Les neurones cholinergiques et adrénergiques efférents du système nerveux cardiaque ‎intrinsèque (SNCI) sont impliqués dans la régulation de la fonction cardiaque chronotrope. ‎La galanine, peptide présent et libéré dans les terminaisons nerveuses cardiaques, est capable ‎de diminuer la libération de l’acétylcholine. Notre hypothèse est d’étudier la possibilité que ‎la galanine puisse agir comme un peptide neuromodulateur inhibiteur dans les neurones du ‎SNCI chez le rat. Les oreillettes et les tissus associés du SNCI sont isolés à partir de rats ‎Sprague-Dawley et perfusés en vitro. Les valeurs de la fréquence auriculaire mesurées d’un ‎battement à l’autre sont tirées à partir d’un enregistrement bipolaire auriculaire. La perfusion ‎de la nicotine en forme de bolus (avec des concentrations dans le bain de 20-250 µm) a ‎induit des réponses biphasiques composées d’une réponse chronotrope négative au début ‎suivie d’une réponse chronotrope positive. Au cours de la superfusion de la galanine ‎‎(170 nM), les réponses chronotropes positives à la nicotine sont réduites chez 12/14 ‎préparations (contrôle : 21,35 ± 17,4 à galanine : 5,46 ± 8,0 bpm, n = 14, p = 0,02), tandis que ‎les réponses chronotropes négatives sont réduites chez 8 /14 préparations et affichent une ‎tendance générale vers la réduction (contrôle : -16,09 ± 14,7, galanine : -12,53 ± 13,2 bpm). ‎On peut conclure que la galanine agit comme un peptide neuromodulateur inhibiteur dans les ‎SNCI chez le rat, en modifiant de préférence la stimulation nicotinique de neurones ‎adrénergiques.‎

Cette étude valide l’efficacité du traitement de céfapirine intra-utérin sur les vaches diagnostiquées comme positives à l’endométrite clinique (PVD+) par vaginoscopie. Un seuil diagnostic des PVD+ a, d’abord, été déterminé selon les effets néfastes significatifs des écoulements vaginaux sur le statut de gestation à 120 jours post-partum. Les observations de 1247 vaches de 18 troupeaux, faites à 34 ± 7 jours post-partum, ont mené à considérer la présence de mucus vaginal trouble avec ou sans flocons de pus comme seuil pour les PVD+. La prévalence de PVD+ était de 21.6%. La chance d’être gestante à la première insémination effectuée à temps fixe sur un protocole de synchronisation était 1.89 fois moindre chez les vaches PVD+. Le traitement de céfapirine a été administré aléatoirement lors de la première visite. Même en absence d’effets significatifs sur la guérison clinique évaluée après 2 semaines, le traitement améliore significativement les performances en reproduction des PDV+. Les PVD+ ayant reçu le traitement avaient 1.85 fois plus de chance d’être gestantes à la première insémination que les PVD+ du groupe contrôle. Elles présentaient aussi une amélioration du jour ouvert médian de 38 jours.

La greffe de cellules souches hématopoïétiques est parfois le seul traitement efficace contre les cancers hématologiques ainsi que plusieurs autres désordres reliés au système hématopoïétique. La greffe autologue est souvent le traitement de choix pour les patients atteints de lymphome ou de myélome. Dans ce cas, les cellules souches hématopoïétiques (CSH) du patient sont récoltées et congelées. Le patient subit ensuite des traitements de chimiothérapie et/ou radiothérapie qui éliminent les cellules malignes, mais détruisent aussi son système hématopoïétique. Ce dernier sera ensuite reconstitué par la greffe de CSH. Ces traitements ont pour conséquence de plonger le patient en état d’aplasie pour une période variant de 2 à 4 semaines. La thrombocytopénie (faible taux de plaquettes) est une complication majeure nécessitant des transfusions plaquettaires répétées et associée à une augmentation de la mortalité hémorragique post-transplantation. Il serait particulièrement intéressant de développer une thérapie accélérant la reconstitution des mégacaryocytes (MK), ce qui aurait pour effet de raccourcir la période de thrombopénie et donc de diminuer les besoins transfusionnels en plaquettes et potentiellement augmenter la survie. HOXB4 est un facteur de transcription qui a déjà démontré sa capacité à expandre les CSH et les progéniteurs multipotents (CFU-GEMM) donnant naissance aux MK. Il est donc un bon candidat pour l’expansion des progéniteurs MK. Comme la protéine HoxB4 a par contre une courte demi-vie (~1.1h), des protéines HoxB4 de deuxième génération avec une plus grande stabilité intracellulaire ont été créées (1423 (HoxB4L7A), 1426 (HoxB4Y23A) et 1427 (HoxB4Y28A)). Nous avons donc étudié la capacité d’HoxB4 sauvage et de deuxième génération à expandre les CSH, ainsi que les MK donnant naissance aux plaquettes. La surexpression rétrovirale de ces protéines HoxB4Y23A et HoxB4Y28A conduit à une expansion des progéniteurs MK murins in vitro supérieure à HoxB4-wt, 1423 et au contrôle GFP. La reconstitution plaquettaire in vivo dans un modèle murin a ensuite été évaluée par des transplantations primaires et secondaires. Les résultats révèlent que la surexpression rétrovirale des différents HoxB4 n’apporte pas de bénéfice significatif à la reconstitution plaquettaire des souris. Lorsque cultivées dans un milieu favorisant la différenciation mégacaryocytaire, le traitement de cellules CD34+ dérivées du sang de cordon ombilical avec les protéines recombinantes TATHoxB4WT ou de seconde génération n’a pas augmenté la production plaquettaire. Par contre, de manière intéressante, les cellules CD34+ provenant de sang mobilisé de patients atteints de myélome et mises en culture dans un milieu favorisant l’expansion des CSH ont montré des différences significatives dans la différenciation des progéniteurs MK en présence de la protéine recombinante TATHoxB4. La protéine HOXB4 possède donc un avenir prometteur quant à une amélioration de l’état thrombocytopénique chez les patients.

Campylobacter jejuni cause gastroenteritis in humans. The main transmission vector is the consumption or handling of contaminated chicken meat, since chicken can be colonized asymptomatically by C. jejuni. However, water has been implicated as the transmission vector in a few outbreaks. One possibility is the contamination of water effluent by C. jejuni originating from chicken farm. The ability of C. jejuni to be transmitted by water would be closely associated to its ability to survive in water. Therefore, in this study, we have evaluated the ability of reference strains and chickenisolated strains to survive in water. Defined water media were used, since the composition of tap water is variable. We showed that some isolates survive better than others in defined freshwater (Fraquil) and that the survival was affected by temperature and the concentration of NaCl. By comparing the ability of C. jejuni to survive in water with other phenotypic properties previously tested, we showed that the ability to survive in water was negatively correlated with autoagglutination. Our data showed that not all chicken isolates have the same ability to survive in water, which is probably due to difference in genetic content.

This study aims to reveal the ability of bacteria isolated from Cochin estuary and the Arabian Sea to produce phosphatases, important characters of the enzymes, its ability to utilize organophosphorus compounds as source of phosphate and also their role in degradation of organophosphorus pesticides. It deals with isolation, identification and screening of bacteria for phosphatase production, and it describes the effect of cultural conditions on growth and phosphatase production. The effect of various factors like pH, NaCl concentration, temperature of incubation, carbon source, period of incubation etc. on growth and phosphatase production by the two selected species were studied to establish suitable environment for phosphatase production by these bacteria. In this study regulation of phosphatase synthesis, characteristics of acid and alkaline phosphatases are discussed.

In the present study, the effects of 5-HT, GABA and Bone Marrow Cells infused intranigrally to substantia nigra individually and in combinations on unilateral rotenone infused Parkinsonism induced rats. Scatchard analysis of DA, DA D1 and D2 receptors in the corpus striatum, cerebral cortex, cerebellum, brain stem and hippocampus showed a significant increase in the Brain regions of rotenone infused rat compared to control. Real Time PCR amplification of DA D1, D2, Bax and ubiquitin carboxy-terminal hydrolase were up regulated in the brain regions of rotenone infused rats compared to control. Gene expression studies of -Synuclien, cGMP and Cyclic AMP response element-binding protein showed a significant down regulation in Rotenone infused rats compared to control. Behavioural studies were carried out to confirm the biochemical and molecular studies.Our study demonstrated that BMC administration alone cannot reverse the above said molecular changes occurring in PD rat. 5-HT and GABA acting through their specific receptors in combination with bone marrow cells play a crucial role in the functional recovery of PD rats. 5-HT, GABA and Bone marrow cells treated PD rats showed significant reversal to control in DA receptor binding and gene expression. 5-HT and GABA have co-mitogenic property. Proliferation and differentiation of cells re-establishing the connections in Parkinson's disease facilitates the functional recovery. Thus, it is evident that 5-HT and GABA along with BMC to rotenone infused rats renders protection against oxidative, related motor and cognitive deficits which makes them clinically significant for cellbased therapy. The BMC transformed to neurons when co-transplanted with 5-HT and GABA which was confirmed with PKH2GL and nestin. These newly formed neurons have functional significance in the therapeutic recovery of Parkinson’s disease.

The temperate, filamentous phage ФMV -5 isolated from Mangalavanam mangrove of Kochi, using the environmental strain of Vibrio sp. MV-5 shares many similar properties with other marine phage isolates, while also remaining unique. The study has revealed that the interaction of temperate phages and the microbial population in the marine environment may contribute significantly to microbial genetic diversity and composition by conversion and transduction and which requires greater study.Prophages contribute a substantial share of the mobile DNA of their bacterial hosts and seem to influence the short-term evolution of pathogenic bacteria. Automated methods for systematic investigation of prophages and other mobile DNA elements in the available bacterial genome sequences will be necessary to understand their role in bacterial genome evolution. In the past, phages were mainly investigated as the simplest model systems in molecular biology. Now it is increasingly realized that phage research will be instrumental in the understanding of bacterial abundance in the environment. One can predict that phage research will impact diverse areas such as geochemistry and medicine. Success will largely depend on integrative multidisciplinary approaches in this field. Clearly, further studies are required to understand how vibriophages interact with Vibrios to promote this organism's acquisition of the critical genes which alter its virulence or adaptation to its environmental niche.It is evident from this study and comparison with those reports cited above that vibriophage ФMV-5 is a previously unreported bacteriophage. It is recommended that the minimum requirement for reporting a new phage should be novel morphological markers and a description of host range, both of which have been achieved in this study.

Considering the importance of diversity of micro algae in our ecosystem and new invasion of many organisms, an attempt was made to monitor the Cochin estuary along the south west coast of India for the qualitative distribution of phytoplankton and to study the growth kinetics and allelopathic effect of the phaeocystis sp. Isolated from the cochin estuary. Phaeocystis blooms are common only in high latitude environments and they rarely occur in low latitude environments such as tropics and subtropics. As phaeocystis is grouped under harmful alga ,in the present study the factors causing the blooms formation in the ecosystem. The nutrient concentration of the water body along with other physiochemical parameters that includes temperature salinity and ph play an important role in triggering the bloom of phaeocystis .The phaeocystis harbor specific bacterial flora associated with it and they exert an important role in the growth ,haemolytic activity and the bloom phases of the alga. The harmful alga mainly depends on the production of alleopathic compounds for the establishment of bloom in the marine environments .These physiological properties of the phaecystis were considered for the study, along with the role of nutrients in the allelopathic and hemolytic activity

Aquatic ecosystem in the south west coast of India is noted for its diversity of habitats. Very often these environments turn bluegreen when the bloom of bluegreen algae (cyanobacteria) appear consequent to eutrophication. This phenomenon occursin these habitats one after the other or simultaneously. This conspicuousness make one curious enough to know more about these nature’s gift bestowed upon mankind. While persuing the literature on the magnificent flora) it is understood that it may provide food fertilizer, chemicals and bioactive substances. These bioactive substances are likely to be involved in regulating natural populations and are potentially useful as biochemical tools and as herbicidal or biocontrol agents. The role of cyanobacteria in the aquatic food chain and contribution in abatement of heavy metals from the natural environment are well documented. Microorganisms distributed in the marine and brackish environments play an important role in the decomposition of organic matter and mineralisation in the system (Seshadri and lgnacimuthu, 2002). Estuary is one of the most productive ecosystems, at the same time one among the least explored ecosystems on earth, which has immense potential as a source of potent microorganisms that produce valuable compounds particularly, enzymes such as proteases. In this scenario, it is very appropriate to embark on finding novel alkaline protease producers from the estuarine system. The area where the present investigation was carried out is a part of the extensive estuarine system of South India viz. Cochin Estuary. There is meagre knowledge regarding the microbial composition, particularly the protease producers of Cochin Estuary. Hence, the present study has been undertaken with the objective of finding novel alkaline protease producing bacteria from Cochin Estuary

We report magnetic and magneto-optical measurements of two Mn12 single-molecule magnet derivatives isolated in organic glasses. Field-dependent magnetic circular dichroism (MCD) intensity curves (hysteresis cycles) are found to be essentially identical to superconducting quantum interference device magnetization results and provide experimental evidence for the potential of the optical technique for magnetic characterization. Cochin, commercial capital of Kerala, located on the west-coast of South India has a large number of chemical and sea food industries. Earlier studies in the past indicated that these industries contribute to heavy metal pollution, particularly mercury, copper, and cadmium, in Cochin backwater. Hence, in the present study, it was desired to isolate cadmium resistant bacteria from effluent discharged by chemical industry with a view to develop an ideal bioremediation process for safe discharge of industrial effluent in to the nearby aquatic environment. Effluent from three industries, located in the industrial belt of Cochin, were collected from the discharge point and cadmium resistant bacteria were screened using standard microbiological techniques

Speech is a natural mode of communication for people and speech recognition is an intensive area of research due to its versatile applications. This paper presents a comparative study of various feature extraction methods based on wavelets for recognizing isolated spoken words. Isolated words from Malayalam, one of the four major Dravidian languages of southern India are chosen for recognition. This work includes two speech recognition methods. First one is a hybrid approach with Discrete Wavelet Transforms and Artificial Neural Networks and the second method uses a combination of Wavelet Packet Decomposition and Artificial Neural Networks. Features are extracted by using Discrete Wavelet Transforms (DWT) and Wavelet Packet Decomposition (WPD). Training, testing and pattern recognition are performed using Artificial Neural Networks (ANN). The proposed method is implemented for 50 speakers uttering 20 isolated words each. The experimental results obtained show the efficiency of these techniques in recognizing speech
