The implementation of various types of marine protected areas is one of several management tools available for conserving representative examples of the biological diversity within marine ecosystems in general and National Marine Sanctuaries in particular. However, deciding where and how many sites to establish within a given area is frequently hampered by incomplete knowledge of the distribution of organisms and an understanding of the potential tradeoffs that would allow planners to address frequently competing interests in an objective manner. Fortunately, this is beginning to change. Recent studies on the continental shelf of the northeastern United States suggest that substrate and water mass characteristics are highly correlated with the composition of benthic communities and may therefore, serve as proxies for the distribution of biological biodiversity. A detailed geo-referenced interpretative map of major sediment types within Stellwagen Bank National Marine Sanctuary (SBNMS) has recently been developed, and computer-aided decision support tools have reached new levels of sophistication. We demonstrate the use of simulated annealing, a type of mathematical optimization, to identify suites of potential conservation sites within SBNMS that equally represent 1) all major sediment types and 2) derived habitat types based on both sediment and depth in the smallest amount of space. The Sanctuary was divided into 3610 0.5 min2 sampling units. Simulations incorporated constraints on the physical dispersion of sampling units to varying degrees such that solutions included between one and four site clusters. Target representation goals were set at 5, 10, 15, 20, and 25 percent of each sediment type, and 10 and 20 percent of each habitat type. Simulations consisted of 100 runs, from which we identified the best solution (i.e., smallest total area) and four nearoptimal alternates. We also plotted total instances in which each sampling unit occurred in solution sets of the 100 runs as a means of gauging the variety of spatial configurations available under each scenario. Results suggested that the total combined area needed to represent each of the sediment types in equal proportions was equal to the percent representation level sought. Slightly larger areas were required to represent all habitat types at the same representation levels. Total boundary length increased in direct proportion to the number of sites at all levels of representation for simulations involving sediment and habitat classes, but increased more rapidly with number of sites at higher representation levels. There were a large number of alternate spatial configurations at all representation levels, although generally fewer among one and two versus three- and four-site solutions. These differences were less pronounced among simulations targeting habitat representation, suggesting that a similar degree of flexibility is inherent in the spatial arrangement of potential protected area systems containing one versus several sites for similar levels of habitat representation. We attribute these results to the distribution of sediment and depth zones within the Sanctuary, and to the fact that even levels of representation were sought in each scenario. The present study was designed to examine the following: (1) the taxonomic. spatial, and temporal patterns of availability of all invertebrate species associated with Macrocystis (excluding protozoans and nematodes); (2) the utilization of this invertebrate assemblage as food by kelp forest fishes within the Macrocystis "foliage- searching" feeding guild, as well as proximal mechanisms leading to observed patterns of resource partitioning; and (3) the dynamic relationship between availability and utilization of this food resource. The approach was largely descriptive. with observations collected during a 19-month period from June 1975 to December 1976. Chapter I is an investigation of the resource utilization patterns of four species of kelp forest fishes with respect to food-related resource dimensions. and tests aspects of current theory involving inter- and intraspecific competition. Chapter II is a detailed examination of the invertebrate assemblage associated with Macrocystis and presents life histories of the fishes examined during this study. Species composition, biomass, density, and diversity of benthic invertebrates from six bard-bottom areas were evaluated. Seasonal collections using a dredge, trawl, and suction and grab samplers yielded 432, 525, and 845 taxa, respectively. Based on collections wltb the different gear types, species composition of invertebrates was found to change bathymetrically. Inner- and mlddle-shelf sites were more similar to each other in terms of invertebrate species composition than they were to outer-shelf sites, regardless of season. Sites on the inner and outer shelf were grouped according to latitude; however, results suggest that depth is apparently a more important determinant of invertebrate species composition than either season or latitude. Sponges generally dominated dredge and trawl collections in terms of biomass. Generally, cnidarians, bryozoans, and sponges dominated at sites In terms of number of taxa collected. The most abundant smaller macrofauna collected in suction and grab samples were polychaetes, amphipods, and mollusks. Densities of the numerically dominant species changed botb seasonally and bathymetrically, with very few of these species restricted to a specific bathymetrlc zone. The high diversity of invertebrates from hard-bottom sites is attributed to the large number of rare species. No consistent seasonal changes in diversity or number of species were noted for individual stations or depth zones. In addition, H and its components showed no definite patterns related to depth or latitude. However, more species were collected at middle-shelf sites than at inner- or outer-shelf sites, which may be related to more stable bottom temperature or greater habitat complexity in that area. Background: Colorectal cancer (CRC) is a disease of complex aetiology, with much of the expected inherited risk being due to several common low risk variants. Genome-Wide Association Studies (GWAS) have identified 20 CRC risk variants. Nevertheless, these have only been able to explain part of the missing heritability. Moreover, these signals have only been inspected in populations of Northern European origin. Results: Thus, we followed the same approach in a Spanish cohort of 881 cases and 667 controls. Sixty-four variants at 24 loci were found to be associated with CRC at p-values <10-5. We therefore evaluated the 24 loci in another Spanish replication cohort (1481 cases and 1850 controls). Two of these SNPs, rs12080929 at 1p33 (P-replication=0.042; P-pooled=5.523x10(-03); OR (CI95%)=0.866(0.782-0.959)) and rs11987193 at 8p12 (P-replication=0.039; P-pooled=6.985x10(-5); OR (CI95%)=0.786(0.705-0.878)) were replicated in the second Phase, although they did not reach genome-wide statistical significance. Conclusions: We have performed the first CRC GWAS in a Southern European population and by these means we were able to identify two new susceptibility variants at 1p33 and 8p12 loci. These two SNPs are located near the SLC5A9 and DUSP4 loci, respectively, which could be good functional candidates for the association signals. We therefore believe that these two markers constitute good candidates for CRC susceptibility loci and should be further evaluated in other larger datasets. Moreover, we highlight that were these two SNPs true susceptibility variants, they would constitute a decrease in the CRC missing heritability fraction.

Background: Poor outcomes of invasive candidiasis (IC) are associated with the difficulty in establishing the microbiological diagnosis at an early stage. New scores and laboratory tests have been developed in order to make an early therapeutic intervention in an attempt to reduce the high mortality associated with invasive fungal infections. Candida albicans IFA IgG has been recently commercialized for germ tube antibody detection (CAGTA). This test provides a rapid and simple diagnosis of IC (84.4% sensitivity and 94.7% specificity). The aim of this study is to identify the patients who could be benefited by the use of CAGTA test in critical care setting. Methods: A prospective, cohort, observational multicentre study was carried out in six medical/surgical Intensive care units (ICU) of tertiary-care Spanish hospitals. Candida albicans Germ Tube Antibody test was performed twice a week if predetermined risk factors were present, and serologically demonstrated candidiasis was considered if the testing serum dilution was >= 1: 160 in at least one sample and no other microbiological evidence of invasive candidiasis was found. Results: Fifty-three critically ill non-neutropenic patients (37.7% post surgery) were included. Twenty-two patients (41.5%) had CAGTA-positive results, none of them with positive blood culture for Candida. Neither corrected colonization index nor antifungal treatment had influence on CAGTA results. This finding could corroborate that the CAGTA may be an important biomarker to distinguish between colonization and infection in these patients. The presence of acute renal failure at the beginning of the study was more frequent in CAGTA-negative patients. Previous surgery was statistically more frequent in CAGTA-positive patients. Conclusions: This study identified previous surgery as the principal clinical factor associated with CAGTA-positive results and emphasises the utility of this promising technique, which was not influenced by high Candida colonization or antifungal treatment. Our results suggest that detection of CAGTA may be important for the diagnosis of invasive candidiasis in surgical patients admitted in ICU.

Background: The aim of this study is to examine the influence of the catechol-O-methyltranferase (COMT) gene (polymorphism Val158 Met) as a risk factor for Alzheimer's disease (AD) and mild cognitive impairment of amnesic type (MCI), and its synergistic effect with the apolipoprotein E gene (APOE). A total of 223 MCI patients, 345 AD and 253 healthy controls were analyzed. Clinical criteria and neuropsychological tests were used to establish diagnostic groups. The DNA Bank of the University of the Basque Country (UPV-EHU) (Spain) determined COMT Val158 Met and APOE genotypes using real time polymerase chain reaction (rtPCR) and polymerase chain reaction (PCR), and restriction fragment length polymorphism (RFLPs), respectively. Multinomial logistic regression models were used to determine the risk of AD and MCI. Results: Neither COMT alleles nor genotypes were independent risk factors for AD or MCI. The high activity genotypes (GG and AG) showed a synergistic effect with APOE epsilon 4 allele, increasing the risk of AD (OR = 5.96, 95% CI 2.74-12.94, p < 0.001 and OR = 6.71, 95% CI 3.36-13.41, p < 0.001 respectivily). In AD patients this effect was greater in women. In MCI patients such as synergistic effect was only found between AG and APOE epsilon 4 allele (OR = 3.21 95% CI 1.56-6.63, p = 0.02) and was greater in men (OR = 5.88 95% CI 1.69-20.42, p < 0.01). Conclusion: COMT (Val158 Met) polymorphism is not an independent risk factor for AD or MCI, but shows a synergistic effect with APOE epsilon 4 allele that proves greater in women with AD.

Background: The presence of EGFR kinase domain mutations in a subset of NSCLC patients correlates with the response to treatment with the EGFR tyrosine kinase inhibitors gefitinib and erlotinib. Although most EGFR mutations detected are short deletions in exon 19 or the L858R point mutation in exon 21, more than 75 different EGFR kinase domain residues have been reported to be altered in NSCLC patients. The phenotypical consequences of different EGFR mutations may vary dramatically, but the majority of uncommon EGFR mutations have never been functionally evaluated. Results: We demonstrate that the relative kinase activity and erlotinib sensitivity of different EGFR mutants can be readily evaluated using transfection of an YFP-tagged fragment of the EGFR intracellular domain (YFP-EGFR-ICD), followed by immunofluorescence microscopy analysis. Using this assay, we show that the exon 20 insertions Ins770SVD and Ins774HV confer increased kinase activity, but no erlotinib sensitivity. We also show that, in contrast to the common L858R mutation, the uncommon exon 21 point mutations P848L and A859T appear to behave like functionally silent polymorphisms. Conclusion: The ability to rapidly obtain functional information on EGFR variants of unknown relevance using the YFP-EGFR-ICD assay might prove important in the future for the management of NSCLC patients bearing uncommon EGFR mutations. In addition, our assay may be used to determine the response of resistant EGFR mutants to novel second-generation TKIs.

ENGLISH: Annual estimates of the number of purse-seine sets made on tunas associated with dolphins are needed to estimate the total number of dolphins killed incidentally by the eastern Pacific tuna fishery. The most complete source of data, the Inter-American Tropical Tuna Commission's logbook data base, was used in this study. In the logbook data base, most sets are identified as being either associated with dolphins or not associated with dolphins. Some sets are not identified in this respect. However, the number of these unidentified sets which were associated with dolphins have been estimated by stratifying the logbook data according to whether or not any tuna were caught, whether or not the nearest identified set was associated with dolphins, and the distance to the nearest identified set. Most of the unidentified sets fell in strata characterized by a proportion of sets on tuna associated with dolphins that was lower than the overall unstratified proportion. Landings data were used to estimate the number of sets on tunas associated with dolphins from fishing trips not included in the logbook data base. Fish cage culture is a rapid aquacultural practice of producing fish with more yield compared to traditional pond culture. Several species cultured by this method include Cyprinus carpio, Orechromis niloticus, Sarotherodon galilaeus, Tilapia zilli, Clarias lazera, C. gariepinus, Heterobranchus bidorsalis, Citharinus citharus, Distochodus rostratus and Alestes dentes. However, the culture of fish in cages has some problems that are due to mechanical defects of the cage or diseases due to infection. The mechanical problems which may lead to clogged net, toxicity and easy access by predators depend on defects associated with various types of nets which include fold sieve cloth net, wire net, polypropylene net, nylon, galvanized and welded net. The diseases problems are of two types namely introduced diseases due to parasites. The introduced parasites include Crustaseans, Ergasilus sp. Argulus africana, and Lamprolegna sp, Helminth, Diplostomulum tregnna: Protozoan, Trichodina sp, Myxosoma sp, Myxobolus sp. the second disease problems are inherent diseases aggravated by the very rich nutrient environment in cages for rapid bacterial, saprophytic fungi, and phytoplanktonic bloom resulting in clogging of net, stagnation of water and low biological oxygen demand (BOD). The consequence is fish kill, prevalence of gill rot and dropsy conditions. Recommendations on routine cage hygiene, diagnosis and control procedures to reduce fish mortality are highlighted

The distinguished character of Particularly Sensitive Sea Areas (PSSAs) is that every application for PSSAs must be accompanied by Associated Protected Measures (APMs) which can make PSSAs efficient in practice.1 That is why APMs are regarded as the core feature of every PSSA.2 APM is “an international rule or standard that falls within the purview of an international maritime organization (IMO) and regulates international maritime activities for the protection of the area at risk.” So far, APMs have been approved by IMO as following: -Compulsory or recommended pilotage -Mandatory ship reporting -An area to be avoided -Traffic separation schemes -Discharge prohibition or regulations -Mandatory no anchoring areas -Deep water routes -Emission control areas (PDF contains 5 pages)

The paper highlights the concept of information and the significance of environmental and occupational hazards associated with pond fish production in Nigeria and discuss the possible options for the ways forward. The major raw material used in fish production system is the organic manure (cow dung, poultry droppings, porcine manure etc) that serves as substrate for heterotrophic production of bacteria and protozoa, which act as food for zooplankton and the fish. The pathogenic organisms (viruses, bacteria, protozoa's, and parasites), are noted for the potential hazard to the fish handlers and consumers. Nine species from seven genera of bacteria associated with fish diseases are found to have association with diseases of human such as typhoid fever, bacillary dysentery and other gastrointestinal tract related problems. Also the environmental contaminants in pond fish production become important because of its significance to consumers' acceptance of the fish products

Samples of plankton were taken from Broom Water in August 1997 after a sighting of medusae on 11th August. Broom Water is about 8 m wide, and extends 250 m from the main channel of the Thames, above the weir at Teddington. On 11th August medusae were so abundant that it was possible to collect 20 in ten minutes. They were rising to the surface in bright sunlight, then sinking slowly down through the water. Examination of a medusa's tentacles under a microscope revealed the presence of a commensal protozoan, a ciliate Trichodina pediculus. Over 20 species of phytoplankton were found in Broom Water. Most of the species are common and widespread, but it was a surprise to find Errerella bornhemiensis with its characteristic pyramidal colonies, which is a relatively rare species. Zooplankters in Broom Water consisted of Rotifera and Crustacea. Zooplankton is the main food of Craspedacusta and it was found that the crustaceans but not the rotifers did undergo significant changes during the period 11-19th August. The major changes were a big increase in the percentage of cyclopoids, and a marked decrease in Bosmina. This could be because the delicate cuticle of Bosmina is much more susceptible to the stinging cells of the medusae compared with the tougher exoskeleton of the cyclopoid.

I. Foehn winds of southern California.
An investigation of the hot, dry and dust laden winds
occurring in the late fall and early winter in the Los Angeles
Basin and attributed in the past to the influences of the desert
regions to the north revealed that these currents were of a
foehn nature. Their properties were found to be entirely due
to dynamical heating produced in the descent from the high level
areas in the interior to the lower Los Angeles Basin. Any dust
associated with the phenomenon was found to be acquired from the
Los Angeles area rather than transported from the desert. It was
found that the frequency of occurrence of a mild type foehn of this
nature during this season was sufficient to warrant its classification
as a winter monsoon. This results from the topography of
the Los Angeles region which allows an easy entrance to the air
from the interior by virtue of the low level mountain passes north
of the area. This monsoon provides the mild winter climate of
southern California since temperatures associated with the foehn
currents are far higher than those experienced when maritime air
from the adjacent Pacific Ocean occupies the region.
II. Foehn wind cyclo-genesis.
Intense anticyclones frequently build up over the high level
regions of the Great Basin and Columbia Plateau which lie between
the Sierra Nevada and Cascade Mountains to the west and the Rocky
Mountains to the east. The outflow from these anticyclones produce
extensive foehns east of the Rockies in the comparatively low
level areas of the middle west and the Canadian provinces of
Alberta and Saskatchewan. Normally at this season of the year very
cold polar continental air masses are present over this territory
and with the occurrence of these foehns marked discontinuity surfaces
arise between the warm foehn current, which is obliged to slide over
a colder mass, and the Pc air to the east. Cyclones are
easily produced from this phenomenon and take the form of unstable
waves which propagate along the discontinuity surface between the
two dissimilar masses. A continual series of such cyclones was
found to occur as long as the Great Basin anticyclone is maintained
with undiminished intensity.
III. Weather conditions associated with the Akron disaster.
This situation illustrates the speedy development and
propagation of young disturbances in the eastern United States
during the spring of the year under the influence of the conditionally
unstable tropical maritime air masses which characterise the
region. It also furnishes an excellent example of the superiority
of air mass and frontal methods of weather prediction for aircraft
operation over the older methods based upon pressure distribution.
IV. The Los Angeles storm of December 30, 1933 to January 1, 1934.
This discussion points out some of the fundamental interactions
occurring between air masses of the North Pacific Ocean in connection
with Pacific Coast storms and the value of topographic and
aerological considerations in predicting them. Estimates of rainfall
intensity and duration from analyses of this type may be made and
would prove very valuable in the Los Angeles area in connection with
flood control problems.
