to note is that factorial analysis did not result in a large reduction in the number of variables, once the best model fit occurred with the addition of 15 of 18 analyzed variables (LC) and 17 of 18 analyzed variables (SC).

Os solos submetidos a sistemas de produção que não incluem o preparo do solo, estão condicionados, em sua sustentabilidade, a um manejo que priorize o acompanhamento da compactação que o tráfego de máquinas provoca. Foram realizados ensaios de campo com o objetivo de avaliar a reologia de um solo Typic Hapludert, submetido ao tráfego de veículos em dois sistemas de preparo do solo (preparo convencional - LC, e semeadura direta - SD). Os subtratamentos foram quatro intensidades de tráfego (0; 80; 160 e 240 Mg km ha-1). As variáveis experimentais dependentes foram: índice de cone (IC) e densidade aparente a seco (DA). Para 160 e 240 Mg km ha-1, encontraram-se diferenças estatísticas significativas no IC em relação à testemunha. Para o LC, com as maiores intensidades de tráfego, encontraram-se valores de DA superiores a 1,83 Mg m-3. O comportamento mecânico do solo sob tráfego é mais diferente em SD que em LC. Existe uma relação direta entre o número de passadas e a profundidade onde se localiza a camada de maior resistência à penetração nos solos sob tratamento com LC.

Frequency converters are widely used in the industry to enable better controllability and efficiency of variable speed AC motor drives. Despite these advantages, certain challenges concerning the inverter and motor interfacing have been present for decades. As insulated gate bipolar transistors entered the market, the inverter output voltage transition rate significantly increased compared with their predecessors. Inverters operate based on pulse width modulation of the output voltage, and the steep voltage edge fed by the inverter produces a motor terminal overvoltage. The overvoltage causes extra stress to the motor insulation, which may lead to a prematuremotor failure. The overvoltage is not generated by the inverter alone, but also by the sum effect of the motor cable length and the impedance mismatch between the cable and the motor. Many solutions have been shown to limit the overvoltage, and the mainstream products focus on passive filters. This doctoral thesis studies an alternative methodology for motor overvoltage reduction. The focus is on minimization of the passive filter dimensions, physical and electrical, or better yet, on operation without any filter. This is achieved by additional inverter control and modulation. The studied methods are implemented on different inverter topologies, varying in nominal voltage and current.For two-level inverters, the studied method is termed active du/dt. It consists of a small output LC filter, which is controlled by an independent modulator. The overvoltage is limited by a reduced voltage transition rate. For multilevel inverters, an overvoltage mitigation method operating without a passive filter, called edge modulation, is implemented. The method uses the capability of the inverter to produce two switching operations in the same direction to cancel the oscillating voltages of opposite phases. For parallel inverters, two methods are studied. They are both intended for two-level inverters, but the first uses individual motor cables from each inverter while the other topology applies output inductors. The overvoltage is reduced by interleaving the switching operations to produce a similar oscillation accumulation as with the edge modulation. The implementation of these methods is discussed in detail, and the necessary modifications to the control system of the inverter are presented. Each method is experimentally verified by operating industrial frequency converters with the modified control. All the methods are found feasible, and they provide sufficient overvoltage protection. The limitations and challenges brought about by the methods are discussed.
Comparison between open and laparoscopic elective cholecystectomy in elderly, in a teaching hospital

Objective: to analyze the differences in mortality rates, length of hospital stay, time of surgery and the conversion rate between elective open cholecystectomies (OC) and laparoscopic ones (LC) in elderly patients. Methods : we evaluated medical records of patients 65 years of age or older undergoing open or laparoscopic cholecystectomy at the Hospital Regional de Mato Grosso do Sul between January 2008 and December 2011. We excluded individuals operated in non-elective scenarios or who underwent intraoperative cholangiography. Results : we studied 113 patients, of whom 38.1% were submitted to the OC and 61.9%, to LC. Women accounted for 69% of patients and men, for 31%. The conversion rate was 2.9%. The mean age and duration of the procudure was 70.1 and 84 minutes, respectively, with no significant difference between OC and LC. Patients undergoing LC had shorter hospital stays (2.01 versus 2.95 days, p=0.0001). We identified operative complications in sixpatients (14%) after OC and in nine (12%) after LC, with no statistical difference. Conclusion : there was no difference in morbidity and mortality when comparing OC with LC. The laparoscopic approach led to shorter hospital stay. Operative time did not differ between the two access routes. The conversion rate was similar to other studies.

O Grupo Mycoplasma mycoides (GMM) foi diagnosticado por PCR-REA e imunoperoxidase indireta (IPI) em amostras de lavados de conduto auditivo de bovinos no Estado do Rio de Janeiro, Brasil. 60 bovinos foram selecionados aleatoriamente. As lavagens foram feitas com uso de seringas estéreis contendo um volume de 60 mL de solução salina tamponada (PBS pH 7.2). As amostras obtidas foram estocadas em glicerol (1:2) e congeladas a 20ºC até uso. Estas amostras foram diluídas até 10-5 e repicadas em meio Hayflick modificado, sólido e líquido, sendo incubados a 37ºC por 48-72 horas. As placas foram mantidas em microaerofilia e observadas diariamente, para visualização das colônias típicas em ovo-frito. Das 60 amostras cultivadas, 48 (80,00%) foram positivas para Mycoplasma spp. A prevalência obtida para o GMM na IPI foi de 20,0% (12/60) enquanto na PCR-REA foi de 41,7% (25/60). Das cepas tipificadas pela IPI 58,3% (7/12) foram M. mycoides subsp. mycoides LC e 41,7% (5/12) foram M. capricolum. Na PCR-REA o grupo M. mycoides foi confirmado pela visualização de um amplicon de 785bp, compatível com este grupo. O valor encontrado no teste Kappa para associação entre estes testes foi de 0,14 (P>0,05. Na clivagem do produto da PCR com a enzima de restrição AluI, de cepas de referências e dos isolados de ouvido os fragmentos obtidos foram de 81, 98, 186 e 236pb, mas não de 370pb, que é específica para o agente da Pleuropneumonia Contagiosa Bovina. A presença de espécies de micoplasmas no conduto auditivo de bovinos assintomáticos representa um risco para propagação de Mycoplasma spp. entre rebanhos bovinos no Brasil.

Esse estudo foi realizado com o objetivo de verificar a associação entre micoplasmas e ácaros (Raillietia auris e R. flechtmanni) no conduto auditivo de bovinos. Foram realizadas lavagens no conduto auditivo externo de 60 bovinos abatidos no Estado do Rio de Janeiro, Brasil. Para a lavagem dos condutos auditivos foi utilizada solução salina tamponada (PBS, pH 7.2) em seringas estéreis de 60mL. Para o isolamento de micoplasmas foram utilizados pools de ácaros por animal, lavados sucessivamente em 1mL de meio Hayflick modificado. Os lavados dos ácaros foram diluídos de 10-1 até 10-5 e repicados em meio Hayflick modificado, sólido e líquido e incubados a 37°C por 48-72 horas em microaerofilia. A identificação das espécies de micoplasmas foi realizada pelo teste da imunoperoxidase indireta (IPI). Verificou-se alta prevalência de ácaros do gênero Raillietia spp. 76,7% (46/60). O parasitismo por ácaros e micoplasmas foi verificado em 40 animais (74,1%), sendo esta associação significativa (p<0,001). Dos ácaros processados para isolamento de micoplasmas, 193 foram fêmeas e 25 machos. A frequência de Mycoplasma em Raillietia spp. foi de 81,2% (177/218) (p<0.001). Das fêmeas identificadas, 52,3% (101/193) foram R. auris e 47,7% (92/193) R. flechtmanni. A frequência de Mycoplasma nas fêmeas de R. auris foi de 75,2% (76/101) e na espécie R. flechtmanni foi de 88% (81/92) (P<0.05). As espécies de micoplasmas tipificadas pela IPI nos ácaros Raillietia auris foram: M. alkalescens 6,9%, M. arginini 3,4%, M. bovirhinis 9,2%, M. conjunctivae 18,4%, M. mycoides mycoides LC 8,0%, M. capricolum 5,7%. Em R. flechtmanni as espécies de micoplasmas identificadas foram: M. alkalescens 12,2%, M. arginini 1,0%, M. bovirhinis 18,9%, M. bovis 2,2%, M. conjunctivae 21,0%, M. mycoides mycoides LC 11,0% e M. capricolum 4,4%. As espécies de micoplasmas identificadas no conduto auditivo externo dos bovinos foram as mesmas presentes nos ácaros R. auris e R. flechtmanni. Os resultados confirmam que o conduto auditivo externo de bovinos é um habitat de Mycoplasma spp., incluindo espécies potencialmente patogênicas para os rebanhos, além dos ácaros R. auris e R. flechtmanni estarem associados com esses molicutes carreando-os em seu organismo.

A Artrite Encefalite Caprina (AEC) e a Linfadenite Caseosa (LC) possuem alta incidência e transmissibilidade em pequenos ruminantes. Como ambas possuem tropismo por monócitos-macrófagos e afetam mecanismos da resposta inata do hospedeiro, acredita-se que a AEC predispõe o animal a infecções por Corynebacteruim pseudotuberculosis, agente etiológico da LC. Para confirmar esta hipótese, avaliou-se a fagocitose de células da série monócito-macrófago de cabras naturalmente infectadas pelo vírus da AEC (VAEC). Para tanto, foram utilizadas 30 cabras da raça Saanen, alocadas em dois grupos distintos, com 15 animais cada, conforme a sororreatividade de anticorpos séricos antivírus da AEC. Células mononucleares de sangue periférico foram isoladas por gradiente de densidade e plaqueadas para isolamento de células da série monócito-macrófago. Posteriormente, o ensaio de fagocitose de C. pseudotuberculosis foi realizado, após incubação por duas horas a 37ºC a 5% de CO2, e a visualização da fagocitose foi identificada por microscopia óptica. O presente estudo não encontrou diferença na porcentagem de monócito-macrófagos que realizaram fagocitose entre os diferentes grupos (P = 0,41). Todavia, a análise quantitativa de bactérias fagocitadas, demonstrou maior capacidade fagocítica pelos macrófagos-monócitos do grupo sororreagente ao vírus da AEC. Correlação entre monócitos fagocitando e macrófagos que fagocitaram mais de 12 bactérias foi observado neste grupo (r = 0,488; P = 0,006), não sendo o mesmo encontrado no grupo de animais sorroreagentes negativos. Os dados demonstram aumento na intensidade da fagocitose de macrófagos de animais infectados com o vírus da AEC.

The study aimed to identify potential biomarkers of mammary gland infection in Santa Inês sheep. Commercial flocks of sheep provided the same hygiene, sanitary, and nutritional management under semi-intensive production systems were monitored during the lactation stage-and assessed 15, 30, 60, and 90 days after delivery (through the end of lactation and weaning). The California Mastitis Test (CMT) was performed on the mammary glands. Milk was collected for bacterial examination and protein analysis. Bacterial culture and biochemical characterization of the samples were performed. Forty-two milk samples from healthy glands (negative CMT and bacterial testing) and 43 milk samples from infected glands (positive CMT and bacterial testing) taken at the predefined time points were assessed. A rennin solution was used to obtain the whey. The proteins analysis was performed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), which allowed for the quantification of nine whey proteins produced in healthy glands: serum albumin, lactoferrin, IgA, IgG heavy-chain (IgG HC), IgG light-chain (IgG LC), total IgG (IgG HC + IgG LC), α-lactalbumin, β-lactoglobulin, protein with MW 15.000 Da, protein with MW 29.000 Da and eleven whey proteins secreted by infected glands, including haptoglobin and α-1-acid glycoprotein. A comparison of whey proteins between healthy and infected glands showed increases (P<0.05) in the secreted and total contents of all proteins, except for IgG LC and α-lactoalbumin. The most significant changes were observed in α-1-acid glycoprotein, lactoferrin and haptoglobin, which showed three-, five-, and seven-fold increases in secretion, respectively. This study showed that haptoglobin, α-1-acid glycoprotein, lactoferrin, albumin, and the IgA and IgG immunoglobulins may serve as potential biomarkers for mammary gland infection in sheep.

O objetivo deste trabalho foi avaliar a intoxicação de planta daninha e cultivares de cana-de-açúcar ao amicarbazone. Para isso, utilizou-se Ipomoea grandifolia como planta daninha representante e os cultivares de cana-de-açúcar PO8862, SP80 3280 e RB83 5486, caracterizados como sensível, intermediário e tolerante aos herbicidas, respectivamente. Foi verificado o consumo de água e quantificada a concentração do amicarbazone em seiva de xilema dos três cultivares de cana-de-açúcar e de I. grandifolia por meio da bomba de Schollander e da cromatografia e espectrometria de massas (LC-MS). A intoxicação das plantas foi verificada através de leituras da fluorescência, com auxílio do fluorômetro portátil, que permitiu a correlação da taxa de transporte de elétrons (ETR) com a concentração de amicarbazone absorvido pelos cultivares de cana-de-açúcar e por I. grandifolia. Verificou-se, através do experimento, que a redução dos valores da ETR pode ser utilizada para indicar o nível de intoxicação de I. grandifolia e de plantas de cana-de-açúcar ao amicarbazone. I. grandifolia destacou-se em relação à cana-de-açúcar pela maior sensibilidade ao amicarbazone. A suscetibilidade diferencial dos cultivares de cana-de-açúcar PO8862, SP80 3280 e RB83 5486 pode ser justificada, possivelmente, pela absorção diferencial do amicarbazone entre os cultivares.
