This paper given at the conference "Donne a Venezia" in May 2008 discloses the complexity of Arcangela Tarabotti (1604-1652), a proto-feminist polemist and writer who lived all her life in a remoted convent, Sant'Anna in Castello, in a remoted part of Venice. Despite having been cloistered at the age of 11, according to her account, she managed to publish her books and become a recognised member of the République des Lettres. How did she manage to develop her incredibly modern ideas, suggesting women should study and work and avoid getting married with astronomical dowries? The article deals with entirely new documents, uncovered in archives and tries to provide an answer to such dearing questions.

The absorption spectra of phytoplankton in the visible domain hold implicit information on the phytoplankton community structure. Here we use this information to retrieve quantitative information on phytoplankton size structure by developing a novel method to compute the exponent of an assumed power-law for their particle-size spectrum. This quantity, in combination with total chlorophyll-a concentration, can be used to estimate the fractional concentration of chlorophyll in any arbitrarily-defined size class of phytoplankton. We further define and derive expressions for two distinct measures of cell size of mixed populations, namely, the average spherical diameter of a bio-optically equivalent homogeneous population of cells of equal size, and the average equivalent spherical diameter of a population of cells that follow a power-law particle-size distribution. The method relies on measurements of two quantities of a phytoplankton sample: the concentration of chlorophyll-a, which is an operational index of phytoplankton biomass, and the total absorption coefficient of phytoplankton in the red peak of visible spectrum at 676 nm. A sensitivity analysis confirms that the relative errors in the estimates of the exponent of particle size spectra are reasonably low. The exponents of phytoplankton size spectra, estimated for a large set of in situ data from a variety of oceanic environments (~ 2400 samples), are within a reasonable range; and the estimated fractions of chlorophyll in pico-, nano- and micro-phytoplankton are generally consistent with those obtained by an independent, indirect method based on diagnostic pigments determined using high-performance liquid chromatography. The estimates of cell size for in situ samples dominated by different phytoplankton types (diatoms, prymnesiophytes, Prochlorococcus, other cyanobacteria and green algae) yield nominal sizes consistent with the taxonomic classification. To estimate the same quantities from satellite-derived ocean-colour data, we combine our method with algorithms for obtaining inherent optical properties from remote sensing. The spatial distribution of the size-spectrum exponent and the chlorophyll fractions of pico-, nano- and micro-phytoplankton estimated from satellite remote sensing are in agreement with the current understanding of the biogeography of phytoplankton functional types in the global oceans. This study contributes to our understanding of the distribution and time evolution of phytoplankton size structure in the global oceans.

Background: Phosphorus (P) is a major limiting nutrient for plant growth in many soils. Studies in model species have identified genes involved in plant adaptations to low soil P availability. However, little information is available on the genetic bases of these adaptations in vegetable crops. In this respect, sequence data for melon now makes it possible to identify melon orthologues of candidate P responsive genes, and the expression of these genes can be used to explain the diversity in the root system adaptation to low P availability, recently observed in this species. Methodology and Findings: Transcriptional responses to P starvation were studied in nine diverse melon accessions by comparing the expression of eight candidate genes (Cm-PAP10.1, Cm-PAP10.2, Cm-RNS1, Cm-PPCK1, Cm-transferase, Cm-SQD1, Cm-DGD1 and Cm-SPX2) under P replete and P starved conditions. Differences among melon accessions were observed in response to P starvation, including differences in plant morphology, P uptake, P use efficiency (PUE) and gene expression. All studied genes were up regulated under P starvation conditions. Differences in the expression of genes involved in P mobilization and remobilization (Cm-PAP10.1, Cm-PAP10.2 and Cm-RNS1) under P starvation conditions explained part of the differences in P uptake and PUE among melon accessions. The levels of expression of the other studied genes were diverse among melon accessions, but contributed less to the phenotypical response of the accessions. Conclusions: This is the first time that these genes have been described in the context of P starvation responses in melon. There exists significant diversity in gene expression levels and P use efficiency among melon accessions as well as significant correlations between gene expression levels and phenotypical measurements.

Background: Monosporascus cannonballus is the main causal agent of melon vine decline disease. Several studies have been carried out mainly focused on the study of the penetration of this pathogen into melon roots, the evaluation of symptoms severity on infected roots, and screening assays for breeding programs. However, a detailed molecular view on the early interaction between M. cannonballus and melon roots in either susceptible or resistant genotypes is lacking. In the present study, we used a melon oligo-based microarray to investigate the gene expression responses of two melon genotypes, Cucumis melo 'Piel de sapo' ('PS') and C. melo 'Pat 81', with contrasting resistance to the disease. This study was carried out at 1 and 3 days after infection (DPI) by M. cannonballus. Results: Our results indicate a dissimilar behavior of the susceptible vs. the resistant genotypes from 1 to 3 DPI. 'PS' responded with a more rapid infection response than 'Pat 81' at 1 DPI. At 3 DPI the total number of differentially expressed genes identified in 'PS' declined from 451 to 359, while the total number of differentially expressed transcripts in 'Pat 81' increased from 187 to 849. Several deregulated transcripts coded for components of Ca2+ and jasmonic acid (JA) signalling pathways, as well as for other proteins related to defence mechanisms. Transcriptional differences in the activation of the JA-mediated response in 'Pat 81' compared to 'PS' suggested that JA response might be partially responsible for their observed differences in resistance. Conclusions: As a result of this study we have identified for the first time a set of candidate genes involved in the root response to the infection of the pathogen causing melon vine decline. This information is useful for understanding the disease progression and resistance mechanisms few days after inoculation.

A detailed study was performed for a sample of low-mass pre-main-sequence (PMS) stars, previously identified as weak-line T Tauri stars, which are compared to members of the Tucanae and Horologium Associations. Aiming to verify if there is any pattern of abundances when comparing the young stars at different phases, we selected objects in the range from 1 to 100 Myr, which covers most of PMS evolution. High-resolution optical spectra were acquired at European Southern Observatory and Observatorio do Pico dos Dias. The stellar fundamental parameters effective temperature and gravity were calculated by excitation and ionization equilibria of iron absorption lines. Chemical abundances were obtained via equivalent width calculations and spectral synthesis for 44 per cent of the sample, which shows metallicities within 0.5 dex solar. A classification was developed based on equivalent width of Li I 6708 angstrom and Ha lines and spectral types of the studied stars. This classification allowed a separation of the sample into categories that correspond to different evolutive stages in the PMS. The position of these stars in the Hertzsprung-Russell diagram was also inspected in order to estimate their ages and masses. Among the studied objects, it was verified that our sample actually contains seven weak-line T Tauri stars, three are Classical T Tauri, 12 are Fe/Ge PMS stars and 21 are post-T Tauri or young main-sequence stars. An estimation of circumstellar luminosity was obtained using a disc model to reproduce the observed spectral energy distribution. Most of the stars show low levels of circumstellar emission, corresponding to less than 30 per cent of the total emission.

In this article we describe and illustrate the new species Paepalanthus hirtellus (Eriocaulaceae, Paepalanthoideae). The species is, as far as known, restricted to Pico do Itambe State Park, in the state of Minas Gerais, Brazil. Paepalanthus hirtellus occurs on the campos rupestres of the Espinhaco Range. We compare it with P. lombensis and P. chrysophorus, the two morphologically most similar species. We provide additional comments on the morphological variability, habitat, geographic distribution and we provide its conservation status.

A checklist of the 14 genera and 34 species of Bromeliaceae from the Parque Estadual do Rio Preto in Sao Goncalo do Rio Preto municipality, Minas Gerais state, southeastern Brazil, is presented. The Tillandsioideae was the most diverse subfamily and was found to be concentrated in rocky field areas. Bromelioideae is also a species rich subfamily, but its taxa have shown a preference to forested areas and savannas at lower altitudes. Pitcairnioideae is highlighted by its level of endemism, but has only four species. Cryptanthus micrus, a new species found in this area is described and illustrated. Our cluster analysis indicated that the Rio Preto State Park has a Bromeliaceae flora more similar to that from Pico do Itambe and Grao Mogol State Parks. Taxa like Dyckia glandulosa, Orthophytum itambense and Vriesea medusa, which were previously considered to be endemic to Pico do Itambe, now have their area of occurrence extended to Rio Preto. These new occurrences highlight the importance to create a corridor joining these neighboring reserves to connect populations of narrowly ranged or rare species. In this work we present pictures of 19 species in their habitats within the park, and we hope that these illustrations will help in the identification and conservation of these taxa.

In this paper we describe a new genus of Bromehaceae, Lapanthus, restricted to the southern portion of the Espinhaco Range, Minas Germs State, in southeastern Brazil Two new combinations to accommodate species previously described in the genera Orthophytum and cryptanthus and one new synonym are proposed Lapanthus has morphological affinities with both Cryptanthus and Orthophytum but nevertheless differs by the combination of margins of the petals ciliate, presence of lanceolate petal appendages and free stamens, and also by molecular data Cryptanthus and Orthophytum have petals entire along the margins, and the filaments of the most internal whorl are adnate to the petals Lapanthus stands out by having a pair of lanceolate petal appendages, which are almost completely adnate to the petals In Orthophytum, however, appendages are cupuhform or sacciform and they are totally absent in the genus Cryptanthus Lapanthus and Orthophytum present meiotic and mitotic chromosome numbers equal to n=25 and 2n=50, 100 and 150 respectively, while Cryptanthus presents meiotic and mitotic chromosome numbers n=17 and 2n=34, 36, 54 respectively, and this difference is considered to be an autapomorphic feature of Cryptanthus Descriptions of the genus and species, identification keys, illustrations, photographs of living specimens, and taxonomic comments are provided

In the microvillar microdomain of the kidney brush border, sodium hydrogen exchanger type 3 (NHE3) exists in physical complexes with the serine protease dipeptidyl peptidase IV (DPPIV). The purpose of this study was to explore the functional relationship between NHE3 and DPPIV in the intact proximal tubule in vivo. To this end, male Wistar rats were treated with an injection of the reversible DPPIV inhibitor Lys [Z(NO(2))]-pyrrolidide (I40; 60 mg center dot kg(-1)center dot day(-1) ip) for 7 days. Rats injected with equal amounts of the noninhibitory compound Lys[ Z(NO(2))]-OH served as controls. Na(+) -H(+) exchange activity in isolated microvillar membrane vesicles was 45 +/- 5% decreased in rats treated with I40. Membrane fractionation studies using isopycnic centrifugation revealed that I40 provoked redistribution of NHE3 along with a small fraction of DPPIV from the apical enriched microvillar membranes to the intermicrovillar microdomain of the brush border. I40 significantly increased urine output ( 67 +/- 9%; P < 0.01), fractional sodium excretion ( 63 +/- 7%; P < 0.01), as well as lithium clearance ( 81 +/- 9%; P < 0.01), an index of end-proximal tubule delivery. Although not significant, a tendency toward decreased blood pressure and plasma pH/HCO(3)(-) was noted in I40-treated rats. These findings indicate that inhibition of DPPIV catalytic activity is associated with inhibition of NHE3-mediated NaHCO(3) reabsorption in rat renal proximal tubule. Inhibition of apical Na(+) -H(+) exchange is due to reduced abundance of NHE3 protein in the microvillar microdomain of the kidney brush border. Moreover, this study demonstrates a physiologically significant interaction between NHE3 and DPPIV in the intact proximal tubule in vivo.

The activity of the Na(+)/H(+) exchanger NHE3 is regulated by a number of factors including parathyroid hormone (PTH). In the current study, we used a renal epithelial cell line, the opossum kidney (OKP) cell, to elucidate the mechanisms underlying the long-term effects of PTH on NHE3 transport activity and expression. We observed that NHE3 activity was reduced 6 h after addition of PTH, and this reduction persisted almost unaltered after 24 h. The decrease in activity was associated with diminished NHE3 cell surface expression at 6, 16, and 24 h after PTH addition, total cellular NHE3 protein at 16 and 24 h, and NHE3 mRNA abundance at 24 h. The lower levels of NHE3 mRNA were associated to a small, but significant, decrease in mRNA stability. Additionally, by analyzing the rat NHE3 gene promoter activity in OKP cells, we verified that the regulatory region spanning the segment -152 to +55 was mildly reduced under the influence of PTH. This effect was completely abolished by the presence of the PKA inhibitor KT 5720. In conclusion, long-term exposure to PTH results in reduction of NHE3 mRNA levels due to a PKA-dependent inhibitory effect on the NHE3 promoter and a small reduction of mRNA half-life, and decrease in the total amount of protein which is preceded by endocytosis of the apical surface NHE3. The decreased NHE3 expression is likely to be responsible for the reduction of sodium, bicarbonate, and fluid reabsorption in the proximal tubule consistently perceived in experimental models of PTH disorders.

O estudo das propriedades mecânicas dos solos estruturados necessita de equipamentos de laboratório com características específicas. No caso de ensaios triaxiais, os equipamentos devem incorporar técnicas especiais, tais como: medição local de deformações e controle automático de trajetórias de tensão. Caso o solo seja ensaiado na condição não saturada, o equipamento deve ser capaz de controlar a sucção imposta ao corpo de prova. Esta dissertação apresenta a montagem de um equipamento triaxial automatizado para o estudo de solos estruturados, saturados e não saturados, sendo que para a execução de ensaios com diferentes trajetórias de tensões, pequenas modificações devem ser introduzidas no programa. O equipamento foi montado com peças encontradas no mercado nacional e de fácil aquisição. Este equipamento dispõe de medição local de deformações baseada em sensores de efeito Hall, com uma resolução de deformações axiais e radiais da ordem de 10-4. O equipamento utiliza um sistema de válvulas proporcionais para o controle automático das pressões aplicadas. O equipamento assim construído, foi utilizado para estudar o comportamento mecânico de um solo residual de arenito Botucatu oriundo de uma jazida na rodovia RS 239. Este solo apresenta um índice de vazios médio de 0,62. Envoltórias de resistência ao cisalhamento para valores de pico e para valores de deformações axiais >10%, foram determinadas a partir de resultados de ensaios triaxiais. Os resultados dos ensaios também permitiram identificar uma superfície de fluência. O efeito da cimentação, tanto na superfície de fluência como no módulo de Young, foi constatado através da análise das curvas tensão desvio x deformação axial, que mostram claramente uma mudança de comportamento associada à progressiva desestruturação do material.

Essa dissertação analisa o potencial de medidas de gerenciamento da demanda no transporte coletivo por ônibus urbano, com foco na tarifa diferenciada por hora do dia. A simultaneidade temporal e espacial das atividades determina picos de demanda por transportes em certos horários do dia, causando congestionamento nas vias e sobrecarga nos transportes coletivos. Neste contexto, o transporte coletivo desempenha importante papel, uma vez que se constitui como único meio de transporte para grande parte da população, além de ser a alternativa mais sustentável para as viagens motorizadas. A conveniência de uma distribuição mais uniforme da demanda por viagens decorre das perdas provocadas pelo congestionamento e do desperdício de recursos que representa a capacidade ociosa do transporte coletivo e do sistema viário urbano nos horários de baixa demanda. Políticas de gerenciamento da demanda por transportes visando diluir os picos são aplicadas com fteqüência em grandes centros urbanos e, entre as medidas que atuam diretamente sobre a demanda por transporte coletivo, destacam-se as políticas tarifárias, programas de horário de trabalho flexível, escalonado, com semana comprimida ou teletrabalho, e, também, melhorias dos níveis de serviço. Tarifa diferenciada por hora do dia no transporte público é prática corrente em diversos países. Com o objetivo de influenciar o usuário quanto à escolha do horário de viagem, adotam-se valores tarifários mais altos para as viagens realizadas nos períodos de pico e mais baixos fora do pico Essa estratégia busca atrair a demanda mais elástica para os horários em que o sistema é menos solicitado. A justificativa para a discriminação apóia-se no fato de que a excessiva concentração de usuários em poucas horas do dia é um dos fatores responsáveis pela elevação dos custos totais do sistema. O estudo de caso permitiu analisar, através de modelagem comportamental, a resposta da demanda do período de pico a políticas de diferenciação tarifária por hora do dia. A metodologia empregada mostrou-se adequada e os resultados encontrados permitem concluir que políticas que contemplem variações tarifárias em troca de alterações nos horários de início da viagem podem ser empregadas no contexto analisado. Ainda, verificou-se que os entrevistados aos quais é proposta a antecipação do horário de viagem são mais elásticos do que aqueles aos quais é proposta a postergação do horário de viagem.
