Because of the role that DNA damage and depletion play in human disease, it is important to develop and improve tools to assess these endpoints. This unit describes PCR-based methods to measure nuclear and mitochondrial DNA damage and copy number. Long amplicon quantitative polymerase chain reaction (LA-QPCR) is used to detect DNA damage by measuring the number of polymerase-inhibiting lesions present based on the amount of PCR amplification; real-time PCR (RT-PCR) is used to calculate genome content. In this unit, we provide step-by-step instructions to perform these assays in Homo sapiens, Mus musculus, Rattus norvegicus, Caenorhabditis elegans, Drosophila melanogaster, Danio rerio, Oryzias latipes, Fundulus grandis, and Fundulus heteroclitus, and discuss the advantages and disadvantages of these assays.

The rust Puccinia psidii infects many species in the family Myrtaceae. Native to South America, the pathogen has recently entered Australia which has a rich Myrtaceous flora, including trees of the ecologically and economically important genus Eucalyptus. We studied the genetic basis of variation in rust resistance in Eucalyptus globulus, the main plantation eucalypt in Australia. Quantitative trait loci (QTL) analysis was undertaken using 218 genotypes of an outcross F2 mapping family, phenotyped by controlled inoculation of their open pollinated progeny with the strain of P. psidii found in Australia. QTL analyses were conducted using a binary classification of individuals with no symptoms (immune) versus those with disease symptoms, and in a separate analysis dividing plants with disease symptoms into those exhibiting the hypersensitive response versus those with more severe symptoms. Four QTL were identified, two influencing whether a plant exhibited symptoms (Ppr2 and Ppr3), and two influencing the presence or absence of a hypersensitive reaction (Ppr4 and Ppr5). These QTL mapped to four different linkage groups, none of which overlap with Ppr1, the major QTL previously identified for rust resistance in Eucalyptus grandis. Candidate genes within the QTL regions are presented and possible mechanisms discussed. Together with past findings, our results suggest that P. psidii resistance in eucalypts is quantitative in nature and influenced by the complex interaction of multiple loci of variable effect.

Os híbridos de Eucalyptus globulus representam uma excelente alternativa para o setor de celulose e papel, em razão dos ganhos em qualidade da madeira para a fabricação de celulose. Entretanto, estes híbridos têm apresentado recalcitrância ao enraizamento adventício. Assim, a micropropagação é apontada como a técnica para o rejuvenescimento desses híbridos adultos, viabilizando a propagação clonal dos mesmos. O presente trabalho avaliou o cultivo in vitro de três clones de Eucalyptus grandis x Eucalyptus globulus e de três clones de Eucalyptus urophylla x Eucalyptus globulus, em relação à multiplicação in vitro, no meio MS suplementado com 0,5 mg L-1 de BAP e 0,01 mg L-1 de ANA, bem como o efeito das concentrações de 0,25; 0,50; 0,75 e 1,0 mg L-1 de AIB e dos meios de cultura MS e JADS no alongamento in vitro das brotações. Os clones diferiram quanto à multiplicação in vitro das brotações e apresentaram uma taxa de multiplicação média dos clones de 3,0 tufos de brotações em cada subcultivo, ao longo dos 25 subcultivos realizados. No alongamento in vitro, os clones diferiram quanto às concentrações de AIB adequadas para provocar o alongamento, bem como em relação aos meios de cultura MS e JADS. O intervalo médio entre 0,40 e 0,80 mg L-1 de AIB proporcionou o maior número e comprimento das brotações alongadas in vitro e com maior vigor.

O nitrogênio e um dos nutrientes mais demandados pelas espécies vegetais, sua presença no solo, sob formas orgânicas ou minerais disponíveis para as plantas, está vinculada à qualidade e quantidade dos resíduos vegetais aportados ao solo. O estudo teve o objetivo de avaliar a influência do cultivo do eucalipto e da acácia na composição das formas orgânicas e inorgânicas de N e, na abundância natural de 15N em um Argissolo Amarelo. Para isso, foram coletadas amostras de solo e serapilheira em monocultivos do Eucalyptus urograndis (clone do Eucalyptus urophylla S. T. Blake x Eucalyptus grandis W. Hill ex Spreng) de ciclo curto (sete anos), sistemas de cultivo de rotação com acácia ( Acacia mangium Willd.) após monocultivo de eucalipto, monocultivo de eucalipto de ciclo longo (24 anos) e mata nativa (Mata Atlântica) como condição original de solo do litoral Norte do Espírito do Santo. Foram avaliados os teores de C orgânico total, N total, N-NH4+, N-NO3-, relação C/N, fracionamento do N orgânico e abundância natural de 15N no solo e serapilheira. Das formas de N-orgânico hidrolisado, o N-amino foi a fração que apresentou maior contribuição (39%), seguida pela fração de N-não identificado (27%), da fração N-amida (18%) e N-hexosamina (15%). O povoamento de acácia promoveu menor abundância natural de 15N e maiores teores de N total e C orgânico no solo e aumentou as formas orgânicas de N-hidrolisado, quando comparado àqueles de eucalipto de ciclo curto. Isso indica o aumento de formas lábeis de N orgânico no solo para as plantas e redução da humificação da matéria orgânica do solo (MOS) de acácia. Nesse sentido, a rotação de cultivos florestais com acácia após eucalipto de ciclo curto contribuiu para o aumento de formas orgânicas no solo, importantes para a nutrição de plantas, por serem potenciais fontes de nutrientes às plantas em curto período de tempo.

One Kunitz-type trypsin inhibitors (PmTI) was purified from Piptadenia moniliformis seeds, a tree of the sub-family Mimosoideae, by TCA precipitation, affinity chromatography on immobilized trypsin-Sepharose, DEAE cellulose (ion exchange) and Superose 12 (molecular exclusion) column FPLC/AKTA. The inhibitor has Mr of 25 kDa by SDS-PAGE and chromatography molecular exclusion. The N-terminal sequence of this inhibitor showed high homology with other family Kunitz inhibitors. This also stable variations in temperature and pH and showed a small decrease in its activity when incubated with DDT in the concentration of 100mM for 120 minutes. The inhibition of trypsin by PmTI was competitive, with Ki of 1.57 x10-11 M. The activity of trypsin was effectively inhibited by percentage of inhibition of 100%, among enzymes tested, was not detected inhibition for the bromelain, was weak inhibitor of pancreatic elastase (3.17% of inhibition) and inhibited by 76.42% elastase of neutrophils, and inhibited in a moderate, chymotrypsin and papain with percentage of inhibition of 42.96% and 23.10% respectively. In vitro assays against digestive proteinases from Lepidoptera, Diptera and Coleoptera pests were carried out. Several degrees of inhibition were found. For Anthonomus grandis and Ceratitis capitata the inhibition was 89.93% and 70.52%, respectively, and the enzymes of Zabrotes subfasciatus and Callosobruchus maculatus were inhibited by 5.96% and 9.41%, respectively, and the enzymes of Plodia. interpunctella and Castnia licus were inhibited by 59.94% and 23.67, respectively. In vivo assays, was observed reduction in the development of larvae in 4rd instar of C. capitata, when PmTI was added to the artificial diet, getting WD50 and LD50 of 0.30% and 0.33%, respectively. The cercospora leaf spot, caused by Cercospora abelmoschi Ellis and Everhart, is quite common in okra culture. Therefore, this study aimed to evaluate the efficiency of aqueous extracts of neem ( Azadirachta indica A. Juss), citronella ( Cymbopogon nardus (L.) Rendle), eucalyptus ( Eucalyptus grandis L.), ecolife, A. indica oil and fungicide cercobin 700 PM® in control of cercospora leaf spot on okra in greenhouse. The extracts and neem oil were tested in concentration 10%, the fungicide cercobin 700PM® in dose 2.5 g.l-1, applied 10 days after pathogen inoculation by leaf spray and the citric biomass extract ecolife in concentration 5.0 ml.l-1, applied 10 days before pathogen inoculation. All treatments, except ecolife, were effective in controlling cercospora leaf spot and may be recommended as alternatives in agroecological systems.

Effects of different forestry covers - “mata de panda” (MP), Pinus patula (PP) Eucalyptus grandis (EG) and Grevillea robusta (GR) - installed between 1965 and 1968 in the Estação Experimental Agrícola da Chianga (Huambo, Angola), on chemical properties of Ferrallitic Soils were assessed up to 30 cm depth, as compared to those observed in nearby conventional agricultural fields. Only the soils of the areas with EG and GR showed a clear improvement in their reaction, content of organic carbon and of extractable non-acid cations and effective cation exchange capacity, usually up to 10 cm depth. The improvement associated with “mata de panda” was less pronounced and that of PP plantations was negligible or nil. The recover capacity of soil fertility may depend on the nature of tree cover as well as on the soil characteristics itself. Results also indicate that the low soil capacity to retain cations exhibited by soils of the Planalto Central of Angola can be increased through both acidity correction and increasing the content of soil organic matter

The aim of this research was to investigate the effects of wood species, particle treatments and mix proportion on the physical (density) and mechanical (compressive strength and dynamicmodulus of elasticity) properties of cement-wood composites. Different mix proportions were investigated, based on the cement: wood ratio of 0.3:0.7, in volume, with Pinus elliottii and Eucalyptus grandis sawdust percentages of 0-100, 25-75, 50-50, 75-25 or 100-0. Sawdust particles were pre-treated with either lime or cement coating to improve cement and wood compatibility. Results show that wood species, particle treatments and mix proportions may influence the physical and mechanical properties of cement-wood composites. In general, results confirm that Eucalyptus sawdust and cement are naturally compatible and do not require any previous particle treatment to avoid compatibility problems.

Este trabalho foi desenvolvido com o objetivo de avaliar e comparar o desempenho e as variações na qualidade da madeirade árvores de eucalipto implantadas em sistema silvipastoril e em monocultivo. Os dados foram coletados em 13 árvores amostras aos 36 meses de idade, selecionadas em função do intervalo de confiança da média dos diâmetros a altura do peito (DAP), e da posição das árvores na faixa de plantio no sistema silvipastoril, com face de exposição sul, norte e central. Foram avaliadas as variáveis DAP, altura total e altura comercial, volume e conicidade do tronco, densidade básica e deslocamento da medula. Concluiu-se que no sistema silvipastoril as árvores apresentaram maior DAP, menor altura total e maior conicidade do tronco que no monocultivo. O DAP não diferiu em relação à posição na faixa de plantio, porém as árvores com face de exposição norte foram mais baixas e cônicas. A densidade básica e o deslocamento da medula não foram influenciados pelo sistema de cultivo.
