Dendritic cells (DCs) are the most efficient antigen presenting cells, they provide co-stimulation, are able to secrete various proinflammatory cytokines and therefore play a pivotal role in shaping adaptive immune responses. Moreover, they are important for the promotion and maintenance of central and peripheral tolerance through several mechanisms like the induction of anergy or apoptosis in effector T cells or by promoting regulatory T cells. The murine CD8α+ (MuTu) dendritic cell line was previously derived and described in our laboratory. The MuTu cell line has been shown to maintain phenotypical and functional characteristics of endogenous CD8α+ DCs. They are able to cross-present exogenous antigens to CD8+ T cells and produce interleukin (IL-) 12 upon engagement of Toll like receptors. The cell line constitutes an infinite source of homogenous, phenotypically well-defined dendritic cells. This allows us to investigate the role and potential of specific molecules in the induction as well as regulation of immune responses by DCs in a rational and standardized way. In a first project the MuTu dendritic cell line was transduced in order to stably express the immunosuppressive molecules IL-10, IL-35 or the active form of TGF-β (termed IL-10+DC, IL-35+DC or actTGFβ+DC). We investigated the capability of these potentially suppressive or tolerogenic dendritic cell lines to induce immune tolerance and explore the mechanisms behind tolerance induction. The expression of TGF-β by the DC line did not affect the phenotype of the DCs itself. In contrast, IL-10+ and IL-35+DCs were found to exhibit lower expression of co-stimulatory molecules and MHC class I and II, as well as reduced secretion of pro-inflammatory cytokines upon activation. In vitro co-culture with IL-35+, IL10+ or active TGFβ+ DCs interfered with function and proliferation of CD4+ and CD8+ T cells. Furthermore, IL-35 and active TGF-β expressing DC lines induced regulatory phenotype on CD4+ T cells in vitro without or with expression of Foxp3, respectively. In different murine cancer models, vaccination with IL-35 or active TGF-β expressing DCs resulted in faster tumor growth. Interestingly, accelerated tumor growth could be observed when IL-35-expressing DCs were injected into T cell-deficient RAG-/- mice. IL-10expressing DCs however, were found to rather delay tumor growth. Besides the mentioned autocrine effects of IL-35 expression on the DC line itself, we surprisingly observed that the expression of IL-35 or the addition of IL-35 containing medium enhances neutrophil survival and induces proliferation of endothelial cells. Our findings indicate that the cytokine IL-35 might not only be a potent regulator of adaptive immune responses, but it also implies IL-35 to mediate diverse effects on an array of cellular targets. This abilities make IL-35 a promising target molecule not only for the treatment of auto-inflammatory disease but also to improve anti-cancer immunotherapies. Indeed, by applying active TGFβ+ in murine autoimmune encephalitis we were able to completely inhibit the development of the disease, whereas IL-35+DCs reduced disease incidence and severity. Furthermore, the preventive transfer of IL-35+DCs delayed rejection of transplanted skin to the same extend as the combination of IL-10/actTGF-β expressing DCs. Thus, the expression of a single tolerogenic molecule can be sufficient to interfere with the adequate activation and function of dendritic cells and of co-cultured T lymphocytes. The respective mechanisms of tolerance induction seem to be different for each of the investigated molecule. The application of a combination of multiple tolerogenic molecules might therefore evoke synergistic effects in order to overcome (auto-) immunity. In a second project we tried to improve the immunogenicity of dendritic cell-based cancer vaccines using two different approaches. First, the C57BL/6 derived MuTu dendritic cell line was genetically modified in order to express the MHC class I molecule H-2Kd. We hypothesized that the expression of BALB/c specific MHC class I haplotype (H-2Kd) should allow the priming of tumor-specific CD8+ T cells by the otherwise allogeneic dendritic cells. At the same time, the transfer of these H-2Kd+ DCs into BALB/c mice was thought to evoke a strong inflammatory environment that might act as an "adjuvant", helping to overcome tumor induced immune suppression. Using this so called "semi-allogeneic" vaccination approach, we could demonstrate that the delivery of tumor lysate pulsed H-2Kd+ DCs significantly delayed tumor growth when compared to autologous or allogeneic vaccination. However, we were not able to coherently elucidate the cellular mechanisms underlying the observed effect. Second, we generated MuTu DC lines which stably express the pro-inflammatory cytokines IL-2, IL-12 or IL-15. We investigated whether the combination of DC vaccination and local delivery of pro-inflammatory cytokines might enhance tumor specific T cell responses. Indeed, we observed an enhanced T cell proliferation and activation when they were cocultured in vitro with IL-12 or IL-2-expressing DCs. But unfortunately we could not observe a beneficial or even synergistic impact on tumor development when cytokine delivery was combined with semi-allogeneic DC vaccination.

O presente trabalho teve por objetivo avaliar cinco seleções de laranjeira 'Valência', quanto ao potencial industrial e de consumo in natura com relação às variedades 'Natal' e 'Valência', enxertadas sobre o citrumeleiro 'Swingle' (experimento I) e a tangerineira 'Sunki' (experimento II). Os experimentos foram conduzidos na Estação Experimental de Citricultura de Bebedouro (E.E.C.B.)-SP. A implantação dos experimentos foi em fevereiro de 2001, sob espaçamentos de 7,0 m entre linhas e 5,0 m entre plantas (285 plantas/ha) e 7,0 m entre linhas e 3,0m entre plantas (476 plantas/ha), respectivamente, para os experimentos I e II. O delineamento experimental foi em blocos ao acaso, duas plantas por parcela, quatro repetições e 7 tratamentos, constituídos pelas seguintes variedades-copa: 1) 'Natal'; 2) 'Valência'; 3) 'Valência-Don João'; 4) 'Valência-late-Burjasot' IVIA 35-2; 5) 'Valência-Rohde-Red' SRA-36; 6) 'Valência-Temprana' IVIA25; 7) 'Valência Camphell'. A seleção 'Valência late' é a que se destaca por ser a que mais se assemelhou às variedades tradicionais Valência e Natal.

O presente trabalho foi realizado com o objetivo de identificar as fases da germinação de sementes de araticum-de-terra-fria (Annona emarginata (Schltdl.) H. Rainer) sob diferentes temperaturas. Para tanto, o trabalho foi dividido em dois experimentos: um para a determinação das fases I e II da germinação, e outro para a fase III. O delineamento experimental foi o inteiramente casualizado, com 3 tratamentos e 4 repetições de 25 sementes por parcela, em ambos os experimentos. Os tratamentos foram constituídos pelas temperaturas constantes de 25ºC e 30ºC e temperatura alternada de 20-30ºC (8-16 h, respectivamente), com luz constante. As variáveis analisadas foram a variação do grau de umidade ao longo do tempo (experimento 1), a porcentagem e a velocidade média de germinação (experimento 2). Os dados foram submetidos à análise de variância, as médias comparadas pelo teste Tukey, a 5% de probabilidade e regressão polinomial. A variação do grau de umidade ao longo do tempo foi estudada através da regressão não linear monomolecular, com os parâmetros das funções comparados conforme os tratamentos, ajustados, submetidos à análise de variância e teste Tukey, a 5% de probabilidade. Observou-se que as sementes mantidas a 30ºC apresentaram maiores valores para velocidade de aquisição de água, atingindo 27,85% de grau de umidade na mudança da fase I para a fase II. Quando as sementes foram submetidas a 25ºC, a fase I teve duração de 60 horas, atingindo 28,35% de grau de umidade e, sob 20/30ºC, a fase durou 72 horas, alcançando 28,33% de grau de umidade. Embora 30ºC tenha promovido a maior velocidade de embebição, não refletiu em maior porcentagem de germinação (fase III), que foi observada nas sementes mantidas a 20/30ºC.

Durante abril de 2005 se realizó una campaña de muestreo de la vegetación acuática macroscópica en las cuencas de los ríos Foix, Besòs y Llobregat, sumando un total de 68 estaciones de muestreo. El objetivo principal fue evaluar el estado trófico de dichas cuencas mediante el índice trófico generado en España denominado IVAM (Índice de Vegetación Acuática Macroscópica). Se identificaron un total de 44 táxones, 8 géneros de Cyanophyta, 5 de Rodophyta, 2 de Xantophyceae, 3 de Bacillariophyceae, 15 de Clorophyta, 8 de Spermatophyta, 1 liquen y 2 briófitos (musgos y hepáticas). La cuenca del Llobregat fue la más diversa (37 t´axones), seguida del Besós (33) y el Foix (25). Los táxones más frecuentes fueron Cladophora, Vaucheria, Oedogonium, Apium, Rorippa y Oscillatoria, además de los musgos. Las fanerógamas fueron escasas, destacando los g´eneros Ranunculus, Zannichellia y Potamogeton. Los táxones que alcanzaron una mayor puntuación del IVAM (valor de tolerancia, vt = 8), aunque poco frecuentes, fueron Ranunculus, Cymbella, Rivularia, Nostoc, Tolypothrix, Chroococcus y Lemanea, todos indicadores de aguas oligotr´oficas. En aguas de buena calidad pero algo eutrofizadas (aguas mesotróficas, vt = 6), los táxones más frecuentes fueron Chara, Monostroma y Hildenbrandia, además de las hepáticas y un liquen. Según la calificación de los tramos en clases de estado trófico realizada por el IVAM, el 38.2% de los tramos presentaron una buena calidad (clases I y II) mientras que el 61.8% presentaron un estado trófico alterado (clases III, IV y V). La respuesta del IVAM a la concentración de nutrientes (fosfato, amonio, nitrito y nitrato) fue siempre significativa, destacando la alta correlación alcanzada con el ión fosfato (r = 0.71, p < 0.001), superior a la alcanzada por otros índices de diatomeas y macrófitos de resolución taxonómica específica. Este hecho confirma la ventaja de utilizar el IVAM en la evaluación del estado trófico de los ríos, ya que sin necesidad de alcanzar un nivel taxonómico de especie se obtiene una respuesta adecuada a dicha perturbación.

BACKGROUND: Studies in bipolar disorder (BD) to date are limited in their ability to provide a whole-disease perspective--their scope has generally been confined to a single disease phase and/or a specific treatment. Moreover, most clinical trials have focused on the manic phase of disease, and not on depression, which is associated with the greatest disease burden. There are few longitudinal studies covering both types of patients with BD (I and II) and the whole course of the disease, regardless of patients' symptomatology. Therefore, the Wide AmbispectiVE study of the clinical management and burden of Bipolar Disorder (WAVE-bd) (NCT01062607) aims to provide reliable information on the management of patients with BD in daily clinical practice. It also seeks to determine factors influencing clinical outcomes and resource use in relation to the management of BD. METHODS: WAVE-bd is a multinational, multicentre, non-interventional, longitudinal study. Approximately 3000 patients diagnosed with BD type I or II with at least one mood event in the preceding 12 months were recruited at centres in Austria, Belgium, Brazil, France, Germany, Portugal, Romania, Turkey, Ukraine and Venezuela. Site selection methodology aimed to provide a balanced cross-section of patients cared for by different types of providers of medical aid (e.g. academic hospitals, private practices) in each country. Target recruitment percentages were derived either from scientific publications or from expert panels in each participating country. The minimum follow-up period will be 12 months, with a maximum of 27 months, taking into account the retrospective and the prospective parts of the study. Data on demographics, diagnosis, medical history, clinical management, clinical and functional outcomes (CGI-BP and FAST scales), adherence to treatment (DAI-10 scale and Medication Possession Ratio), quality of life (EQ-5D scale), healthcare resources, and caregiver burden (BAS scale) will be collected. Descriptive analysis with common statistics will be performed. DISCUSSION: This study will provide detailed descriptions of the management of BD in different countries, particularly in terms of clinical outcomes and resources used. Thus, it should provide psychiatrists with reliable and up-to-date information about those factors associated with different management patterns of BD. Työn aiheena on siistausmassan saostuksessa käytettävä kiekkosuodatinväliaine ja tarkoituksena selvittää lankamateriaalien ja pinnoitusaineiden vaikutusta väliaineen likaantumisherkkyyteen ja suodatusteknisiin ominaisuuksiin. Siistauslaitosten ongelmana on suodatinväliaineen likaantuminen massassa olevien tahmo- ja painoväripartikkelien johdosta. Tällä hetkellä siistauslaitosten kiekkosuodatinpussien valmistuksessa käytetään pääasiassa polypropeeni- ja polyvinylideenifluoridimonofilamenttilankoja, joiden lianhylkivyys on osoittautunut huonoksi. Uusiksi lankamateriaaleiksi tutkimuksiin valittiin Easy Kleen I, Easy Kleen II, Hyflon, Halar ja Polyesteri. Tulosten perusteella Easy Kleen I ja II monofilamenttilangoista kudotun suodatinkankaan lianhylkivyys on polypropeeni- ja polyvinylideenifluoridimonofilamenttilangoista kudottua kangasta parempi. Suodatusteknisissä ominaisuuksissa ei materiaalien välillä havaittu merkittävää eroa. Easy Kleen monofilamenttilangat näyttävät soveltuvan myös keittokutistumiltaan ja mekaanisilta ominaisuuksiltaan kiekkosuodatinpussien materiaaleiksi. Polypropeeni- ja polyesterimonofilamenettilangoista kudottujen suodatinkankaiden pinnoittamisessa käytettiin hydrofiilisiä ja hydrofobisia pinnoitusaineita. Tulosten perusteella on mahdollista parantaa suodatinkankaan lianhylkivyyttä pinnoittamalla. Pinnoitettu kangas on myös suodatusteknisiltä ominaisuuksiltaan toimiva. Ongelmaksi suodatinkankaiden pinnoittamisessa saattaa kuitenkin muodostua pinnoituslämpötila, jossa kangas kutistuu asennuskelvottomaksi.

O raleio promove aumento no tamanho dos frutos, equilíbrio entre o crescimento vegetativo e produtivo e evita alternância da produção. O objetivo deste trabalho foi avaliar o efeito de diferentes concentrações de ethephon no raleio químico da ameixeira cv. Irati, com 4 anos de idade, sobre o porta-enxerto A-9, em espaçamento 5,0 x 2,5 m, no município de Arapoti - PR. Os tratamentos foram cinco concentrações de ethephon, 0,1; 0,125; 0,15; 0,2 mg L-1, aplicados 30 dias após a plena florada. As seguintes características foram avaliadas: número médio de frutos por ramo, número total de frutos na colheita, produção por planta, peso médio de frutos, diâmetro dos frutos, teor de sólidos solúveis, firmeza, acidez titulável, pH e coloração. O aumento na concentração de ethephon reduziu linearmente o número de frutos calibre I, calibre II e calibre III, aumentando o número de frutos de calibre IV, frutos maiores, obtidos com a concentração de 0,093 mg L-1 de ethephon . A seca ocorrida entre os meses de agosto e setembro pode ter afetado o bom desenvolvimento dos frutos.

Doxorubicin (DOX) is a widely used, potent chemotherapeutic agent; however, its clinical application is limited because of its dose-dependent cardiotoxicity. DOX's cardiotoxicity involves increased oxidative/nitrative stress, impaired mitochondrial function in cardiomyocytes/endothelial cells and cell death. Cannabidiol (CBD) is a nonpsychotropic constituent of marijuana, which is well tolerated in humans, with antioxidant, antiinflammatory and recently discovered antitumor properties. We aimed to explore the effects of CBD in a well-established mouse model of DOX-induced cardiomyopathy. DOX-induced cardiomyopathy was characterized by increased myocardial injury (elevated serum creatine kinase and lactate dehydrogenase levels), myocardial oxidative and nitrative stress (decreased total glutathione content and glutathione peroxidase 1 activity, increased lipid peroxidation, 3-nitrotyrosine formation and expression of inducible nitric oxide synthase mRNA), myocardial cell death (apoptotic and poly[ADP]-ribose polymerase 1 [PARP]-dependent) and cardiac dysfunction (decline in ejection fraction and left ventricular fractional shortening). DOX also impaired myocardial mitochondrial biogenesis (decreased mitochondrial copy number, mRNA expression of peroxisome proliferator-activated receptor γ coactivator 1-alpha, peroxisome proliferator-activated receptor alpha, estrogen-related receptor alpha), reduced mitochondrial function (attenuated complex I and II activities) and decreased myocardial expression of uncoupling protein 2 and 3 and medium-chain acyl-CoA dehydrogenase mRNA. Treatment with CBD markedly improved DOX-induced cardiac dysfunction, oxidative/nitrative stress and cell death. CBD also enhanced the DOX-induced impaired cardiac mitochondrial function and biogenesis. These data suggest that CBD may represent a novel cardioprotective strategy against DOX-induced cardiotoxicity, and the above-described effects on mitochondrial function and biogenesis may contribute to its beneficial properties described in numerous other models of tissue injury.

O objetivo deste trabalho foi avaliar a distância genética entre 37 acessos da espécie cultivada Psidium guajava, L. (goiaba) e de araçás do gênero Psidium do banco de germoplasma da Universidade Estadual do Norte Fluminense (UENF), via marcadores moleculares ISSR. Nos 17 marcadores selecionados, foram obtidas 216 bandas polimórficas. Pelo método de agrupamento UPGMA, houve a formação de cinco principais grupos. Os acessos de araçá da espécie P. cattleyanum Sabine , ficaram alocados nos grupos I e II. No grupo II, foi observada, dentro da espécie P cattleyanum, maior proximidade com a goiabeira. No grupo III, ficou alocado o acesso da espécie P. guineense Sw (araçá-do-campo) e dentre os araçás, foi o que ficou mais próximo da goiaba. Os genótipos de goiabeira ficaram alocados do grupo IV e V, confirmando sua alta divergência. Os marcadores moleculares foram eficientes em estimar a distância genética intra e interespecífica.

Annonaceae seeds are known by presenting dormancy mechanisms, whose reports ranging from coating impermeable to the physiological dormancy. By this way, the present study aimed to evaluate water uptake in Annona diversifolia Saff and Annona purpurea Moc & Sessé ex Dunal seeds. For this study, seeds were placed under immersion in distilled water, and used four replicates of 25 seeds of each species, which were weighed during the 480 hours that were immersed. To determine the place of purchase of water, Annona diversifolia seeds were sealed with paraffin at different locations. Based on the results, seeds from both species reached the phases I and II of water uptake, which indicates they are not hard; however, germination (Phase III) was not reached. Annona diversifolia seeds completed Phase I with, 50h and Annona purpurea with 70h from imbibitions begin, which shows that even slowly, water is acquire.

This is the first TEM examination of vitellogenesis in the cestode Aporhynchus menezesi, a parasite of the velvet belly lanternshark Etmopterus spinax and a member of a little-studied trypanorhynch family, the Aporhynchidae. The synthetic activity of vitellocytes plays two important functions in the developmental biology of cestodes: (1) their shell-globules serve in eggshell formation; and (2) their accumulated reserves of glycogen and lipids represent a food source for the developing embryo. In A. menezesi, vitelline follicles consist of cells at various stages of development, from peripheral, immature cells of the gonial type to mature cells towards the centre of the follicle. These stages are: (I) immature; (II) early differentiation; (III) advanced maturation; and (IV) mature. Gradual changes involved in this process occur within each stage. Vitellogenesis involves: (1) an increase in cell volume; (2) the development of a smooth endoplasmic reticulum and an accelerated formation and accumulation of both unsaturated and saturated lipid droplets, along with their continuous enlargement and fusion; (3) the formation of individual β-glycogen particles and their accumulation in the form of glycogen islands scattered among lipid droplets in the cytoplasm of maturing and mature vitellocytes; (4) the rapid accumulation of large, moderately saturated lipid droplets accompanied by dense accumulations of β-glycogen along with proteinaceous shell-globules or shell-globule clusters in the peripheral layer during the advanced stage of maturation; (5) the development of cisternae of granular endoplasmic reticulum that produce dense, proteinaceous shell-globules; (6) the development of Golgi complexes engaged in the packaging of this material; and (7) the progressive and continuous enlargement of shell-globules into very large clusters in the peripheral layer during the advanced stage of maturation. Vitellogenesis in A. menezesi, only to some extent, resembles that previously described for four other trypanorhynchs. It differs in: (i) the reversed order of secretory activities in the differentiating vitellocytes, namely the accumulation of large lipid droplets accompanied by glycogenesis or β-glycogen formation during early differentiation (stage II), i.e. before the secretory activity, which is predominantly protein synthesis for shell-globule formation (stage III); (ii) the very heavy accumulation of large lipid droplets during the final stage of cytodifferentiation (stage IV); and (iii) the small number of β-glycogen particles present in mature vitellocytes. Ultracytochemical staining with PA-TCH-SP for glycogen proved positive for a small number of β-glycogen particles in differentiating and mature vitellocytes. Hypotheses, concerning the interrelationships of patterns of vitellogenesis, possible modes of egg formation, embryonic development and life-cycles, are commented upon.

OBJETIVO: Avaliar, no coração, por imuno-histoquímica, a localização das proteínas TGFbeta1 latente e TGFbeta1 ativa, se ocorre ativação radioinduzida da proteína TGFbeta1 latente, e a distribuição das fibras colágenas em diversos períodos de tempo após irradiação. MATERIAIS E MÉTODOS: Trinta e dois camundongos isogênicos (C57BL) foram divididos em dois grupos: GI (não irradiado), com 12 animais, e GII (irradiado), com 20 animais. Os animais do GII receberam radiação gama (telecobaltoterapia, 60Co, com rendimento de 0,97 Gy/min., dose única de 7 Gy em corpo inteiro). Os camundongos dos grupos I e II foram sacrificados por estiramento cervical nos períodos de 1, 14, 30 e 90 dias após irradiação. RESULTADOS: Os corações irradiados apresentaram: 1) alterações nucleares e diminuição das estriações das células musculares cardíacas; 2) aumento significante da deposição de fibras colágenas aos 90 dias depois da irradiação; 3) ativação da proteína TGFbeta1 latente em cardiomiócitos e células do conjuntivo depois da irradiação. CONCLUSÃO: Nossos resultados mostram a importância da proteína TGFbeta1 no processo de fibrose cardíaca radioinduzida e sugerem que células do parênquima (cardiomiócitos) e do conjuntivo podem participar deste mecanismo atuando como fontes da proteína TGFbeta1 ativa.

This is the first TEM examination of vitellogenesis in the cestode Aporhynchus menezesi, a parasite of the velvet belly lanternshark Etmopterus spinax and a member of a little-studied trypanorhynch family, the Aporhynchidae. It differs in: (i) the reversed order of secretory activities in the differentiating vitellocytes, namely the accumulation of large lipid droplets accompanied by glycogenesis or β-glycogen formation during early differentiation (stage II), i.e. before the secretory activity, which is predominantly protein synthesis for shell-globule formation (stage III); (ii) the very heavy accumulation of large lipid droplets during the final stage of cytodifferentiation (stage IV); and (iii) the small number of β-glycogen particles present in mature vitellocytes. Ultracytochemical staining with PA-TCH-SP for glycogen proved positive for a small number of β-glycogen particles in differentiating and mature vitellocytes. Hypotheses, concerning the interrelationships of patterns of vitellogenesis, possible modes of egg formation, embryonic development and life-cycles, are commented upon.

Most fishes produce free-living embryos that are exposed to environmental stressors immediately following fertilization, including pathogenic microorganisms. Initial immune protection of embryos involves the chorion, as a protective barrier, and maternally-allocated antimicrobial compounds. At later developmental stages, host-genetic effects influence susceptibility and tolerance, suggesting a direct interaction between embryo genes and pathogens. So far, only a few host genes could be identified that correlate with embryonic survival under pathogen stress in salmonids. Here, we utilized high-throughput RNA-sequencing in order to describe the transcriptional response of a non-model fish, the Alpine whitefish Coregonus palaea, to infection, both in terms of host genes that are likely manipulated by the pathogen, and those involved in an early putative immune response. Embryos were produced in vitro, raised individually, and exposed at the late-eyed stage to a virulent strain of the opportunistic fish pathogen Pseudomonas fluorescens. The pseudomonad increased embryonic mortality and affected gene expression substantially. For example, essential, upregulated metabolic pathways in embryos under pathogen stress included ion binding pathways, aminoacyl-tRNA-biosynthesis, and the production of arginine and proline, most probably mediated by the pathogen for its proliferation. Most prominently downregulated transcripts comprised the biosynthesis of unsaturated fatty acids, the citrate cycle, and various isoforms of b-cell transcription factors. These factors have been shown to play a significant role in host blood cell differentiation and renewal. With regard to specific immune functions, differentially expressed transcripts mapped to the complement cascade, MHC class I and II, TNF-alpha, and T-cell differentiation proteins. The results of this study reveal insights into how P. fluorescens impairs the development of whitefish embryos and set a foundation for future studies investigating host pathogen interactions in fish embryos.

The increase in seafood production, especially in mariculture worldwide, has brought out the need of continued monitoring of shellfish production areas in order to ensure safety to human consumption. The purpose of this research was to evaluate pathogenic protozoa, viruses and bacteria contamination in oysters before and after UV depuration procedure, in brackish waters at all stages of cultivation and treatment steps and to enumerate microbiological indicators of fecal contamination from production site up to depuration site in an oyster cooperative located at the Southeastern estuarine area of Brazil. Oysters and brackish water were collected monthly from September 2009 to November 2010. Four sampling sites were selected for enteropathogens analysis: site 1- oyster growth, site 2- catchment water (before UV depuration procedure), site 3 - filtration stage of water treatment (only for protozoa analysis) and site 4- oyster's depuration tank. Three microbiological indicators ! were examined at sites 1, 2 and 4. The following pathogenic microorganisms were searched: Giardia cysts, Cryptosporidium oocysts, Human Adenovirus (HAdV), Hepatitis A virus (HAV), Human Norovirus (HnoV) (genogroups I and II), JC strain Polyomavirus (JCPyV) and Salmonella sp. Analysis consisted of molecular detection (qPCR) for viruses (oysters and water samples); immunomagnetic separation followed by direct immunofluorescence assay for Cryptosporidium oocysts and Giardia cysts and also molecular detection (PCR) for the latter (oysters and water samples); commercial kit (Reveal-Neogee (R)) for Salmonella analysis (oysters). Giardia was the most prevalent pathogen in all sites where it was detected: 36.3%, 18.1%, 36.3% and 27.2% of water from sites 1, 2, 3 and 4 respectively; 36.3% of oysters from site 1 and 54.5% of depurated oysters were harboring Giardia cysts. The huge majority of contaminated samples were classified as Giardia duodenalis. HAdv was detected in water and o! ysters from growth site and HnoV GI in two batches of oysters ! (site 1) in huge concentrations (2.11 x 10(13), 3.10 x 10(12) gc/g). In depuration tank site, Salmonella sp., HAV (4.84 x 10(3)) and HnoV GII (7.97 x 10(14)) were detected once in different batches of oysters. Cryptosporidium spp. oocysts were present in 9.0% of water samples from site four. These results reflect the contamination of oysters even when UV depuration procedures are employed in this shellfish treatment plant. Moreover, the molecular comprehension of the sources of contamination is necessary to develop an efficient management strategy allied to shellfish treatment improvement to prevent foodborne illnesses.
