Foram estudadas, pelo método da assinatura digital, 35 biópsias esofágicas provenientes de pacientes da província de Linxian, China, classificadas por dois observadores com ampla experiência em patologia gastrointestinal como normais, displasias ou carcinomas (8 casos normais, 6 displasias leves, 8 displasias moderadas, 4 displasias acentuadas, 4 carcinomas suspeitos de invasão e 5 carcinomas invasores). O objetivo do trabalho foi caracterizar os núcleos das populações celulares desses casos de forma que permitisse a derivação de informações diagnósticas e de possível implicação prognóstica a partir do estudo quantitativo das características nucleares de cada caso ou categoria diagnóstica. As biópsias foram coradas pelo método de Feulgen, sendo então selecionados e digitalizados 48 a 50 núcleos de cada uma delas. De cada núcleo foram extraídas 93 características cariométricas, arranjadas arbitrariamente em histograma designado como assinatura nuclear. Da média aritmética de cada característica dos núcleos de uma mesma biópsia resultou a assinatura digital do caso. A análise de funções discriminantes, baseada nas 15 características cariométricas que ofereceram melhor discriminação entre as categorias diagnósticas, mostrou que o grupo classificado como normal foi claramente distinto das demais categorias. A densidade óptica total aumentou progressivamente segundo a classificação das biópsias, do normal à displasia acentuada, sendo o valor do carcinoma semelhante ao da displasia moderada. A matriz de comprimento de seqüência apresentou o mesmo perfil, ou seja, ambas as características ofereceram discriminação clara entre as categorias diagnósticas, com exceção do carcinoma invasor, cujos valores foram superponíveis aos da displasia moderada. O estudo demonstrou a viabilidade da quantificação de características nucleares através das assinaturas nucleares digitais, que demonstraram diferenças estatisticamente significativas entre diferentes categorias diagnósticas e a elevação progressiva dos valores mensurados relacionados com o espectro das lesões, apresentando-as como um histograma (assinatura digital nuclear).

Recent progress in the technology for single unit recordings has given the neuroscientific community theopportunity to record the spiking activity of large neuronal populations. At the same pace, statistical andmathematical tools were developed to deal with high-dimensional datasets typical of such recordings.A major line of research investigates the functional role of subsets of neurons with significant co-firingbehavior: the Hebbian cell assemblies. Here we review three linear methods for the detection of cellassemblies in large neuronal populations that rely on principal and independent component analysis.Based on their performance in spike train simulations, we propose a modified framework that incorpo-rates multiple features of these previous methods. We apply the new framework to actual single unitrecordings and show the existence of cell assemblies in the rat hippocampus, which typically oscillate attheta frequencies and couple to different phases of the underlying field rhythm

The subthalamic nucleus (STN) is a key area of the basal ganglia circuitry regulating movement. We identified a subpopulation of neurons within this structure that coexpresses Vglut2 and Pitx2, and by conditional targeting of this subpopulation we reduced Vglut2 expression levels in the STN by 40%, leaving Pitx2 expression intact. This reduction diminished, yet did not eliminate, glutamatergic transmission in the substantia nigra pars reticulata and entopeduncular nucleus, two major targets of the STN. The knock-out mice displayed hyperlocomotion and decreased latency in the initiation of movement while preserving normal gait and balance. Spatial cognition, social function, and level of impulsive choice also remained undisturbed. Furthermore, these mice showed reduced dopamine transporter binding and slower dopamine clearance in vivo, suggesting that Vglut2-expressing cells in the STN regulate dopaminergic transmission. Our results demonstrate that altering the contribution of a limited population within the STN is sufficient to achieve results similar to STN lesions and high-frequency stimulation, but with fewer side effects.

Little is known about genetic exchanges in natural populations of bacteria of the spore-forming Bacillus cereus group, because no population genetics studies have been performed with local sympatric populations. We isolated strains of Bacillus thuringiensis and B. cereus from small samples of soil collected at the same time from two separate geographical sites, one within the forest and the other at the edge of the forest. A total of 100 B. cercus and 98 B. thuringiensis strains were isolated and characterized by electrophoresis to determine allelic composition at nine enzymatic loci. We observed genetic differentiation between populations of B. cereus and B. thuringiensis. Populations of a given Bacillus species-B. thuringiensis or B. cereus-were genetically more similar to each other than to populations of the other Bacillus species. Hemolytic activity provided further evidence of this genetic divergence, which remained evident even if putative clones were removed from the data set. Our results suggest that the rate of gene flow was higher between strains of the same species, but that exchanges between B. cereus and B. thuringiensis were nonetheless possible. Linkage disequilibrium analysis revealed sufficient recombination for B. cereus populations to be considered panmictic units. In B. thuringiensis, the balance between clonal proliferation and recombination seemed to depend on location. Overall, our data indicate that it is not important for risk assessment purposes to determine whether B. cereus and B. thuringiensis belong to a single or two species. Assessment of the biosafety of pest control based on B. thuringiensis requires evaluation of the extent of genetic exchange between strains in realistic natural conditions.

The lacewings appear in many agroecosystems, preying several species of agricultural pests. They have great search capability, high voracity, high reproductive potential and are easily maintained in laboratory conditions. In laboratory rearing, to avoid problems in the mass production are recommended adjustments in the type and quality of prey to be used in order to obtain individuals with desirable characteristics. It is necessary special care with the laboratory populations, avoiding problems from inbreeding. Larvae were reared individually in Petri dishes (9.0 cm diameter) and fed with eggs of Sitotroga cerealella (Olivier, 1789) (Lepidoptera: Gelechiidae), in the amount of 25 mg / larva, while the adults were kept in PVC cylindrical cages (10 cm x 30 cm). Thus, the study analyzed the influence of the size of the population of Chrysoperla externa (Hagen, 1861) (Neuroptera: Chrysopidae) on the pre-imaginal period (egg to adult) and reproductive capacity of this specie come from different populations and generations of laboratory. To this end, we used two populations, one of Jaboticabal (F-8 and F-21) and one of Piracicaba (F-6 and F-15), and subpopulations of 1, 5, 10, 15 and 20 couples, analyzing the incubation of eggs and the number of eggs per female in each population, generation and subpopulation. The pre-imaginal period (egg to adult) and the number of eggs per female of C. externa are influenced by the generation and the number of founding individuals, being these parameters favored when laboratory populations are established with the largest number of couples.

Despite recent advances, the mechanisms of neurorespiratory control in amphibians are far from understood. One of the brainstem structures believed to play a key role in the ventilatory control of anuran amphibians is the nucleus isthmi (NI). This nucleus is a mesencephalic structure located between the roof of the midbrain and the cerebellum, which differentiates during metamorphosis; the period when pulmonary ventilation develops in bullfrogs. It has been recently suggested that the NI acts to inhibit hypoxic and hypercarbic drives in breathing by restricting increases in tidal volume. This data is similar to the influence of two pontine structures of mammals, the locus coeruleus and the nucleus raphe magnus. The putative mediators for this response are glutamate and nitric oxide. Microinjection of kynurenic acid (an ionotropic receptor antagonist of excitatory amino acids) and L-NAME (a non-selective NO synthase inhibitor) elicited increases in the ventilatory response to hypoxia and hypercarbia.
