The mycotoxin zearalenone (ZEN) is a secondary metabolite of fungi which is produced by certain species of the genus Fusarium and can occur in cereals and other plant products. Reporter gene assays incorporating natural steroid receptors and the H295R steroidogenesis assay have been implemented to assess the endocrine disrupting activity of ZEN and its metabolites -zearalenol (-ZOL) and -zearalenol (-ZOL). -ZOL exhibited the strongest estrogenic potency (EC50 0.022 ± 0.001 nM), slightly less potent than 17- estradiol (EC50 0.015 ± 0.002 nM). ZEN was ~70 times less potent than -ZOL and twice as potent as -ZOL. Binding of progesterone to the progestagen receptor was shown to be synergistically increased in the presence of ZEN, -ZOL or -ZOL. ZEN, -ZOL or -ZOL increased production of progesterone, estradiol, testosterone and cortisol hormones in the H295R steroidogenesis assay, with peak productions at 10 M. At 100 M, cell viability decreased and levels of hormones were significantly reduced except for progesterone. -ZOL increased estradiol concentrations more than -ZOL or ZEN, with a maximum effect at 10 M, with -ZOL (562 ± 59 pg/ml) > -ZOL (494 ± 60 pg/ml) > ZEN (375 ± 43 pg/ml). The results indicate that ZEN and its metabolites can act as potential endocrine disruptors at the level of nuclear receptor signalling and by altering hormone production.

In this paper we investigate gas-phase chemistry in the remnant 'superwind' of a carbon-rich red giant star, during its transition to a planetary nebula. The interacting stellar winds model is used. It is found that during the first few hundred years of transition, significant abundances of a few small molecules and ions (e.g. CH+, CH2+, CH3+, CH, CH2, NH) may occur in the thin, dense, shocked shell of gas predicted by thiS model, but that most molecules observed in protoplanetary nebulae will be rapidly destroyed, through photodissociation by strong UV from the central star. If dense clumps are present during transition, they may allow the gas-phase formation and/or survival of small amounts of some molecules, such as HCN, CN, C2H2, and HC3N, until about 2000 yr after termination of the superwind; and young, fully developed planetary nebulae may show observable amounts of polyatomic molecules by this means. Such clumping may explain the existence of, e.g., HCN in NGC 7027.

We have investigated the effects of depletion of the elements C, N and O on the chemical composition of dark clouds, using both isothermal and isochoric cloud models. Our work differs from previous approaches in that we have considered a much larger range of CNO depletions. We have included the chemistry of the ortho-and para-forms of H2 and the exothermic reaction between N+ and ortho-H2, which synthesizes NH3. In the isothermal models, the ortho:para ratio is very small at large depletions, but NH3 formation is still efficient owing to reactions between He+ and CN or HCN. In the isochoric models, the equilibrium temperature of the gas is larger, and a thermal ortho:para ratio, which is large enough to drive NH3 formation, results. In all cases, the fractional abundance of NH3 is close to 10(-8) and this may help to explain the puzzling observation that, in dark clouds, the column density of NH3 is always close to 10(15) cm-2.

A survey was carried out on the occurrence of dinitrocarbanilide (DNC), the marker residue for nicarbazin, in poultry produced in Ireland during 2002-2004. Liver (n = 736) and breast muscle samples (n = 342) were tested. DNC residues were found in 40 and 26% of liver and breast muscle samples at levels greater than 12.5 and 5 mu g kg(-1), respectively. DNC residues were found at >200 mu g kg(-1) in 12 and 0% of liver and muscle samples, respectively. Samples of breast muscle (n = 217) imported from 11 countries were also tested for DNC residues. A lower incidence of DNC residues (6%) was found in imported breast muscle. Egg samples (n = 546) were tested and DNC residues were found in nine samples, with levels ranging between 14 and 122 mu g kg(-1). Analysis of poultry, carried out as part of official food inspection in the period 2004-2006, indicated a reduction in the number of broiler liver samples containing DNC at >200 mu g kg(-1), to approximately 7%. Bromate in drinking water, at a level of microgrammes/litre, is a problem in ozonated waters but can be adsorbed, to a certain extent, by granular activated carbon. The adsorption capacity of granular activated carbon for bromate is significantly lowered when there are high concentrations of other anions, most notably chloride and sulphate, present in the water.

Rhodococcus rhodochrous NCIMB13064 can dehalogenate and use a wide range of 1-haloalkanes as sole carbon and energy source. The 1-chloroalkane degradation phenotype may be lost by cells spontaneously or after treatment with Mitomycin C. Two laboratory derivatives of the original strain exhibited differing degrees of stability of the chloroalkane degradation marker. Plasmids of approximately 100 kbp (pRTL1) and 80 kbp (pRTL2) have been found in R. rhodochrous NCIMB13064. pRTL1 was shown to be carrying at least some genes for the dehalogenation of 1-chloroalkanes with short chain lengths (C-3 to C-9). However, no connection was found between the utilization of 1-chloroalkanes with longer chain lengths (C-12 to C-18) and the presence of pRTL1. Three separate events were observed to lead to the inability of NCIMB13064 to dehalogenate the short-chain 1-chloroalkanes; the complete loss of pRTL1, the integration of pRTL1 into the chromosome, or the deletion of a 20-kbp fragment in pRTL1. High-frequency transfer of the 1-chloroalkane degradation marker associated with pRTL1 has been demonstrated in bacterial crosses between different derivatives of R. rhodochrous NCIMB13064, (C) 1995 Academic Press, Inc.
