The recovery of sperm from the epididymal cauda may be the last chance to obtain genetic material when sudden death or serious injuries occur in valuable stallions. However, the lack of technical knowledge regarding the storage and transportation of the epididymis often prevents the preservation of the sperm. Therefore, the aim of this study was to compare sperm parameters of sperm obtained immediately after orchiectomy with sperm recovered from epididymal cauda at different times after storage at 5°C and at room temperature (RT). For that, 48 stallions of different breeds were used. In group 1 (control group), eight stallions were used, and the harvest of the epididymal sperm was performed immediately after orchiectomy. In group 2, 40 stallions were used, which were divided into five groups according to the storage time of the epididymis after orchiectomy (6, 12, 18, 24, or 30 hours), making a total of eight stallions per group. One epididymis of each stallion was stored at 5°C, and the contralateral epididymis was stored at RT, both for the same period. The sperm parameters of total motility, progressive motility, progressive linear velocity, curvilinear velocity, percentage of rapid sperm, and plasma membrane integrity were evaluated in all the groups after sperm recovery, resuspension in a sperm freezing diluent, and thawing. In conclusion, the storage of the testis-epididymis complex at 5°C provided better preservation of epididymal sperm than the storage at RT, and regardless of the temperature, the progressive motility is the sperm parameter that is most sensitive to storage time. © 2013 Elsevier Inc.

The use of cooling, without using adequate hygienic practices in primary milk production, allows for the growth of psychrotrophic microorganisms that produce the thermoresistant lipases that give milk a rancid flavor. This study aimed to verify how the variation in temperature influences the lipolytic metabolism of the psychrotrophic organisms. Samples of raw milk were collected and submitted to laboratorial analysis as follows: psychrotrophic bacteria count, lipolytic bacteria count, and free fatty acids dosage. Each sample was divided into 3 aliquots and then incubated at 4, 8, and 12 °C, respectively. For each temperature, analyses were repeated after 12, 24, and 48 h of storage. Despite the psychrotrophs growth increase, according to temperature rise, the lipolytic metabolism was not consistent and presented the lower index at 8 °C, suggesting an intensification of the proteolytic compensatory activity at this temperature. © 2013 Institute of Food Technologists®.

This study evaluated the physicochemical changes in Nile tilapia (n = 82, 373.71 ± 61.91 g) refrigerated for up to 92 h and in the frozen fillets. The tilapias were captured with nets, slaughtered by ice and water shock (1:1) in a temperature of approximately 2°C for 30 min, and stored refrigerated at 4°C in polystyrene boxes containing ice. The fish were filleted, and filets were weighed and frozen. The drip loss and protein were determined after 23 days of frozen storage. After 4 h of storage, all fish were in full rigor mortis. The pH of the muscles decreased for up to 45 h of the storage period. The fillets obtained from tilapia stored for more than 72 h lost more weight and protein. Thus, the filleting or processing of tilapia should be done before 72 h of cold storage, since deterioration of the fish starts to occur after this period. Copyright © Taylor & Francis Group, LLC.
Antioxidant activity of basil and oregano extracts added to soybean oil for accelerated storage test

The objective of this research was to evaluate the antioxidant activity of extract of basil and oregano, and its synergistic effect when added to soybean oil and subjected to accelerated storage test. Extracts of basil, oregano and mixtures of both were applied to soybean oil at a concentration of 2.000mg/kg, then the extracts were heated in an oven at 60C for a period of 10 days. Samples were taken every 2 days and analyzed for concentrations of peroxides and conjugated dienes. Synthetic antioxidant tert-butylhydroquinone (TBHQ) at a concentration of 50mg/kg and soybean oil free antioxidant (control) subject to the same conditions were used as benchmarks. The results showed that the antioxidant effect decreases according to oil heating for all treatments. However, the synthetic antioxidant showed superior protection to the soybean oil during the formation of primary oxidation compounds, followed by the natural extracts, which showed no synergism. © 2012 Wiley Periodicals, Inc.

We evaluated the effects of constant low-temperature storage on Diaeretiella rapae (McIntosh) (Braconidae, Aphidiinae). Diaeretiella rapae mummies were stored at 5 ± 1°C for 0-36 days. The percentage of D. rapae emergence varied (100-83%) after 0-32 days of storage. After 32 days, emergence reduced to 55%. According to our Probit analysis, 50% mortality (LT50) of the population of D. rapae was reached after 40 days of storage at 5°C. Storage for up to 32 days did not negatively affect emergence and survival. Cold exposure of D. rapae for 36 days did not influence morphological malformations, sex ratio, and emergence of the F1 generation. After 4-36 days of storage, D. rapae showed a gradual decrease in emergence, longevity, reproductive capacity, and F1 sex ratio. Diaeretiella rapae can be stored for up to 24 days at 5°C, at which time the percentage of parasitism and the F1 sex ratio remain above 38% and at 0.50, respectively. © 2013 Sociedade Entomológica do Brasil.

Although sugarcane consumption is very popular worldwide there are few studies regarding the postharvest storage of stalks that focus on controlling enzymatic browning. The objective of this study was to evaluate the quality of sugarcane stalks stored at 10±1°C in controlled atmosphere with various oxygen (O2) levels (1kPa, 5kPa, 10kPa, 15kPa, and 21kPa) and to verify the effect of this storage on the activities of polyphenoloxidase (PPO) and peroxidase (POD). At 1kPa and 5kPa O2, the stalks' respiratory rate, POD activity, and reducing sugar content were significantly less (P<0.05) than at higher O2 concentrations. In contrast, low O2 levels did not affect PPO activity or influence cane coloration. These results can be used to guide the choice of plastic films, making it possible to store sugarcane stalks in a controlled atmosphere. © 2013 Elsevier B.V.

Cachaça is a distiled beverage obtained from the fermentation of sugar cane syrup that, depending on the production procedures, may be susceptible to contamination by polycyclic aromatic hydrocarbons (PAHs). These compounds present carcinogenic and/or mutagenic properties and offer a risk to human health. Sixteen PAHs were determined in cachaças that had been stored in glass bottles and in polyethylene tank by gas chromatography coupled with mass spectrometry. The quantification of the PAHs utilised an internal standard. The limits of detection and quantification varied from 0.05 to 0.10 μg L−1 and 0.20 to 0.30 μg L−1, respectively. A total PAH concentration of 51.57 μg L−1 was found in the beverages that were stored in the tank, while the concentration in the cachaça stored in glass jugs was 6.07 μg L−1. These results indicate that the polyethylene tank is a source for PAHs in cachaça.

Fatores de transcrição desempenham importantes funções em vários processos fisiológicos. Nos últimos anos, muitos fatores de transcrição têm sido isolados de plantas, emergindo como poderosas ferramentas na manipulação de características agronômicas. No presente trabalho, iniciamos estudos para isolar fatores de transcrição de mandioca (Manihot esculenta Crantz), importante cultura tropical e subtropical. Nossos resultados revelaram três tipos de proteínas diferencialmente expressas na raiz de reserva de mandioca (Manihot esculenta Crantz):e imunologicamente relacionadas com o fator de transcrição opaco-2 de milho. Experimentos de Southwestern mostraram duas proteínas capazes de interagir in vitro com uma seqüência de DNA do gene be2S1 de castanha-do-brasil.

A mandioca é cultivada como "mandioca mansa" para consumo in natura e "mandioca para indústria" como fonte de amidos e farinhas. Raças locais foram utilizadas para descoberta de "mutações espontâneas" e desenvolvimento de abordagem evolutiva e de melhoramento para estudos de função gênica. Recursos de Genômica e Proteômica foram obtidos. Análises de expressão gênica por blot de RNA e microarranjos foram desenvolvidos para identificação de expressão diferencial. "Mandioca açucarada" foi identificada, sendo relacionada com falta de expressão do gene da BEI e de uma mutação "nonsence" na sequência do gene GBSSI causando a formação do amido serose. "Mandioca avermelhada" apresentou falta de expressão do gene CasLYB, e a "amarela" uma regulação de repressão do gene CasHYb. Análise Proteômica do complexo carotenóide-proteína, juntamente com a análise de expressão de gene da CAP4, revelaram uma dupla fita de cDNA associada ao elevado acúmulo de carotenóide. Sequenciamento do gene da GBSSI identificou 22 haplótipos e grande diversidade de nucleotídios. Populações segregantes de cruzamentos de fenótipos bioquímicos diferenciados com cultivares elites dos Cerrados foram obtidas.

The spin injector part of spintronic FET and diodes suffers from fatigue due to rising heat on the depletion layer. In this study the stiffness of Ga1-xMnxAs spin injector in terms of storage modulus with respect to a varying temperature, 45 degrees C <= T <= 70 degrees C was determined. It was observed that the storage modulus for MDLs (Manganese Doping Levels) of 0%, 1% and 10% decreased with increase in temperature while that with MDLs of 20% and 50% increase with increase in temperature. MDLs of 20% and 50% appear not to allow for damping but MDLs <= 20% allow damping at temperature range of 45 degrees C <= T <= 70 degrees C. The magnitude of storage moduli of GaAs is smaller than that for ferromagnetic Ga1-xMnxAs systems. The loss moduli for GaAs were found to reduce with increase in temperature. Its magnitude of reducing gradient is smaller than Ga1-xMnxAs systems. The two temperature extremes show a general reduction in loss moduli for different MDLs at the study temperature range. From damping factor analysis, damping factors for ferromagnetic Ga1-xMnxAs was found to increase with decrease in MDLs contrary to GaAs which recorded the largest damping factor at 45 degrees C <= T <= 70 degrees C Hence, MDL of 20% shows little damping followed by 50% while MDL of 0% has the most damping in an increasing trend with temperature.
