A low-cost electrochemical method was developed for the determination of trace-level of methyl parathion (MP) based on the properties of graphite-modified basal plane pyrolytic graphite electrode (graphite-bppg). A combination of graphite-bppg with square-wave voltammetric (SWV) analysis resulted in an original, sensitive and selective electrochemical method for determination of MP pesticide in drinking water. The electrode was constructed and the electrochemical behavior of MP was studied. Immobilization is achieved via film modification from dispersing graphite powder in deionized water and through pipeting a small volume onto the electrode surface allowing the solvent to volatilize. The strong affinity of the graphite modifier for the phosphorous group of the MP allowed the deposition of a significant amount of MP in less than 60 seconds. The cyclic voltammetric results indicate that the graphite-bppg electrode can enhance sensitivity in current intensity towards the quasi-reversible redox peaks of the products of the cathodic reduction of the nitro group at negative potential (peak I = 0.077 V and peak II = –0.062 V) and that the cathodic irreversible peak (peak III = –0.586 V) in comparison with bare bppg electrode and is also adsorption controlled process. Under optimized conditions, the concentration range and detection limit for MP pesticide are respectively 79.0 to 263.3 mmol L-1 and 3.00 mmol L-1. The proposed method was successfully applied to MP determination in drinking water and the performance of this electrochemical sensor has been evaluated in terms of analytical figures of merit.

The mercury rejected in the water system, from mining operations and lixiviation of soils after deforestation, is considered to be the main contributors to the contamination of the ecosystem in the Amazon Basin. The objectives of the present study were to examine cytogenetic functions in peripheral lymphocytes within a population living on the banks of the Tapajós River with respect to methylmercury (MeHg) contamination, using hair mercury as a biological indicator of exposure. Our investigation shows a clear relation between methylmercury contamination and cytogenetic damage in lymphocytes at levels well below 50 micrograms/gram, the level at which initial clinical signs and symptoms of mercury poisoning occur. The first apparent biological effect with increasing MeHg hair level was the impairment of lymphocyte proliferation measured as mitotic index (MI). The relation between mercury concentration in hair and MI suggests that this parameter, an indicator of changes in lymphocytes and their ability to respond to culture conditions, may be an early marker of cytotoxicity and genotoxicity in humans and should be taken into account in the preliminary evaluation of the risks to populations exposed in vivo. This is the first report showing clear cytotoxic effects of long-term exposure to MeHg. Although the results strongly suggest that, under the conditions examined here, MeHg is both a spindle poison and a clastogen, the biological significance of these observations are as yet unknown. A long-term follow-up of these subjects should be undertaken.

Sacarose. ABSTRACT Study of antimicrobial activity of sucrose picolinate against Escherichia coli Sucrose esters are generally used as surfactants, fat substitutes and antibiotics. The aim of the present study was to synthesize new sucrose esters derived from picolinic acid (2-pyridine carboxylic acid) and study in vitro antimicrobial activity on the Gramnegative pathogenic bacterium Escherichia coli. The synthesis of sucrose picolinate was performed through the transesterification of sucrose with methyl picolinate under anhydrous conditions at 80 oC using dimethyl sulfoxide (DMSO) as the solvent and K2CO3as the catalyst. The separation of the formed isomers was performed by HPLC in a semi-preparative chromatograph system. Five fractions were collected and applied to a disc-diffusion antibiogram in solidmedium tests at concentrations of 150, 300, 450, 600, 750 and 900 μg/mL. Fractions 2, 4 and 6 were active against E. coli. Fraction 4 (900 μg/mL) was the most efficient and was selected for the determination of antimicrobial activity in synergistic tests with EDTA at concentrations of 250, 500 and 750 μg/mL. The best result was obtained with 750 μg/mL of EDTA. Fraction 4 was characterized as a monosubstituted sucrose ester by FTIR and mass spectrometry (ESI-MS). Keywords: Sucrochemistry. Sucrose. Chromium picolinate sucrose. Escherichia coli. Susceptibility testing by disk diffusion.
