Designers who want to manufacture a hardenable steel component need to select both the steel and its heat treatment. This project aims to develop a selection methodology for steels and process routes as an aid to designers. Three studies were conducted: - production of software to calculate the "equivalent diameter" and "equivalent Jominy distance" for simple shapes of a steel component; - prediction of semi-empirical Jominy curves (as-cooled) using CCT diagrams and process modelling methods, which were validated by experiment on plain carbon steels; - investigation of tempering of Jominy bars to explore the potential for semi-empirical models for the hardness after tempering.

We aim to characterize fault slip behavior during all stages of the seismic cycle in subduction megathrust environments with the eventual goal of understanding temporal and spatial variations of fault zone rheology, and to infer possible causal relationships between inter-, co- and post-seismic slip, as well as implications for earthquake and tsunami hazard. In particular we focus on analyzing aseismic deformation occurring during inter-seismic and post-seismic periods of the seismic cycle. We approach the problem using both Bayesian and optimization techniques. The Bayesian approach allows us to completely characterize the model parameter space by searching a posteriori estimates of the range of allowable models, to easily implement any kind of physically plausible a priori information and to perform the inversion without regularization other than that imposed by the parameterization of the model. However, the Bayesian approach computational expensive and not currently viable for quick response scenarios. Therefore, we also pursue improvements in the optimization inference scheme. We present a novel, robust and yet simple regularization technique that allows us to infer robust and somewhat more detailed models of slip on faults. We apply such methodologies, using simple quasi-static elastic models, to perform studies of inter- seismic deformation in the Central Andes subduction zone, and post-seismic deformation induced by the occurrence of the 2011 Mw 9.0 Tohoku-Oki earthquake in Japan. For the Central Andes, we present estimates of apparent coupling probability of the subduction interface and analyze its relationship to past earthquakes in the region. For Japan, we infer high spatial variability in material properties of the megathrust offshore Tohoku. We discuss the potential for a large earthquake just south of the Tohoku-Oki earthquake where our inferences suggest dominantly aseismic behavior.

Two overrun effects in the Coulomb explosion dynamics of heteronuclear clusters have been investigated theoretically by the use of a simplified electrostatic model. When the charge-to-mass ratio of light ions is higher than that of heavy ions, the light ions can overtake the heavy ions inside the cluster and acquire a higher kinetic energy. Further, if the charge density of the heavy ions is twice as high as that of the light ions, i.e. a proposed competitive parameter xi = rho BqB/rho AqA > 2, the inner light ions can overtake those light ions on the surface of the cluster and form a shock shell during the explosion, which might drive the intracluster collision and fusion of the light ions. Different regimes of nuclear fusion are discussed and the corresponding neutron yields are estimated. Our analysis indicates that the probability of intracluster fusion is quite low even if deuterated heteronuclear clusters such as (DI)(n) with large size and high competitive parameter are employed. However, heteronuclear clusters are still a better candidate compared with homonuclear clusters for enhancing the total intercluster fusion yield because both a higher energy region and a higher proportion of deuterons distributing in the energy region can be created in the deuterated heteronuclear clusters.

This thesis describes a series of experimental studies of lead chalcogenide thermoelectric semiconductors, mainly PbSe. Focusing on a well-studied semiconductor and reporting good but not extraordinary zT, this thesis distinguishes itself by answering the following questions that haven’t been answered: What represents the thermoelectric performance of PbSe? Where does the high zT come from? How (and how much) can we make it better? For the first question, samples were made with highest quality. Each transport property was carefully measured, cross-verified and compared with both historical and contemporary report to overturn commonly believed underestimation of zT. For n- and p-type PbSe zT at 850 K can be 1.1 and 1.0, respectively. For the second question, a systematic approach of quality factor B was used. In n-type PbSe zT is benefited from its high-quality conduction band that combines good degeneracy, low band mass and low deformation potential, whereas zT of p-type is boosted when two mediocre valence bands converge (in band edge energy). In both cases the thermal conductivity from PbSe lattice is inherently low. For the third question, the use of solid solution lead chalcogenide alloys was first evaluated. Simple criteria were proposed to help quickly evaluate the potential of improving zT by introducing atomic disorder. For both PbTe1-xSex and PbSe1-xSx, the impacts in electron and phonon transport compensate each other. Thus, zT in each case was roughly the average of two binary compounds. In p-type Pb1-xSrxSe alloys an improvement of zT from 1.1 to 1.5 at 900 K was achieved, due to the band engineering effect that moves the two valence bands closer in energy. To date, making n-type PbSe better hasn’t been accomplished, but possible strategy is discussed.

In this thesis, we discuss 3d-3d correspondence between Chern-Simons theory and three-dimensional N = 2 superconformal field theory. In the 3d-3d correspondence proposed by Dimofte-Gaiotto-Gukov information of abelian flat connection in Chern-Simons theory was not captured. However, considering M-theory configuration giving the 3d-3d correspondence and also other several developments, the abelian flat connection should be taken into account in 3d-3d correspondence. With help of the homological knot invariants, we construct 3d N = 2 theories on knot complement in 3-sphere for several simple knots. Previous theories obtained by Dimofte-Gaiotto-Gukov can be obtained by Higgsing of the full theories. We also discuss the importance of all flat connections in the 3d-3d correspondence by considering boundary conditions in 3d N = 2 theories and 3-manifold.

The investigations presented in this thesis use various in vivo techniques to understand how trans-acting factors control gene expression. The first part addresses the transcriptional regulation of muscle creatine kinase (MCK). MCK expression is activated during the course of development and is found only in differentiated muscle. Several in vivo footprints are observed at the enhancer of this gene, but all of these interactions are limited to cell types that express MCK. This is interesting because two of the footprints appear to represent muscle specific use of general transcription factors, while the other two correspond to sites that can bind the myogenic regulator, MyoD1, in vitro. MyoD1 and these general factors are present in myoblasts, but can bind to the enhancer only in myocytes. This suggests that either the factors themselves are post-translationally modified (phosphorylation or protein:protein interactions), or the accessibility of the enhancer to the factors is limited (changes in chromatin structure). The in vivo footprinting study of MCK was performed with a new ligation mediated, single-sided PCR (polymerase chain reaction) technique that I have developed.

The second half of the thesis concerns the regulation of mouse metallothionein (MT). Metallothioneins are a family of highly conserved housekeeping genes whose expression can be induced by heavy metals, steroids, and other stresses. By adapting a primer extension method of genomic sequencing to in vivo footprinting, I've observed both metal inducible and noninducible interactions at the promoter of MT-I. From these results I've been able to limit the possible mechanisms by which metal responsive trans-acting factors induce transcription. These interpretations correlate with a second line of experiments involving the stable titration of positive acting factors necessary for induction of MT. I've amplified the promoter of MT to 10^2-10^3 copies per cell by fusing the 5' and 3' ends of the MT gene to the coding region of DHFR and selecting cells for methotrexate resistance. In these cells, there is a metal-specific titration effect, and although it acts at the level of transcription, it appears to be independent of direct DNA binding factors.

Although there have been a number of studies on aquatic conditions and the flora and fauna of Lake Titicaca over many decades, most of this work has been centred on the offshore regions of the main lake. Water quality there has been degrading and abundant growth of Lemna spp. has been developing. Lemna spp., commonly called floating duck-weed or ‘lenteja de agua’ in Puno, occurs perennially in most parts of the inner Puno Bay shore-line. In this article, the authors compare water quality changes over recent decades in shore-line regions of Inner Puno Bay and their possible effects on the distribution, abundance and biomass of Lemna spp..

This project analyses the influence of the futures market on middle and low income countries. In it, I attempt to show that investments made by large investment funds in this market, as well as by certain pension plans, bring major consequences whose effects are more evident in less developed countries. The cornerstones of the work are as follows; to attempt to see the existing relationship between the commodity futures market and its underlying assets; analysing products such as wheat, rice and corn in-depth, because these are the most basic foodstuffs at a global level; to determine how an increase in trading in these markets can affect the lives of people in the poorest countries; to analyse investor concern regarding the consequences that their investments may have. Throughout the project we will see how large speculators use production forecasting models to determine the shortage of a commodity in order to take a position in the futures market to profit from it. In addition we will see how an increase in trading in this market causes an increase in the price of the underlying asset in the spot market. As for investor concern, I can say it is negligible, but the idea of running pension plans or investment funds that follow some social criteria has been welcomed by those interviewed, which makes me think that different legislation is possible. This legislation will only come into existence if it is demanded by the people. A fact that now becomes complicated because without a minimum financial basis, they cannot even know how the large investment funds trade with hunger in the world. The day when most people understand how large speculators profit from famine will be the day to put pressure on governments to begin to put limits on speculation. This makes financial awareness necessary in order to achieve a curb in excessive speculation.

We know very little about the role of birds in different ecosystems, despite numerous interesting works of researchers and amateur ornithologist scattered throughout the world. Less attention is paid to freshwater ecosystems because of the difficulties encountered in the experimental methods as well as a lack of interest on behalf of hydrobiologists, for the activities of birds in these ecosystems. This article summarises types and morphoecological forms of waterfowl and their place in the lake ecosystem.

Fuzzy-reasoning theory is widely used in industrial control. Mathematical morphology is a powerful tool to perform image processing. We apply fuzzy-reasoning theory to morphology and suggest a scheme of fuzzy-reasoning morphology, including fuzzy-reasoning dilation and erosion functions. These functions retain more fine details than the corresponding conventional morphological operators with the same structuring element. In a 1955 paper, Ky Fan, Olga Taussky, and John Todd presented discrete analogues of inequalities of Wirtinger type, and by taking limits they were able to recover the continuous inequalities. We generalize their techniques to mixed and higher derivatives and inequalities with weight functions in the integrals. We have also considered analogues of inequalities of Müller and Redheffer and have used these inequalities to derive a necessary and sufficient condition on ordered pairs of numbers so that the first number is the square norm of the kth derivative of some periodic function and the second number is the square norm of the mth derivative of the same periodic function.

When salmonid redds are disrupted by spates, the displaced eggs will drift downstream. The mean distance of travel, the types of locations in which the eggs resettle and the depth of reburial of displaced eggs are not known. Investigation of these topics under field conditions presents considerable practical problems, though the use of artificial eggs might help to overcome some of them. Attempts to assess the similarities and/or differences in performance between real and artificial eggs are essential before artificial eggs can validly be used to simulate real eggs. The present report first compares the two types of egg in terms of their measurable physical characteristics (e.g. dimensions and density). The rate at which eggs fall in still water will relate to the rate at which they are likely to resettle in flowing water in the field. As the rate of fall will be influenced by a number of additional factors (e.g. shape and surface texture) which are not easily measured directly, the rates of fall of the two types of egg have been compared directly under controlled conditions. Finally, comparisons of the pattern of settlement of the two types of egg in flowing water in an experimental channel have been made. Although the work was primarily aimed at testing the value of artificial eggs as a simulation of real eggs, several side issues more directly concerned with the properties of real eggs and the likely distance of drift in natural streams have also been explored. This is the first of three reports made on this topic by the author in 1984.

The presence of fungi on the phytoplankton of Lake Windermere was first noted by the author in 1943. Surveys into the fungal and protozoan parasites of the planktonic algae of the English Lake District and elsewhere were carried out. This article discusses the descriptive studies, epidemics and culture work, which was carried out in connection with these studies.

This dissertation is divided into three parts.

The first section is concerned with protein synthesis in cellfree systems from reticulocytes. The sub-cellular reticulocyte fractions, reagents, etc. have been examined for the presence of traces of ribonuclease, using. an assay based upon the loss of infectivity of RNA fran bacteriophage MS2. This assay is sensitive to 5 x 10-7 γ RNase/ml. In addition, the loss of synthetic capacity of an 80S ribosome on dissociation has been studied, and can be attributed to loss of messenger RNA when the monomer is separated into subunits. The presence of ribonuclease has been shown to be a major cause of polyribosome disintegration during cell-free protein synthesis.

The second section concerns the changes in ribosomes and polyribosomes which occur during the maturation of a reticulocyte into an erythrocyte. With increasing age, the cells lose a large proportion of the ribonucleoprotein, but the percentage of ribosomes present as polyribosomes is only slightly altered. The loss of hemoglobin synthesis on maturation is probably due to both the loss of total ribosomes and to the lessened specific activity of the polyribosomes.

The third section contains analytical ultracentrifugation data on 80S ribosomes, polyribosomes, and ribosomal RNA from reticulocytes. The 60s and 40s subunits, obtained by dissociation of the 80s particle with inorganic pyrophosphate, were also studied. The RNA from reticulocyte ribosomes has been examined under a variety of denaturing conditions, including dimethyl sulfoxide treatment, formaldehyde reaction and thermal denaturation. From these studies we can conclude that the 28S and 16S RNA's are single polynucleotide chains and are not made up of smaller RNA subunits hydrogen-bonded together.
