Oilseeds are a high-value natural resource, due to its use as a substitute for petroleum. However, the storage time can reduce seed viability and oil quality. Therefore, scientific efforts have been made to provide a increment of storage time, germination rates and plant establishment of high-value oilseeds. The seedling establishment depends of the plant pass over the functional transition stage, characterized by a metabolic change from heterotrophic condition to autotrophic one. The storage oil mobilization is performed by β-oxidation process and the glyoxylate cycle. Also, the functional transition involves acclimation to photosynthetic condition, which generally includes the participation of antioxidant system and the reactive oxygen species, the latter are produced in various reactions of primary and secondary metabolism. In the present study, Catalase was inhibited during the functional transition of sunflower and safflower, after were performed many analyzes to elucidate the effects caused on the SOD and APX antioxidant systems. Also, were checked the changes in expression pattern of the glyoxylate cycle enzymes markers, ICL and MLS. It was observed that after CAT inhibition, the SOD and APX antioxidant systems allow the seedling establishment. Besides, was verified that both oilseeds can be accelerate the reverse mobilization and the photosynthetic establishment when Catalase activity has dramatically decreased

The brazilian-plum (Spondias tuberosa, His) is a tropical fruit tree that has been consolidated in the market for agribusiness processing, due to its characteristic flavor of fruit. Accordingly, studies to optimize the propagation of plants are necessary for production of seedlings with agronomic and quality assurance measures. This study aimed at determining the efficient techniques for uniform seed germination, as brazilian-plum seed present mechanical dormancy, and establish optimal culture media for multiplication of shoots from the in vitro micropropagation. Firstly, in a greenhouse at the Universidade Federal do Rio Grande do Norte, was evaluated the influence of different methods of breaking dormancy in the emergence of seedlings of brazilian-plum and speed of germination (IVG) of seeds. After 60 days of cultivation, it was found that splay in the distal portion of the seed was the best treatment, with rates of 85.33% in germinability and 3.415 of IVG, compared with the treatment of seed-soaking in water for 12h + humus and the control group. Subsequently, new sources of seedling explants were obtained in studies of tissue culture. Laboratory of Plant Biotechnology that the university, was used stem apex, nodal segments and internodes in search of decontamination with various concentrations of calcium hypochlorite [Ca(OCl)2] and micropropagation, inoculating them in half WPM (1980) with various concentrations of 6-benzylaminopurine (BAP). We used 10 sample units with three replications for different concentrations of [Ca(OCl)2], BAP and explants type. After thirty days, which was observed for the control of contamination, during the establishment in vitro, concentrations of [Ca(OCl)2] between 0.5% and 2.0% were effective in combating exogenous contamination of the apex. In nodal segments and internodes, concentrations of [Ca(OCl)2] between 1.0% and 2.0% and 1.5% and 2.0% were respectively, sufficient to reduce the percentage of losses in these infestations explants. For micropropagation, the culture medium supplemented with 0.1 mg.L-1 BAP promotes better development of multiple shoots per explants from nodal segment. However, success does not get to shoot training in internodal segment

Flowering is controlled by several environmental and endogenous factors, usually associated with a complex network of metabolic mechanisms. The gene characterization in Arabidopsis model has provided much information about the genetic and molecular mechanisms that control flowering process. Some of these genes had been found in rice and maize. However, in sugarcane this processe is not well known. It is known that early flowering may reduce its production up to 60% at northeast conditions. Considering the impact of early flowering in sugarcane production, the aim of this work was to make the gene characterization of two cDNAs previously identified in subtractive cDNA libraries: scPKCI and scSHAGGY. The in silico analysis showed that these two cDNAs presented both their sequence and functional catalytic domains conserved. The results of transgenic plants containing the overexpression of the gene cassette scPKCI in sense orientation showed that this construction had a negative influence on the plant development as it was observed a decrease in plant height and leaf size. For the scPKCI overexpression in antisense orientation it was observed change in the number of branches from T1 transgenic plants, whereas transgenic T2 plants showed slow development during germination and initial stages of development. The other cDNA analyzed had homology to SHAGGY protein. The overexpression construct in sense orientation did not shown any effect on development. The only difference observed it was an increase in stigma structure. These results allowed us to propose a model how these two genes may be interact and affect floweringdevelopment.

O objetivo deste trabalho foi avaliar o efeito da temperatura sobre a germinação e o armazenamento de sementes de Erythroxylum ligustrinum DC. As sementes foram colocadas germinar caixas de plásticas sobre duas folhas de papel, umedecidas com Nistatina 0, 2%, mantidas em germinadores nas temperaturas constantes de 5, 10, 15, 20, 25, 30, 35, 40 e 45ºC e com fotoperíodo de 12 horas. Para o armazenamento, as sementes foram colocadas em sacos de papel, mantidos à temperatura ambiente e em geladeira. em seguida, mensalmente, foi avaliada a germinação das sementes. A temperatura mínima, a faixa de temperatura ideal e a temperatura máxima para germinação de sementes de coca estão entre 10 e 15ºC, 25 e 30ºC e 40 e 45ºC, respectivamente. O armazenamento de sementes coca por longo período é inviável em embalagens de papel na geladeira (10ºC ± 2) e em temperatura ambiente (27ºC ± 5).

The present work describes myxozoans found in Cyphocharax nagelli (Characiformes: Curimatinae) commonly called saguiru collected from Rio do Peixe Reservoir, Sao Jose do Rio Pardo, São Paulo, Brazil. From a total of 38 examined fish, 24 were infected with Henneguya garavelli n. sp. (63% prevalence) and two with Myxobolus peculiaris n. sp. (5% prevalence) in the gills. Spores were studied by staining and fresh spores were observed by differential interference contrast optics. Henneguya garavelli n. sp. differs from Henneguya iheringi , Henneguya occulta, Henneguya cesarpintoi, Henneguya santae, Henneguya pisciforme, Henneguya amazonica, Henneguya striolata, Henneguya leporinicola and Henneguya chydadea in spore length and from Henneguya travassosi, Henneguya adherens, Henneguya malabarica, Henneguya piaractus and also Henneguya chydadea in polar capsule length and tail length. Myxobolus peculiaris n. sp. was very different when compared to other species of Myxobolus in its morphology and the biggest size of spore body. This work was divided in two experiments, the first one to evaluate the lettuce productivity (cv Lucy Brown - American group) influenced by increasing and decreasing periods of coexisting with harmful plants, and a second moment, aiming at to evaluate the allelopathic potential of nutsedge (Cyperus rotundus L.) on the initial lettuce development.The experimental treatments constituted of six increasing and decreasing periods of coexisting with harmful plants, or control of it in the culture, considered from the plantation of the lettuce; being separate in two groups: Weeds and Clean weeds.In the second moment of the experiment, purple nutsedge aerial parts had been collected in the area of experiment 1, that drying had been after triturated and immersed in methylic alcohol P. A in ratios 10, 5,0 and 2.5% (w/ v) and later impregnated in germination paper, where had been placed lettuce seeds.The analyses of results, one concludes that the presence of C. rotundus plants can intervene with the germination, growth and development of the plants of lettuce cv Lucy Brown, and that initial periods of coexisting between the culture and the harmful are associate to an induction of the foliar area development, and can not express the reduction of productivity in the end of the cycle. Drawn out periods of competition induce the reduction of the lettuce foliar area, and then they will intervene with the yield productivity.

Little is known about genetic exchanges in natural populations of bacteria of the spore-forming Bacillus cereus group, because no population genetics studies have been performed with local sympatric populations. We isolated strains of Bacillus thuringiensis and B. cereus from small samples of soil collected at the same time from two separate geographical sites, one within the forest and the other at the edge of the forest. A total of 100 B. cercus and 98 B. thuringiensis strains were isolated and characterized by electrophoresis to determine allelic composition at nine enzymatic loci. We observed genetic differentiation between populations of B. cereus and B. thuringiensis. Populations of a given Bacillus species-B. thuringiensis or B. cereus-were genetically more similar to each other than to populations of the other Bacillus species. Hemolytic activity provided further evidence of this genetic divergence, which remained evident even if putative clones were removed from the data set. Our results suggest that the rate of gene flow was higher between strains of the same species, but that exchanges between B. cereus and B. thuringiensis were nonetheless possible. Linkage disequilibrium analysis revealed sufficient recombination for B. cereus populations to be considered panmictic units. In B. thuringiensis, the balance between clonal proliferation and recombination seemed to depend on location. Overall, our data indicate that it is not important for risk assessment purposes to determine whether B. cereus and B. thuringiensis belong to a single or two species. Assessment of the biosafety of pest control based on B. thuringiensis requires evaluation of the extent of genetic exchange between strains in realistic natural conditions.

Sunflower is an annual dicotyledonous plant, herbaceous, erect and native of North America. It is thermo- and photo-insensitive, hence, can be grown round the year in sub-tropical and tropical countries. Only two spp. H. annuus and H. tuberosum are cultivated for food, remaining spp. are ornamentals, weeds and wild plants. However, H. annuus is allelopathic and inhibit the growth and development of other plants thus reducing their productivity. Much information is available about the allelopathic effects of sunflower crop on following crops in crop rotations. Although it is harmful to all crops, but, is less harmful to crops of Graminae family than other families. It seems that the harmful effects of sunflower in crop rotations are due to release and accumulation of root exudates during crop growth in soil. Soil incorporation of its fresh (green manure) or dry biomass in soil is inhibitory to both crops and weed spp. Several allelochemicals have been characterized from the H. annuus, which inhibit the seed germination and seedling growth of A. albus, A. viridis, Agropyron repens (Elymus repens), Ambrosia artemsiifolia, Avena fatua, Celosia crustata, Chenopodium album, Chloris barbara, Cynodon dactylon, D. sanguinalis, Dactyloctenium ageyptium, Digitaria ciliaris, Echinochloa crus-galli, Flaveria australasica, Parthenium hysterophorus, Portulaca oleracea, Sida spinosa, Trianthema portulacastrum, Veronica perisca the inhibitory effects of this crop may be used for weed management with less herbicides for sustainable agriculture.

Several Alternaria cassiae isolates were recovered from diseased sicklepod plants (Senna obtusifolia) in the southern regions of Brazil. A representative isolate (Cenargen CG593) was tested for its host range under greenhouse conditions. The fungus promoted symptoms in sicklepod, cassava (Manihot dulce), tomato (Lycopersicon esculentum) and eggplant (Solanum melongena) when tested at a spore concentration of 10(6) spores ml(-1). When the plants were inoculated with a suspension of 10(5) spores ml(-1) and held at a dew period of 12 h (cassava) or 18 h (tomato and eggplant), the plants showed symptoms of the disease, but they recovered and continued their normal vegetative growth. These results show that the fungus A. cassiae is safe to use for the control of S. obtusifolia under Brazilian conditions, because it did not cause excessive damage in the three plants tested.

The external morphology of seeds and post-germination developmental stages of Angelonia salicariifolia Bonpl. (Scrophulariaceae) were investigated using scanning electron microscopy. Some structural features of the seed exotesta and seedling in Angelonia are presented for the first time and are of potential taxonomic value for this neotropical genus. The seeds are very small (0.9-1.7 mm long and 0.5-0.9 mm wide), ovate, with a reticulate-crested exotesta, reticules arranged uniformly in longitudinal rows, with a high density of microcilia-like projections on the cell wall of the reticule base and on the edge of the crests. The hilum is located beside the micropyle at the narrow end of the seed. Germination is epigeal. During germination the radicle develops, followed by elongation of the hypocotyl and primary root. At this stage dense root hairs develop on the lower part of the hypocotyl. The apical bud-located between the cotyledons-begins to develop after the cotyledons have unfolded. The cotyledons are equal in size, sessile and ovate. The seedlings have two types of trichomes, one characteristic of the cotyledons and first pair of leaves (glandular, sessile, four-celled head with quadrangular shape) and the other characteristic of the hypocotyl and epicotyl (stalked, erect, elongate and three-celled with dome-shaped unicellular head). Frutos, sementes e plântulas de Crotalaria lanceolata, conhecida popularmente como guizo-de-cascavel, chocalho-de-cobra, xique-xique ou feijão-de-guizo, planta tóxica infestante que ocorre no Estado de São Paulo, foram estudadas morfologicamente e citogeneticamente. Os frutos são secos, deiscentes, polispérmicos e do tipo legume. As sementes são reniformes e o embrião é constituído de eixo embrionário e dois cotilédones. A testa pode apresentar variadas tonalidades de castanhos. A germinação é epígea e fanerocotiledonar. A espécie apresenta número cromossômico diplóide 2n = 16 com formulação cariotípica 12M + 4SM e comprimento cromossômico médio geral de 3,340 ± 0,689.

O conhecimento da composição específica do banco de sementes de plantas daninhas e a sua correlação com a flora estabelecida são importantes para nortear o manejo a ser adotado e a escolha dos herbicidas. A colheita mecanizada de cana-de-açúcar acarretou mudanças significativas na composição da flora infestante, quando proporcionou a manutenção de uma camada de palha, reduziu a movimentação do solo e dispensou a prática da queimada. Foram realizados levantamentos do banco de sementes e da flora de plantas daninhas que se estabeleceu em 28 talhões colhidos mecanicamente, sem queima prévia da palha. Com base nos dados de banco de sementes, efetuaram-se estudos fitossociológicos e de correlação entre a composição do banco de sementes e a flora emergida. As principais espécies presentes no banco de sementes foram as pertencentes à classe das dicotiledôneas anuais, com destaque para Amaranthus spp. e diversas espécies de Euphorbiaceae e Convolvulaceae. As sementes de gramíneas tradicionais da cultura tiveram pouca participação. O banco de sementes apresentou correlação não-significativa com a flora emergente, independentemente da época de colheita do talhão, da metodologia de quantificação do banco de sementes e das espécies de plantas daninhas.

O potencial alelopático de quatro diferentes extratos de plantas medicinais (Eclipta alba, Gomphrena globosa, Tabernaemontana catharinensis e Tithonia diversifolia) foi avaliado na germinação de Bidens pilosa e Lactuca sativa. Dentre os resultados obtidos no teste de germinação, notou-se um maior poder inibitório do extrato de G. globosa sobre sementes e plântulas de Bidens pilosa e Lactuca sativa independente da concentração utilizada e aumento da atividade inibitória de extratos T. catharinensis e de T. diversifolia com aumento da concentração destes. Quanto ao Índice de Velocidade de Germinação, os dados demonstraram redução do índice devido a elevada mortalidade das plântulas submetidas aos extratos de G. globosa e de T. catharinensis e a germinação tardia das sementes submetidas ao extrato de T. diversifolia. Quanto ao vigor das sementes, as menores porcentagens de sobrevivência ocorreram extratos à 2,5% para B. pilosa e 10% para L. sativa.

Estudos conduzidos em laboratório ou em condições de campo demonstram o potencial alelopático da espécie Sorghum bicolor L., seja inibindo a germinação e o desenvolvimento de plantas daninhas ou até mesmo de plantas cultivadas em sucessão. Com o objetivo de avaliar os efeitos potencialmente alelopáticos de extratos aquosos das folhas, caules e raízes de cinco híbridos de sorgo (SARA, DKB860, DKB 599, XBG00478 e XBG06020), sobre a germinação e o desenvolvimento de plântulas de soja, cv. MG/BR 46 (Conquista), foram conduzidos dois bioensaios no período de março a junho de 2002. Foram estabelecidas como testemunhas: água destilada (com pH 8,0); água destilada com pH ajustado para 4,5; água destilada com pH ajustado para 6,0 e solução de polietilenoglicol (PEG) equivalente ao potencial osmótico de 0,1 MPa. Dentre as características avaliadas, houve efeito significativo apenas no comprimento de radícula das plântulas de soja, sendo que aquelas tratadas com os extratos de sorgo apresentaram menor radícula, diferindo significativamente das plântulas testemunhas. Observou-se que, com os extratos de raízes dos híbridos SARA, DKB860, XBG00478 e XBG06020 ocorreram os menores comprimentos de radículas de soja, diferindo do DKB 599. O extrato de folhas do XBG00478 resultou em menor média, não diferindo dos extratos de DKB860, DKB599 e XBG06020. No entanto, para o extrato de caule, o SARA resultou em menor radícula, diferindo apenas do XBG00478. Verificou-se, também, que, para os extratos de DKB860 e XBG00478, os extratos de folhas proporcionaram menor média, diferindo dos extratos de raiz e caule, enquanto para DKB599, XBG06020 e SARA, não houve diferença significativa entre os extratos de folha e caule.
