The work highlights the first Global Comparison of Zooplankton Time Series. ► Variation of the peak in abundance is affected by annual temperature anomalies. ► Results show no global-scale synchrony in zooplankton time-series. ► There are spatial autocorrelations over substantial distances (1000–3000 km). ► There remains considerable uncertainty about the relative causes of shifts in distributions.

Regime shifts are sudden changes in ecosystem structure that can be detected across several ecosystem components. The concept that regime shifts are common in marine ecosystems has gained popularity in recent years. Many studies have searched for the step-like changes in ecosystem state expected under a simple interpretation of this idea. However, other kinds of change, such as pervasive trends, have often been ignored. We assembled over 300 ecological time series from seven UK marine regions, covering two to three decades. We developed state-space models for the first principal component of the time series in each region, a common measure of ecosystem state. Our models allowed both trends and step changes, possibly in combination. We found trends in three of seven regions and step changes in two of seven regions. Gradual and sudden changes are therefore important trajectories to consider in marine ecosystems.

Mangroves are highly productive environments that play important ecological and socioeconomic roles; however, they have been impacted to different degrees in most countries worldwide. The knowledge of which organisms inhabit this environment and their ecological interactions is the first step towards its conservation. The natural variability of environmental factors in mangroves provides numerous niches available to different species. Meiofauna have patchy patterns of distribution that are related to the availability of resources. Hence, meiofauna are expected to present a high diversity of different taxa occupying the different microhabitats offered by mangroves. This work aims to test the hypothesis that the assemblage structure of Nematoda varies significantly among mangrove microhabitats and to contribute knowledge on the meiofauna diversity in mangrove environments. This work was carried out in a mangrove region at Pernambuco state, Northeastern Brazil. Qualitative samples were collected in nine microhabitats which show different characteristics mainly in terms of presence of vegetation or another organism and sediment grain size. Univariate and multivariate analysis were applied to Nematoda genera abundance data. Our results demonstrate the existence of significant differences among microhabitats regarding nematode assemblage structure corroborating the hypothesis. Different Nematoda assemblages are present in at least seven microhabitats. These assemblages are composed of nematode genera with different trophic and morphological features, demonstrating a strong relationship between morphological diversity and ecological plasticity. Furthermore, this study also demonstrates the importance of the conservation of this ecosystem and its attributes.

A multi-sensor satellite approach based on ocean colour, sunglint and Synthetic Aperture Radar imagery is used to study the impact of interacting internal tidal (IT) waves on near-surface chlorophyll-a distribution, in the central Bay of Biscay. Satellite imagery was initially used to characterize the internal solitary wave (ISW) field in the study area, where the “local generation mechanism” was found to be associated with two distinct regions of enhanced barotropic tidal forcing. IT beams formed at the French shelf-break, and generated from critical bathymetry in the vicinities of one of these regions, were found to be consistent with “locally generated” ISWs. Representative case studies illustrate the existence of two different axes of IT propagation originating from the French shelf-break, which intersect close to 46°N, − 7°E, where strong IT interaction has been previously identified. Evidence of constructive interference between large IT waves is then presented and shown to be consistent with enhanced levels of chlorophyll-a concentration detected by means of ocean colour satellite sensors. Finally, the results obtained from satellite climatological mean chlorophyll-a concentration from late summer (i.e. September, when ITs and ISWs can meet ideal propagation conditions) suggest that elevated IT activity plays a significant role in phytoplankton vertical distribution, and therefore influences the late summer ecology in the central Bay of Biscay.

The Channel Catchments Cluster (3C) aims to capitalise on outputs from some of the recent projects funded through the INTERREG IVa France (Channel) England programme. The river catchment basins draining into the Channel region drain an area of 137,000km2 and support a human population of over 19M. Throughout history, these catchments, rivers and estuaries have been centres of habitation, developed through commerce and industry, providing transport links to hinterland areas. These catchments also provide drinking water and food through provision of agriculture, fisheries and aquaculture. In addition, many parts of the region are also economically important now for the tourism and leisure industries. Consequently, there is a need to manage the balance of these many and varied human activities within the catchments, rivers, estuaries and marine areas to ensure that they are maintained or restored to good environmental condition . This document highlights some of the recent work carried out by projects within the INTERREG IVa programme that provide tools and techniques to assist in the achievement of these goals.

Ocean Virtual Laboratory is an ESA-funded project to prototype the concept of a single point of access for all satellite remote-sensing data with ancillary model output and in situ measurements for a given region. The idea is to provide easy access for the non-specialist to both data and state-of-the-art processing techniques and enable their easy analysis and display. The project, led by OceanDataLab, is being trialled in the region of the Agulhas Current, as it contains signals of strong contrast (due to very energetic upper ocean dynamics) and special SAR data acquisitions have been recorded there. The project also encourages the take up of Earth Observation data by developing training material to help those not in large scientific or governmental organizations make the best use of what data are available. HSP70 chaperones mediate protein folding by ATP-dependent interaction with short linear peptide segments that are exposed on unfolded proteins. The mode of action of the Escherichia coli homolog DnaK is representative of all HSP70 chaperones, including the endoplasmic reticulum variant BiP/GRP78. DnaK has been shown to be effective in assisting refolding of a wide variety of prokaryotic and eukaryotic proteins, including the -helical homodimeric secretory cytokine interferon- (IFN-). We screened solid-phase peptide libraries from human and mouse IFN- to identify DnaK-binding sites. Conserved DnaK-binding sites were identified in the N-terminal half of helix B and in the C-terminal half of helix C, both of which are located at the IFN- dimer interface. Soluble peptides derived from helices B and C bound DnaK with high affinity in competition assays. No DnaK-binding sites were found in the loops connecting the -helices. The helix C DnaK-binding site appears to be conserved in most members of the superfamily of interleukin (IL)-10-related cytokines that comprises, apart from IL-10 and IFN-, a series of recently discovered small secretory proteins, including IL-19, IL-20, IL-22/IL-TIF, IL-24/MDA-7 (melanoma differentiation-associated gene), IL-26/AK155, and a number of viral IL-10 homologs. These cytokines belong to a relatively small group of homodimeric proteins with highly interdigitated interfaces that exhibit the strongly hydrophobic character of the interior core of a single-chain folded domain. We propose that binding of DnaK to helix C in the superfamily of IL-10-related cytokines may constitute the hallmark of a novel conserved regulatory mechanism in which HSP70-like chaperones assist in the formation of a hydrophobic dimeric "folding" interface.

The non-beta-amyloid (Aß) component of Alzheimer's disease amyloid (NAC) and its precursor a-synuclein have been linked to amyloidogenesis in several neurodegenerative diseases. NAC and a-synuclein both form ß-sheet structures upon ageing, aggregate to form fibrils, and are neurotoxic. We recently established that a peptide comprising residues 3±18 of NAC retains these properties. To pinpoint the exact region responsible we have carried out assays of toxicity and physicochemical properties on smaller fragments of NAC. Toxicity was measured by the ability of fresh and aged peptides to inhibit the reduction of the redox dye 3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide (MTT) by rat pheochromocytoma PC12 cells and human neuroblastoma SHSY-5Y cells. On immediate dissolution, or after ageing, the fragments NAC(8±18) and NAC(8±16) are toxic, whereas NAC(12±18), NAC(9±16) and NAC(8±15) are not. Circular dichroism indicates that none of the peptides displays ß-sheet structure; rather all remain random coil throughout 24 h. However, in acetonitrile, an organic solvent known to induce ß sheet, fragments NAC(8±18) and NAC(8±16) both form ß-sheet structure. Only NAC(8±18) aggregates, as indicated by concentration of peptide remaining in solution after 3 days, and forms fibrils, as determined by electron microscopy. These findings indicate that residues 8±16 of NAC, equivalent to residues 68±76 in a-synuclein, comprise the region crucial for toxicity.
