Emphysema is a chronic obstructive pulmonary disease characterized abnormal dilatation of alveolar spaces, which impairs alveolar gas exchange, compromising the physical capacity of a patient due to airflow limitations. Here we tested the effects of G-CSF administration in pulmonary tissue and exercise capacity in emphysematous mice. C57Bl/6 female mice were treated with elastase intratracheally to induce emphysema. Their exercise capacities were evaluated in a treadmill. Lung histological sections were prepared to evaluate mean linear intercept measurement. Emphysematous mice were treated with G-CSF (3 cycles of 200 μg/kg/day for 5 consecutive days, with 7-day intervals) or saline and submitted to a third evaluation 8 weeks after treatment. Values of run distance and linear intercept measurement were expressed as mean ± SD and compared applying a paired t-test. Effects of treatment on these parameters were analyzed applying a Repeated Measures ANOVA, followed by Tukey's post hoc analysis. p < 0.05 was considered statistically significant. Twenty eight days later, animals ran significantly less in a treadmill compared to normal mice (549.7 ± 181.2 m and 821.7 ± 131.3 m, respectively; p < 0.01). Treatment with G-CSF significantly increased the exercise capacity of emphysematous mice (719.6 ± 200.5 m), whereas saline treatment had no effect on distance run (595.8 ± 178.5 m). The PCR cytokines genes analysis did not detect difference between experimental groups. Morphometric analyses in the lung showed that saline-treated mice had a mean linear intercept significantly higher (p < 0.01) when compared to mice treated with G-CSF, which did not significantly differ from that of normal mice. Treatment with G-CSF promoted the recovery of exercise capacity and regeneration of alveolar structural alterations in emphysematous mice. © 2013.

Given that cancer is one of the main causes of death worldwide, many efforts have been directed toward discovering new treatments and approaches to cure or control this group of diseases. Chemotherapy is the main treatment for cancer; however, a conventional schedule based on maximum tolerated dose (MTD) shows several side effects and frequently allows the development of drug resistance. On the other side, low dose chemotherapy involves antiangiogenic and immunomodulatory processes that help host to fight against tumor cells, with lower grade of side effects. In this review, we present evidence that metronomic chemotherapy, based on the frequent administration of low or intermediate doses of chemotherapeutics, can be better than or as efficient as MTD. Finally, we present some data indicating that noncytotoxic concentrations of antineoplastic agents are able to both up-regulate the immune system and increase the susceptibility of tumor cells to cytotoxic T lymphocytes. Taken together, data from the literature provides us with sufficient evidence that low concentrations of selected chemotherapeutic agents, rather than conventional high doses, should be evaluated in combination with immunotherapy. The antibody and cellular immune responses against infectious bronchitis virus (IBV) were evaluated at mucosal sites of chickens after immunization with various doses of an attenuated vaccine at 1 day of age. The correlation of these immune responses with protection of tracheal tissues was evaluated after experimental infection of these birds. Significantly reduced tracheal pathologic effects, measured according to ciliostasis and histology lesions, and reduced viral load were observed only in the full-dose vaccinated group at 5 days post-infection (dpi), while incomplete protection was observed for the subdose vaccinated groups. Moreover, birds of vaccinated groups, especially with full dose, developed higher levels of lachrymal IBV-specific IgG and IgA and increased the expression of cell-mediated immunity (CMI) genes, such as gamma interferon (IFNγ), CD8+ T cell marker, and granzyme homolog A more rapidly. In addition, these humoral and cellular immune responses evaluated at mucosal sites correlated significantly with tracheal protection against homologous IBV challenge in a vaccine dose-dependent manner. The results indicate that IgG, IgA and CD8+ T cell responses developed at mucosal sites after IBV vaccination of day-old chicks, could be taken as good correlates of protection against this virus. © 2013, Mary Ann Liebert, Inc.

Amphibians have melanin-containing cells in visceral organs that are similar to pigmentary cells from the epidermis. Both of them are derived from the ectodermal neural crest. Epidermal cells respond to α-melanocyte stimulating hormone (α-MSH), which is associated to the dispersion of melanin granules within melanocytes. Therefore, our aim was to test whether a non-degradable analogue of the α-MSH changes the superficial colouration of organs of Eupemphix nattereri. The hormone rapidly increases (within 12 hours) the colouration on the surface of the pericardium, heart, testes, nerves of the lumbar plexus, and lumbosacral parietal peritoneum. Colouration increased late (after 24 hours) in the kidneys and mesentery following hormone administration. However, this hormone did not change colouration of intestine, rectum and lungs. Our findings could be explained by the similarities between epidermal and visceral melanocytes, since both cells have a common embryonic origin. Furthermore, the increase in visceral colouration may be related to the dispersion of melanosomes within melanocytes, which causes the darkening of organs. Our results demonstrate for the first time that the visceral colouration is responsive, thereby altering the internal pattern of organs' colouration in anurans. © 2013 Copyright 2013 Unione Zoologica Italiana.

Breeder feed restriction may negatively affect broiler progeny immunity. Sources of trace minerals (TM) with higher bioavailability in breeder diets have been reported to enhance humoral and cellular immunity in broiler progeny. An experiment was conducted to examine the effects of breeder feeding programs and TM dietary sources on maternal antibody transfer and humoral immune response of progeny to a live vaccine against Newcastle disease virus (NDV). Cobb 500 breeders were fed according to 2 feed allocation programs, sigmoid late fast and sigmoid late slow, from 14 to 29 wk of age. From 56 to 62 wk of age, breeders were fed with either inorganic TM or an organic source (OTM) to replace 30% of Cu, Zn, and Mn. Progeny broilers were vaccinated intraocularly with La Sota NDV vaccine at 7 d of age. Blood samples were collected at hatching, 4, and 14 d postvaccination. Serum antibody levels against NDV were assessed by ELISA and cytokine expression by real time PCR. At hatching, late slow breeder progeny fed diets with 30% OTM had higher antibody titers as compared with progeny of breeders fed 100% inorganic TM. Similar results were observed 2 wk postvaccination. Breeder feeding programs and TM sources affected the expression level of IL-4 in NDV vaccinated broiler progeny. It was concluded that breeder feeding programs influenced humoral immune response to NDV vaccine in the broiler progeny, and 30% OTM may increase these responses. © 2013 Poultry Science Association, Inc.

The human skin not only provides passive protection as a physical barrier against external injury, but also mediates active surveillance via epidermal cell surface receptors that recognize and respond to potential invaders. Primary keratinocytes and immortalized cell lines, the commonly used sources to investigate immune responses of cutaneous epithelium are often difficult to obtain and/or potentially exhibit changes in cellular genetic make-up. Here we investigated the possibility of using salivary epithelial cells (SEC) to evaluate the host response to cutaneous microbes. Elevated secretion of IFN-γ and IL-12 was observed in the SEC stimulated with Staphylococcus aureus, a transient pathogen of the skin, as mono species biofilm as compared to SEC stimulated with a commensal microbe, the Staphylococcus epidermidis. Co-culture of the SEC with both microbes as dual species biofilm elicited maximum cytokine response. Stimulation with S. aureus alone but not with S. epidermidis alone induced maximum toll-like receptor-2 (TLR-2) expression in the SEC. Exposure to dual species biofilm induced a sustained upregulation of TLR-2 in the SEC for up to an hour. The data support novel application of the SEC as efficient biospecimen that may be used to investigate personalized response to cutaneous microflora. © 2013 Wiley Periodicals, Inc.

O mercúrio pode ser encontrado em diversas formas, sendo a orgânica como metilmercúrio (MeHg), considerada a mais tóxica. Facilmente absorvido por via oral, se acumula na cadeia trófica e se amplifica em carnívoros aquáticos, principalmente em peixes, daí o risco maior para as populações que deles se alimentam preferencialmente, como os ribeirinhos Amazônidas. O efeito neurotóxico dessa forma de mercúrio tem sido amplamente demonstrado através de estudos epidemiológicos e experimentais. Alguns desses estudos também mostraram que hormônios e substâncias antioxidantes podem agir protegendo o organismo contra a ação deletéria do mercúrio. A prolactina é um destes hormônios que apresenta ação protetora, mas age também como citocina pró-inflamatória. Desde que o MeHg pode também agir como uma substância imunotóxica, procuramos neste trabalho estudar a ação citoprotetora da PRL em cultivos contínuos de linhagem B95-A de linfócitos de primata afim de avaliar sua fragilidade ao MeHg e sua reatividade a ação da PRL. Com o objetivo de avaliar a integridade funcional dos linfócitos expostos ao MeHg utilizou-se teste de reação colorimétrica para 3-(4,5-dimetiltiazol-2-yl)-2,5-difenil tetrazólio bromide (MTT), o qual detecta atividade metabólica mitocondrial. Para avaliar a resposta imune do linfócito, medidas da concentração do fator de necrose tumoral alfa (TNF α) no sobrenadante do cultivo, foram realizadas por ELISA. É uma citocina pró-inflamatória liberada em resposta a agressão celular de diferentes causas, incluindo estresse oxidativo, um dos efeitos agudos mais evidentes do MeHg, além disso, esta citocina também poder responder a regulação prolactinérgica em linfócitos humanos. Após 18 horas de exposição do cultivo a crescentes concentrações do metal (0,1; 1, 5, 10 e 50 μM) verificou-se significativa diminuição do tipo dose-dependente da viabilidade celular a partir de 1 μM (35%) e progressivamente até 50 μM (80%), quando poucas células íntegras foram encontradas nos cultivos. Um efeito bifásico em forma de “sino” ocorreu na liberação de TNF α, onde concentrações mais baixas de MeHg inibiram (0,1 e 1 μM), a intermediária estimulou (5 μM) e as duas maiores (10 e 50 μM) voltaram a inibir. A prolactina também diminuiu a viabilidade celular, em cerca de 30%, somente na dose mais elevada (10 nM). Por outro lado, na dose de 1 nM a PRL preveniu a diminuição de 40% da viabilidade celular resultante a exposição ao MeHg a 5 μM. Esta dose de 1 nM de PRL foi a única a estimular a liberação de TNF α, mas curiosamente, reverteu a liberação desta citocina quando associada a 5 μM de MeHg, concentração que igualmente estimulou a secreção de TNF α. Os resultados confirmaram a toxidade do MeHg para linfócitos de primatas (linhagem B95-A) e sua reversão por uma possível ação protetora da PRL. Um efeito bifásico na secreção de TNF α resultou da exposição ao MeHg, sugerindo a presença de diferentes mecanismos citotóxicos resultantes a ação mercurial. Por outro lado, a PRL foi pouco efetiva em estimular a secreção daquela citocina, invertendo esta resposta quando associada ao MeHg. No entanto, estes resultados são preliminares e carecem de um estudo mais acurado para sua completa elucidação.
