Esta tesis es resultado de un intenso trabajo de investigación realizado de agosto de 2014 a agosto de 2015, con financiación de la Fundação de Amparo à Pesquisa do Estado de São Paulo (Fapesp). El objetivo se centró en la realización de un análisis, aunque preliminar, del papel de la TV pública con tecnología digital, como también de las tendencias, desafíos y perspectivas para consolidarla, tanto en Brasil como en Argentina, países con diversas similitudes en contextos económicos, sociales, políticos y culturales. Para cumplir los objetivos, estudiamos los recientes proyectos gubernamentales de televisión pública en los dos países suramericanos. El gobierno argentino invirtió en modernización en Canal 7 (antiguo Argentina Televisora Color-ATC), una emisora de casi seis décadas de funcionamiento, que ha sido prácticamente refundada a partir de los investimentos públicos del gobierno nacional argentino, realizados para digitalizar los sistemas de transmisión y de recepción de televisión abierta. El nuevo Canal 7 pasó a servir de cabeza de red del sistema de televisión pública de Argentina. En Brasil, el gobierno creó a fines del 2007, la Empresa Brasil de Comunicación (EBC), estructura administrativa que opera la red de televisión pública de TV Brasil. Ambos proyectos prometen construir y gestionar sistemas nacionales abiertos, modernos y competitivos de TV Pública Digital Abierta (TVPDA). Esta tesis también describe y compara de modo sucinto el desarrollo histórico de la radiodifusión - evidenciando la televisión como una plataforma en permanente disputa comunicacional, política, ideológica, publicitaria, cultural y económica -, tal como los modelos de televisión pública y comercial en cada uno de los dos países analizados (no entiendo la expresión). The Brazilian Federal Constitution promulgated in 1988 created the concept of Social Welfare, which is based on the triad: Health, Social Security and Social Assistance. The Unified Health System (SUS) was then instituted. SUS is a conquest of a society that seeks social justice, integrality, equalitarian and universal access to health services. In the present essay, I succinctly discourse on the various meanings of integrality. I present the theoretical basis of complexity and transdisciplinarity by opposing to reductionism, aiming at showing that, by means of transdisciplinarity and intersectoriality, integrality can be achievable.

The Amazon region has a large sociobiodiversity, where lives people with traditional knowledge about the uses of its natural resources. The Convention on Biological Diversity (CBD) in 1992 recognized the autonomy of each nation over its genetic resources and the rights of traditional populations about the knowledge of the use of these natural resources. In 2001 it was published the law M.P. 2.186-16/2001 which created the Conselho Nacional de Gestão do Patrimônio Genético (CGEN) and the first rules about access to genetic resources and associated traditional knowledge in Brazil. Since his creation, the actions of CGEN have received criticism from various sectors, both private and public, and, unlike its initial proposal, has become one of the main obstacles of ethnobotanical studies, the bureaucracy, the long process analysis and communication difficulties with this organ.

Are wild bearded capuchin monkeys selective about where they place nuts on anvils, specifically the anvil pits, during nut cracking? In the present study, we examined (1) whether capuchins` preferences for particular pits are influenced by the effectiveness of the pit in cracking the nut and/or by the stability of the nut during striking, (2) how capuchins detect the affordances of novel pits and (3) the influence of social context on their selections. Anvil pits varied in horizontal dimension (small, medium and large) in experiment 1 and in depth (shallow, medium and deep) in experiment 2. In both experiments, three different pits were simultaneously presented, each on one anvil. We coded the capuchins` actions with the nut in each pit, and recorded the outcome of each strike. In both experiments, capuchins preferred the most effective pit, but not the most stabilizing pit, based on the number of first strikes, total strikes and nuts cracked. Their choice also reflected where the preceding individual had last struck. The capuchins explored the pits indirectly, placing nuts in them and striking nuts with a stone. The preference for pits was weaker than the preference for nuts and stones shown previously with the same monkeys. Our findings suggest that detecting affordances of pits through indirect action is less precise than through direct action, and that social context may also influence selection. We show that field experiments can demonstrate embodied cognition in species-typical activities in natural environments. (C) 2010 The Association for the Study of Animal Behaviour. Modern sugarcane cultivars are complex hybrids resulting from crosses among several Saccharum species. Traditional breeding methods have been employed extensively in different countries over the past decades to develop varieties with increased sucrose yield and resistance to pests and diseases. Conventional variety improvement, however, may be limited by the narrow pool of suitable genes. Thus, molecular genetics is seen as a promising tool to assist in the process of developing improved varieties. The SUCEST-FUN Project (http://sucest-fun.org) aims to associate function with sugarcane genes using a variety of tools, in particular those that enable the study of the sugarcane transcriptome. An extensive analysis has been conducted to characterise, phenotypically, sugarcane genotypes with regard to their sucrose content, biomass and drought responses. Through the analysis of different cultivars, genes associated with sucrose content, yield, lignin and drought have been identified. Currently, tools are being developed to determine signalling and regulatory networks in grasses, and to sequence the sugarcane genome, as well as to identify sugarcane promoters. This is being implemented through the SUCEST-FUN (http://sucest-fun.org) and GRASSIUS databases (http://grassius.org), the cloning of sugarcane promoters, the identification of cis-regulatory elements (CRE) using Chromatin Immunoprecipitation-sequencing (ChIP-Seq) and the generation of a comprehensive Signal Transduction and Transcription gene catalogue (SUCAST Catalogue).

Bacteria activate a regulatory network in response to the challenges imposed by DNA damage to genetic material, known as the SOS response. This system is regulated by the RecA recombinase and by the transcriptional repressor lexA. Leptospira interrogans is a pathogen capable of surviving in the environment for weeks, being exposed to a great variety of stress agents and yet retaining its ability to infect the host. This study aims to investigate the behavior of L. interrogans serovar Copenhageni after the stress induced by DNA damage. We show that L. interrogans serovar Copenhageni genome contains two genes encoding putative LexA proteins (lexA1 and lexA2) one of them being potentially acquired by lateral gene transfer. Both genes are induced after DNA damage, but the steady state levels of both LexA proteins drop, probably due to auto-proteolytic activity triggered in this condition. In addition, seven other genes were up-regulated following UV-C irradiation, recA, recN, dinP, and four genes encoding hypothetical proteins. This set of genes is potentially regulated by LexA1, as it showed binding to their promoter regions. All these regions contain degenerated sequences in relation to the previously described SOS box, TTTGN 5CAAA. On the other hand, LexA2 was able to bind to the palindrome TTGTAN 10TACAA, found in its own promoter region, but not in the others. Therefore, the L. interrogans serovar Copenhageni SOS regulon may be even more complex, as a result of LexA1 and LexA2 binding to divergent motifs. New possibilities for DNA damage response in Leptospira are expected, with potential influence in other biological responses such as virulence

MMP-2 and MMP-14 process extracellular matrix proteins,cytokines, growth factors and adhesion molecules to generatefragments with enhanced or reduced biological activity.In this study, a vectorsystem was developed for theconditional expression of MMP-2 and MMP-14 in the liver oftransgenic mice. For this vectorsystem the murine albuminpromotor was chosen together with the cre/lox system toachieve an inducible MMP-expression in the liver.Only one of the MMP-14 transgenic lines expressed highamounts of active MMP-14 protein after recombination of thelox-P sites. In these mice MMP-14 was able to activate MMP-2and MMP-13 in vivo. However, none of the livers of MMP-14overexpressing mice showed no differences in liverweight,amount of extracellular matrixproteins and rate ofproliferation, apoptosis and tumor-induction when comparedto the liver of wildtype mice.On the other hand overexpression of MMP-2 was embryoniclethal in all MMP-2 transgenic lines. After crossing theMMP-2 transgenic mice with cre deleter mice, a cre mediatedrecombination could be shown at day 6.5 post coitum (pc).Some of the double transgenic embryos of one of thetransgenic lines had severe deformations of the head,especially of the telencephalon and the mesencephalon.It could be shown in this study that disregulation of MMP-2in early embryonic development is lethal but anoverexpression of MMP-14 has no influence on the embryonicdevelopment or the homeostasis of the adult liver.With this conditional vectorsystem it is to possible studythe influnce of MMP-2 and MMP-14 on fibrogenesis,regeneration and tumorgenesis in the liver of mice.

Nucleic acid biosensors represent a powerful tool for clinical and environmental pathogens detection. For applications such as point-of-care biosensing, it is fundamental to develop sensors that should be automatic, inexpensive, portable and require a professional skill of the user that should be as low as possible. With the goal of determining the presence of pathogens when present in very small amount, such as for the screening of pathogens in drinking water, an amplification step must be implemented. Often this type of determinations should be performed with simple, automatic and inexpensive hardware: the use of a chemical (or nanotechnological) isothermal solution would be desirable. My Ph.D. project focused on the study and on the testing of four isothermal reactions which can be used to amplify the nucleic acid analyte before the binding event on the surface sensor or to amplify the signal after that the hybridization event with the probe. Recombinase polymerase amplification (RPA) and ligation-mediated rolling circle amplification (L-RCA) were investigated as methods for DNA and RNA amplification. Hybridization chain reaction (HCR) and Terminal deoxynucleotidil transferase-mediated amplification were investigated as strategies to achieve the enhancement of the signal after the surface hybridization event between target and probe. In conclusion, it can be said that only a small subset of the biochemical strategies that are proved to work in solution towards the amplification of nucleic acids does truly work in the context of amplifying the signal of a detection system for pathogens. Amongst those tested during my Ph.D. activity, recombinase polymerase amplification seems the best candidate for a useful implementation in diagnostic or environmental applications.

RNAi (RNA interference) is a powerful technology for sequence-specific targeting of mRNAs. This thesis was aimed at establishing conditions for conditional RNAi-mediated silencing first in vitro and subsequently also in transgenic mice. As a target the basic helix-loop-helix transcription factor encoding gene SCL (stem cell leukaemia also known as Tal-1 or TCL5) was used. SCL is a key regulator for haematopoietic development and ectopic expression of SCL is correlated with acute T-lymphoblastic leukaemias. Loss of SCL function studies demonstrated that ab initio deletion of SCL resulted in embryonic lethality around day E9 in gestation. To be able to conditionally inactivate SCL, RNAi technology was combined with the tetracycline-dependent regulatory system. This strategy allowed to exogenously control the induction of RNAi in a reversible fashion and consequently the generation of a completely switchable RNAi knockdown. First a suitable vector allowing for co-expression of tetracycline-controlled shRNAs (small hairpin RNAs) and constitutively active EGFP (enhanced green fluorescent protein) was generated. This novel vector, pRNAi-EGFP, was then evaluated for EGFP expression and tetracycline-mediated expression of shRNAs. Four sequences targeting different regions within the SCL mRNA were tested for their efficiency to specifically knockdown SCL. These experiments were performed in M1 murine leukaemia cells and subsequently in the HEK 293 cell line, expressing an engineered HA-tagged SCL protein. The second assay provided a solid experimental method for determining the efficiency of different SCL-siRNA knockdown constructs in tissue culture. Western blotting analyses revealed a down regulation of SCL protein for all four tested SCL-specific target sequences albeit with different knockdown efficiencies (between 25% and 100%). Furthermore, stringent tetracycline-dependent switchability of shRNA expression was confirmed by co-transfecting the SCL-specific pRNAi-EGFP vector (SCL-siRNA) together with the HA-tagged SCL expression plasmid into the HEK 293TR /T-REx cell line constitutively expressing the tetracycline repressor (TetR). These series of experiments demonstrated tight regulation of siRNA expression without background activity. To be able to control the SCL knockdown in vivo and especially to circumvent any possible embryonic lethality a transgenic mouse line with general expression of a tetracycline repressor was needed. Two alternative methods were used to generate TetR mice. The first approach was to co-inject the tetracycline-regulated RNAi vector together with a commercially available and here specifically modified T-REx expression vector (SCL-siRNA T-REx FRT LoxP mouse line). The second method involved the generation of a TetR expressor mouse line, which was then used for donating TetR-positive oocytes for pronuclear injection of the RNAi vector (SCL-siRNA T-REx mouse line). As expected, and in agreement with data from conditional Cre-controlled adult SCL knockout mice, post-transcriptional silencing of SCL by RNAi caused a shift in the maturation of red blood cell populations. This was shown in the bone marrow and peripheral blood by FACS analysis with the red blood cell-specific TER119 and CD71 markers which can be used to define erythrocyte differentiation (Lodish plot technique). In conclusion this study established conditions for effective SCL RNAi-mediated silencing in vitro and in vivo providing an important tool for further investigations into the role of SCL and, more generally, of its in vivo function in haematopoiesis and leukaemia. Most importantly, the here acquired knowledge will now allow the establishment of other completely conditional and reversible knockdown phenotypes in mice.

A range of vectors were made in which the EYFP gene or the Cre gene were inserted in the start codon of the NG2 gene. The NG2-EYFP vectors were used to generate NG2-EYFP “knockin” mice by homologous recombination. The F1 generation showed lack of EYFP expression, due to NeoR cassette interference. Excision of the NeoR, by breeding the F1 generation to ELLA-Cre mice allowed proper expression of EYFP. NG2-EYFP heterozygous mice were characterized in detail for astrocytic, neurogenic and oligodendrocytic properties through antibody labeling. NG2-EYFP+ cells did not label for the astrocyte marker GFAP, but some cells did express S100 Beta. The cells did not label with any neuronal markers like Beta III tubulin, Neun, and double cortin, but many of the NG2-EYFP+ cells made intimate contacts to the neurons. These contacts are widespread throughout the grey and white matter of the brain. The NG2-EYFP+ cells did label for oligodendrocyte markers like PDGFα-R, NG2, Olig2, O4, and Sox 10. There were a few cells termed phantom cells that did label for NG2, but had no EYFP expression. This could have been caused by improper excision of the NeoR cassette in the F2 generation. The heterozygous mouse is a tool to allow the characterization of the in vivo properties of the NG2+ cells. Breeding of these mice to homozygosity yielded an NG2-knockout mouse, which was also subjected to initial characterization. The NG2-EYFP homozygous showed equivalent cell labeling results to the NG2-EYFP heterozygous mouse, but the phantom cells disappeared in the knockout. The results show that the NG2 cells are a heterogenous population that does not express astrocytic or neuronal markers. The homozygous mouse is an ideal tool to further dissect the properties of the cells, lacking NG2 in vivo.

Diese Arbeit befasst sich mit der Rolle des Fibronektins im Knochen sowie in der diabetischen Nephropathie. Fibronektin im Knochen: Es war bekannt, dass Osteoblasten für ihre Differenzierung in vitro Fibronektin benötigen, dass Fibronektin für die Ausbildung einer Kollagenmatrix erforderlich ist und für die Matrixintegrität eine kontinuierliche Fibronektin-Versorgung gewährleistet sein muss. Um die Rolle des Fibronektins im Knochen, dessen Matrix zu 90% aus Kollagen besteht, näher zu untersuchen, wurde das Fibronektin der Osteoblasten spezifisch über das Cre/loxP-System in Mäusen ausgeschaltet. Dies führte zu einer erhöhten Anzahl an Osteoblasten, deren Fähigkeit die Matrix zu mineralisieren jedoch beeinträchtigt war. Dennoch zeigte sich kein Einfluss auf die Eigenschaften der Knochenmatrix. Insbesondere war der Fibronektingehalt nicht vermindert, entgegen der allgemeinen Annahme, dass die Osteoblasten die Produzenten des Fibronektins der Knochenmatrix seien. Im Gegensatz dazu stellte sich durch Untersuchungen an anderen genetisch veränderten Mäusen heraus, dass eine Ausschaltung des Plasmafibronektins im Blut zu einer deutlichen Verringerung des Fibronektingehalts des Knochens sowie zu einer Verminderung des Mineralgehalts bezogen auf die Proteinmenge führte. Auch die Komposition des Minerals war verändert. Da es jedoch keinen nennenswerten Effekt auf die Knochenzellen gab, lässt sich schlussfolgern, dass die Osteoblasten-spezifische Fibronektin-Isoform für eine regelgerechte Funktion der Osteoblasten notwendig ist, während das von der Leber produzierte Plasmafibronektin die Zusammensetzung der Knochenmatrix beeinflusst. Fibronektin in der diabetischen Niere: Mit der diabetischen Nephropathie geht eine Ausdehnung des Mesangiums in den Glomeruli einher, die mit dem Ausmaß des Nierenschadens korreliert ist. Fibronektin ist ein Bestandteil dieses expandierten Mesangiums. Vorarbeiten hatten gezeigt, dass injiziertes Fibronektin durch die Blutzirkulation in die Niere gelangt und in der Mesangialmatrix der Glomeruli eingelagert wird. Daher wurden in konditionellen Knockout-Mäusen das Plasmafibronektin bzw. das Fibronektin der Mesangialzellen und das Plasmafibronektin zugleich ausgeschaltet. In diesen Mäusen wurde ein Diabetes mellitus induziert und die Tiere für 22 Wochen mit Diabetes gehalten. Die Ausschaltung des Fibronektins hatte eine geringere Ausbreitung der Mesangialmatrix sowie eine geringere Mortalität der Tiere zur Folge. Interessanterweise schien das Plasmafibronektin alleine bereits grob ein Drittel der Ausdehnung des Mesangiums zu verursachen. Die kombinierte Ausschaltung von zirkulierendem und lokalem Fibronektin vermochte die Expansion der Mesangialmatrix sogar beinahe zu halbieren. Zusammengefasst zeigten sich neue Rollen eines traditionellen Proteins der Extrazellulärmatrix in physiologischen und pathologischen Zuständen. Einige dieser Aspekte demonstrieren die große Bedeutung der Fibronektin-Produktion durch die Leber.

Elf3 gehört zur Familie der Ets-Transkriptionsfaktoren und wird unter nicht-entzündlichen Bedingungen ausschließlich in epithelialen Zellen exprimiert, vor allem in den Enterozyten des gastrointestinalen Traktes. Um die Rolle des Transkriptionsfaktors Elf3 in Hinblick auf potentielle Zielgene und Einflüsse auf die Darm-Morphologie zu untersuchen, wurde ein Vektorsystem für die konditionelle Expression eines dominant-negativen Elf3 (dnElf3) in Darmepithelzellen generiert. Regulatorische Elemente des humanen Keratin 20 Gens in Kombination mit dem Cre/loxP-System ermöglichten eine induzierbare, darmepithel-spezifische Expression in transgenen Mäusen. Die Expression von dnElf3 führt zu einem signifikanten Gewichtsverlust und deutlichen morphologischen Veränderungen des Darmepithels. Im Dünndarm konnte ein erhöhte Anzahl von Becherzellen und eine verstärkte Mukusproduktion nachgewiesen werden. Sowohl die Keratin 8 Expression, als auch die Expression des Zellmembranproteins Claudin 7 waren signifikant herab reguliert. Im Rahmen dieser Arbeit konnte erstmals eine Regulation der Claudin 7 Expression durch Elf3 im Darm gezeigt werden.
