A 19 cal ka BP pollen and charcoal record from Lake Shaman (44°S; 71°W, Chile) was analyzed to establish vegetation, fire and climate dynamics of the forest-steppe ecotone in Central Chilean Patagonia. Lake Shaman record indicates that the upper Río Cisnes valley was free of ice at around 19 cal ka BP. From this date and until 14.8 cal ka BP, a grass steppe with high proportions of shrubs associated to colder and drier conditions than present developed in this area. A continuous increase of Nothofagus accompanied by a decline in the steppe shrubs and sudden dominance of paludal over aquatic plants from 11 cal ka BP was associated to effective moisture increase but still under modern values. The replacement of the cold-dry grass-shrub steppe by a similar-than-present forest-steppe ecotone suggests an increase in temperature indicating the onset of the Holocene. At the same time, moderate fire activity suggested by the charcoal record could be related to major fuel availability as consequence of Nothofagus forest expansion. Between 8 and 3 cal ka BP, the record indicates the easternmost position of the forest-steppe ecotone suggesting the highest effective moisture with the establishment of seasonality between 5 and 3 cal ka BP. From 3 cal ka BP, the record indicates a retraction of the forest-steppe ecotone accompanied by a high pollen record variability and an increased fire activity. These late changes suggest decreased effective moisture associated with a high climatic variability. At regional and extra-regional scale, climatic changes at Lake Shaman's record are mostly associated to changes (latitudinal shifts and/or strengthening/weakening) of past Southern Westerlies that were previously recorded along Patagonia from the Lateglacial to the mid-Holocene. During the Late Holocene, a regional pattern characterized by high record variability emerges throughout Central Chilean Patagonia. This variability would be related to (1) low magnitude Southern Westerlies changes probably associated to ENSO and/or SAM or (2) the complex relationships between vegetation, fire and human occupations during the last 3 cal ka.

Callosobruchus maculatus has for years remained a serious menace in cowpea in Sub-Sahara Africa. The objective of this study was to investigate the effect of genotypic cowpea (Vigna unguiculata (L.) Walp) varieties, time and dose on C. maculatus exposed to powders of Piper guineense and Eugenia aromatica. Irrespective of duration and botanicals, bruchid reared on KDV showed the highest tolerance to both plant materials; while their counterparts from IAR48V were the most susceptible. Median lethal time (LT50) also varied according to the plant materials; with the highest in KDV reared bruchid [P. guineense: KDV (18.31), IAR48V (9.27), IFBV (13.17); E. aromatica: KDV (76.01), IAR48V (5.59), IFBV (6.49)]. There was a significant impact of cowpea variety (V), exposure time (T) and dose (D) on the tolerance of C. maculatus to both plant materials. The effect of all two-way (VxT, VxD, DxT) and three way interactions (V×T×D) on the tolerance of C. maculatus to both plant materials was also significant. Varietal effect was more pronounced in bruchids exposed to E. aromatica; while exposure time was more pronounced in bruchids exposed to P. guineense.

Background Vascular hyperproliferative disorders are characterized by excessive smooth muscle cell (SMC) proliferation leading to vessel remodeling and occlusion. In pulmonary arterial hypertension (PAH), SMC phenotype switching from a terminally differentiated contractile to synthetic state is gaining traction as our understanding of the disease progression improves. While maintenance of SMC contractile phenotype is reportedly orchestrated by a MEF2C-myocardin (MYOCD) interplay, little is known regarding molecular control at this nexus. Moreover, the burgeoning interest in microRNAs (miRs) provides the basis for exploring their modulation of MEF2C-MYOCD signaling, and in turn, a pro-proliferative, synthetic SMC phenotype. We hypothesized that suppression of SMC contractile phenotype in pulmonary hypertension is mediated by miR-214 via repression of the MEF2C-MYOCD-leiomodin1 (LMOD1) signaling axis. Methods and Results In SMCs isolated from a PAH patient cohort and commercially obtained hPASMCs exposed to hypoxia, miR-214 expression was monitored by qRT-PCR. miR-214 was upregulated in PAH- vs. control subject hPASMCs as well as in commercially obtained hPASMCs exposed to hypoxia. These increases in miR-214 were paralleled by MEF2C, MYOCD and SMC contractile protein downregulation. Of these, LMOD1 and MEF2C were directly targeted by the miR. Mir-214 overexpression mimicked the PAH profile, downregulating MEF2C and LMOD1. AntagomiR-214 abrogated hypoxia-induced suppression of the contractile phenotype and its attendant proliferation. Anti-miR-214 also restored PAH-PASMCs to a contractile phenotype seen during vascular homeostasis. Conclusions Our findings illustrate a key role for miR-214 in modulation of MEF2C-MYOCD-LMOD1 signaling and suggest that an antagonist of miR-214 could mitigate SMC phenotype changes and proliferation in vascular hyperproliferative disorders including PAH.

Four hundred and forty-eight samples of total blood from wild monkeys living in areas where human autochthonous malaria cases have been reported were screened for the presence of Plasmodium using microscopy and PCR analysis. Samples came from the following distinct ecological areas of Brazil: Atlantic forest (N = 140), semideciduous Atlantic forest (N = 257) and Cerrado (a savannah-like habitat) (N = 51). Thick and thin blood smears of each specimen were examined and Plasmodium infection was screened by multiplex polymerase chain reaction (multiplex PCR). The frequency of Plasmodium infections detected by PCR in Alouatta guariba clamitans in the Sao Paulo Atlantic forest was 11.3% or 8/71 (5.6% for Plasmodium malariae and 5.6% for Plasmodium vivax) and one specimen was positive for Plasmodium falciparum (1.4%); Callithrix sp. (N = 30) and Cebus apella (N = 39) specimens were negative by PCR tests. Microscopy analysis was negative for all specimens from the Atlantic forest. The positivity rate for Alouatta caraya from semideciduous Atlantic forest was 6.8% (16/235) in the PCR tests (5.5, 0.8 and 0.4% for P. malariae, P. falciparum and P. vivax, respectively), while C apella specimens were negative. Parasitological examination of I he samples using thick smears revealed Plasmodium sp. infections in only seven specimens, which had few parasites (3.0%). Monkeys from the Cerrado (a savannah-like habitat) (42 specimens of A. caraya, 5 of Callithrix jacchus and 4 of C. apella) were negative in both tests. The parasitological prevalence of P. vivax and P. malariae in wild monkeys from Atlantic forest and semideciduous Atlantic forest and the finding of a positive result for P.falciparum in Alouatta from both types of forest support the hypothesis that monkeys belonging to this genus could be a potential reservoir. Furthermore, these findings raise the question of the relationship between simian and autochthonous human malaria in extra-Amazonian regions. (C) 2008 Elsevier B.V. All rights reserved.

Rationale: Major coronary vessels derive from the proepicardium, the cellular progenitor of the epicardium, coronary endothelium, and coronary smooth muscle cells (CoSMCs). CoSMCs are delayed in their differentiation relative to coronary endothelial cells (CoEs), such that CoSMCs mature only after CoEs have assembled into tubes. The mechanisms underlying this sequential CoE/CoSMC differentiation are unknown. Retinoic acid (RA) is crucial for vascular development and the main RA-synthesizing enzyme is progressively lost from epicardially derived cells as they differentiate into blood vessel types. In parallel, myocardial vascular endothelial growth factor (VEGF) expression also decreases along coronary vessel muscularization. Objective: We hypothesized that RA and VEGF act coordinately as physiological brakes to CoSMC differentiation. Methods and Results: In vitro assays (proepicardial cultures, cocultures, and RALDH2 [retinaldehyde dehydrogenase-2]/VEGF adenoviral overexpression) and in vivo inhibition of RA synthesis show that RA and VEGF act as repressors of CoSMC differentiation, whereas VEGF biases epicardially derived cell differentiation toward the endothelial phenotype. Conclusion: Experiments support a model in which early high levels of RA and VEGF prevent CoSMC differentiation from epicardially derived cells before RA and VEGF levels decline as an extensive endothelial network is established. We suggest this physiological delay guarantees the formation of a complex, hierarchical, tree of coronary vessels. (Circ Res. 2010;107:204-216.)

In recent years, reactive oxygen species (ROS) derived from the vascular isoforms of NADPH oxidase, Nox1, Nox2, and Nox4, have been implicated in many cardiovascular pathologies. As a result, the selective inhibition of these isoforms is an area of intense current investigation. In this study, we postulated that Nox2ds, a peptidic inhibitor that mimics a sequence in the cytosolic B-loop of Nox2, would inhibit ROS production by the Nox2-. but not the Noxl- and Nox4-oxidase systems. To test our hypothesis, the inhibitory activity of Nox2ds was assessed in cell-free assays using reconstituted systems expressing the Nox2-, canonical or hybrid Nox1- or Nox4-oxidase. Our findings demonstrate that Nox2ds, but not its scrambled control, potently inhibited superoxide (O(2)(center dot-)) production in the Nox2 cell-free system, as assessed by the cytochrome c assay. Electron paramagnetic resonance confirmed that Nox2ds inhibits O(2)(center dot-) production by Nox2 oxidase. In contrast, Nox2ds did not inhibit ROS production by either Nox1- or Nox4-oxidase. These findings demonstrate that Nox2ds is a selective inhibitor of Nox2-oxidase and support its utility to elucidate the role of Nox2 in organ pathophysiology and its potential as a therapeutic agent. Nowadays, noninvasive methods of diagnosis have increased due to demands of the population that requires fast, simple and painless exams. These methods have become possible because of the growth of technology that provides the necessary means of collecting and processing signals. New methods of analysis have been developed to understand the complexity of voice signals, such as nonlinear dynamics aiming at the exploration of voice signals dynamic nature. The purpose of this paper is to characterize healthy and pathological voice signals with the aid of relative entropy measures. Phase space reconstruction technique is also used as a way to select interesting regions of the signals. Three groups of samples were used, one from healthy individuals and the other two from people with nodule in the vocal fold and Reinke`s edema. All of them are recordings of sustained vowel /a/ from Brazilian Portuguese. The paper shows that nonlinear dynamical methods seem to be a suitable technique for voice signal analysis, due to the chaotic component of the human voice. Relative entropy is well suited due to its sensibility to uncertainties, since the pathologies are characterized by an increase in the signal complexity and unpredictability. The results showed that the pathological groups had higher entropy values in accordance with other vocal acoustic parameters presented. This suggests that these techniques may improve and complement the recent voice analysis methods available for clinicians. (C) 2008 Elsevier Inc. All rights reserved.

Diffusion coefficients and retardation factors of two metal cations (Cd2+ and Pb2+) were measured for a compacted Brazilian saprolitic soil derived from gneiss, aiming to assess its geoenvironmental performance as a liner for waste disposal sites. This soil occurs extensively all over the country in very thick layers, but has not been used in liners because of its hydraulic conductivity, higher than 10(-9) m/s when compacted at optimum water content of standard Proctor energy, but which can be reduced by means of appropriate compaction techniques or additives. Batch, column, and diffusion tests were carried out with monospecies synthetic solutions at pH 1, 3, and 5.5. Measured diffusion coefficients varied between 0.5 and 4 X 10(-10) m(2)/s. Retardation factors show that cadmium, a very mobile cation, is not adsorbed at pH I but is significantly retained at pH 3 and pH 5.5, whereas lead is retained at all tested pH values though slightly at pH 1. Estimated retardation factors from batch tests were 1.3-2.3 times those resulting from column tests and at its highest when obtained by diffusion tests; whereas batch tests allow a more complete exposure of the soil grains to the solution, time-dependent nonspecific adsorption may take longer to occur. The importance of contact time was observed and should be considered in further investigations. Its significant retention of metals suggests a promising utilization of this soil as a bottom liner for wastes landfills.

The present work demonstrates the successful application of automated biocompatible in-tube solid-phase microextraction coupled with liquid chromatography (in-tube SPME/LC) for determination of interferon alpha(2a) (IFN alpha(2a)) in plasma samples for therapeutic drug monitoring. A restricted access material (RAM, protein-coated silica) was employed for preparation of a lab-made biocompatible in-tube SPME capillary that enables the direct injection of biological fluids as well as the simultaneous exclusion of macromolecules by chemical diffusion barrier and drug pre-concentration. The in-tube SPME variables, such as sample volume, draw/eject volume, number of draw-eject cycles, and desorption mode were optimized, to improve the sensitivity of the proposed method. The IFN alpha(2a) analyses in plasma sample were carried out within 25 min (sample preparation and LC analyses). The response of the proposed method was linear over a dynamic range, from 0.06 to 3.0 MIU mL(-1), with correlation coefficient equal to 0.998. The interday precision of the method presented coefficient of variation lower than 8%. The proposed automated method has adequate analytical sensitivity and selectivity for determination of IFN alpha(2a) in plasma samples for therapeutic drug monitoring. (C) 2010 Elsevier B.V. A simple, fast, and sensitive liquid-liquid extraction method followed by nonaqueous capillary electrophoresis (LLE/NACE) was developed and validated for Simultaneous determination of four antidepressants (fluoxetine, sertraline, citalopram and paroxetine) in human plasma. Several experimental separation conditions using aqueous and nonaqueous media separation were tested by varying the electrolyte pH value (for aqueous medium) and the ionic strength concentration considering the similar mobility of the compounds. High-resolution separation was achieved with a mixture of 1.25 mol L(-1) of phosphoric acid in acetonitrile. The quantification limits of the LLE/CE method varied between 15 and 30 ng mL(-1), with a relative standard deviation (RSD) lower than 10.3%. The method was successfully applied in therapeutic drug monitoring and should be employed in the evaluation of plasma levels in urgent toxicological analysis. (C) 2009 Elsevier B.V. O conhecimento da vegetação em regiões de ocorrências minerais de cobre e ouro torna-se de grande importância para o desenvolvimento de tecnologias limpas na reabilitação de áreas degradadas pela mineração (fitorremediação) e na bioprospecção mineral no Estado. Buscou-se verificar a relação entre a organização espacial e fitossociológica das unidades e subunidades de vegetação na mina Volta Grande, Lavras do Sul, RS e a presença de cobre nas mineralizações. Com base em Ecologia de Paisagem, realizou-se investigações fitossociológicas, avaliação do conteúdo de cobre em raízes e folhas de Axonopus affinis, Eugenia uniflora, Heterothalamus alienus, Saccharum angustifolium, Schizachyrium microstachyum e Schinus lentiscifolius e o zoneamento da vegetação com sobreposição ao mapa de estruturas mineralizadas conhecidas. De acordo com os resultados obtidos, sugere-se que a distribuição das unidades e subunidades de vegetação (unidade de vegetação Eugenia uniflora – Scutia buxifolia, com subunidades Eugenia uniflora – Cupania vernalis e Eugenia uniflora – Allophylus edulis; unidade de vegetação Schinus lentiscifolius – Heterothalamus alienus; unidade de vegetação Eryngium horridum – Saccharum angustifolium, com subunidade Eryngium horridum – Piptochaetium montevidense e unidade de vegetação Axonopus affinis – Paspalum pumilum) pode estar relacionada à posição geomorfológica, à declividade, à umidade do solo e ao manejo para o uso do gado A unidade de vegetação Schinus lentiscifolius – Heterothalamus alienus parece estar relacionada às ocorrências de mineralizações (filões) de cobre e ouro, sendo necessária uma comparação com outras áreas mineralizadas e não-mineralizadas no Estado para utilizar esse dado em prospecção mineral. A espécie Axonopus affinis possui em suas raízes um conteúdo de cobre muito maior do que o considerado normal em plantas, sendo indicada para estudos mais detalhados quanto a sua aplicabilidade na reabilitação de áreas degradadas pela mineração no Rio Grande do Sul.

A dissertação analisa o posicionamento dos médicos gaúchos frente ao princípio da liberdade profissional que vigorava no Rio Grande do Sul durante a Primeira República. A Constituição estadual de 1891 permitia o exercício da medicina por indivíduos não portadores de diploma acadêmico. Neste contexto, o estudo demonstra que os médicos diplomados procuravam legitimar cientificamente suas posições nas instituições que ocupavam. A Faculdade de Medicina de Porto Alegre ocupou um lugar privilegiado neste processo de afirmação da supremacia do conhecimento científico face a práticas de cura derivadas de outros princípios. Desta maneira, a dissertação acompanha a produção intelectual, expressa em conferências e artigos publicados na imprensa especializada, de um grupo de médicos ligados a esta instituição. Descreve também a organização dos serviços de saúde pública no Estado e o crescente envolvimento do discurso médico com as questões sociais. O conceito de “medicina social”, veiculado pelos médicos no período analisado, reuniu as noções de higiene, eugenia e educação sanitária e contribuiu para a formulação de normas para regular a vida nas cidades. Através da defesa da medicina social foi possível articular os interesses corporativos aos da sociedade como um todo, apresentando a regulamentação profissional como uma necessidade social.

O estudo a seguir apresenta um levantamento bibliográfico e exploratório sobre o estresse do profissional que cursa o MBA (Master in Business Administration) devido à sobrecarga de informações a que é exposto. Após examinar o estilo de vida desse profissional durante a fase em que está em contato com novas informações, de diversas áreas e em grande volume, este documento passa a analisar o estresse em relação à motivação, e à luz de várias teorias. O levantamento exploratório sobre o modo de vida do profissional em estudo, sugere que esse profissional está exposto a diversos fatores estressores, que demonstram forte probabilidade desse profissional vir a apresentar sintomas de estresse durante, ou mesmo, logo após o curso do MBA. Finalmente, este estudo apresenta algumas idéias de como esse profissional e o próprio meio acadêmico podem evitar, ou minimizar a vulnerabilidade dele ao estresse, devido à presença de fatores estressores

A cidade de Porto Alegre, devido à localização, abriga espécies vegetais procedente de dois corredores principais: o corredor Atlântico e o corredor do Alto Uruguai. Os morros graníticos sobressaem-se na paisagem porto-alegrense abrigando matas nas suas encostas sul e campos nos topos e encostas nortes, devido às condições criadas pela exposição solar diferenciada. O Morro Santana, com 311 metros é o mais alto deles. Com o objetivo de caracterizar a composição e a estrutura e aspectos da dinâmica de regeneração desta mata, é realizada amostragem do componente arbóreo-arbustivo, dividindo-se os indivíduos em três componentes, de acordo com suas medidas de alturas e de DAP (diâmetro à altura do peito): Componente 3 (0,20 m ≤ h < 1 m); Componente 2 (h ≥ 1 m e DAP < 3 cm) e Componente 1 (DAP≥ 3 cm), sendo que, para a estimativa de regeneração natural por classes e total (RN e RNT), a qual é dada em porcentagem, os componentes 2 e 3 foram distribuídos em três classes de altura: Classe 1 (0,20m ≤ h < 1m); Classe 2 (1m ≤ h < 3m); Classe 3 ( h ≥ 3m e DAP < 5cm).O método utilizado é o de parcelas de 100 m2 (Componente1), 25 m2 (Componente 2), e 4 m2 (Componente 3). Através do software MULVA 5, foram realizadas análises de agrupamento e ordenação (PCoA) para os três componentes. O levantamento resultou em: 505 indivíduos, pertencentes a 63 espécies, 51 gêneros e 30 famílias no Componente 1; 470 indivíduos, distribuídos em 44 espécies, 33 gêneros e 19 famílias no Componente 2; 191 indivíduos, distribuídos em 30 espécies, 26 gêneros e 18 famílias no Componente 3. No Componente 1 destacam-se Guapira opposita (Nyctaginaceae), Pachystroma longifolium (Euphorbiaceae) e Eugenia rostrifolia (Myrtaceae). Nos Componentes 2 e 3 destacam-se Psychotria leiocarpa e Faramea montevidensis (Rubiaceae), Mollinedia elegans (Monimiaceae) e Gymnanthes concolor (Euphorbiaceae). A composição florística nos três componentes, em ordem decrescente de importância, foi dada por espécies de ampla distribuição, espécies Atlânticas e espécies do Alto Uruguai. As espécies secundárias perfizeram a maioria absoluta da amostra nos três componentes, sendo que as secundárias tardias superaram as secundárias inicias. As espécies pioneiras foram infimamente representadas nos três componentes inventariados. Quanto às síndromes de dispersão, mais de 80 por cento das espécies de ambos os componentes apresentou síndrome zoocórica. O restante das espécies apresenta as síndromes autocórica e anemocórica em proporções semelhantes.A análise fitossociológica indicou tratar-se de uma mata baixa, sem estratificação definida, com dominância de um número reduzido de espécies de ocorrência comum na região, sendo que a grande maioria das espécies conbribui com um ou dois indivíduos. A diversidade específica (H’) foi estimada em 3,30 (Componente 1); 2,81 (Componente 2) e 2,86 (Componente3) e a equabilidade (J’) em 0,80 (Componente 1) ; 0,74 (Componente 2) e 0,84 (Componente 3). As análises de agrupamento e de ordenação não evidenciaram diferenças entre as unidades de amostragem da borda e aquelas do interior da mata. Os maiores valores na estimativa de regeneração natural total (RNT) foram concentrados pelas espécies de sub-bosque com maiores densidades e freqüência (Psychotria leiocarpa, Faramea montevidensis, Mollinedia elegans e Gymnanthes concolor). Os representantes que compõem o dossel da mata com maiores valores de regeneration foram: Guapira opposita, Eugenia rostrifolia e Cupania vernalis (Sapindaceae). Os resultados da estimativa de regeneração natural indicaram que a maioria das espécies parece estar obtendo sucesso quanto ao recrutamento de novos indivíduos, o que remete a um bom estado de conservação da mata, a qual, permanecendo as condições atuais, deverá no futuro manter uma composição florística semelhante a atual.
