Improved methods for the detection of Histoplasma capsulatum are needed in regions with limited resources in which the organism is endemic, where delayed diagnosis of progressive disseminated histoplasmosis (PDH) results in high mortality rates. We have investigated the use of a loop-mediated isothermal amplification (LAMP) assay to facilitate rapid inexpensive molecular diagnosis of this disease. Primers for LAMP were designed to amplify the Hcp100 locus of H. capsulatum. The sensitivity and limit of detection were evaluated using DNA extracted from 91 clinical isolates of known geographic subspecies, while the assay specificity was determined using DNA extracted from 50 other fungi and Mycobacterium tuberculosis. Urine specimens (n = 6) collected from HIV-positive individuals with culture- and antigen-proven histoplasmosis were evaluated using the LAMP assay. Specimens from healthy persons (n = 10) without evidence of histoplasmosis were used as assay controls. The Hcp100 LAMP assay was 100% sensitive and specific when tested with DNA extracted from culture isolates. The median limit of detection was <= 6 genomes (range, 1 to 300 genomes) for all except one geographic subspecies. The LAMP assay detected Hcp100 in 67% of antigen-positive urine specimens (4/6 specimens), and results were negative for Hcp100 in all healthy control urine specimens. We have shown that the Hcp100 LAMP assay is a rapid affordable assay that can be used to expedite culture confirmation of H. capsulatum in regions in which PDH is endemic. Further, our results indicate proof of the concept that the assay can be used to detect Histoplasma DNA in urine. Further evaluation of this assay using body fluid samples from a larger patient population is warranted.

There is little research on the practices of sanitary inspection in the chain of production of vegetables to the consumer, especially those eaten raw, they are liable to serve microorganism such as bacteria, fungi and parasites, contributing to possible health hazards. The aim of this study was to assess qualitatively contamination by parasites and / or commensals of medical interest in lettuce leaves (Lactuca sativa) fresh market in the municipality of Quata Sao Paulo. A total of 15 random samples were analyzed every other day of the three different places that sell vegetables a grocery store, a supermarket and a vegetable garden during the month of May 2011. The parasites and / or commensals found in lettuce were Entamoeba coli (67%), Entamoeba histolytica (20%), Giardia sp (13%) and Ascaris lumbricoides (7%). The analysis showed the presence of parasites and / or commensals in all samples, except in the cultivated garden which showed poor sanitary conditions, probably due to contamination in the shipping and handling by third parties in supermarket and grocery store. The parasite monitoring sanitary conditions of vegetables sold in urban environments becomes relevant for preventive measures to avoid the continued parasitic cycle and possible future health complications.

The intervertebral disc disease is considered a major neurological disorder involving the spinal cord, being a disease commonly seen in dogs in clinical practice of small animals. The pathophysiology of most discopathies have not been fully elucidated, but it is known that the symptoms observed will depend on the severity of spinal cord compression and location of spinal injury, ranging from localized pain to complete paralysis. Through a detailed clinical examination and laboratory tests, we choose the most appropriate treatment for each patient, which can be conservative, through the use of anti-inflammatories, analgesics and acupuncture or surgical, through decompression techniques. Acupuncture, therapeutic method of ancient Chinese medicine, uses the stimulation of specific points on the body in order to balance the body's physiological. The action mechanism of acupuncture in discopathies has not been fully elucidated, but its analgesic effects, anti-inflammatory and restoration of sensory and motor function are known, showing results as good as surgical therapy

The present study is about data characterization and evaluation related to private urban users (legal entity) of the Watersheds of the rivers Piracicaba, Capivari, and Jundiai (PCJ), with the PCJ Collection System, sustained by the Department of Water and Power (DAEE), in order to: provide quantitative numbers about this sector, identifying the cities and economic activities corresponding to the largest water consumers, and determine the sector's share in the total charged contribution. The charge for water use is the most recent and the last instrument which has been implemented for the management of water resources, provided by the institution of the State and National Policy of Water Resources in 1991 and 1997, respectively, regarded as an important step towards the preservation and restoration of water resources. According to the data collected in the PCJ Collection System, the urban sector is the private sector that has the highest number of users in these watersheds (52,95% of users), but with less representation in the financial recovery (only 7%), due to its low water demand in their uses compared to the uses of other sectors. The collected data will also serve as both a parameter for comparing the amount of water used by different economic activities and municipalities in the PCJ watersheds and other locations, and as a tool for water resources management

The Atlantic Forest biome, reduced to 12% of its original coverage, holds a significant portion of brazilian biodiversity, with high levels of endemism. Among the few remaining areas of the Atlantic, there is the region between the Complex of Cantareira and the municipality of Nazaré Paulista, which will be the object of study of this work. Using the concepts of Landscape Ecology, and analysis of thematic maps, this study aims to characterize the region in a ecologically scaled perspective, identifying two types of hotspots: i) for conservation and ii) for maintain habitat connectivity. Concepts of Landscape Ecology as landscape structure, patches, matrix, corridors (structural and functional) and connectivity, as well as their effects on local biodiversity was adopted and applied in the study region. We also analyzed the effects of roads, hydrographic system, Permanent Protection Areas (PPAs), and Conservation Units for flora and fauna maintenance at regional scale. This allowed us to define strategies and priority areas for the conservation and restoration of forest fragments of Atlantic Forest remnants within the study region. This study generated relevant knowledge for a better planning the region in order to create best conditions for the maintenance of regional biodiversity, and consequently, allowing to improve the quality of life for local population

The knowledge of the dynamics of soil seed bank and seed rain is fundamental to understand the forest succession process, as well as for its conservation and restoration. This paper aimed at studying the role of the soil seed bank and seed rain in the dynamics of a riparian tropical seasonal forest fragment located on the hinterland of São Paulo State. The seed rain was studied by 30 litter traps installed at 50 cm from the floor, with an area of 50 cm x 50 cm. Between November of 2008 and October of 2009, 11364 seeds of 82 species belonging to 33 different families were collected. The deposition density of seeds was 126.27 individuals.m². With the data obtained from this research, the two seasons couldn’t be differed from each other hence 2009 was an atypical year considering the precipitation, which was constantly elevated. For the soil seed bank, fifty superficial soil samples will be taken at the end of both the rainy and dry seasons. The germination method was applied for the data analysis. It was observed a predominance of herbaceous habit species, with an average of 71% of the germinations in both seasons. The Sφrensen similarity index between the seasons was low (0,27). The density and species diversity were higher after the rainy season, unlike other published researches

The bee Apis mellifera has a great importance because it is the most economically valuable pollinator for crops worldwide, ensuring cross-pollination and increasing fruit yield. Moreover, agriculture increasingly develops chemicals to control weeds, fungi and insect pests to ensure productivity. Insecticides are used on a large scale in the state of São Paulo, in cultures of citrus for control of greening. Applications are usually made by aircraft and as a result of the effect derives a significant mortality is observed in apiaries near the plantations. Honey bees can get in contact with such chemical agent through their activities of water harvesting, plant resins, pollen and nectar. Intoxication resulting from this exposure can be lethal, which is easily detectable, or cause effects on the physiology and behavior of the insect. These, in turn are hardly detectable, such as paralysis, disorientation, behavioral changes, but can compromise the entire social structure of the colony, therefore aimed to study the effects of the insecticide Thiamethoxam behavior of honeybees A. mellifera. Newly emerged individuals and with 10 days of age were tested. Applications of 1 μL de Thiamethoxam, diluted in acetone, were made on the dorsal thorax with a microapplicator. Preliminarily, it was observed LD50 twenty four hours after topical treatment of Thiamethoxam. It was found that the LD50 for newly emerged honeybees is 8 ng/bee and for honeybees with 10 days of age is 18 ng/bee. The behaviors were analyzed 1 hour after application of insecticide at doses corresponding to LD50/100, LD50/50, LD50/10 and LD50, besides the control group. In the test reflex proboscis extension, there was impairment of behavior at doses of 8 and 18 ng/bee workers in newly emerged workers and 10 days of age, respectively. And in locomotor behavior was no change only at a dose of 18 ng/bee workers at 10 days of age... (Complete abstract click electronic access below)

The oil and biodiesel are oily substances which, if spilled or thrown out in the environment, can cause serious damages. In this context, bioremediation techniques can be used to the recovering of sites degradated by mineral or vegetable oil, besides them, the biodegradation. However, it is necessary that there are microorganism strains used to degradate the contaminant, previously selected for the fast, efficient and minimum impact remediation. The main objective of this study was to isolate microorganism strains coming from different ecological recesses and verify their potentiality on biodegradating oil and biodiesel. Strains of bacteria, fungi and yeasts were isolated from soils contaminated by hydrocarbons (diesel, gasoline, grease) before, by doing striations on culture plates. On the biodegradation tests, it was used the redox indicator, 2,6-dichlorophenol indophenol (DCPIP), then, after the indicador’s discoloration, the absorbance of the suspension obtained was measured. 46 different microorganism strains were isolated, and the oil, when it was used as a carbon resource, it was degradate easier than the biodiesel. The experiments showed that, in a period, the site impacted by oil spills recovers itself by the biodegradation, in aerobic conditions, by the redox activity.

Habitat loss and fragmentation of landscapes endanger the planet’s biodiversity. Strategies for identify priority areas for conservation and restoration of biodiversity rich areas becomes essential for the planning of the management of these landscape become successful. This study aims to propose a novel, transparent and replicable method for choosing priority areas for restoration, and includes the following steps: (a) identification of regional biodiversity hotspots for conservation; (b) identification of priority areas for restoration with the greatest potential to increase the connectivity of the fragments already existing; (c) estimate the potential richness of understory birds before and after restoration, analyzing the gain of species for the future scenario. In order to identify the corridors to be restored in a future scenario we considered the approach of multiple corridors, which aimed to connect the main fragments within the region through analysis of multi-paths. Already existing regression models were applied to estimate the richness of the landscape considering three models: a) species richness as a function of patch area of the fragment selected as hotspots; b) richness as a function of areas connected by structural corridors and c) connected area for species which are able to access nearby fragments within 20m. The gain of species for future scenario which consider the potential restoration of selected areas was estimated. Based on our results we observed that species that use corridors showed the highest increment of species richness of understory birds. As a result it had to restore corridors to model species with the ability to use forest corridors was the highest gain potential species richness of understory birds.
